Opis:
The genetic continuity of the potato spindle tuber viroid (PSTVd) genome was analysed after infection of tomato plants with cloned cDNAs of parental strains. During the six weeks of the experiment, several new sequence variants appeared. The sequence variants detected in the progeny population induced sequence-specific disease symptoms. The PSTVd genome therefore follows the pattern expected for typical pseudo-strains propagating in plants as a population of similar sequences. Assessing further the replicon continuity, a PSTVd cDNA mutant with a deletion in the central conserved region was constructed and proven to be non-infectious. Surprisingly, in a sub-population of potato transformants expressing the same deleted PSTVd RNA an infectious viroid was detected. This suggests specific transcript conversion followed by recovery of the full-length pathogen genome.

Opis:
Twenty-eight isozymic loci were studied in the Beskid Mts., in four populations of common silver-fir (Abies alba): one in Beskid Makowski (BM) and three populations in Beskid Sądecki (BS). Their genetic variation and diversity were analyzed, and Nei's genetic distances between the populations were calculated. The results show that the geographical distance between the BM population and the three BS populations is reflected in genetic distances. The BM population is clearly distinct from the others. It has the lowest genetic diversity (I = 0.42), percentage of polymorphic loci (%PoL = 64.29) and number of rare alleles (NoRa = 5). Besides, the BM population has the highest observed heterozygosity (Ho = 0.291), which exceeds the expected heterozygosity (He = 0.254), estimated on the basis of the Hardy-Weinberg Principle. On the contrary, BS populations are in the state of equilibrium, which is manifested, in similar values of He = 0.262 and Ho = 0.264.

Opis:
Genetic diversity is one of the key requirements for the adaptive potential of forest trees. This study presents the analysis of 30 Polish Scots pine provenances. According to the RAPD method, three 10−mer primers: OPE−08, OPE−09 and OPF−07 (Operon Technologies) were used to generate most polymorphic bands of amplified DNA among 450 individuals. The genetic similarity index was calculated after Nei (1987) matching coefficient and results were presented as dendrogram of genetic distances obtained from cluster analysis (UPGMA) for all populations. The Scots pine provenances were also characterized by height and diameter at the breast height „DBH” traits and were compared with the genetic diversity level. All studies populations were characterized by genetic diversity GST=0,215. This coefficient of inter−popu− lation variation was lower than the level of the differentiation within populations (HT=0,262), as it was reported for other Pine species. The results from these studies may be useful to the forest practice to help with decisions on the establishment of stable forest stands; they are also used in the forest genetic resources conservation programmes.

Opis:
Seeds collected from individual trees in the 16 Carpathian and 2 Sudeten silver-fir (Abies alba) populations were studied with the starch gel electrophoresis in megagametophytes using 14 enzyme systems with 28 loci. The results show that the geographical distance between populations are in a small part reflected in genetic distances. There are two main groups of populations: Sudeten and Carpathian with a very big genetic distance between them. Other populations consist of a few small groups with low gene flow between them (Nm = 3.286). About 80% of genetic variation is located within populations (FST = 0.223). Average values for genetic multiplicity and diversity for Carpathian populations are as follows: number of alleles per locus: Na = 2.308, with effective number of alleles Ne = 1.552 and proportion of polymorphic loci 71.21%. The mean number of alleles per locus (Na) varied from 2.107 to 2.607 in population. The mean effective number of alleles per locus (Ne) ranged from 1.429 to 1.662. Average Fis for Carpathian populations was -0.021, which means that there is small excess of heterozygotes. The average observed heterozygosity amounted to Ho = 0.275 and expected heterozygosity was He = 0.269. The dendrogram structure and presence of rare alleles found in silver-fir of Czech, and Slovakian populations allow for a hypothesis that in postglaciation the silver-fir moved into the Polish Carpathians not westward from the east but from the south along river valleys from some Balkan refuges, getting North bypassing the High Tatra Range. This way, a highly diversified set of populations originated, differ in the presence of rare alleles. This differentiation is not prevented by a relatively small flow of genes between populations. The calculated gene flow Nm = 3.286 also indicates isolation between the populations. It means 3.3 immigrants per generation into the studied populations.

