The increase in the number of filamentous bacteria in activated sludge causes complications in the functioning of the sewage treatment plant. The paper presents the results of investigation of activated sludge coming from two sewage treatment plants - municipal and industrial. In both treatment plants the increase in the number of filamentous bacteria in winter has been noticed. In order to reduce the number of filamentous bacteria in activated sludge, the sludge samples were subjected to ultrasonic disintegration at a frequency of 40 kHz at 1, 2, 3, 5, 10 and 20 minutes. The use of low frequency ultrasounds has contributed to the defragmentation and destruction of most filamentous bacteria in activated sludge after 10 minutes of their operation. Necessary time to almost complete defragmentation of filamentous bacteria is more than 20 minutes.
Przyrost liczebności bakterii nitkowatych w osadzie czynnym powoduje komplikacje w funkcjonowaniu oczyszczalni ścieków. W pracy przedstawiono wyniki badań osadu czynnego pochodzącego z dwóch oczyszczalni ścieków - komunalnej i przemysłowej. W obu oczyszczalniach stwierdzono wzrost liczebności bakterii nitkowatych w okresie zimowym. W celu ograniczenia liczebności bakterii nitkowatych w osadzie czynnym próbki osadu poddano dezintegracji ultradźwiękami o częstotliwości 40 kHz w czasie 1, 2, 3, 5, 10 i 20 minut. Zastosowanie ultradźwięków o niskiej częstotliwości przyczyniło się do defragmentacji i zniszczenia większości obecnych w osadach bakterii nitkowatych po czasie 10 min. Do całkowitej defragmentacji bakterii nitkowatych niezbędny jest czas powyżej 20 min.

This work is a comprehensive study of a full-scale municipal wastewater treatment plant (WWTP) in the city of Zgierz (Poland) aiming at the estimation of the impact of seasonal changes of temperature on flocs morphology, biomass concentration, quantity of filamentous bacteria, content of polyhydroxybutyrate (PHB) and composition of microinvertebrates in the activated sludge system. Two short-term measurement campaigns, in winter and in summer, were carried out under dry weather conditions. It occurred that lower temperatures (11.1–14.6 °C) had hardly any effect on flocs size or concentration of activated sludge biomass in the full-scale activated sludge system treating municipal wastewater. They did not aggravate the biosynthesis of PHB either. However, decreasing temperature favored the growth of filamentous bacteria and led to the elevation of the sludge volume index (SVI). In spite of this, it did not induce any bulking events. Irrespective of the season, the groups of crawling ciliates, attached ciliates and testate amoebae, indicating good performance of the full-scale plant tested, were predominant.

Bulking and foaming of activated sludge are related to excessive proliferation of a specific group of activated sludge biocenosis – filamentous bacteria. The research was carried out to compare filamentous bacteria populations in foam and activated sludge in a full-scale municipal wastewater treatment plant located near Warsaw (Poland). Fluorescent in situ hybridization (FISH) – a quantitative, culture-independent, molecular method was applied to evaluate the structure of filamentous populations. Activated sludge and foam were examined for the abundance of eleven groups of these microorganisms, which occur in wastewater treatment plants in Europe. Filamentous bacteria constituted 18% and 24% of all bacteria detected in sludge and foam, respectively. The structures of filamentous bacteria populations in sludge and foam were different, although the same bacteria were found in both types of samples. The most abundant filaments belonged to phylum Chloroflexi (targeted by the CFXmix probe) and genus Microthrix (targeted by the MPAmix probe) in sludge and foam, respectively. The third significantly abundant bacteria was Haliscomenobacter hydrossis (targeted by HHY654).

W pracy omówiono możliwości zastosowania cyfrowej analizy obrazu do oceny morfologii i składu osadu czynnego wraz z przedstawieniem przykładowych, uproszczonych procedur jej wykonania. Zautomatyzowane procedury analizy obrazu są najczęściej wykorzystywane do (1) pomiarów wielkości kłaczków osadu, (2) oszacowania stężenia biomasy, (3) oznaczenia udziału bakterii nitkowych, (4) oszacowania udziału bakterii akumulują-cych polifosforany, (5) identyfikacji pierwotniaków i metazoa.
The application of digital image analysis for the evaluation of activated sludge is discussed in the paper. Also the simplified procedures of image analysis were presented as examples. Automated image analysis procedures were the most often used for the following purposes: (1) size measurements of sludge floes, (2) estimation of biomass concentration, (3) determination of filamentous bacteria fraction, (4) estimation of polyphosphate accum ing bacteria fraction, (5) identification of protozoans and metazoans.

