W artykule przedstawiono techniczne i organizacyjne aspekty związane z produkcją na wielką skale samochodów osobowych, zasilanych z baterii akumulatorów nowej generacji. Przedstawiono techniczne możliwości w zakresie produkcji samochodów zasilanych energią elektryczną, w tym sposoby szybkiego uzupełniania energii akumulatorów i ich zalety oraz wady. Szczególną uwagę zwrócono na aspekt energetyczny, bezpieczeństwo w czasie eksploatacji i procesu ładowania baterii.
Some technical and organizational aspects of series production of electric cars, driven from a set of new generation rechargeable cells, are presented in the paper. Technical capabilities in the scope of a production of cars driven with electric energy, including methods of a quick charging of rechargeable cells as well as their advantages and disadvantages, are described. Special attention is paid to power aspect, operational safety and battery loading process.

Introduction. Treatment and diagnostic process in solid tumors like lung cancer are still based on invasive methods such as bronchoscopy, solid biopsy et cetera. One of the less invasive methods is a proposed “liquid biopsy” which is based on capturing of tumor cells circulating in the blood. Aim. The aim of the study was to standardize conditions and to assess the sensitivity of the identification of circulating tumor cells (CTCs) with the use of flow cytometry and qRT-PCR. Material and methods. In the first model of CTCs, cells from the A549 lung cancer cell line were suspended in 1 ml of healthy donors’ blood in 5 spikes increasingly: 0, 10, 50, 100 and 200 and the cells were detected in flow cytometer. In the second model, cells from the A549 and H1975 lung cancer cell lines were used. Spikes were prepared as in the first model, but cells were suspended in 400 µl of healthy donors’ blood and were detected with the use of qRT-PCR. Results. An increasing number of detected cytokeratin positive events from the 1st spike “0” to the last one - “200” was observed by flow cytometry. Median value in the negative control was 0 false positive cells. In tubes from “10” to “200” the median was 5, 43.5, 58 and 78, respectively. Mean sensitivity of flow cytometry was 63.79%. In qRT-PCR, correlation between increasing number of sorted cells in several spikes and the level of mRNA expression for KRT19 gene was not observed. Conclusion. Commonly available methods like flow cytometry and qRT-PCR seem to be attractive solutions for CTCs detection, but they need pre-enrichment procedures and standardization

The aim of this paper it to review various scale approaches to the blood flow modelling. Blood motion may be described by three types of mathematical models according to the observed scales or resolutions, namely microscopic, mesoscopic and macroscopic descriptions. The above approaches are discussed together with their advantages and disadvantages. Several results of mesoscopic simulations are presented with particular attention paid to mesoscale semi-continuum models suitable for real-time blood flow visualisation.

The development of electromobility is focused on the design and implementation of increasingly more effective electric drives. In such a system, apart from energy recovery, it is not possible to recharge the batteries while driving. Electric vehicles equipped with fuel cells and a battery (FCHEV – fuel cell hybrid electric vehicle) in a parallel configuration boast increased energy transfer capabilities. The article presents an energy flow analysis in a parallel hybrid drive system with fuel cells and a battery. The research was carried out on a 30 W vehicle made in 1:10 scale with a NiMH battery and a fuel cell with a proton exchange membrane (PEM). Increasing driving dynamics causes a 29% increase in energy consumption, 43.6% reduction of energy transfer from a fuel cell and a 23% increase of in the energy share intended for battery charging. Continuous operation of the system in full power mode ensures a much greater efficiency of energy transmission to the drive train (95%) compared to the system operating in dynamic driving conditions – 64–75%.

