Opis:
Jatropha curcas plant is greatly impaired by mosaic disease, caused by the viruses (Begomovirus), transmitted by whiteflies, which act as the vector. Roguing (i.e. removal of infected plant) and spraying of insecticides are common methods, employed in order to get rid of the disease. In this article, a mathematical model has been developed to study the mosaic disease dynamics while considering preventive measures of roguing and insecticide spraying. Sufficient conditions for the stability of equilibrium points of the system are among the results obtained through qualitative analysis. We obtain the basic reproduction number R0 and show that the disease free system is stable for R0 < 1 and unstable for R0 > 1. The region of stability of equilibrium points in different parameter spaces have also been analysed. Hopf bifurcation at the endemic steady state has been studied subsequently, as well. Finally, by formulating an optimal control problem, optimal application of roguing and spraying techniques has been determined, keeping in mind the cost effective control of the mosaic disease. Pontryagin minimum principle has been utilized to solve the optimal control problem. Numerical simulations illustrate the validity of the analytical outcomes.

Opis:
Nanoparticles are gaining ever−wider application in plant production (for both agriculture and forestry), in the role of pesticides, as well as stimulators of plant growth and resistance. We sought to determine the efficacy of silver and copper nanoparticles (AgNPs and CuNPs respectively), used as seed dressings or subjected to foliar application, in affording protection from parasitic damping−off disease among soil−grown seedlings of Scots pine in a forest nursery. Experiments also assessed the influence of the nanoparticles on the growth of the plants potentially safeguarded in this way. Nanoparticles were used in seed soaking and foliar spraying at 50 ppm concentration. Pines treated with fungicides (Zaprawa Nasienna T 75 DS, Acrobat MZ 69 WG, Topsin M 500 SC, Gwarant 500 SC, Thiram Granuflo 80 WG and Signum 33 WG) or unprotected at all were used for comparison. In each treatment (AgNPs, CuNPs, fungicides or unprotected), seedlings were inventoried 6 weeks after the sowing and at the end of the growing season, while all individuals on 80 1−m−long segments of seed row were counted. At the end of the growing season, shoot length, root−collar diameter, root length and dry mass of shoots and roots were determined. The seedlings treated with nanoparticles had longer root systems of greater dry mass, but also only more weakly−developed above−ground parts (both height and dry mass being limited) in comparison with young Scots pines that had been fungicide−treated or were unprotected. It resulted in a significantly more favourable ratio between shoot and root masses, where nanoparticle treatment had been applied. The effectiveness of the protection extended to the germination and first−growth stages up to 6 weeks from the time of sowing was furthermore shown to be greatest where AgNPs had been applied, while at the season end there were comparable results among pines treated with either nanoparticles or fungicides. Our results thus indicate that nanoparticles limit damping−off disease in pine seedlings (AgNPs more effectively than CuNPs), with that effectiveness also proving comparable with that noted for the fungicides applied traditionally.

Opis:
Fungal antagonist, Talaromyces flavus, is one of the most important biological agents of soil-borne fungal diseases including Verticillium and Fusarium wilt. In this study, to increase the effectiveness of T. flavus isolates obtained from greenhouse cucumbers and field grown tomatoes five chemical stabilizers were evaluated. Based on the results of previous studies, the most effective substrate for the growth, sporulation and stability of T. flavus isolates related to the above-mentioned plants was a mix of rice bran and peat-moss. Different chemical stabilizers were mixed with the above-mentioned substrate containing spore suspensions of various T. flavus isolates. For each plant, a completely randomized experiment was conducted under greenhouse conditions with seven treatments and three replications. The results of this study indicated that treatments containing sodium nitrate and D-cycloserine were more effective than those containing other stabilizers. The overall results of this study suggest that the use of some chemical stabilizers may enhance the biocontrol potential of fungal antagonists in controlling different plant diseases including Verticillium and Fusarium wilt.

