Opis:
Neuroblastoma, the most common extracranial solid tumour of childhood, is a malignancy of unknown origin and non-specific symptoms. One of the markers of the disease is GD2 ganglioside (disialoganglioside), which is abundantly expressed on the surface of neuroblastoma cells. Gangliosides are known to be shed by tumour cells and this phenomenon can be significant in cancer progression as they inhibit a number of immune responses both in vitro and in vivo. In search for novel markers useful in monitoring and prognosis of neuroblastoma, we developed and validated a new quantitative method of GD2 ganglioside analysis in human blood plasma. We evaluated the level of gangliosides in blood serum of 34 neuroblastoma patients using high-performance liquid chromatography. The technique was used to detect fluorescently labelled oligosaccharides derived from serum glycosphingolipids by enzymatic digestion with ceramide glycanase. The developed method allowed determination of GD2 concentrations at the picomole level and required only 40 µl of plasma, which should be particularly useful when the quantity of clinical material is limiting. Moreover, this method can be applied to study concentration of other gangliosides, as shown for GD3 ganglioside. Analysis of plasma samples from the 34 neuroblastoma patients did not reveal any correlations between the concentration of GD2 ganglioside and clinical parameters, including the results of therapy; it showed, however, that the concentration of GD2 ganglioside in the plasma of neuroblastoma patients decreased substantially in the course of treatment.

In gilts, the period of early pregnancy occurring from the time of fertilization to the beginning of implantation is sensitive to any environmental disruptions, including an unbalanced diet of a future mother. Previously, we found that due to the undernutrition in gilts during this period, the endocrine intrauterine microenvironment and DNA methylation in the uterus have been changed. These distortions may diminish the success of pregnancy. In this study we focused on the influence of a restricted diet used in gilts during the first days of pregnancy on their biochemical and haematological parameters in peripheral blood. The applied restrictive diet vs. normal diet covered only 70% of the nutritional demands of early pregnant gilts. Normal (n = 4 gilts) or restrictive (n = 5 gilts) diets were used from the day of the first signs of the estrus until day 9 of pregnancy and biochemical and haematological parameters in blood plasma were determined during peri-implantation period, e.g. on days 15 to16 of pregnancy. In restrictive vs. normal fed gilts significantly lower plasma phosphorus, calcium and total cholesterol as well as the tendency to increasing concentrations of triglicerydes and asparate aminotranserase were found. Haematological parameters did not differ between the studied gilts. Thus, it seems that the availability of nutritional factors became suboptimal in restrictively fed early pregnant gilts. Even short-lasting undernutrition of females during the peri-conceptional period may cause a disruption of biochemical homeostasis during the peri-implantation period and probably affect the success of pregnancy.

W badaniach in vitro wykazano, że chlorfenwinfos w dawce 8,5 µg/cm3 osocza lub homogenatu wątrobowego szczura nie wykazuje istotnego wpływu na aktywność aminotransferaz w osoczu i w cytozolu wątrobowym.
In vitro experiments the effect of chlorfenvinphos on aminotransferases activities in rat plasma and liver homogenate cytoplasmic fraction was studied. As amino groups donors in the transamination reactions with plasma enzymes the next eight amino acids were used: alanine, aspartic acid, cysteine, phenylalanine, leucine, lysine, valine and asparagine, and with the liver cytoplasmatic enzymes - the same above mentioned without asparagine. In all these reactions as an amino groups acceptor alfa-ketoglutaric acid was used. To the incubation mixtures were added: 1 cm3 of the plasma or liver homogenate catoplasmic fraction; 0,05 cm3 of chlorfenvinphos solution in ethyl acetate (0,17 mg/cm3) or 0,05 cm3 ethyl acetate. Aminotransferase activity was expressed as the amount of glutamate developing durign 1 h incubation. Glutamic acid was determined spectrophotometrically after chromatographic separation on filter paper. Both in rat plasma and in liver cytoplasmic fraction all used amino acids as amino groups donors in the transamination reactions were shown. In the reactions with the blood plasma enzymes the most active donors were: alanine, aspartic acid and cysteine and with the participation of liver transaminases: alanine, aspartic acid and phenylalanine. As well in plasma, as in liver the greater acitivity of AIAT than of AspAT was observed. In the reaction with alanine and aspartate there was formed in the case of plasma 1,51 µmol Glu/cm3 and 1,00 µmola Glu/cm3 respectively and in the case of liver - 69,07 µmol Glu/g and 53,26 µmol Glu/g respectively. Results of statistical analysis revealed that small plasma and liver aminotransferases inhibition was caused by the solvent, while the insecticide under test had no influence on the efficiency of transamination.
