Vessel traffic systems provide a high level of safety on coastal waters due to coastal radar stations and industrial cameras transmitting information to traffic supervision centers, as well. To improve a vessel traffic services is very important to ensure the speed and secrecy for the transmission of video images. The paper presents the basic issues of the multimedia data protection by digital watermarking and fingerprinting methods. Main applications for such digital marking were described in the paper as well as its requirements. Furthermore, the importance of multicast transmission for fingerprinting methods was presented by comparing the scalability of methods using only unicast transmissions and methods using multicast transmissions. The paper also presents the greatest threat to fingerprinting methods, which are attacks performed by more than one pirate. These attacks are called collusion attacks. The criteria that should be followed during identifying rogue users taking part in the collusion attack on the security systems has been presented. The paper also contains description of the extended Hillcast method, which belongs to the group of JFD (Joint Fingerprinting and Decryption) methods. The method provides a cryptographic security and digital fingerprinting of multimedia content, while maintaining high scalability. Main purpose of this method is VoD (Video on Demand) service, but it can also be used in vessel traffic supervision systems, such as VTS and AIS. In the last part of paper, there are results of studies which indicate high resistance to most common collusion attacks. Method proposed by authors can enhance the security of visual data transmission in vessel traffic systems.

Invasive alien species are non-native plant species that displace native species and pose adverse effects to environment, ecosystem, economy, and human health by diminishing the growth of native plants and by exhibiting higher stress tolerance. In our present study, four invasive alien species, namely Lantana camara, Parthenium hysterophorous, Ricinus communis, and Ageratum conyzoides, were studied from different locations. Plants growing under natural environmental conditions were sampled at random in the vicinity of Jalandhar. To gain insights into the biochemical basis of invasiveness of these plants, the samples were subjected to chemical fingerprinting by using UV-Vis, fluorescent, and Fourier transform-infrared (FT-IR) techniques under natural abiotic stress conditions (moderate and hot conditions). Indices of oxidative stress, such as malondialdehyde (MDA), were also studied. MDA levels were enhanced under hot conditions. Elevated peaks (major and minor) were observed in UV fingerprinting during adverse abiotic conditions. Fluorescent spectroscopy also validated the enhanced levels of secondary metabolites. FT-IR spectroscopy confirmed the presence of alkaloids and phenolics during stress conditions. Peaks were identified as rutin, vanillic acid, ascorbic acid, and glutathione reduced. The obtained results showed that under stressful conditions, the studied plants may produce an increased level of metabolites that might play a role in minimizing the oxidative stress faced by these plants. It was concluded that the studied plants, namely P. hysterophorus, L. camara, R. communis, and A. conyzoides, have the potential to cope with abiotic stress such as high temperature, which could be the reason for their invasiveness and vast adaptability.

As a result of their unpredictable ability to adapt to varying environmental conditions, microorganisms inhabit different types of biological niches on Earth. Owing to the key role of microorganisms in many biogeochemical processes, trends in modern microbiology emphasize the need to know and understand the structure and function of complex microbial communities. This is particularly important if the strategy relates to microbial communities that cause biodeterioration of materials that constitute our cultural heritage. Until recently, the detection and identification of microorganisms inhabiting objects of cultural value was based only on cultivation-dependent methods. In spite of many advantages, these methods provide limited information because they identify only viable organisms capable of growth under standard laboratory conditions. However, in order to carry out proper conservation and renovation, it is necessary to know the complete composition of microbial communities and their activity. This paper presents and characterizes modern techniques such as genetic fingerprinting and clone library construction for the assessment of microbial diversity based on molecular biology. Molecular methods represent a favourable alternative to culture-dependent methods and make it possible to assess the biodiversity of microorganisms inhabiting technical materials and cultural heritage objects.

