Image analysis is widely used for monitoring seeds during germination, and it is often the final phase of germination that is subjected to the greatest attention. However, the initial phase of germination (the so-called imbibition) also exhibits interesting behaviour. This work shows that image analysis has significant potential in the imbibition. Herein, a total of 120 seeds were analysed during germination tests, and information about seed size and shape was stored and analysed. It was found that the imbibition can be divided into two newly defined parts. The first one (‘abrupt imbibition’) consists mainly of the swelling of the seed embryo part and lasts approximately one hour. The second one, referred to as ‘main imbibition’, consists mainly of spatial expansion caused by imbibition in the other parts of the seed. The results presented are supported by the development of seed cross area and shape parameters, and by direct observation.

This study investigated the influence of cadmium (Cd), copper (Cu), iron (Fe), lead (Pb), selenium (Se), and zinc (Zn) in follicular fluid on the effectiveness of in vitro fertilization (IVF) treatment and the dynamics of embryo development. The study involved 221 women aged 25–35 years in whom intracytoplasmic sperm injection (ICSI) was performed. Analysis of the effects of the average concentrations of Se, Zn, and Cu on the developmental dynamics of embryos showed that higher concentrations of these metals were accompanied by shorter lead times for each of the various stages of development, while the reverse was true in the case of cadmium and lead. No relationship was observed between the mean concentrations of Fe and the dynamics of the human embryo. In order to discover whether the fact of becoming pregnant is affected by the investigated variables, a logistic regression model was applied. The dependent variable was the fact of achieving pregnancy, and the independent variable tested was the level of trace metals. Ultimately, only one variable remained statistically significant in the model: the level of Se.

Opis:
White lupin is an important source of proteins, the content of which amounts to 36–40% in its seeds. Breeding of new varieties usually takes several years. The present study aimed to shorten the breeding cycle by applying the single-seed descent technique together with in vitro culture of embryos dissected from immature seeds. The effects of temperature on embryo development were studied in indeterminate vs. determinate varieties in in vitro culture using the following temperature regimes:(1) 8 °C; (2) 16 °C in the first week, then 8 °C; (3) 20 °C in the first week, then 20/18 °C; and (4) 16 °C throughout the culture period. The embryos were cultured on the standard Murashige and Skoog medium. The observed growth rate of plantlets as well as the survival of plants ex vitro showed that at lower temperatures (8–16 °C) the plants developed slowly, but their ex vitro survival rate was higher compared to those grown at a higher temperature (20 °C). In addition, the results indicated that the in vitro embryo development rate of determinate varieties is significantly lower than that of indeterminate varieties.

Opis:
The effect of additional extremely low frequency (ELF) magnetic field (50 Hz) on the development of chick embryo was investigated. The experiment was carried out in three variants for induction values 15, 10 or 5 μT. Magnetic field (MF) in the experimental incubator was generated by a set of three Helmholtz coils. The following analyses were performed: embryopathological analysis; evaluation of the hatched chicks for quality; analysis of the course and synchronization of hatching Early embryo mortality in groups exposed to MF 10 and 15 μT compared to the control was higher by about 3 percentage points. Simultaneously, in these groups the hatching processes accelerated, but the quality of hatched chicks was much lower.

The completely new strategy of pseudophysiological transcomplementary (transcytoplasmic) activation (PP-TCA) of nuclear-transferred oocytes, which had been derived from pWAPhGH-GFPBsd transfected foetal fibroblast cells, was recently applied to the somatic cell cloning of pigs. It resulted in the considerable enhancing not only the cleavage activity of cultured cloned embryos, but also their morula and blastocyst formation rates as compared to the use of standard simultaneous fusion and electrical activation of reconstituted oocytes (77% vs. 57%, 63% vs. 46% and 40% vs. 27%, respectively). Altogether, the use of cytosolic components descended from heterologous (rabbit) zygotes as the agents for stimulation of porcine clonal cytoplasmic hybrids (cybrids) turned out to be reliable and feasible strategy for the generation of transgenic blastocysts by somatic cell nuclear transfer (SCNT). Furthermore, to our knowledge, no previous study has reported the preimplantation developmental outcome of transgenic nuclear-transferred pig embryos following the PP-TCA that was developed and optimised in our laboratory.