The aim of the study was to determine the relation between the cytotoxic and cytostatic effects of tezacitabine and cladribine on a HL-60 cell line and the time of exposure of cells to these drugs. Cell viability and induction of apoptosis were assessed using flow cytometry methods. Apoptosis was confirmed by direct microscopic observation. Growth inhibition was examined by cell counting. After 24 h incubation tezacitabine was equally or less toxic compared to cladribine. However, toxicity of tezacitabine strongly rose after 48 h incubation leading to massive cell death at doses much lower than those of cladribine. Assessment of the effect of increased exposure time on the clinical efficacy of tezacitabine is indicated.

Opis:
Characterization of free radical-induced cell injury processes of placenta cells is of vital importance for clinical medicine for the maintenance of intrauterine fetal life. The present study has analyzed cell injury processes in cells of the choriocarcinoma cell line JAR treated with menadione, an anticancer drug, and Hg2O2 in comparison to osteosarcoma 143B cells using electron microscopic and flow cytometric techniques. Flow cytometry on JAR cells exposed to 100 μM menadione and double-stained with Annexin V and propidium iodide (PI) detected apoptotic cells reaching the maximum after 4 h of incubation with a rapid decrease thereafter. Viable cells became decreased to 46% of the control after 2 h of incubation, reaching 5% after 4 h. Cells stainable with both Annexin V and PI began to increase distinctly after 2 h of incubation, reaching 55% after 4 h. Electron microscopy showed that cells stainable with both dyes specified above had condensed nuclei and swollen cytoplasm, suggesting that they were undergoing a switch of the cell death mode from apoptosis to necrosis. On the other hand, 90% of 143B cells remained intact after 4 h of menadione treatment although the intracellular levels of superoxide were always higher than those of JAR cells treated with the drug. In contrast, JAR cells were more resistant than 143B cells to H2O2-induced cytotoxicity. These results may suggest that cytotoxicity of menadione cannot be explained simply by oxygen free radicals generated from the drug. The resistance of JAR cells to oxygen free radical-induced cytotoxicity may be advantageous for intrauterine fetal life.

Magnetic-activated cell sorting (MACS) has become a standard method for the isolation of hematopoietic stem/progenitor cells (HSC/HPC) in human or mouse model using CD34 antibodies. However, at the present there is no useable CD34 antibody that could be successfully used for the selection of rabbit HSC/HPC. Therefore, the aim of this preliminary study was to remove all mature cells (CD45+) from the heterogeneous mixture of rabbit peripheral blood and bone marrow mononuclear cells (PBMCs and BMMCs) in order to enrich these cell populations for the CD34+ cells. Briefly, cells were incubated with a CD45 antibody and proper magnetic microbeads. Three different MACS sorting strategies were used in the experiment that differed mainly in the sample loading rate and the number of used magnetic columns. Control (unsorted) and sorted cells were assessed for the sorting efficiency (% of double positive cells for CD45 and Labelling Check Reagent - LCR) by flow cytometry and for the relative expression of CD34 antigen by qPCR. According to flow cytometry, Depl025 mode showed the best sorting efficiency in terms of the lowest percentages of CD45+LCR+ cells for rabbit PBMCs as well as BMMCs. qPCR analysis confirmed this mode as the best in terms of the relative CD34 expression for rabbit PBMCs. However, higher relative expression of CD34 in BMMCs was obtained by other mode - Posselds. In conclusion, this study demonstrates a possible enrichment of rabbit (CD34+) HSC/HPC by the magnetic depletion of mature hematopoietic (CD45+) cells.

