A rapid, accurate and precise LC-MS method is described for the quantitative determination of pramipexole in human plasma matrix using ropinirole as internal standard. Pramipexole and ropinirole were extracted from plasma by liquid-liquid extraction technique. The method was validated over the concentration range of 100-2514 pg/mL. The method was found to have acceptable accuracy, precision, linearity and selectivity. The mean extraction recovery from spiked plasma samples was in the range of 79.415-87.00 %. The intra-day accuracy of the assay ranged from 98.924 to 112.236 % and intra-day precision ranged from 3.489 to 6.756 %. Inter-day accuracy and precision results for quality control samples ranged between 100.340 and 107.443% of nominal and precision is observed to be 3.970-5.714 %. The pramipexole was found to be stable after several stability studies. The proposed method yielded a quick, simple and reliable protocol for estimating pramipexole concentrations in human plasma.

Opis:
Simple and rapid reverse phase high-performance liquid chromatography (RP-HPLC) method was developed and validated using Phenomenex Gemini c18 (4.6 x 250 mm, 5 μ) reverse phase column for the determination of LAF in human plasma, Solid Phase Extraction (SPE) technique was used for the extraction of analyte, detection was carried out by Photo Diode Array detector at 216 nm. Chromatographic resolution of the LAF was achieved within 4.6 min by using mobile phase Methanol and 5 mM Di-Potassium Hydrogen Phosphate Buffer (pH 9.5) (80:20, v/v), flow rate was 1.0 mL/min. Calibration curve was linear with correlation coefficient of 0.9996 in the range of 50-1000 ng/mL, Limit of Detection (LOD) and Limit of Quantitation (LOQ) were 10 ng/mL and 30 ng/mL respectively, intra and inter-day deviations were lower than 3.92 % and 3.98 % respectively. The overall mean recovery of LAF was 94.57 %. No any endogenous constituents were found to interfere at retention time of the analyte. This new RP-HPLC method was successfully validated and may be applied to conduct bioavailability & bioequivalence studies of LAF.

Neuroblastoma, the most common extracranial solid tumour of childhood, is a malignancy of unknown origin and non-specific symptoms. One of the markers of the disease is GD2 ganglioside (disialoganglioside), which is abundantly expressed on the surface of neuroblastoma cells. Gangliosides are known to be shed by tumour cells and this phenomenon can be significant in cancer progression as they inhibit a number of immune responses both in vitro and in vivo. In search for novel markers useful in monitoring and prognosis of neuroblastoma, we developed and validated a new quantitative method of GD2 ganglioside analysis in human blood plasma. We evaluated the level of gangliosides in blood serum of 34 neuroblastoma patients using high-performance liquid chromatography. The technique was used to detect fluorescently labelled oligosaccharides derived from serum glycosphingolipids by enzymatic digestion with ceramide glycanase. The developed method allowed determination of GD2 concentrations at the picomole level and required only 40 µl of plasma, which should be particularly useful when the quantity of clinical material is limiting. Moreover, this method can be applied to study concentration of other gangliosides, as shown for GD3 ganglioside. Analysis of plasma samples from the 34 neuroblastoma patients did not reveal any correlations between the concentration of GD2 ganglioside and clinical parameters, including the results of therapy; it showed, however, that the concentration of GD2 ganglioside in the plasma of neuroblastoma patients decreased substantially in the course of treatment.

