Opis:
The article presents the achievements in DNA forensic science practice, the latest developments as well as future trends. The article concerns also other than forensic DNA applications as well as expectations, concerns and obstacles. DNA identification technology unlike other biometric techniques requires the collection of biological material and the identification is not performed in real time. DNA utilized in most of forensic identification, is present almost in every living cell in the body. What is more, each cell of the same body has the same DNA molecule which means that it is possible to compare the DNA sampled from different sources, for example saliva with blood or semen from the same person. Rapid development and reliability of DNA technology contribute to the fact that the analysis of polymorphic DNA sequences constitutes a very important evidence used in the court. The unique properties of DNA and rapid development of DNA analytical devices allow to claim that DNA may assume a more important position amongst biometric data in the future.

The adaptation of an athlete to systematic physical exercise has been shown to be determined by a combination of great many genes. The aim of our study was to investigate the dependence of the aerobic capacity parameters in sport on the set of gene polymorphisms. Cardio-respiratory system (CRS) adaptation reactions to exercise of 72 endurance athletes were assessed using the gas analysis. The analysis of the obtained results has shown both single and combined effect of the gene polymorphisms on the aerobic capacity. The impact of 6 polymorphisms on the aerobic performance level was analyzed: Т–786→С polymorphism of the promoter of еNOS gene as well as АСЕ I/D polymorphism, Рго/Ala polymorphism of PPARG gene, G/C polymorphism of PPARA gene, Pro582Ser polymorphism of HIF1α gene, and Ala203Pro polymorphism of PPARGC1B. It was found that a single impact on the HRmax providing АСЕ I/D polymorphism. Individual influence of АСЕ gene accounts for 2% of this index dissipation. Results showed that there is a dependence between the amount the maximum volume of consumed oxygen (VO2max) from the set of gene polymorphisms. Cumulative impact of these polymorphisms in the combination with the individual parameters (gender; qualification; kind of sport) stipulates 71% of dispersion of VO2max value.

Opis:
All tree breeders cope with the same challenge of the very long time interval of a single breeding cycle. What is more, trees are long−lived, with desirable breeding traits expressing late during their life cycle. Increasing problems with climate change, globalization or economic growth have forced us to accelerate tree breeding and improve selection precision, both of which can be achieved by genomic selection (GS). The idea of GS was introduced nearly 20 years ago as an extension of marker−assisted selection (MAS) in order to advance breeding technologies using genetic markers. Unlike MAS, which exploits only a set of marker−trait associations, GS relies on a high number of genetic markers that are spread throughout the entire length of the genome. All markers effects are assessed simultaneously in order to build a precise model that allows prediction of genetic estimated breeding value of a particular individual using genetic data only. GS has already revolutionized dairy cattle breeding resulting in remarkable improvements across multiple traits and is becoming more and more common in crop production. We now know that genetic architecture of quantitative traits is complex, but recent advances in genomics have made it possible to deal with this problem in an unprecedented way. There are certain concerns regarding GS in forest tree species that include genotype−environment (G×E) interaction and the usefulness of the predictive model built up by GS in the next generation of trees. Nevertheless, experimental results obtained so far have shown that the genetic gain per unit time as well as selection precision can be substantially increased. Here we present the basic principles of GS for forest tree species, giving examples of studies carried out so far and discussing problems and future possibilities that GS may soon open up for forest tree breeders.