The South-Western highlands of Ethiopia are considered to be the centre of origin and diversity of the arabica coffee, Coffea arabica. More than 80 accessions of arabica coffee collected from Ethiopia are avail-able in Indian gene bank. However, the genetic diversity of these accessions is not studied in detail. In the present study, genetic diversity analysis of 48 accessions collected from eight provinces of Ethiopia was car-ried out using Sequence-related amplified Polymorphism (SRAP) marker. Among the thirty two SRAP primer combinations tested, 14 primer pairs were polymorphic and generated 203 distinct fragments. The number of fragments ranged from 7 to 21 with a mean of 14.5 fragments per primer combination. Of the total 203 ampli-fied fragments, 182 (89.65%) were polymorphic and the percent of polymorphism ranged from 53.84% to a maximum of 100% using different primers. The average resolving power (Rp) and average polymorphism information content (PIC) of the 14 SRAP primer combinations was 14.31 and 0.648 respectively. A total of 13 rare alleles were obtained from SRAP assays, of which six rare alleles were obtained from the accessions collected from Shoa province.The UPGMA clustering algorithm from SRAP analysis grouped the 48 coffee accessions into two major clusters. The accessions collected from particular province clustered together which could be attributed to the substantial gene flow between adjacent population and the influence of geographical origin on genetic diver-sity. The study demonstrated the existence of substantial genetic variation in Ethiopian germplasm which could be utilized in coffee germplasm conservation and improvement program.

Species of Gleditsia show considerable morphological variability that makes them difficult to distinguish using either vegetative or floral characters. Honeylocusts, especially the thornless cultivars, are popular ornamental, shade, street, attractive landscape trees. In this study the ISSR technique was used to evaluate the range of genetic variability between seven genotypes of Gleditsia cultivated in Polish dendrological collections [Gleditsia caspica Desf., Gleditsia japonica Miq., Gleditsia japonica Miq. var. korainensis (= G. korainensis Nakai), Gleditsia triacanthos L., Gleditsia triacanthos L. (bulk), Gleditsia triacanthos f. inermis (L.) Zabel (bulk). Forty ISSR primers were tested and 18 were selected for their ability to produce clear and reproducible patterns of multiple bands.A total of 177 loci of 260-2600 bp were amplified, of which 89 (50%) were polymorphic, 14 (8%) monomorphic and 74 (42%) were accession-specific. Accession-specific ISSR loci were obtained for all of the seven accessions tested. A dendrogram generated using the UPGMA, based on a similarity measure of total character difference, showed that the Gleditsia accessions were clustered into two main groups (‘a’ and ‘b’). The first grup –‘a’– included: Gleditsia triacanthos L., Gleditsia triacanthos L. (bulk) and Gleditsia triacanthos f. inermis (L.) Zabel (similarity 0.61–0.75), the second –‘b’– included 2 species: Gleditsia japonica and Gleditsia japonica var. korainensis (similarity 0.43). Analysis of the phylogenetic similarity dendrogram has shown wide range of diversity between studied accessions. The clustering pattern obtained in our experiment was in agreement with the data based on morphological, allozyme and ITS analysis.

Opis:
A biosynthetic human insulin precursor displayed enhanced susceptibility to deamidation at one particular site. The present study was undertaken to monitor progress of precursor deamidation at successive manufacturing stages. MALDI-TOF/TOF MS in combination with controlled endoproteinase Glu-C and endoproteinase Asp-N proteolysis was used for rapid and unambiguous determination of deamidated residue within the investigated structure. Close inspection of isotopic distribution patterns of peptides resulting from enzymatic digestion enabled determination of distinct precursor forms occurring during the production process. Asn, Asp, isoAsp and succinimide derivatives of the amino acid at position 26 were unambiguously identified. These modifications are related to the leader peptide of a precursor encompassing amino acid sequence corresponding to that of superoxide dismutase [Cu-Zn] (SOD1 1, EC=1.15.1.1). Monitoring of precursor deamidation process at successive manufacturing stages revealed that the protein folding stage was sufficient for a prominent replacement of asparagine by aspartic and isoaspartic acid and the deamidated human insulin precursor constituted the main manufactured product. Conversion proceeded through a succinimide intermediate. Significant deamidation is associated with the presence of SNG motif and confirms results achieved previously on model peptides. Our findings highlight an essential role of the specific amino acid sequence on accelerated rate of protein deamidation. To our knowledge, this is the first time that such a dramatic change in the relative abundance of Asp and isoAsp resulting from protein deamidation process is reported.

Fingerprinting of corpses is always problematic even for experienced forensic experts and technicians. This publication is a compendium of the knowledge about the physical and chemical methods in dactyloscopy and improving the readability of the friction ridges of corpses. It mentions the possibility of a very broad use of not only dactyloscopic paints, but also fingerprinting powders, films, as well as silicone casts, latex matrices etc. The publication also highlights the necessity of photographic documentation of fingerprints in every case and after every completed procedure, with the aim of improving the readability of the corpse's friction ridges.

