Opis:
Background. The influence of infection with two species of entomopathogenic nematodes of Steinernematidae family on the activity of superoxide dismutase (SOD) of the host was studied. Material and methods. Last instar larvae of Galleria mellonella were experimentally infected with Steinernema affinis and S. feltiae at 20 invasive juveniles per insect. At 6, 12, 18, 24 and 36 h after infection activity of SOD was determined in extracts from infected and control insects. Results. The activity of SOD decreased gradually in the controls during the experiment. The activity of enzyme was 2-4-times higher in insects from both infected groups than in the control. During the first 12 h of infection the activity of SOD in insects infected with S. feltiae was higher than in those infected with S. affinis, then the activity of enzyme in the insects of both infected groups stayed at a similar level. A significant decrease of SOD activity in infected was recorded in second day of the infection.

Opis:
Concentration of glycogen, maltose, and trehalose as well as the activity of hydrolases catabolising them: «-amylases, glucoamylase, maltase, and trehalase were studied during a 48 h infection of Galleria mellonella (Lepidoptera) instar larvae infected with infective Heterorhabditis zealandica (20 nematodes/insect). The content of trehalose in the infected insects during the first day of infection was higher (p < 0.05) than in the controls. The concentration of maltose and glycogen were similar in both groups despite the fact that the activity of all examined o-glycosidases was generally much higher in the infected insects than in the controls.

Similar patterns of hydrolases were observed in three species representing two genera of entomopathogenic nematodes: Steinernema affinis, S. feltiae, and Heterorhabditis zealandica. The same enzymes were present in the studied nematodes but they differed in the level of activity of individual sub-classes of enzymes. A higher activity of esterases and proteolytic enzymes could be observed for H. zealandica than for S. affinis and S. feltiae. On the other hand, the activity of glycosidases in steinernematids was generally much higher than in H. Zealandica.

This study focused on the effect of heavy metal cobalt ions (at concentrations of 1–1000 ppm) on the development and enzymatic activity of four entomopathogenic fungi: Beauveria bassiana, Beauveria brongniartii, Isaria fumosorosea and Metarhizium robertsii, commonly used in biological plant protection. It was found that each of the tested species of fungi reacted individually to contact with the Co2+ ions at their various concentrations. Depending on the variants of the experiment carried out, there were changes in the development of the mycelia (mainly growth inhibition) and their morphological features (color and structure) in comparison to the control samples. Co2+ ions had a fungistatic effect on all fungal strains, whereas a fungicidal effect was noted at concentrations of 750 ppm and 1000 ppm against M. robertsii and I. fumosorosea, respectively. In addition, there was a discrepancy in enzymatic activity between the tested fungal species developing in the medium with varying concentrations of metal salt. The inhibitory effect of Co2+ ions on lipase production was observed in I. fumosorosea. Protease production was stimulated in B. bassiana at all Co2+ concentrations, whereas in M. robertsii this effect was noted at 1 ppm. The changing dynamics of extracellular fungal hydrolases, due to the action of Co2+ ions, may translate into the role of these microorganisms in the processes of insect pathogenesis. This work suggests that severe pollution of the environment by cobalt could be a restrictive factor for the development and pathogenicity of entomopathogenic fungi and must be taken into account for their successful application in biological plant protection.

In this in vitro study, the influence of tin ions at concentrations of 1–1,000 ppm on the development and enzymatic activity of four entomopathogenic fungi (Beauveria bassiana, B. brongniartii, Isaria fumosorosea, and Metarhizium robertsii), that are commonly used in biological plant protection, are examined. Each of the fungal species tested reacted differently to contact with the Sn2+ ions at the tested concentrations. Exposure to Sn2+ ions affected the rate of development, morphology, and enzymatic activity of fungi. Of the four fungal species studied, M. robertsii was the most resistant and showed complete growth inhibition at the highest Sn2+ concentration tested (1,000 ppm). For the other entomopathogenic fungi, the fungicidal effect of Sn2+ ions was noted at the concentration of 750 ppm. Exposure to Sn2+ ions (up to 500 ppm) resulted in enhanced biochemical activity; and all entomopathogens that were tested showed increased production of N-acetyl-β-glucosaminidase (NAG) as well as several proteases. Moreover, B. brongniartii and M. roberstii showed increased lipases synthesis. These changes may increase the pathogenicity of the fungi, thereby making them more effective in limiting the population of pest insects. The exposure of the entomopathogenic fungi to a medium containing Sn2+ ions, at concentrations that were appropriate for each species, induced hyperproduction of hydrolases, which might be involved in aiding the survival of entomopathogenic fungi in the presence of heavy metals. This study shows that the fungistatic effect of Sn2+ on entomopathogenic fungi did not restrict their pathogenicity, as evidenced by the stimulation of the production of enzymes that are involved in the infection of insects.
