An antibody (Ab), also known as immunoglobulin (Ig), is a large Y-shape protein consisting of two heavy chains and two light chains (Figure 1) which form a functionally bivalent monomer that is produced by B cells receptor used by the immune system to identify and neutralize foreign objects such as bacteria and viruses. Current widely used rapid detection systems exploit antibodies for recognition, identification and quantification of target analytes [68]. Abs played an important role in the advancement of diagnostic assays, making these indispensable in diagnostic tests that are commonly used routinely in clinics in classical immunological methods such as ELISA, dot blot immunobinding assays, electro-chemiluminescence, flow cytometry and several microscopic techniques like fluorescence, as well as in the construction of numerous immunosensors. Immunoglobulin M (IgM) usually exists as a pentamer in mammals that predominates in primary immune responses to most antigens, and is the most efficient complement fixing immunoglobulin. It comprises approximately 10% of all normal human serum Ig content. IgM is mainly produced by the immune system for protection against numerous viral, bacterial, fungal and parasitic infections.

Nineteen canine lymphomas were included in this study. Tumors were classified according to the updated Kiel classification adapted for canine lymphomas by Fournel-Fleury et al. Immunoglobulin light chains (κ and λ) and IgM and IgG expression were determined by immunohistochemical method. In all examined cases neoplastic cells were positive for one of the immunoglobulin light chains. Expression of λ light chains and κ light chains was observed in 18/19 and 1/19 tumors, respectively. In the majority of neoplastic cells in each examined specimen this reaction had a membranous pattern (sκ/sλ). In all examined cases the presence of immunoglobulin light chains was also observed in the cytoplasm of some neoplastic cells (cκ/cλ). These cells were usally rare and never constituted a dominant population. The expression of immunoglobulin was found in 13/19 cases. Most lymphomas were sIgM positive (11/13 cases). In one case expression of IgG was found, and in another lymphoma two populations of neoplastic cells with different expression of examined immunoglobulins (cells with IgM+ and IgG+ phenotypes) were observed. The reaction also had a membranous pattern. The cells containing cytoplasmic immunoglobulins were rare, and in most cases were of the same type as the surface immunoglobulins. Our study has confirmed that canine lymphomas are a monoclonal proliferation of B-cells usually expressing immunoglobulin λ light chains and that the vast majority of tumors deriving from B-cells express IgM. Our study also indicates a possibility of occurence of biclonal lymphomas in canine species.

Introduction and aim. Kawasaki disease (KD) is an acute vasculitis with unknown etiology, usually occurring in children under 5 years old. This article will summarize the three consensuses formulated in China about KD. Material and methods. English databases for consensus search include UpToDate, BMJ Clinical Evidence, National Guideline Clearinghouse, Joanna Briggs Institute Library, Cochrane Library, and PubMed, etc.; Chinese databases include China Biomedical Literature Service, China Knowledge Network, Wanfang database, etc. All literature searches ended on February 28, 2022. Analysis of the literature. KD is a common acquired heart disease in children and can lead to severe complications such as coronary injury. However, intravenous immunoglobulin (IVIG) combined with oral aspirin (Asp) is currently recognized as the most effective treatment in KD acute stage and the first-line treatment to prevent cardiovascular complications. Glucocorticoid (GC) is mainly used for KD patients with a high risk of coronary artery aneurysm (CAA), no immunoglobulin response, and confirmed CAA. There are already consensus guidelines on diagnosing and treating KD in different countries. This article summarizes the relevant expert consensus on aspirin, glucocorticoids and IVIG for the treatment of Kawasaki disease in China. Conclusion. Still, there are inconsistent opinions in the literature on the mechanism, optimal timing, and dosage of medication for KD.

There is growing evidence suggesting that the magnitude of cortisol awaking response (CAR), which is characterized by a profound increase of salivary cortisol secretion after awakening, plausibly reflects the level of a chronic stress, social stress, anxiety, etc. In this study the alternation of CAR at the start and at the end of two weeks session of self-instruction for good sleep was investigated; by which we anticipated that the self-instruction for good sleep would bring-forth a positive affection for the participants, and would result in decline of cortisol awakening response (CAR). Nevertheless, as a result unexpectedly, subjects did not change their sleep and dietary habits along with the instruction, moreover the increased CAR was observed. This result implies that the suggestion of an impractical instruction would solely be taken as a stressful task for participants, even though they know that it is effective to improve their sleep. On the contrary, if one develops an instruction with practicable indication for daily life, it is highly possible to observe a positive effect of the instruction on CAR.

