A membrane-bound phospholipid : steryl glucoside acyltransferase from Solanum melongena leaves was partially purified and its specificity and molecular as well as kinetic properties were defined. Among the steryl glycosides tested (e.g. typical plant steryl glucosides, steryl galactosides and cholesteryl xyloside) the highest activity was found with cholesteryl glucoside, but some structurally related compounds such as sito- and stigmasteryl glucoside or galactoside as well as cholesteryl galactoside were also acylated, albeit at lower rates. The investigated enzyme was able to use all classes of phosphoglycerolipids (phosphatidylethanolamine, phosphatidylcholine, phosphatidylserine, phosphatidylinositol, phosphatidylglycerol) as an acyl source for biosynthesis of acylated steryl glucoside. Among them 1,2-dimirystoylphosphatidylic acid appeared to be the best acyl donor. Apart from phosphoglycerolipids, 1,2-diacylglycerols were also used as acyl donor for steryl glucoside acylation, although at a distinctly lower rate. The acyl moiety was transferred from the C-1 position of phospholipid molecule. The investigated acyltrasferase activity was stimulated by 2-mercaptoethanol, Triton X-100, 1-monoacylglycerols and inhibited in the presence of divalent cations such as Ca2+, Mn2+, Zn2+ or Co2+, some lipids (MDGD, ceramide), detergents (Tween 20, 40, 60 and 80, Tyloxapol, sodium deoxycholate) and high ionic strength.

In our research, the concentration of lotaustralin in the roots of two species Rhodiola kirilowii and Rhodiola rosea were compared. Aqueous and hydroalcoholic extracts from those plants were analyzed too. To determine the content of this compound the ultra performance liquid chromatography – tandem mass spectrometry (UPLC-MS/MS, Waters) was used. The obtained results showed that the content of measured lotaustralin depends on the species of Rhodiola. R. rosea roots are the richer source of lotaustralin then R. kirilowii. The same situation was observed in the extracts. A hydroalcoholic extract from R. rosea contains up to 135.276 mg of lotaustralin in 100 g of dry powdered material. In the case of R. kirilowii extracts, an aqueous extract contained more lotaustralin (74.791 mg/100 g of dry powdered material) then a hydroalcoholic extract.
W badaniach przeprowadzonych w Instytucie Włókien Naturalnych i Roślin Zielarskich porównano zawartość lotaustraliny w dwóch gatunkach roślin: Rhodiola kirilowii i Rhodiola rosea, jak również w przygotowanych z tych roślin dwóch wyciągach: wodnym oraz wodnoalkoholowym (50% etanolowy wyciąg). W celu wyznaczenia zawartości lotaustraliny wykorzystano ultrasprawny chromatograf cieczowy sprzężony z tandemowym spektrometrem mas (UPLC-MS/MS, Waters). Wykazano, że zawartość cyjanogennego glukozydu zależna jest od gatunku. Przeprowadzona walidacja metody pozwala na satysfakcjonujące wykorzystanie tej metody w badaniach zawartości lotaustraliny.

A membrane-bound UDP-glucose : sterol glucosyltransferase from Solanum melongena (eggplant) leaves was partially purified and its specificity as well as molecular and kinetic properties were defined. Among a wide spectrum of 3-OH steroids (i.e. typical plant sterols, androstane, pregnane and cholestane derivatives, steroidal alkaloids and sapogenins) and triterpenic alcohols, the highest activity was found with 22-oxycholesterol. UDP-glucose appeared to be the best sugar donor. The enzyme preparation was also able to utilize UDP-galactose, TDP-glucose and CDP-glucose as a sugar source for sterol glucosylation, however, at distinctly lower rates. The investigated glucosyltrasferase was stimulated by 2-mercaptoethanol, Triton X-100 and negatively charged phospholipids, and inhibited in the presence of UDP, mono-, di- and triacylglycerols, divalent cations such as Zn2+, Co2+, high ionic strength, cholesteryl glucoside, galactoside and xyloside and some amino acid-modifying reagents (SITS, DIDS, PLP, DEPC, pCMBS, NEM, WRK and HNB). Our results suggest that unmodified residues of lysine, tryptophan, cysteine, histidine and dicarboxylic amino acids are essential for full enzymatic activity and indicate that a glutamic (or aspartic) acid residue is necessary for the binding of sugar donor, i.e. UDP-glucose in the active site of the GT-ase while histidine and cysteine residues are both important for the binding of the nucleotide-sugar as well as of the steroidal aglycone.

