In tomato fruits, chlorophyll, lycopene and ß-carotene are mostly responsible for the color. During ripening of tomato fruits, the color of the pericarp changes from green to red as chlorophyll is degraded and carotenoids accumulate. These changes are associated with an increase in respiration and ethylene production. Carotenoid biosynthesis pathway in plants can be disturbed by herbicide fluridone (1-methyl-3-phenyl-5-[3-trifluoromethyl(phenyl)]- 4(1H)-pyridinone), which inhibits the activity of phytoene desaturase, an enzyme responsible for conversion of phytoene to phytofluene. Fluridone is also used as an inhibitor of biosynthesis of abscisic acid (ABA) and strigolactones, and it reduces chlorophyll production in plants. In our research we studied the effect of fluridone on some physiological parameters, such as color, firmness, ethylene production, lycopene and chlorophyll content during ripening of the tomato fruit. Tomato plants cv. Altadena (Syngenta) were cultivated in a greenhouse in controlled temperature and both immature and mature fruits were used for the experiments, performed between August and November 2016. Fluridone at concentrations of 0.1% and 1.0% in lanolin paste was applied as a 2-3 mm stripe from the top to the base of tomato fruits, and as a control a stripe of lanolin was applied in the same way on the opposite side of the fruits. Fluridone at a concentration of 1.0% greatly inhibited lycopene accumulation in the pericarp of tomato fruits from the treated side. The measurements of fruit firmness have shown no significant differences between firmness of the part of the tomato fruits treated with fluridone, and the non-treated ones. Tomato fruits treated with fluridone produced amounts of ethylene similar to those found in control tissues on the opposite side of the same fruit. Fluridone delayed chlorophyll degradation in tomato fruits. The metabolic significance of these findings is discussed with the role of carotenogenesis inhibition in tomato fruit ripening.

Opis:
The effects of fluridone on the content and composition of fatty acids and some other properties of tomato fruits during a 14-day period of ripening were studied. Mature green tomato fruits were used for the experiments. Fluridone at a 1.0% concentration in lanolin paste was applied as a 2-3 mm stripe from the top to the base of a tomato fruit, and a stripe of lanolin was applied in the same way on the opposite side of the frui as control. After 14 days of the experiment, the treated (yellow) and untreated (red) halves of the tomato were separately freeze-dried and powdered before analysis. Determination of the profile and content of fatty acids was carried out in a micro-HPLC-MS/MS system including a 5600 QTOF mass spectrometer. Fatty acids were identified, based on retention times of available standards and the MS/MS spectra. The degree of lipid peroxidation, DPPH radical scavenging activity, proline and soluble protein content were determined using spectrophotometric methods. The use of fluridone did not cause significant changes in the content of unbound fatty acids, although there was a tendency to increased amounts of unsaturated acids. Concerning total fatty acids, the use of fluridone significantly increased the ratio of oleic acid to stearic, as well as the ratio of total unsaturated acids to saturated acids in the treated pericarp of tomato fruits. Fluridone had not effect on the content of proline, total phenolic acids, MDA and antioxidant activity, but inhibited soluble protein accumulation and enhanced the content of total flavonoids.

We examined the response of maturing tomato fruit exposed for 7 days to fluridone (1-methyl-3-phenyl-5-[3-trifluoromethyl(phenyl)]-4(1H)-pyridinone). Fluridone was applied in lanolin paste in the form of a 2–3 mm wide strip from the top to the base of the fruit. As a control, a similar stripe of lanolin was applied in the same way on the opposite side of the same fruit. The content of major carotenoids, as well as flavonoids, and free and bound phenolic acids were determined using a HPLC and HPLC-MS-MS methods. Fluridone almost completely blocked the biosynthesis of lycopene and substantial declined content of ß-carotene and lutein in the tomato fruit. The fluridone caused a decreased content of quercetin, rutin and naringenin, and increased level of epicatechin. The herbicide did not affect the content of p-coumaric acid, but reduced the level of caffeic acid, both free and ester form, and declined the content of free ferulic and chlorogenic acids. Changes in phenolics composition observed for the first time indicate that fluridone interferes with the biosynthesis of further products of the metabolism of p-coumaric acid, both flavonoids and phenolic acids.

The experiments aimed to investigate the conversion of somatic embryos of the tulip ‘Apeldoorn’ variety in an in vitro culture supplemented with some chosen compounds. Tulip somatic embryos in torpedo stage, obtained by indirect somatic embryogenesis were placed for 1 week on media containing growth regulators (5 ěM Picloram, 1 ěM 6-benzylaminopurine (BAP) – control) and abscisic acid (ABA), abscisic acid + fluridone, Etephon and Etephon + salicylic acid (SA). Then, the embryos were maintained in the dark or under light for 10 weeks. After time of experiment, the greatest percent of leaf-forming embryos (40 %) and the greatest number of leaves (3.6 leaves) were observed when the embryos were treated simultaneously with Etephon and salicylic acid. Light did not influence the number of newly developed leaves but significantly increased (vs dark) percentage of leaf-forming embryos on control media (from 0 to 24 %) and on Etephon-supplemented medium (from 4 to 24 %). Abscisic acid and abscisic acid + Fluridone supplementation inhibited organogenesis of leaves but only in the cultures maintained under light. Regeneration of leaves was not observed on the control media maintained in the dark and on the abscisic acid + fluridone-supplemented media under light.
W przeprowadzonym doświadczeniu badano konwersję zarodków somatycznych tulipana 'Apeldoorn' pod wpływem wybranych czynników kultury in vitro. Zarodki somatyczne tulipana, w stadium torpedy, uzyskane drogą pośredniej embriogenezy somatycznej, wykładano na okres 1 tygodnia, na pożywki zawierające substancje wzrostowe (5 M Picloram, 1 M 6-benzyloaminopuryna (BAP) - kontrola) oraz: kwas abscysynowy (ABA), kwas abscysynowy+Fluridon, Etefon i Etefon+kwas salicylowy (SA). Następnie zarodki umieszczano na 10 tygodni w warunkach zaciemnienia lub światła. Po upływie czasu trwania doświadczenia zaobserwowano, że pod wpływem jednoczesnego działania Etefonu i kwasu salicylowego uzyskano największy udział zarodków formujących liście (40 %) oraz największą liczbę wytworzonych liści (3,6 szt.). Światło nie miało wpływu na liczbę powstałych liści, natomiast istotnie zwiększyło udział tworzących je zarodków na pożywkach kontrolnych (z 0 do 24 %) i po zastosowaniu Etefonu (z 4 do 24 %) w porównaniu z zarodkami utrzymywanymi w ciemności. Dodatek kwasu abscysynowego oraz kwasu abscysynowego i Fluridonu hamował organogenezę liści, ale tylko w obecności światła. Nie obserwowano regeneracji liści na pożywkach kontrolnych w warunkach zaciemnienia oraz pod wpływem kwasu abscysynowego i Fluridonu na świetle.