The research was conducted on clinically healthy mares (n=40) and foals (n=78) duringY. pseudotuberculosis associated enzootics. The animals were divided into groups: I to IV – mares, IA to IVA – their offsprings, IB to IVB – foals which mothers were not treated with any medicaments. The animals in group I, IA and IB were injected with PBS; in group II, IIA and IIB – with Y. pseudotuberculosis strain-based vaccine, in group III, IIIA and IIIB – with P. acnes strain-based immunostimulator; in group IV, IVA and IVB – with P. acnes strain-based immunostimulator and (5 days after the immunostimulator injection)Y. pseudotuberculosis strain-based vaccine. The presence of antibodies was determined by means of ELISA. The study revealed anti-Yersinia pseudotuberculosis IgG only in 19 mares before, and in 25 mares and 26 foals 3 weeks after vaccination. The mean extinction 3 weeks after vaccination amounted to: II-0.489, IV-2.578, IIA-0.572, IVA-0.974, IIB-0.312, IVB-0.418. The cut-off extinction value was 0.154. The presence of anti-Yersinia pseudotuberculosis IgG before vaccination in the sera of clinically healthy mares may suggest that Y. pseudotuberculosis infection occurs definitely more often than is expected. Vaccination preceded by immunostimulation appeared to be the most efficient method of treatment against yersiniosis.

The self-assembling tendency and protein complexation capability of dyes related to Congo red and also some dyes of different structure were compared to explain the mechanism of Congo red binding and the reason for its specific affinity for β-structure. Complexation with proteins was measured directly and expressed as the number of dye molecules bound to heat-aggregated IgG and to two light chains with different structural stability. Binding of dyes to rabbit antibodies was measured indirectly as the enhancement effect of the dye on immune complex formation. Self-assembling was tested using dynamic light scattering to measure the size of the supramolecular assemblies. In general the results show that the supramolecular form of a dye is the main factor determining its complexation capability. Dyes that in their compact supramolecular organization are ribbon-shaped may adhere to polypeptides of β-conformation due to the architectural compatibility in this unique structural form. The optimal fit in complexation seems to depend on two contradictory factors involving, on the one hand, the compactness of the non-covalently stabilized supramolecular ligand, and the dynamic character producing its plasticity on the other. As a result, the highest protein binding capability is shown by dyes with a moderate self-assembling tendency, while those arranging into either very rigid or very unstable supramolecular entities are less able to bind.

The aim of the study was the determination of IgA, IgM and IgG concentrations in porcine serum and colostrum, in order to evaluate their variations in the perinatal period, as well as to clarify whether there is a correlation between colostrum intake, initial level of immunoglobulins (Ig) in piglet serum and development of their own immunity. The mean IgA, IgM and IgG concentrations in sow serum 10 days before parturition were 1.58, 6.12 and 39.56 mg/ml, respectively. Seven days later only the IgG level was insignificantly lower (34.94 mg/ml, p=0.55), while concentrations of IgA and IgM increased to 2.25 and 7.25 mg/ml, respectively (p=0.23 and 0.62, respectively). The mean initial IgG concentration in colostrum at farrowing was 118.5 mg/ml and differed between sows. The average value of IgA in colostrum at birth was 23.8 mg/ml and decreased to 7.85 mg/ml at 6 hours (h) and to 4.59 mg/ml at 24 h after the onset of farrowing. IgM concentration at birth was 12.1 mg/ml and decreased to 4.23 mg/ml at 24 h postpartum. Positive relationships were found between concentrations of IgM and IgA in serum of piglets at 14 and 56 days of life (r = 0.41 and 0.80, respectively, p≤0.05) as well as for IgG concentration in the piglets serum at 7 days and 56 days of age (r = 0.48, p≤0.05). The above observations suggest that there is a correlation between the level of Ig in piglet serum in the first days of life and improvement of their own immunity.