The actin and the tubulin cytoskeleton organization during the differentiation of the embryo-suspensor in Alisma plantago-aquatica was studied in comparison with the development of embryo, using immunofluorescence detection and rhodamine-phalloidin assay. At the early stage of the suspensor basal cell development (from 2- to ~10-celled embryos) microfilaments form an abundant network in the cytoplasm of the basal cell, while the microtubules form a delicate network. At the mature stage of development (from a dozen to several dozen-celled embryos), in the suspensor basal cell, the microfilaments and microtubules were localized from micropylar to chalazal pole of the cell. At the micropylar end of the basal cell a high amount of actin and tubulin material was observed. The microfilaments were mainly arranged parallel whereas numerous bundles of microtubules distributed longitudinally or transversally to the long axis of the cell. At this stage of basal cell functioning, some bundles of microtubules appeared to pass close to the nucleus surface. Microtubules were also observed distributed at the chalazal pole of the basal cell. At the senescence stage of the suspensor basal cell (>100-celled embryos) the actin and tubulin filaments disorganize, some disrupted microfilaments and microtubules were observed in the cytoplasm of the basal cell. At all stages of the suspensor basal cell development in the embryo cells an extensive actin and tubulin network was observed.

The article presents the main discoveries of Prof. Andrzej K. Tarkowski, which proved to be fundamental for modern mammalian developmental biology and also for progress in animal breeding and assisted reproduction. Among his achievements the most important are: the demonstration of regulative abilities of blastomeres isolated from early mammalian embryos, generation of first chimaeric mice, studies on mammalian parthenogenesis and establishment of blastomere electrofusion technique for production of tetraploid embryos. Studies on nucleo- cytoplasmic interactions in germ cells and early embryos contributed substantially to the development of mammalian cloning. Prof. Tarkowski’s work and discoveries provided a tremendous input to the contemporary developmental biology of mammals.

Exceptionally well−preserved embryonic skulls of Upper Cretaceous (Campanian Anacleto Formation) sauropods from Auca Mahuevo (Neuquén Province, Argentina) provide important insights into the ontogeny and evolution of titanosaurian neosauropods. The most important cranial modifications occurring during titanosaurian ontogeny appear to be centered on the infraorbital and narial regions, which exhibit a substantial degree of “mosaic” evolution. On one hand, the Auca Mahuevo embryos show a large jugal that forms part of the lower margin of the skull and unretracted external nares, as indicated by the position and orientation of the lacrimals as well as the anterior extension of the frontals. Both of these features are ancestral for neosauropods, being present in prosauropods. On the other hand, the embryonic skull exhibits a large ventral notch, tentatively interpreted as homologous to the neosauropod preantorbital fenestra, that opens ventral to the jugal and between the maxilla and the quadratojugal, and a temporal region that closely resembles the adult neosauropod condition. This mosaic of character states indicates that different regions of the skull of titanosaurian neosauropods acquired their characteristic morphology at substantially different rates during their ontogenetic development.

Opis:
Microscopic analysis of embryogenic tissues of Picea abies and P. omorika showed that the tissues differed in proportions of various types of proembryogenic masses, and in presence/absence of early somatic embryos, which suggests that they differed in embryogenic potential. On the basis of this study, embryogenic competence of tissues can be assessed at early stages of somatic embryogenesis, and thus the most efficient lines can be selected (one Picea abies and two P. omorika). In the course of development of somatic embryos embryogenic tissue was placed on a medium with abscisic acid (ABA), which affects an increase in osmotic pressure at the stage of maturation of somatic embryos and finally on the regeneration of cotyledonary embryos. In this stage of embryos development, storage materials were accumulated, which are used in specific metabolic processes during embryo development.

This article discusses the provisions for the legal protection of the human genome at the embryonic stage. I examine both the Polish and international regulations. I also look at the threats resulting from the development of genetic technologies and the need to introduce appropriate legal regulations for effective protection of the human being at the embryonic stage.
W artykule podjęta została próba analizy regulacji dotyczących prawnej ochrony ludzkiego genomu na etapie embrionalnym. Analiza objęła zarówno unormowania krajowe, jak i międzynarodowe. Wskazane zostały również zagrożenia wynikające z rozwoju technologii genetycznych oraz związana z tym potrzeba wprowadzenia stosownych regulacji prawnych, pozwalających na zapewnienie skutecznej ochrony istocie ludzkiej na etapie embrionalnym