The results of contemporary studies confirm the formation of two neural networks in the brain during the period of adolescence. The first is defined as emotional, located in the limbic system, develops earlier, quicker, and more intensively than the second one in the prefrontal cortex, called the judgement network, which fulfils the role of control and inhibition of emotional reactions. The domination of the emotional network in adolescence is manifested by hyperactivity of the limbic system, accompanied by intensified undertaking of courageous, reckless, risky, or even sometimes dangerous actions, so very characteristic in the maturation. The aim of the article is to present the state of the art in the field of latest achievements in experimental neurophysiology related to the maturation of the structural end functional processes in adolescents, and to alcohol vulnerability. Alcohol effect initiation starts in early adolescence, and therefore is connected with alcohol abuse and addiction in adulthood, which confirms the necessity for provision of an early prophylactic protection for juveniles, even before entering the phase of early adolescence. Some electrophysiological characteristics, such as low P3 amplitude of the Event-Related Potential (ERP) and Event-Related Oscillations (EROs), are manifested by their high risk offspring, and are considered to be biological markers (endophenotypes) of a predisposition to develop alcohol use disorders. Electroencephalographic oscillations induced within the range of the theta and delta waves (Event-Related Oscillation- ERO), considered as endophenotypes and markers of increased vulnerability for addiction, present three groups of genes and three types of neurotransmitters, with gamma aminobutyric acid, acetylcholine and glutamate as neurotransmitters in the central nervous system. A new research approach consisting in the application of electroencephalographic methods and techniques in developmental and genetic studies of the conditioning of varied vulnerability, and especially increased preferences for alcohol tasting and abuse in adolescence, provide unique possibilities for comprehensive and deepened studies which may contribute to the prevention of alcohol addiction, the genesis of which, to a great extent, is related with the effect of causative environmental and genetic factors during adolescent development.

The occurrence of hernias in pigs is a serious breeding and veterinary problem. Hernia is a condition in which internal organs (most often intestines), protrude through an opening in the layers of muscle and connective tissue of the abdominal wall. There are two major types of hernia - umbilical and inguinal. This pathology is a serious problem in pig production, leading to economic loss through the cost of surgical intervention, increased mortality rate, and reduced carcass value. The overall incidence of hernias in a normal swine population is low but may increase due to matings to a boar that transmits inguinal hernias. The heritability of this trait oscillates around 0.3. Current knowledge of hernias in pigs is scarce, and mostly based on genome-wide association studies. In this review, we indicate the recent studies concerning the genetic markers association analysis in terms of heritability of hernias in pigs. Moreover, the new approach focused on differently expressed genes in swine hernias etiology is here discussed.

Genetic markers in mitochondrial and chloroplast DNA were analysed to determine the native status and possibly trace the origin of the silver fir (Abies alba Mill.) growing outside this species natural range in Osusznica Forest District (northern Poland). The provenance of the extralimital populations were determined through comparison with reference samples from the native range of silver fir in Poland and Ukraine. Our results suggest that the Sudety Mts. are the most probable region of the origin of fir from Osusznica. It also seems that the plus seed stands in Osusznica can be treated as a valuable archive of the genetic resources of the fir from the Sudety and, if necessary, may support program of this species restitution.

Forty eight inbred lines of winter rye, of various origin and pedigree, were analysed using 19 RAPD (random amplified polymorphic DNA) primers, 8 ISSR (inter-simple sequence repeats) primers and 13 SSR (simple sequence repeats) primer pairs. On the basis of particular marker types, there were created three separate dendrograms and one combined similarity tree, prepared on account of the whole data. Correlation coefficients for individual technique based on genetic similarity matrices were not significant. By comparing the GS data obtained on the basis of singular methods with collective matrix, it was observed that the highest correlation rate was for ISSR method (r=0.68). The utility of each marker technique was compared by using marker index MI. Genetic diversity of eight genotypes of Carthamus tinctorius L., two populations of C. oxyacanthus, and one population of C. lanatus was investigated using inter-simple sequence repeat (ISSR) markers. All samples were uniquely distinguished by 10 ISSR primers with 144 bands which generated 100% polymorphism. Furthermore, the ISSR markers could separate three safflower species properly, that highlights the effectiveness of this marker system for phylogenetic studies. The most and least informative primers were ISSR9 (PIC=0.367) and ISSR2 (PIC=0.254), and some primers were more efficient in detecting polymorphism in one species than for the others. Unweighed pairgroup method with arithmetical averages (UPGMA) cluster analysis enabled construction of a dendrogram  for estimating genetic distances among different populations. The result of cluster analysis suggested that cultivated and wild populations of C. oxyacanthus had close relationship with each other and far relationship with C. lanatus. The extreme genetic dissimilarity was observed between genotypes of C. tinctorius and C. lanatus populations. Based on the results, C. oxyacanthus could introduce favorable genes to cultivated safflower via inter-specific hybridization in breeding programs. Nei’s gene diversity index, Shannon’s index and percent of polymorphic loci showed that Isfahan ecotype of C. oxyacanthus had the highest variation at DNA level in relation to populations of other species. The ISSRs developed in this research along with those recently studied by other researchers will contribute to construct genetic map with a density sufficient for safflower molecular breeding.