Opis:
Multidrug resistance-associated protein (MRP1) is a transmembrane pump protein responsible for the efflux of chemotherapeutic drugs, an important cause of anticancer treatment failure. Trying to circumvent MRP-mediated resistance we designed and synthesized hairpin loops forming antisense oligodeoxyribonucleotides (ODNs), both phosphodiesters (PO-ODNs) and their phosphorothioate analogues (PS-ODNs), to reduce the protein expression by targeting its mRNA in a sequence specific manner. Melting temperature measurements as well as polyacrylamide gel electrophoresis supported the preferential formation of a secondary structure, which was expected to protect ODNs against 3'-exonuclease degradation. ODNs and PS-ODNs designed in this work were successfully tested as antisense inhibitors of the expression of MRP1 in the leukaemia HL60/ADR cell line. Foreseeing the necessity to perform clinical studies with such ODNs we investigated their stability against the 3'-exonuclease activity of fetal calf serum and human plasma. Under the conditions, corresponding to physiological ones, we observed high stability of hairpin loop forming ODNs, especially those containing longer (e.g. 7 base pair) stems. Comparative studies on the stability of chemically unmodified hairpin loop forming ODNs and their PS-counterparts indicated that endonuclease activity did not play any important role in the process of their nucleolytic degradation. Our studies provide strong evidence for high stability of chemically unmodified hairpin loop ODNs, making them an attractive alternative to phosphorothioate analogues commonly used in antisense strategy.

Objectives: The aim of the study was to evaluate effects of Finnish sauna bathing on lipid profile in healthy, young men. Material and Methods: Sixteen male subjects (20–23 years) were subjected to 10 sauna bathing sessions in a Finnish sauna every 1 or 2 days. The mean sauna temperature was 90±2°C, while humidity was 5–16%. Each session consisted of three 15-minute parts and a 2-minute cool-down between them. The following measurements were taken before and after the sauna sessions: body mass, heart rate, body skinfold thickness. The percentage fat content and then, the lean body mass were calculated. Total cholesterol, triacylglycerols, lipoprotein cholesterol LDL and HDL were measured in blood samples. Results: A statistically significant decrease of total cholesterol and LDL cholesterol was observed during 3 weeks of sauna treatment and in the week afterwards. A significant decline in triacylglycerols was found directly after the 1st and 24 h directly after the 10th sauna session. After the 10th sauna session the level of HDL cholesterol remained slightly increased, but this change was not statistically significant. A decrease in blood plasma volume was found directly after the 1st and the last sauna bathing session due to perspiration. An adaptive increase in blood plasma volume was also found after the series of 10 sauna sessions. Conclusions: Ten complete sauna bathing sessions in a Finnish sauna caused a reduction in total cholesterol and LDL cholesterol fraction levels during the sessions and a gradual return of these levels to the initial level during the 1st and the 2nd week after the experiment. A small, statistically insignificant increase in HDL-C level and a transient decline in triacylglycerols were observed after those sauna sessions. The positive effect of sauna on lipid profile is similar to the effect that can be obtained through a moderate-intensity physical exercise.

Background. Researchers suspect that the accepted adequate ascorbic acid plasma concentration is not being met even after dietary intake of the recommended amount of vitamin C. Current dietary intake recommendation in Poland is 60 mg per day for women and 75 mg per day for man (EAR), while in Western Europe and North America is higher and amounts to 75-90 mg per day. Objective. The paper aimed at studying a correlation between composition of nutrients in daily diet and plasma vitamin C levels in university students. Materials and methods. This study examined diet composition and the nutritional status of ascorbic acid in plasma of 120 university students in Szczecin, Poland. Ascorbic acid was determined in blood plasma using HPLC method. The information concerning diet composition was collected using the method of “7-days food record” prior to blood collection. Results. Plasma ascorbic acid deficiency (<40 μmol/L) was observed in 23% of women and 28% of men. The average plasma ascorbic acid concentration was 48.65 μmol/L in women and 45.61 μmol/L in men. The average intake of vitamin C in women with observed deficiency was average 46.55 mg/day, whereas in men it was 48.56 mg/day. Conclusions. The recommendation of dietary intake of vitamin C in Poland is low in comparison to other countries. Population- based studies are necessary to determine the actual demand for vitamin C in various population groups in Poland.
Wprowadzenie. Badacze podejrzewają, że przyjęte odpowiednie stężenie kwasu askorbinowego w osoczu nie jest spełnione nawet po spożyciu zalecanej ilości witaminy C. Aktualna norma spożycia witaminy C w Polsce wynosi 60 mg/dzień dla kobiet i 75 mg/dzień dla mężczyzn (EAR), podczas gdy w Europie Zachodniej i Ameryce Północnej jest większa i wynosi 75-90 mg dziennie. Cel badań. Celem pracy było zbadanie korelacji między składnikami pokarmowymi całodziennej racji pokarmowej a witaminą C u studentów. Materiał i metody. Badano skład diety oraz zawartość witaminy C w osoczu 120 studentów ze Szczecina. Stężenie kwasu askorbinowego oznaczano w surowicy krwi za pomocą HPLC. Informacje dotyczące żywienia zebrano stosując metodę 7 dniowego zapisu żywieniowego przed pobraniem krwi. Wyniki. Niedobór kwasu askorbinowego w osoczu (< 40 μmol/L) wystąpił u 23% kobiet i 28% mężczyzn. Średnie stężenie kwasu askorbinowego w osoczu wynosiło 48.65 μmol/L u kobiet i 45,61 μmol/L u mężczyzn. U kobiet, u których stwierdzono niedobór witaminy C, jej średnie spożycie z dietą wynosiło 46,55 mg/dzień, podczas gdy u mężczyzn 48,56 mg/dzień. Wnioski. Zalecana w Polsce norma dziennego spożycia witaminy C jest niska w porównaniu z innymi krajami. Konieczne są badania populacyjne w celu określenia rzeczywistego zapotrzebowania na witaminę C wśród różnych grup społecznych w Polsce.