Multivariate characterization of breeding value and evaluation of genetic divergence in five blackcurrant (Ribes nigrum L.) varieties are presented. In the study general combining ability effects (GCA) estimated over two years in a full-diallel cross design (Griffing’s method 1) for fruit yield per plant, yield-contributing traits and resistance traits have been elaborated. The multivariate analysis of variance (MANOVA) according to the fixed Griifing’s model and canonical variate analysis for GCA effects have been used. The suggested statistical approach delivers information for two known and commonly accepted criterions of successful selecting parents for crosses in breeding programs, both multivariate estimates of GCA effects of parents and their genetic distances, here based on multivariate GCA effects. GCA effects for fruit yield per plant, bush vigour, spring frost damage to flowers, infestation by white pine blister rust and powdery mildew had the largest discrimination power in multivariate GCA divergence of the parents. It was proved that the breeding value of potential parents regarding productivity attributes could be the predominant criterion of proper parents selecting for crosses in blackcurrant breeding. Genetic distance evaluated on the basis of multivariate GCA effects has not been a powerful predictor of progenies productivity. The paper is thought mainly as experimental, methodological study on effective using and usefulness of the suggested statistical procedure in researches for plant breeding purposes. The showed results could be also meaningful for blackcurrant breeding.

Opis:
The study was carried out in three Scots pine (Pinus sylvestris L.) single tree plot half-sib progeny tests planted in 1984–86. Phenotypic selection was done in two ways: F about 0.5% to 1% of the total number of trees were chosen by the author on the basis of their appearance only, and Fbis on the basis of diameters at breast height and heights measurements and observations scored for individual trees. The best trees were selected on the basis of the index value weighing traits by their economic value. Genetic selection was performed based on both family index value (Wf) and tree index value (WT), weighing traits by heritability and economic value. This was done in four different ways: (A) The best trees were selected only on the basis of their index value (WT) with no attention paid to the family; (B) About 10 to 12 trees were selected in the best families, (both (Wf) and (WT) were taken into consideration); (C) The best 30% of the families (Wf) were selected and then equal numbers of best trees were selected (WT); (D) The same number families (Wf) as in the phenotypic selection were selected and within these families equal numbers of best trees (WT). The number of trees that were genetically selected was always the same as the number of phenotypically selected ones (selection type F). The selections were compared using a calculated expected genetic gain and the relative loss of effective population size. As expected, phenotypic selection resulted in the lowest genetic gain. Phenotypic selection generally conserved genetic variability, while genetic selection reduced it, especially when genetic gain was maximised. Phenotypic selection type F generally identifies the good families. The agreement of genetic selection with the phenotypic selection is low (0% to 19%) at the individual tree level. This is slightly better with phenotypic selections using measurements data (Fbis). Phenotypic selection is recommended only where information concerning pedigrees is not available. It can provide a simple and cheap way of obtaining material for future selection. Genetic selection method (A) where best trees were selected without any restrictions provided highest genetic gain and cause the greatest loss of genetic diversity.

Opis:
We have investigated genetic diversity and the mating system of Scots pine plus trees from the Tuchola Forests – Poland, using allozymes as genetic markers. The studied plus trees possess high genetic diversity (expected heterozygosity He = 0.427) and low inbreeding (Wright’s index F = –0.028), which is comparable to Scots pine trees observed in other studies in natural populations and seed orchards. The mating system analyses revealed that almost all offspring produced by the plus trees were due to outcrossing (tm = 0.983), with no apparent bi-parental inbreeding. The estimate of correlation of paternity indicated that each of mother trees is pollinated on average by a relatively large number of effective males (Nep = 30.3). The observed genetic diversity and mating system patterns indicate that the seeds produced by the studied plus trees possess high genetic variation needed in reforestation processes.

For the conservation strategy, information about the distribution of the genetic variation within and among populations plays very important role. Thus, extensive study in other populations of dwarf birch is needed.

An extremely endangered population of Viola uliginosa Besser at the classical locality of this taxon has been studied. The AFLP analysis was based on 18 specimens of V. uliginosa (about 10% of preserved individuals); additionally, two individuals of V. riviniana were included in the data set as the out group. A high genetical uniformity of the whole population (similarity indexes close to 1) was detected. It was not correlated significantly with the spatial distribution of the plants. The study serves as a basis for practical conservation measures and at the same time as a starting point for a more extensive research on the genetical variability of the species throughout its range.
