The genetic continuity of the potato spindle tuber viroid (PSTVd) genome was analysed after infection of tomato plants with cloned cDNAs of parental strains. During the six weeks of the experiment, several new sequence variants appeared. The sequence variants detected in the progeny population induced sequence-specific disease symptoms. The PSTVd genome therefore follows the pattern expected for typical pseudo-strains propagating in plants as a population of similar sequences. Assessing further the replicon continuity, a PSTVd cDNA mutant with a deletion in the central conserved region was constructed and proven to be non-infectious. Surprisingly, in a sub-population of potato transformants expressing the same deleted PSTVd RNA an infectious viroid was detected. This suggests specific transcript conversion followed by recovery of the full-length pathogen genome.

Opis:
Salinity is one of the most important abiotic stresses which can negatively affect the plant metabolic processes in the world. This can impact the plant production, either for economic or sustenance benefits. The salinity stress can cause many physiological and biochemical changes in the plants. β-glucans are important polysaccharides, which are present in the cell walls of various cereal grains. They protect the plant responses and occur in plant suspensions. In this study, the researchers attempted to investigate various physiological mechanisms and determine the role of the β-glucans in the NaCl-mediated stress conditions on the Ocimum basilicum L. seedlings. For this purpose, they carried out an experiment for assessing various shoot and root parameters along with the antioxidant enzyme activities, proline levels and the ISSR markers. When the seedlings were exposed to the NaCl stress conditions, they showed a significant decrease in the growth parameters and an increase in the antioxidant and proline levels compared to the control seedlings grown under normal saline conditions. On the other hand, the β-glucantreated seeds, when grown under the saline stress conditions, showed better growth parameters as well as high antioxidant enzyme activities and proline levels, compared to the control and NaCl-treated plants. Furthermore, a PCR analysis was carried out using the ISSR-marker technology, which could help in evaluating the DNA fingerprints and genetic variations in the plants. The results indicated that the exogenous application of the β-glucans could protect the antioxidant enzyme activities and protect the plants against the salinity stresses, without affecting the DNA-markers without affecting the genetic variations and could be a better choice for use in DNA-markers.

Opis:
Identification of cultivars is essential both in breeding and to settle cultivar disputes. The purpose of the study has been to examine cultivar identities based on absorption spectra of plant pigments and to confirm a genetic stability with SCoT and RAPD molecular markers in new Polish lines of Chenopodium quinoa Willd. Spectral analysis of pigments extracted from plant inflorescences in quinoa gives an opportunity to confirm the cultivar identity and identification of ‘Faro’ and ‘Titicaca’ cultivars and their new lines. Spectral analysis is an effective method of confirming cultivar identity and it should be used in practice for the identification of cultivars or cultivars lines in Chenopodium quinoa Willd. Analysis of molecular markers indicated by RAPD as well as SCoT technique revealed a high genetic stability of the derivative lines of ‘Faro’ and ‘Titicaca’, while variation was detected in plants representing original cultivars: banding pattern different than predominant was present in three plants of ‘Titicaca’ (genetic distnaces from 7.5% to 55.9%) and in a single plant of ‘Faro’(genetic distance 61.2% as indicated by SCoT technique).

Opis:
Ex vitro rooting and acclimatization of two cultivars ‘Wojtek’ and ‘Zojka’ of blue honeysuckle (Lonicera caerulea var. kamtschatica Sevast.) were studied. To the ex vitro conditions were transferred rooted and unrooted shoots. The post-effect of auxin type and concentration as well as microcutting and soil substrate types were tested. The genetic stability of the plantlets in relation to the mother plants by using amplified fragment length polymorphism (AFLP) and inter simple sequence repeat (ISSR) markers has been also determined. It has been found that in vitro rooted cuttings of both cultivars showed a higher survival rate (max. 88%) and better growth and development when they were rooted on a medium containing a low auxin level (1.0 mg·dm-3). The results of the second experiment showed successful ex vitro rooting of blue honeysuckle shoots without auxin treatment. Higher ex vitro rooting and survival rate in the greenhouse have been observed for ‘Wojtek’ (max. 96%) than ‘Zojka’ (max. 88%). Better growth and development of shoots and roots were observed on peat alone or a mixture of peat and perlite as compared to a mixture of peat and sand. The micropropagated plantlets appeared similar to mother plants. Molecular analysis confirmed a high level of genetic stability of blue honeysuckle after 2 years of in vitro propagation. However, among the cultivars studied, ‘Wojtek’ showed slightly higher genetic stability than ‘Zojka’ (99.5% and 97.7%, respectively). For ‘Zojka’ plants, the degree of variation was comparable for AFLP and ISSR markers. For ‘Wojtek’, no polymorphism was detected using the ISSR analysis in contrast to the AFLP analysis.

