Seeds of sesame (Sesamum indicum L), genotypeTMV3 were treated with various concentrations of EMS and Colchicine like 0.2, 0.4, 0.6, 0.8, 1.0, 1.2, 1.4, 1.6, 1.8 and 2.0 % and the LD50 values observed at 1.0 % of EMS and 0.6 % of Colchicine. Based on their LD50 values appreciable concentrations only selected for further study and their effects on various morphological characters such as plant height per plant, number of branches per plant, number of leaves per plant, days to first flowering, number of capsules per plant, number of seeds per capsule and seed yield per plant (g) were measured quantitatively and the results showed that all the characters were consequently or significantly reduced when compared to control and also EMS showed more effective than colchicine.

Opis:
Effect of EMS (ethyl methane sulphonate) on induced morphological mutants and chromosomal variation in cowpea was studied using five different doses of mutagen along with a control in randomized blocked design with three replications. The morphological mutants there are two types of viable and chlorophyll mutants. Viable mutant contains tall, dwarf, early maturity, late maturity, leaf mutants pod mutant and flower mutants. The frequency of chlorophyll mutant contains albino, xantha and viridis. This concentration can damage or modify important components of plant cells and have been reported to affect the morphology, anatomy, biochemistry and physiology of plants differentially depending on the concentration level. These effects include changes in the cellular structure and metabolism of the plants e.g., dilation of thylakoid membranes, alteration in photosynthesis, modulation of the antioxidative system and accumulation of phenolic compounds. The morphological and chromosomal variation was found to be mutagen sensitive in somatic cells of cowpea. It was found to increase with increasing the concentration of EMS in Cowpea plants. The chemical mutagen like ethyl methane sulphonate induces high frequency of chromosomal changes like anaphasic bridge; anaphasic laggard, anaphasic bridge and clumbing of chromosome were including control plants also observed.

Opis:
In recent years, the demand of chilli has tremendously increased due to its attractive market price and multifarious used in cooked and processed forms. At present people are much concerned about the fruit quality and yield. Therefore, attention is being paid for development of genotypes having high yield potential with desirable fruit quality characters in a short period of time. For this purpose, seeds of chilli were mutagenised with ethyl methane sulphonate (EMS) and diethyl sulphate (DES) to determine their mutagenic sensitivity in M1 generation. The increasing concentration of EMS and DES decreased in morphological and yield characters. The spectrum of mutation and induced variability for various quantitative traits were observed in M1 generation such as germination (%), plant height, primary and secondary branches per plant, days to first flowering, fruit length (cm), fruit girth (cm), total number of fruits per plant, number of seeds per fruit, seed weight per fruit (g), 100 seed weight (g) and pericarp: seed ratio showed variability in chilli with the effect of EMS and DES. The percentage of chromosomal abnormalities in different mitotic stages was significantly higher than that of the control in all the treatment concentrations.

The effect of gamma irradiation and EMS treatment on seed germination and seedling height of Chick pea (Cicer arietinum. L.). In this regard Co-4 variety of chick pea was subjected to different doses/concentrations of gamma rays (20, 30, 40, 50, and 60 kR) and EMS (10, 20, 30, 40 and 50 mM) for inducing mutation. The effect of mutagen was observed on the basis of percentage of seed germination, seedling height reduction at 15th day and survivability. From the result it was observed that, the percentage of seed germination, seedling height reduction at 15th day and survivability were significantly decreased with increasing doses/concentrations of mutagen. The effective doses/ concentrations which caused 50 % growth reduction were 40kR in gamma rays and 30 mM in EMS.

Opis:
Beauveria bassiana is an entomopathogenic fungus that is widely used in Thailand to control pest insects. However, the increasing temperature has influenced the insect control efficiency of the fungus. Therefore, determination of thermotolerant isolates of B. bassiana that can grow and remain pathogenic at higher temperatures than its current optimum temperature may be a better way to control pest insects in a high temperature environment. Three isolates of B. bassiana obtained from the Rice Department, Thailand were selected for mutagenesis using ethyl methanesulfonate (EMS) with subsequent screening at high temperatures (33 and 35°C). In addition, the recovery of fungal growth after exposure to a high temperature for a period of time (5–15 days) and then transferring to 25°C was evaluated. No isolates were found that grew at 35°C but one mutant isolate (BCNT002MT) produced larger diameter colonies and more spores than the corresponding wild type (WT) at 33°C. Growth and spore production of the BCNT002MT isolate were greater than its WT when incubated at 25°C for 14 days following exposure to 33°C for 7 days. In addition, the spore germination level (%) of BCNT002MT was significantly higher than its WT during culture at 25°C after prior exposure to 33°C for 5, 10 and 15 days. The pathogenicity against the brown planthopper, Nilaparvata lugens (Stål), of this mutant isolate was also prominent.