In gilts, the period of early pregnancy occurring from the time of fertilization to the beginning of implantation is sensitive to any environmental disruptions, including an unbalanced diet of a future mother. Previously, we found that due to the undernutrition in gilts during this period, the endocrine intrauterine microenvironment and DNA methylation in the uterus have been changed. These distortions may diminish the success of pregnancy. In this study we focused on the influence of a restricted diet used in gilts during the first days of pregnancy on their biochemical and haematological parameters in peripheral blood. The applied restrictive diet vs. normal diet covered only 70% of the nutritional demands of early pregnant gilts. Normal (n = 4 gilts) or restrictive (n = 5 gilts) diets were used from the day of the first signs of the estrus until day 9 of pregnancy and biochemical and haematological parameters in blood plasma were determined during peri-implantation period, e.g. on days 15 to16 of pregnancy. In restrictive vs. normal fed gilts significantly lower plasma phosphorus, calcium and total cholesterol as well as the tendency to increasing concentrations of triglicerydes and asparate aminotranserase were found. Haematological parameters did not differ between the studied gilts. Thus, it seems that the availability of nutritional factors became suboptimal in restrictively fed early pregnant gilts. Even short-lasting undernutrition of females during the peri-conceptional period may cause a disruption of biochemical homeostasis during the peri-implantation period and probably affect the success of pregnancy.