Sludge sedimentation depends on various factors like: air temperature, quality of wastewater (BOD), characteristics of a bioreactor (sludge age, oxygen concentration in nitrification chamber, recirculation rate, concentration of sludge in a bioreactor), dosage of coagulants for phosphorus removal and a presence of filamentous bacteria. However, still there is a lack of a mathematical model that would established the dependency between all these inputs, because of their number, and sludge sedimentation. In the presented study, such model is proposed, moreover its structure allows for a direct interpretation of its parameters, making it especially useful for operators of treatment plants. The article presents the application of the logistic regression for the estimation of sludge sedimentation potential, using the characteristic of the sludge (concentration and temperature) in a respect of the presence of filamentous bacteria. The sludge deterioration was measured using a threshold a value of the Sludge Volume Index (SVI = 150 mg/dm3). Results suggests that Sphaerotilus natans and Nostocoida sp. bacteria are the most noticeable factor for the sludge deterioration and the sediment potential.

The activated sludge method is most frequently used at the stage of biological waste water treatment. The main reason for improper sludge sedimentation is the excessive growth of filamentous bacteria leading to disturbances in technological processes of sewage treatment plants, such as formation of swollen, flowing out sludge, foam transforming in to a thick surface scum, which consequently contributes to a decrease in the effectiveness of removing pollutions from sewage. There is a growing tendency to make attempts aiming at optimizing the chemical methods used to eliminate filamentous bacteria which often constitute a factor limiting proliferation of those bacteria. Accordingly, research was undertaken to estimate the effect of some iron and aluminum coagulants and oxidizing compounds (chlorine dioxide and sodium hypochlorite) on the elimination of filamentous bacteria in the activated sludge in the aspect of sewage treatment effectiveness. Activated sludge was collected from the mechanical and biological sewage treatment plant situated in the Kuyavian-Pomeranian voivodeship. The study was carried out using a laboratory biological reactor graduated to reflect the technological parameters of the sewage treatment plant. Analyses involved determination of physicochemical parameters (total suspensions, sludge volumetric index, sludge density index) and the microscopic evaluation of activated sludge (floc parameters, the composition of microfauna, the Sludge Biotic Index, the quantitative and species composition of filamentous bacteria). At the next stage of the experiment, water solution of iron chloride, water solution of polyaluminum chloride, chlorine dioxide and sodium hypochlorite were proportioned to the activated sludge. At the same time, samples of activated sludge were analyzed without chemical reagents. After a week, the effect of applied reagents on the quantitative and species composition of filamentous bacteria and the other components of activated sludge biocenosis was evaluated. It was proved that activated sludge subjected to analyses was characterized by poor sedimentation properties, resulted from the excessive proliferation of filamentous bacteria of which the most numerous, dominant group was the species Microthrix parvicella. Best effects of filamentous bacteria elimination were recorded after the use of aluminum coagulant, which selectively affected filamentous microorganisms, effectively limiting their excessive proliferation and at the same time not exerting the negative impact on the other organisms of activated sludge. The preparation affected favorable values of the Sludge Biotic Index, which did not substantially differ from that parameter in the control sample. Polyaluminum chlorite caused elimination of the bacteria Sphaerotilus natans and a considerable decrease in the numbers of type 0041micro-organisms. At the same time that preparation contributed to improving sedimentation properties of activated sludge flocs, as a result of which the volumetric index values were decreased in successive research samples, whereas the density index increased. The applied sodium hypochlorite contributed to the elimination of filamentous bacteria, and at the same time it slightly decreased the other parameters of activated sludge. It was indicated that chlorine dioxide contributed to a considerable decrease in diversity of activated sludge microfauna, causing a decrease in the number of crawling ciliates on the flocs and sedentary forms of those macroorganisms. Moreover an increase in the number of bacteriocidal free-floating protozoa and small flagellates resulted in a substantial decrease in the IBO value. The highest doses of reagent caused impoverishment of the biocenosis, which resulted in a decrease in the biotic index values by 7 units in relation to the control sample, activated sludge in those samples was qualified as IV class of sludge quality. The best effects in filamentous bacteria elimination were recorded after the use of aluminum preparation and chlorine compounds (chlorine dioxide, sodium hypochlorite).

Excessive growth of filamentous bacteria, inducing activated sludge bulking, presents a serious problem in many wastewater treatment plants (WWTPs). The idea of using Lecane inermis rotifers as a tool for controlling the filamentous bacteria density in activated sludge requires developing a method of culturing rotifers at high concentrations. The objective of this work was to determine the effect of two culture media on the growth of three Lecane inermis strains. The growth of rotifers from a single individual (parthenogenetic female) fed the tested culture media was observed for 10 days. The rotifers showed different individual growth, depending on the strain and type of culture medium. The results of the studies suggest that by using the tested culture media, it may be possible to select a Lecane inermis strain with the highest culturing capacity at high density of rotifers in order to inoculate activated sludge with these organisms in wastewater treatment plants.