Opis:
The occurrence of syringin in eight out of ten investigated plant species of the subtribe Centaureinae (Asteraceae) was reported. The compound was isolated from aerial parts of the plants by silica gel column chromatography of methanol extracts. Contents of syringin varied from 0.001 to 0.1% of the dried plant material. Centaurea bella Trautv. appeared to be the best source of this compound (0.1% dry wt.). Syringin has been reported for the first time from these plant species.

The anthocyanins are present in the grapes as monomers, but aged red wines have only linked anthocyanins in the different polymers. One of the possible reaction of red wine colourant is the forming of linkage between the anthocyanins and flavanols by (CH2CH) bridge of acetaldehyde. The formation of coloured products between malvidin 3-glucoside, acetaldehyde and (-)epicatechin, its dimer B2 and oligomers was analysed by liquid chromatography (HPLC). These polymeric compounds were hydrolyzed in the presence of toluen-α-thiol (thiolysis) for the determination the linkage place of anthocyanins with flavanols. The model studies showed, that the procyanidins were linked by "top molecule" across of aldehyde bridge with malvidine, because the coloured toluen-α-thiol derivatives were obtained. The spectrum of these compounds had the maximum at 240 nm corresponding to toluen-α-thiol molecule and other maximum in visible region had shift to longer wavelength. This is characteristic for the polymers of anthocyanins with flavanols. In the red wine after thiolysis, the compounds were identified by the HPLC method with the same retention time as in the model solution, but only in the presence of acetyladehyde being previously added.
Metodą chromatografii cieczowej wysokosprawnej badano proces tworzenia się kompleksów antocyjanów, flawanoli z aldehydem octowym w roztworach modelowych i czerwonym winie. Kompleksy te są jednymi z możliwych barwnych substancji czerwonych dojrzałych win. Produkty kondensacji pomiędzy malwidyno 3-glukozydem, aldehydem octowym, (-)epikatechiną oraz jej dimerem B2 i oligomerami poddano hydrolizie w obecności tolueno-α-tiolu, aby określić miejsca powstawania wiązań w tych kompleksach z procy- janidynami. Doświadczenia wykazały, że procyjanidyny łączą się swą górną cząsteczką poprzez mostek aldehydowy z malwidyną. Uzyskano drogą tiolizy barwne pochodne tiolowe (Rysunek 2). Widmo tych związków posiada maksimum przy 240 nm odpowiadające cząsteczce tolueno-alfa-tiolu oraz ma przesunięte maksimum w zakresie światła widzialnego w kierunku fal dłuższych (Rysunek 3). Jest to charakterystyczne dla kompleksów antocyjanów z flawanolami. W czerwonym winie po tiolizie stwierdzono obecność podobnych związków jak w roztworze modelowym, ale tylko po uprzednim dodaniu aldehydu octowego (Rysunek 5).

2,3,5,4'-tetrahydroxy stilbene-2-O-ß-D-glucoside (THSG) exerts multiple pharmacodynamic actions, found in Fallopia multiflora, but the biosynthesis pathway of THSG is still unclear. To clear this ambiguity, we constructed suppression subtractive hybridization (SSH) libraries to screen the genes involved in THSG biosynthesis from two F. multiflora varieties, which vary significantly in THSG content. Twelve non-redundant differentially expressed sequence tags were obtained and the full lengths of 4 unreported fragments were amplified by rapid amplification of cDNA ends. We totally got 7 fulllength transcripts, and all of them were aligned to the transcriptome and digital gene expression tag profiling database of four F. multiflora tissues (root, stem and leaf from Deqing F. multiflora and another root from Chongqing F. multiflora; data unpublished) using local BLAST. The results showed that there was a significant, organ specific difference in the expression of fragments and full-length sequences. All the sequences were annotated by aligning to nucleotide and protein databases. Kyoto Encyclopedia of Genes and Genomes pathway analysis indicated that THSG biosynthesis was correlated with multiple life activities.