Introduction: IgG4-related disease (IgG4-RD) is a newly recognised disorder of unknown etiology and pathogenesis, characterised by dense IgG4+ cells infiltration and fibrosis. IgG4-RD can affect various organs, but gastrointestinal tract involvement is rare. First case of isolated gastric IgG4-RD reported in polish population was diagnosed in our Clinic and became the reason for conducting a literature review. Materials and methods: A literature review was performed using PubMed database. Eight studies of isolated gastric IgG4-RD, published between 2011 and 2017, and a case diagnosed by the authors were included. Results: Three out of nine analysed patients had gastrointestinal complaints. In other cases lesions were detected accidentally. The majority of them were submucosal tumors while only one was a gastric ulcer. The most commonly affected was the stomach body. In all cases malignancy had been suspected, and the lesions were surgically removed. Diagnosis was based on the histopathology image and immunohistochemical staining. Only one patient had elevated IgG4 serum level. No case of recurrence or other organ involvement was reported. Conclusion: IgG4-related disease may manifest as an isolated gastric lesion and should be taken in consideration in differential diagnosis. Making an ultimate diagnosis without histopathological specimen examination seems to be difficult and can lead to misdiagnosis followed by inappropriate treatment. IgG4-RD responds well to steroid therapy. However, on this matter further studies are needed.

W poniższej pracy przedstawiono opis przypadku 4-letniej dziewczynki chorującej na przewlekłą zapalną polineuropatię demielinizacyjną (Chronic inflammatory demyelinating polyneuropathy, CIDP) o ciężkim, nawrotowym przebiegu wymagającej wieloletniej politerapii skojarzonej. CIDP jest neuropatią autoimmunologiczną charakteryzująca się wieloogniskową demielinizacją włókien ruchowych i czuciowych. Objawy rozwijają się najczęściej w ciągu >8 tygodni. Na obraz kliniczny składają się niedowład mięśni ksobnych i odsiebnych kończyn i/lub zaburzenia czucia oraz hipo-/arefleksja. Lekami pierwszego rzutu u dzieci są dożylne wlewy immunoglobulin (intravenous immunoglobulin, IVIg). W razie ciężkiego przebiegu i braku długotrwałej poprawy należy dołączyć glikokortykosteroidy, a następnie jeden z leków immunosupresyjnych. Rokowanie w CIDP u dzieci jest szczególnie dobre, z remisją lub minimalnymi objawami ubytkowymi u prawie wszystkich pacjentów, jednak czas potrzebny do długotrwałej poprawy bywa zróżnicowany.
In this report we present a case of a 4-year-old girl suffering from chronic inflammatory demyelinating polyneuropathy (CIDP) with a severe recurrent course requiring long-term combined polytherapy. CIDP is an autoimmune neuropathy characterised by a multifocal demyelination of motor and sensory fibers. Symptoms develop during >8 weeks in most cases. The clinical presentation is weakness of proximal and distal muscles of limbs and/or sensory symptoms and hipo-/ areflexia. First line treatment in childhood CIDP is intravenous immunoglobulin (IVIg). In case of a severe course and lack of long-term improvement, corticosteroids should be added, followed by one of the immunosuppressive drugs. CIDP in children has a good prognosis with a remission or minimal deficits in almost all cases, although the time needed for a long- -term improvement may be varied.

The Recent rapid growth of the information technology aid people to improve the performance in processing their job activities. These changes, in turn, force them to carry out their job activities using computers in the workplace; that results in the increase of somatic and/or mental stresses. In this study, we conducted a preliminary experiment to estimate an impact of web-browsing on human mind and body. Two types of web-browsing tasks, which are a 18-minuetes of continuous web-browsing and an intermittent web-browsing (first 45 second of each 1-minutes interval), were given to the subjects (10 healthy male aged from 20-23) with within-subjects experimental design. With regard to physiological measures, two prominent stress biomarkers, salivary immunoglobulin A (IgA) and cortisol, were employed. Comparing among the task conditions, relatively larger and long-lasting increase of IgA and smaller decrease of cortisol was observed with an intermittent web-browsing despite there was no difference in the psychological state. These results illustrate the difficulty of estimating the mental workload caused by web-browsing and the importance of employing physiological indices.