Opis:
Non-point source pollution is a primary cause for concern globally. Various models have been developed to tackle this situation with much emphasis placed on best management practices. This practice has, however, proven to be insufficient to solve the NPS pollution situation. Existing non-point source models are watershed- -based and complicated both in operation, parameter estimation and data requirements. A non-point source model is proposed using the concept of the Hybrid Cells in Series model. The model is a three-parameter model made up of three zones, which describes pure advection through time delay in a plug zone, with combined advection and dispersion occurring when the other two zones are considered as thoroughly mixed. The proposed model is tested using synthetic data and field data from the Snake River, Colorado, USA, obtained from literature. Simulations were performed at four sample points; two from the tracer injection point along the Snake River before a confluence and two further downstream after the confluence. A regression analysis was carried out to determine the model’s capability to simulate pollutant transport for the four sampling points. The coefficients of determination are 0.98, 0.94, 0.84 and 0.97 while the standard error for each reach is 2.28E-2, 2.70E-2, 2.32E-2 and 9.35E-3 respectively. The results show good agreement between the measured and the simulated data. The response of the C-t profiles produced by the proposed model for both synthetic and field data demonstrates its ability to effectively simulate pollutant transport in natural rivers subject to non-point source pollution.

The aim of this paper was to demonstrate the feasibility of using a Computational Fluid Dynamics tool for the design of a novel Proton Exchange Membrane Fuel Cell and to investigate the performance of serpentine micro-channel flow fields. A three-dimensional steady state model consisting of momentum, heat, species and charge conservation equations in combination with electrochemical equations has been developed. The design of the PEMFC involved electrolyte membrane, anode and cathode catalyst layers, anode and cathode gas diffusion layers, two collectors and serpentine micro-channels of air and fuel. The distributions of mass fraction, temperature, pressure drop and gas flows through the PEMFC were studied. The current density was predicted in a wide scope of voltage. The current density – voltage curve and power characteristic of the analysed PEMFC design were obtained. A validation study showed that the developed model was able to assess the PEMFC performance.

Opis:
Flotation enrichment is a complex process due to the influence of many factors on the course of this process. In macroscopic terms, it is assumed that the quantity that takes into account the influence of all important factors on the enrichment process is the flotation velocity coefficient k. In industrial practice the flotation process is followed by heterogeneous material in terms of flotation properties. Therefore, the flotation velocity coefficient k will be characterized by a certain distribution. Thus, the distribution can be described as the function of the density distribution of the particles flotability f(k). Characteristics of this type are used in the design and optimization of flotation technological circuits. The paper presents a three cells flotation machine with a specific structure of connections of individual cells and the methods of selecting their parameters. The proposed circuit of the three cells flotation machine is characterized by a different connection structure and the specific parameters of each cell than the serial (cell-to-cell) flotation machine. The defined structure of connections between the cells and proper selection of their parameters ensures the maximum slope of the “separation characteristics” of the machine for the velocity coefficient k. Conducting the process of mineral enrichment in the proposed system enables the most efficient separation of the useful feed components. [...]
Wzbogacanie flotacyjne jest procesem złożonym ze względu na wpływ wielu czynników na przebieg tego procesu. W ujęciu makroskopowym przyjmuje się, że wielkością która uwzględnia wpływ wszystkich istotnych czynników na proces wzbogacania, jest współczynnik prędkości flotacji k ziaren. W praktyce przemysłowej do procesu flotacji kieruje się materiał niejednorodny pod względem właściwości flotacyjnych. W związku z tym współczynnik prędkości flotacji k będzie się charakteryzował pewnym rozkładem. Nadawę można zatem opisać funkcją gęstości rozkładu flotowalności ziaren f(k). Charakterystyki tego typu stosuje się przy projektowaniu i optymalizacji układów technologicznych flotacji. W artykule przedstawiono maszynę flotacyjną trójkomorową o określonej strukturze połączeń poszczególnych komór i sposobie doboru ich parametrów. Proponowany układ połączeń maszyny flotacyjnej trójkomorowej charakteryzuje się inną od szeregowej strukturą połączeń oraz ściśle określonymi parametrami każdej komory. Zdefiniowany sposób połączeń komór oraz odpowiedni dobór ich parametrów zapewnia maksymalne nachylenie charakterystyki rozdziału maszyny dla współczynnika prędkości k rozdziału. Prowadzenie procesu wzbogacania, w proponowanym układzie, umożliwia najdokładniejszy rozdział składników użytecznych nadawy. [...]