The F2-type population derived from the cross between DS2 and RXL1O inbred lines was used for interval mapping of five growth related traits i.e. plant height, spike length, thousand grain weight, kernel length and kernel thickness. Scanning of the whole 1,140 cM length of rye genetic map consisting of 286 marker loci revealed the existence of 6 regions containing QTL5 on chromosomes 1R-5R. Plant height was strongly affected by 1-3 linked dwarfing genes from a distal region of the chromosome 5RL and by 1 gene on the chromosome 3RL, tightly linked to a marker loci Xpsr4 75. These same genes regulated also thousand grain weight and kernel length and thickness. Spike length was determined only by the QTL from chromosome 5RL. In addition a single QTL from chromosome 2R affecting thousand grain weight and kernel thickness was identified, near the molecular marker locus Xrsq8OS. 1. Kernel length and kernel thickness were additionally controlled by QTL5 on chromosomes 2R and 1R and 4R, respectively.

Habitat fragmentation is one of serious threats to biodiversity of nature in today's world. The present study of a typical steppe species Iris pumila L. (Iridaceae) has analyzed the impacts of geographical isolation and population size on genetic diversity and population structure in conditions of habitat fragmentation. The key indices of population genetic variability calculated from the ISSR markers data were on average as follows: Shannon diversity index (S) – 0.188; unbiased Nei’s gene diversity (He ) – 0.123; and the average measure of Jaccard’s genetic distances between individuals within populations – 58.4%. Although the largest population had significantly higher values of S and He, the small and marginal populations also showed a comparable level of variation. Most of the genetic variation of I. pumila was distributed within the populations. A strong correlation was found between Nei’s genetic distances and geographic distances between the populations. According to the Bayesian analysis, genetic structure of the populations was highly homogeneous; however, the presence of admixed genotypes indicated the possibility of gene flow between the populations at present.

Assessment of genetic diversity using molecular markers is one of the primary and important steps in breeding programs. In this study, genetic diversity of 52 barley lines evaluated using 68 SSR primer pairs and 47 primer pairs produced clear and polymorphic banding pattern. In general, 153 polymorphic alleles de-tected. The number of observed polymorphic alleles varied from 2 to 9, with an average of 3.26 alleles per locus. Polymorphic Information Content (PIC) ranged from 0.07 to 0.81, with an average of 0.45. In this research, SSR markers differentiated the studied lines efficiently. Using cluster analysis, studied barley lines divided into two groups. Genetic diversity was relatively corresponding with geographical origins, because the lines related to a country somewhat diverged from each other. Two-rowed Iranian and Chinese barleys classified in one subgroup. Also, most six-rowed barleys classified in one subgroup. Association mapping analysis was used to identify candidate genes for drought escape in barley lines and 16 informative markers were identified after which confirmation in other tests could be suitable for marker assisted breeding drought escape.