The article presents a method of calibration of material parameters of a numerical model based on a genetic algorithm, which allows to match the calculation results with measurements from the geotechnical monitoring network. This method can be used for the maintenance of objects managed by the observation method, which requires continuous monitoring and design alterations. The correctness of the calibration method has been verified on the basis of artificially generated data in order to eliminate inaccuracies related to approximations resulting from the numerical model generation. Using the example of the tailing dam model the quality of prediction of the selected measurement points was verified. Moreover, changes of factor of safety values, which is an important indicator for designing this type of construction, were analyzed. It was decided to exploit the case of dam of reservoir, which is under continuous construction, that is dam height is increasing constantly, because in this situation the use of the observation method is relevant.

This article presents an efficient search method for representative circular and unconstrained slip surfaces with the use of the tailored genetic algorithm. Searches for unconstrained slip planes with rigid equilibrium methods are yet uncommon in engineering practice, and little publications regarding truly free slip planes exist. The proposed method presents an effective procedure being the result of the right combination of initial population type, selection, crossover and mutation method. The procedure needs little computational effort to find the optimum, unconstrained slip plane. The methodology described in this paper is implemented using Mathematica. The implementation, along with further explanations, is fully presented so the results can be reproduced. Sample slope stability calculations are performed for four cases, along with a detailed result interpretation. Two cases are compared with analyses described in earlier publications. The remaining two are practical cases of slope stability analyses of dikes in Netherlands. These four cases show the benefits of analyzing slope stability with a rigid equilibrium method combined with a genetic algorithm. The paper concludes by describing possibilities and limitations of using the genetic algorithm in the context of the slope stability problem.

This paper presents a new approach in the field of trajectory tracking for nonholonomic mobile robot in presence of disturbances. The proposed control design is constructed by a kinematic controller, based on PD sliding surface using fuzzy sliding mode for the angular and linear velocities disturbances, in order to tend asymptotically the robot posture error to zero. Thereafter a dynamic controller is presented using as a sliding surface design, a fast terminal function (FTF) whose parameters are generated by a genetic algorithm in order to converge the velocity errors to zero in finite time and guarantee the asymptotic stability of the system using a Lyapunov candidate. The elaborated simulation works in the case of different trajectories confirm the robustness of the proposed approach.

W badaniach wykorzystano wyniki oceny odmian ziemniaka dotyczące udziału bulw zdeformowanych, uzyskane w doświadczeniach odmianowych w Zakładzie Agronomii Ziemniaka Instytutu Hodowli i Aklimatyzacji Roślin-PIB, Oddział w Jadwisinie. Celem badań była ocena interakcji genotypowo-środowiskowej dotycząca deformacji bulw ziemniaka. Analizę wykonano za pomocą programu SERGEN 4. W badaniach wykorzystano 21 odmian ziemniaka uprawianych przez pięć lat na glebie lekkiej. Jako środowiska do analizy uwzględniono poszczególne lata badań (warunki klimatyczne: opad + temperatura powietrza). Na podstawie analizy genotypowo-środowiskowej wydzielono odmiany stabilne w zmieniających się warunkach środowiska oraz odmiany niestabilne. Przeprowadzono ocenę wpływu poszczególnych lat na uprawę wybranych odmian ziemniaka. Stwierdzono, że środowisko w dużym stopniu modyfikuje udział deformacji bulw w plonie.
The study was conducted with the use of data regarding the evaluation of potato tuber deformations, collected during varietal experiments at the Potato Agronomy Department, Plant Breeding and Acclimatization Institute - National Research Institute, Division in Jadwisin, Poland. The purpose of the study was the estimation of tuber deformations in the genotype x environment interaction. The analysis was performed using the SERGEN 4 software. The study included 21 potato cultivars, cultivated for a period of five years on a light soil. Particular years of the investigations were taken as different environments (weather conditions → rainfall + air temperature). Among the cultivars tested in variable environmental conditions, stable and unstable cultivars were identified. Moreover, an estimation of the influence of the particular years on the cultivation of the chosen potato cultivars was performed. It was found, the environment profoundly modifies the share of tuber deformation in the yield.
