Opis:
We studied the shell and penis morphology, and cytochrome oxidase subunit I (COI) gene sequences, in minute, valvatiform hydrobiid gastropods from Slovensky Kras, Slovakia. The morphology confirmed the assignment of the studied snails to the genus Hauffenia, while in the molecular tree they were placed within the Hydrobiidae, and not close to Hauffenia. The results indicate that the Slovak valvatiform hydrobiids are two taxa which presumably represent two genera: Hauffenia Pollonera, 1898 and Lobaumia Haase, 1993. More molecular data on the Austrian and Hungarian taxa are needed.

Opis:
Gene promoters contain several classes of functional sequence elements (cis elements) recognized by protein agents, e.g. transcription factors and essential components of the transcription machinery. Here we describe a common DNA regulatory element (tandem TCTCGCGAGA motif) of human TATA-less promoters. A combination of bioinformatic and experimental methodology suggests that the element can be critical for expression of genes involved in enhanced protein synthesis and the G1/S transition in the cell cycle. The motif was identified in a substantial fraction of promoters of cell cycle genes, like cyclins (CCNC, CCNG1), as well as transcription regulators (TAF7, TAF13, KLF7, NCOA2), chromatin structure modulators (HDAC2, TAF6L), translation initiation factors (EIF5, EIF2S1, EIF4G2, EIF3S8, EIF4) and previously reported 18 ribosomal protein genes. Since the motif can define a subset of promoters with a distinct mechanism of activation involved in regulation of expression of about 5% of human genes, further investigation of this regulatory element is an emerging task.

Opis:
Hereditary neuropathy with liability to pressure palsies (HNPP) is manifested by a spectrum of phenotypes, from the classical HNPP course associated with intermittent nerve palsies to a neuropathy resembling Charcot-Marie-Tooth type 1 (CMT1) disease. The majority of HNPP cases are associated with submicroscopical deletions in the 17p11.2-p12 region containing the PMP22 gene, while PMP22 point mutations are rare, representing about 15% of HNPP cases. In this study, we present a patient manifesting with atypical HNPP phenotype associated with a new Thr99fsX110 mutation in the PMP22 gene. We conclude that all patients who fulfill the electrophysiological criteria of HNPP, even if they lack the typical HNPP phenotype, should be tested for point mutations in the PMP22 gene.

Opis:
The structure of the EDA1 gene was investigated in a patient with anhidrotic ectodermal dysplasia. Sequence analysis revealed a novel A1270G transition in exon 9 of the EDA1 gene in the patient and his uncle, whereas the patient's mother and grandmother were heterozygotes. This mutation resulted in Tyr343Cys substitution in the extracellular domain of the EDA1 gene product - ectodysplasin-A. The additional Cys343 was located between Cys332 and Cys346 and formed with Cys352 a cluster of four closely situated residues that could potentially form disulfide bonds. This mutation might affect the tertiary structure of the receptor-binding domain of ectodysplasin-A and precipitate the clinical symptoms of anhidrotic ectodermal dysplasia.

Opis:
The methylotrophic yeast Pichia pastoris GS115, a widely used strain in production of various heterologous proteins, especially membrane-bound enzymes, can also produce linoleic and linolenic acids, which indicates the existence of membrane-bound Δ12 and Δ15-fatty acid desaturases. This paper describes the cloning and functional characterization of a novel Δ12-fatty acid desaturase gene from this methylotrophic yeast. The open reading frame of the gene (named Pp-FAD12) is 1263 bp in size and encodes a 420-amino-acid peptide. The deduced Pp-FAD12 protein shows high identity (50-67%) with Δ12-fatty acid desaturases from other fungi. It also shows a high identity (57%) with Δ15-fatty acid desaturase (named Sk-FAD15) from Saccharomyces kluyveri. Expression of Pp-FAD12 in polyunsaturated fatty acids non-producing yeast Saccharomyces cerevisiae demonstrated that its product converted oleic acid (18 : 1) to linoleic acid (18 : 2). This result suggests that Pp-FAD12 encodes a novel Δ12-fatty acid desaturase in P. pastoris GS115. This is the first report about the cloning and functional characterization of Δ12-fatty acid desaturase gene in methylotrophic yeast.

Opis:
Effective control of Leptinotarsa decemlineata remains an urgent problem for agriculture worldwide. Minimization of the use of non-selective neonicotinoid insecticides, such as thiomethoxam, is an actual vector of development of potato cultivation. In this rapid communication, we show the prospect of the topical use of short unmodified antisense fragment of L. decemlineata CYP6B gene as a DNA insecticide. Investigated parameters, namely, number of larvae per plant, aboveground biomass, yield and number of potatoes produced per plant indicate the possibility of this post-genomic approach as a safe and effective method of L. decemlineata control.

The present work describes a rapid and easy way to study foreign gene expression in wheat through microparticle bombardment. Transient expression of the GUS reporter gene was evaluated in different tissues of the cultivar Buck Ombú. Transformation was carried out employing a helium gun microparticle accelerator and several plasmids. Embryogcnic calli, immature embryos and immature inflorescences showed a higher number of transformed cells per Petri dish. Basal leaf segments were the least efficient. When vectors were tested in scutellar tissue of immature embryos, the maize ubiquitin promoter (Ubil) produced the highest level of transient GUS expression, followed by the alcohol dehydrogenase promoter of maize plus its first intron (Adh1 Adh-intron 1) and the sunflower ubiquitin promoter. The CaMV35S promoter was the least effective. These results were in agreement with previous reports carried out in cell suspensions or embryogenic calli and indicate that wheat immature embryos and immature inflorescences are suitable for rapid test of promoter sequences or transformation conditions. Moreover, efficient transformation of these explants could help in the stable transformation of agronomically important genotypes.
