An efficient in vitro regeneration and conservation system depend on the healthy culture induction from the suitable explants. Culture induction is a significant stage when very small plant material exists from the rare species. It is necessary to develop culture induction protocol from various explants to conserve the valuable plant species. Atalantia monophylla is a rare species with life giving properties. Shoots were induced from the shoot tip, node and internode explants. Among these the nodal shoot segments were reported most appropriate explant for the induction of shoots from the nodal meristems. MS medium (Murashige and Skoog) proved better than the Woody Plants (WP) medium in bud breaking. Sucrose at 3% level was optimum concentration for the establishment of cultures than the other concentrations evaluated. Shoot tips responded on MS medium supplemented with 0.5 mg L-1 each of BAP and Kin, nodal shoot segments responded better on MS medium augmented with 1.0 mg L-1 BAP . Half strength MS medium supplemented with 2,4-D (1.0 mg L-1) induced maximum responsive callus (87%) from the internode explants.

Plants have been an important source of medicine for thousands of years. Medicinal plants are the most important source of life saving drugs. Even today, the WHO estimates that up to 80 per cent of people still rely mainly on traditional remedies such as herbs for their medicines. Medicinal plants are resources of new drugs. Approximately one quarter of prescribed drugs contain plant extracts or active ingredients obtained from plant substances. Medicinal plants are the most important source of life saving drugs for the majority of the world’s population. The biotechnological tools are important to select, multiply and conserve the critical genotypes of medicinal plants. In-vitro regeneration holds tremendous potential for the production of high-quality plant-based medicine. An efficient protocol was developed for high frequency and rapid regeneration was achieved in Withania somnifera (L.) Dunal. Invitro cultures were initiated by meristematic explants on M.S–B5 medium with various growth hormones individually as well as combinations. Highest shoot multiplication rates were observed when explants grown in the medium supplemented with 2.0 mg/l of BAP. M.S–B5 medium fortified with 1.0 mg/l BAP was found suitable for optimum growth and elongation of shoots. The best rooting response were observed when the medium containing 1.0 mg/l of IBA. The rooted plantlets were successfully established in soil.

In vitro micropropagation was achieved from Nodal and leaf explants of Vernonia cinerea L. on MS medium supplemented with different concentrations of BAP (1.0 to 3.0 mg/l), GA3 (1.0 to 5.0 mg/l) and KN (0.5 mg/l) for direct shoot induction. Maximum numbers of shoots were obtained from nodal and leaf explants at BAP (2.5 mg/l) GA3 (2.0 mg/l) and KN (0.5 mg/l) respectively. The multiple shoots elongation and their development were better on MS medium supplemented with 1.5 mg/l BAP and 3.0 mg/l GA3 for both the explants. The regenerated shoots were successfully rooted on MS medium supplemented with IBA 2.0 mg/1, after sequential hardening, survival rate was 91%. The in vitro raised plantlets were acclimatized and established successfully in the field.