Przedmiotem badań była analiza podobieństwa genetycznego form dzikich z rodzaju Secale. Materiałem roślinnym były gatunki żyta: Secale cereale subsp. afghanicum, Secale cereale subsp. dighoricum, Secale cereale subsp. segetale, Secale strictum subsp. africanum. Secale strictum subsp. anatolicum, Secale strictum subsp. ciliatoglume, Secale strictum subsp. kuprijanovii, Secale strictum, Secale sylvestre oraz Secale vavilovii. Analiza prążków RAPD pozwoliła na wytypowanie starterów generujących polimorficzne prążki, pozwalające zróżnicować badane gatunki i podgatunki z rodzaju Secale. Przeprowadzone badania pozwoliły na podział roślin na trzy grupy o różnym stopniu podobieństwa genetycznego. Wieloletnie gatunki z rodzaju Secale utworzyły jedną grupę podobień- stwa, a gatunki jednoroczne dwie grupy, w których podobieństwo wynosiło od 11 do 21%.
The aim of this study was the analysis of genetic similarity among Secale species. The plant material included: Secale cereale subsp. afghanicum, Secale cereale subsp. dighoricum, Secale cereale subsp. segetale, Secale strictum subsp. africanum, Secale strictum subsp. anatolicum, Secale strictum subsp. ciliatoglume, Secale strictum subsp. kuprijanovii, Secale strictum, Secale sylvestre and Secale vavilovii. Based on the RAPD (random amplified polymorphic DNA) analysis primers generating polymorphic products were selected, which made possible to differentiate species and subspecies of Secale. Three groups of the Secale species were distinguished according to the different degree of genetic similarity. Perennial species were classified in one group of similarity, and annual species in two groups, in which the similarity ranged from 11 to 21%.

Silver fir (Abies alba) is a common tree species in the mountainous areas in Europe. A number of natural stands in the hilly regions of northern Europe represent relic populations. The aim of the research was to evaluate the diversity present in Italian populations of the species. Genetic diversity was assessed in 45 silver fir populations covering the species’ distribution range in Italy, based on the allelic variation present at seven microsatellite loci (SSRs). A consistent level of intra-population variability was present. Several of the populations displayed signs of ongoing genetic erosion, and evidence for a recent bottleneck in some was identified. Populations from the eastern Alps and the Apennines were more variable than those sampled from the western Alps. About 8% of the overall genetic variance was found between populations, with the remainder representing variation present within the populations. The data suggested that the southern Apennines acted as a refugium during the most recent Ice Age, and that many of the populations from this area have remained isolated over a prolonged period. Smaller and more isolated populations have experienced genetic drift, whereas the larger ones have preserved a high level of diversity. Identification of genetically homogeneous regions could be informative for the management of genetic resources.

Salinity is one of the most important abiotic stresses which can negatively affect the plant metabolic processes in the world. This can impact the plant production, either for economic or sustenance benefits. The salinity stress can cause many physiological and biochemical changes in the plants. β-glucans are important polysaccharides, which are present in the cell walls of various cereal grains. They protect the plant responses and occur in plant suspensions. In this study, the researchers attempted to investigate various physiological mechanisms and determine the role of the β-glucans in the NaCl-mediated stress conditions on the Ocimum basilicum L. seedlings. For this purpose, they carried out an experiment for assessing various shoot and root parameters along with the antioxidant enzyme activities, proline levels and the ISSR markers. When the seedlings were exposed to the NaCl stress conditions, they showed a significant decrease in the growth parameters and an increase in the antioxidant and proline levels compared to the control seedlings grown under normal saline conditions. On the other hand, the β-glucantreated seeds, when grown under the saline stress conditions, showed better growth parameters as well as high antioxidant enzyme activities and proline levels, compared to the control and NaCl-treated plants. Furthermore, a PCR analysis was carried out using the ISSR-marker technology, which could help in evaluating the DNA fingerprints and genetic variations in the plants. The results indicated that the exogenous application of the β-glucans could protect the antioxidant enzyme activities and protect the plants against the salinity stresses, without affecting the DNA-markers without affecting the genetic variations and could be a better choice for use in DNA-markers.