Wprowadzenie w spektrometrach jonizacji typu MALDI zrewolucjonizowało proces identyfikacji białek. Automatyzacja procesu identyfikacji oraz bezpośrednie połączenie analizy spektrometrem masowym z separacją białek dwuwymiarową elektroforezą żelową (2D-GE) pociągnęły za sobą znaczny rozwój proteomiki. Późniejszy rozrost proteomicznych baz danych pozwolił na zwiększenie dokładności identyfikacji, z wykorzystaniem pierwszej w historii techniki wydajnej identyfikacji białek – peptide mass fingerprinting, w skrócie: PMF. Metoda peptide mass fingerprinting pozwala identyfikować białka z widm masowych uzyskanych w wyniku analizy próbki spektrometrem masowym. Przez wzgląd na powszechność stosowania metody, jak i ciągle obserwowane jej ulepszenia, autorzy postanowili podsumować obecny stan wiedzy w tym zakresie. Praca została podzielona na dwie części: w pierwszej znajduje się opis historii powstania metody PMF wraz z charakterystyką części eksperymentalnej i opisem najpopularniejszych baz danych stosowanych przy identyfikacji, natomiast druga część pracy jest poświęcona zagadnieniom algorytmicznym, związanym z wyszukiwaniem w bazie danych protein najlepiej odzwierciedlających białko analizowane w próbce. Specyfikacja eksperymentu w pierwszej części pracy uwzględnia zarówno opis metody separacji, trawienia białek w próbce, jak i późniejszej ich analizy z wykorzystaniem spektrometru masowego. Eksperymentalne fazy metody PMF są opisane z uwzględnieniem ich cech biochemicznych, mających wpływ na dalsze etapy schematu identyfikacji.
The development of MALDI ionization method in mass spectrometers, had revolutionized the protein identification procedure. The automation of an identification procedure and the mass spectrometry direct connection to the protein separation with the two-dimensional gel electrophoresis (2D-GE) implicated the significant proteomics development. The later growth of the proteomics databases contributed to the enhancement of the identification accuracy, by using the first method of effective protein identification in the history: the peptide mass fingerprinting (PMF). The peptide mass fingerprinting enabled the protein identification from the mass spectra acquired by the mass spectrometry sample analysis. Due to the common use of method and its continuous improvements, the authors decided to summarize the current state of the knowledge in this field of science. The publication is divided into two parts. The first one is devoted to the origins of PMF scheme, the characteristics of its experimental part and a description of the most popular databases used in the identification procedure. The second part relates to the algorithmic issues of searching the database protein, which reflects the sample content in the best way. The experiment specification in the first part takes into the consideration the description of separation and sample digestion methods, as well as the later protein sample analysis by the mass spectrometer. The experimental steps of the PMF method are described according to their biochemical properties, having an impact for the later stages of the identification procedure.

What is the microbe that we are dealing with? Whether it is cholera or anthrax, we want to know the disease-causing microorganism as quickly as possible since prompt identification of the causative organism would help control disease spread - and potentially save lives through provision of appropriate care and medication. Yet, despite the advent of rapid microbial identification tools – particularly those based on mass spectrometry – most undergraduate curricula continue to focus on culture- and nucleic acid-based identification techniques since they are widely used for detecting and identifying microbes in clinical and environmental samples. Mass spectrometry-based methods, however, have increasingly complemented traditional approaches in clinical and research laboratories - but are rarely featured in undergraduate curricula. Motivated by the desire to address the curriculum gap, the author of this study developed an inquiry-based laboratory exercise for introducing students to the operating principles and methodology of mass spectrometry-based microbial identification. By requiring students to identify microbes in environmental water samples – a real-life problem with unknown answers – the exercise piqued the students’ interest in learning, while helping to stir their curiosity through an interesting field activity in which they could put on a scientist’s hat in solving a mystery. This synopsis article summarizes a piece of published educational research and expands on the discussion of concepts underlying matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS)-based microbial identification. Herein, the article discusses the relative advantages and disadvantages of the pattern recognition and proteome database search approaches for analyzing mass spectra data. Additionally, the effect of general and tailored sample preparation protocols on identification accuracy is also elaborated. Finally, the pedagogical utility of field- and inquiry-based educational tools is also discussed in greater detail from a post-publication perspective.