Opis:
Digestive system neoplasms pose a serious health problem both in Poland and abroad. Neoplasms are frequently considered to be caused by impaired homeostasis in the human body. Development of neoplasms may be linked to disturbances in concentration of elements, including magnesium as a major intracellular cation. The objective of this study was to evaluate the concentration of magnesium in plasma and tissue samples taken from patients suffering from neoplasms of the stomach or the large intestine. The study involved 35 patients, including 20 affected by stomach cancer and 15 suffering from large intestine cancer. The patients were in the age rage of 36-77. The material included blood samples taken from patients before and seven days after surgery, as well as samples of cancerous and healthy tissues. The colorimetric method with a Genesis spectrophotometer was used for determination of magnesium concentration. A statistically significant difference was observed between plasma magnesium concentration in patients affected by stomach cancer and the normal range. Elevated values of magnesium concentration measured on the seventh day after the procedure as compared to the concentration before the procedure was noted, however, the difference was statistically insignificant. No significant differences were observed in magnesium concentration measured before and after the procedure, or in comparison to the normal range in patients with large intestine cancer. Determination of tissue magnesium showed that magnesium concentration was higher in cancerous than in healthy tissue. Obtained results demonstrate that magnesium homeostasis is impaired in patients, which may be important in the pathoghenesis of digestive system neoplasms.
Nowotwory przewodu pokarmowego stanowią istotny problem zdrowotny zarówno w Polsce, jak i na całym świecie. Przyczyny nowotworów często upatruje się w zaburzeniach homeostazy organizmu. Rozwój nowotworów może wiązać się z zaburzeniem stężenia pierwiastków, w tym również magnezu będącego najważniejszym kationem wewnątrzkomórkowym. Celem pracy była ocena stężenia magnezu w osoczu i wycinkach tkanek nowotworowych chorych na raka żołądka i raka jelita grubego. Badania wykonano u 35 pacjentów, w tym u 20 chorych na raka żołądka, oraz 15 chorych na raka jelita grubego. Badani byli w przedziale wiekowym od 36 do 77 lat. Materiał do badań stanowiła krew chorych pobierana przed zabiegiem operacyjnym oraz w 7. dobie po zabiegu, a także wycinki tkanek zmienionych nowotworowo i zdrowych. Stężenie magnezu oznaczano metodą kolorymetryczną z użyciem spektrofotometru firmy Genesis. U chorych na raka żołądka wykazano istotną statystycznie różnicę w stężeniu magnezu w osoczu w porównaniu z normą. Zaobserwowano również wyższe stężenie magnezu w 7. dobie po zabiegu w porównaniu ze stężeniem przed operacją, jednak różnica ta była nieistotna statystycznie. Natomiast w przypadku raka jelita grubego nie wykazano różnic istotnych statystycznie zarówno w stężeniu magnezu przed i po zabiegu, jak i przy porównaniu wyników stężeń magnezu z normą. Badając stężenie magnezu w tkankach, stwierdzono wyższe stężenie magnezu w tkance nowotworowej w porównaniu z tkanką zdrową. Wyniki wskazują na istnienie zaburzeń homeostazy magnezu u chorych, co może mieć znaczenie w patogenezie nowotworów przewodu pokarmowego.

Opis:
W badaniach in vitro wykazano, że chlorfenwinfos w dawce 8,5 µg/cm3 osocza lub homogenatu wątrobowego szczura nie wykazuje istotnego wpływu na aktywność aminotransferaz w osoczu i w cytozolu wątrobowym.
In vitro experiments the effect of chlorfenvinphos on aminotransferases activities in rat plasma and liver homogenate cytoplasmic fraction was studied. As amino groups donors in the transamination reactions with plasma enzymes the next eight amino acids were used: alanine, aspartic acid, cysteine, phenylalanine, leucine, lysine, valine and asparagine, and with the liver cytoplasmatic enzymes - the same above mentioned without asparagine. In all these reactions as an amino groups acceptor alfa-ketoglutaric acid was used. To the incubation mixtures were added: 1 cm3 of the plasma or liver homogenate catoplasmic fraction; 0,05 cm3 of chlorfenvinphos solution in ethyl acetate (0,17 mg/cm3) or 0,05 cm3 ethyl acetate. Aminotransferase activity was expressed as the amount of glutamate developing durign 1 h incubation. Glutamic acid was determined spectrophotometrically after chromatographic separation on filter paper. Both in rat plasma and in liver cytoplasmic fraction all used amino acids as amino groups donors in the transamination reactions were shown. In the reactions with the blood plasma enzymes the most active donors were: alanine, aspartic acid and cysteine and with the participation of liver transaminases: alanine, aspartic acid and phenylalanine. As well in plasma, as in liver the greater acitivity of AIAT than of AspAT was observed. In the reaction with alanine and aspartate there was formed in the case of plasma 1,51 µmol Glu/cm3 and 1,00 µmola Glu/cm3 respectively and in the case of liver - 69,07 µmol Glu/g and 53,26 µmol Glu/g respectively. Results of statistical analysis revealed that small plasma and liver aminotransferases inhibition was caused by the solvent, while the insecticide under test had no influence on the efficiency of transamination.
