Opis:
Conventional therapeutic approaches like chemotherapy, surgery and radiation for cancer and other malignancies have limited success rate. Several factors such as drug resistance, non-penetrability into solid tumors and unwanted toxicity due to non-specific action contribute to treatment failure. To overcome these issues, immunotoxins were developed having a target specific action. Immunotoxins are chimeric cytotoxic proteins which are formed by linking a toxin moiety to intact antibody or its fragment. Bacterial and plant based toxins are used in immunotoxins construction but they have many drawbacks in terms of immunogenicity and non-specific toxicity. Recently, a novel class of immunotoxins of human origin has been introduced. Immunotoxins fight back cancers by targeting the neoplastic cells and cause cells death by inhibiting protein synthesis. They have been categorized into two main classes, chemically conjugated immunotoxins and recombinant immunotoxins. This review summarizes recent advances in immunotoxins development and present a comprehensive illustration of sources and generations of immunotoxins. Moreover, it also highlights future directions to be explored. This review also focuses the limitations in immunotoxins development and its use.

Introduction. Vaccinations are one of the most effective medical interventions that protect people against infectious diseases. It should be noted that a new vaccine licensing is always preceded by clinical trials assessing its safety and efficacy. Anti-vaccine propaganda carried out by vaccination opponents has become an international problem with a global reach. Aim. To review the literature on vaccinations of patients from high-risk groups. Material and methods. A literature review of the following databases has been conducted: EBSCO, PubMed, Science Direct, and Springer Link. Results. High-risk groups in the paediatric population include pre-term born infants, patients after stem cell transplantations, children with allergies and other chronic diseases. Vaccinations in the examined groups are generally safe and are an effective method of preventing infections. Conclusion. At a time when the level of vaccine skepticism is high and the epidemiological situation of many diseases is unstable, patients who are more susceptible to infection are particularly endangered. High level of knowledge of health care professionals and their personal positive attitude towards vaccinations are important for improving the vaccination coverage rates. In the light of measles epidemic outbreaks and an almost geometric increase in the number of pertussis cases noted recently, actions are needed to achieve herd immunity.

Due to the increasing incidence of gluten intolerance, researchers are focusing on finding ways to eliminate immunotoxicity of wheat, this would allow the use of wheat products for gluten-intolerant consumers. The article reviews recent studies on biotechnological methods to eliminate and reduce the immunogenicity of wheat products. So far, many gluten removal methods have been proposed, but their efficacy levels were quite different. Enzymatic treatment of gluten fragments can be considered the simplest and non-invasive tool to eliminate the toxicity of gliadins and glutenins. For this purpose, various endogenous enzymes derived from cereals, and also those of bacterial, fungal, plant, and animal origin can be used in food processing. Some of the enzymes hydrolyze gluten, others block the action of toxic protein fragments. The majority of studies were carried out using lactic acid bacteria cultures, as single strains or in consortia. Satisfactory results have been achieved using bacterial and plant enzymes, but the complete elimination of gluten immunogenicity is still possible by using fungal proteases, engineered enzymes or combining several treatments, for example, by using lactic acid fermentation or germination with fungal proteases. However, the question of how degradation of gluten affects the quality of flour (dough) in practice remains unanswered. It is not clear whether the products of such wheat flour are better and safer than those made from starches and whether their price and quality are acceptable to consumers. The insights presented in this review will be helpful to other researchers and cereal-based food producers in choosing ways to reduce gluten immunogenicity.

Biomedical data are difficult to interpret due to their large amount. One of the solutions to cope with this problem is to use machine learning. Machine learning can be used to capture previously unnoticed dependencies. The authors performed random forest classifier with entropy and Gini index criteria on immunogenicity data. Input data consisted of 3 columns: epitope (8-11 amino acids long peptide), major histocompatibility complex (MHC) and immune response. Presented model can predict the immune response based on epitope-MHC complex. Achieved results had accuracy of 84% for entropy and 83% for Gini index. The results are not fully satisfying but are a fair start for more complexed experiments and could be used as an indicator for further research.
Dane biomedyczne są trudne do interpretacji ze względu na ich dużą ilość. Jednym z rozwiązań radzenia sobie z tym problemem jest wykorzystanie uczenia maszynowego. Techniki te umożliwiają wychwycenie wcześniej niezauważonych zależności. W artykule przedstawiono wykorzystanie klasyfikatora Random Forest z kryterium entropii i indeksem Gini na danych dotyczących immunogenności. Dane wejściowe składają się z 3 kolumn: epitop (peptyd o długości 8-11 aminokwasów), główny kompleks zgodności tkankowej (MHC) i odpowiedź immunologiczna. Zaprezentowany model przewiduje odpowiedź immunologiczną na podstawie kompleksu epitop-MHC. Uzyskane wyniki osiągnęły dokładność na poziomie 84% (entropia) i 83% (indeks Gini). Wyniki nie są w pełni satysfakcjonujące, ale stanowią dobry początek dla bardziej złożonych eksperymentów i wyznacznik do dalszych badań.