Opis:
The purpose of the study was to suppress a known natural focus of fi eld fever exerting an infl uence on animal hosts of leptospires – small terrestrial mammals (s.t.m.) by rodenticide. After repeated application of the zinc phosphide rodenticide, the s.t.m. were regularly live-trapped and checked for leptospirosis by dark-fi eld microscopy and culture of their renal tissue and serological examination. Isolated leptospira strains were typed by help of rabbit factor sera. The deratization infl uenced the s.t.m. structure considerably: the proportion of the dominant Microtus arvalis subjects – the main reservoirs of Leptospira kirschneri serovar Grippotyphosa – were gradually and substantionally reduced and, contrarily, the percentage of the potetial hosts subjects – Clethrionomys glareolus and Apodemus fl avicollis – rose decisively over time. Changes in culture and serological positivity for leptospirosis of s.t.m. have also been enregistered. The highest original infestation of M. arvalis have slowly but strongly decreased while that of C. glareolus and A. fl avicollis has increased decisively and reached its maximum within the last years of investigation. It is probable that these two animal species have undertaken the leading role in the maintenance of the natural focus of the fi eld fever. In other animal species analogous trends were not registered. Based on these long-term fi ndings, there exists the possibility to suppress only partially and temporarily the epizootic process of leptospirosis in a natural focus that can be desirable in some circumstances (building or free time activities, etc.).

Opis:
Cry4Aa 678 amino acids fragment (60 kDa) was cloned into Escherichia coli. After induction with IPTG the 60 kDa Cry4Aa fragment was purified by Ni chromatography, separated by SDS PAGE and identified by mass spectrometry (MS/MS). The 60 kDa Cry4Aa fragment exhibited high toxicity towards Ae. aegypti larvae. The earlier results [1] show that Pichia pastoris yeast cells expressing tmfA (synthetic gene coding for the Trypsin Modulating Oostatic Factor of Ae. aegypti) together with E. coli cells expressing Bti toxin genes (cry4Aa, cry11Aa, cyt1Aa and p20) are synergistic. Therefore, P. pastoris, which synthesizes high amounts of heterologous proteins was genetically engineered to produce TMOF and Cry4Aa. Codon-optimized synthetic genes, cry4Aa-tmfA, gst-cry4Aa-tmfA, tmfA and gfptmfA that were expressed by P. pastoris and fed to Ae. aegypti larvae caused 90% mortality. GST (glutathione-S-transferase) enhanced the activity of Cry4Aa-TMOF and protected it from heat denaturation and GFP (Green Fluorescent Protein)- TMOF allowed us to follow yeast cells consumption by individual larva using fluorescent microscopy. This report shows for the first time that 60 kDa Cry4Aa and TMOF expressed together in P. pastoris are highly toxic to Ae. aegypti larvae.

The external appearance and quality of table potatoes are determined, among other factors, by the health status of the plants during the growing season. Chemical control methods are often combined with biocontrol agents to effectively fight potato pathogens. Potatoes of the very early cultivar Rosara were grown in experimental plots. The plots were located in Tomaszkowo (NE Poland, 2007–2009). The experiment involved the following treatments: 1) biological control − mycorrhizal Glomus spp. inoculum was applied to the roots, − tubers were dressed and plants were sprayed with Polyversum three times during the growing season, 2) chemical control – at two-week intervals, plants were sprayed with the following fungicides: Infinito 687.5 SC and Tanos 50 WG, Valbon 72 WG and Tanos 50 WG. In the control treatment, potato plants were not protected against pathogens. During the growing season, the severity of late blight and early blight was evaluated on a nine-point scale. The composition of fungal communities colonising potato stems was analysed. The fungistatic properties of the fungicides used in the field experiment were evaluated in an in vitro test. The symptoms of infections caused by Phytophthora infestans and Alternaria spp. were significantly reduced in the treatment which used the integrated chemical and biological control. The least diverse fungal community was isolated from fungicide-treated plants. In the in vitro test, fungicides at all analysed concentrations inhibited the linear mycelial growth of selected pathogens.

Opis:
Introduction: Diabetes is a disease whose course is strongly determined by self-monitoring and self-care. Ensuring metabolic control of diabetes is not only important for the current state of health, it also allows to maintain the body in the best condition despite the existing chronic disease. Purpose: To assess the relationship between sociodemographic factors and depression symptoms and the level of disease acceptance in diabetes. Materials and methods: The study used a diagnostic survey method with a questionnaire, the Beck Depression Inventory, and the Acceptance of Illness Scale. The study group consisted of 43 women (51%) and 41 men (49%). A total of 67% of respondents had diagnosed type 2 diabetes, 24% type 1 diabetes, and 6% LADA. Average disease duration was 15 years. Results: In the studied group, 15% of patients had diagnosed symptoms of depression; while according to the Beck Depression Inventory, depressive episodes occurred more often in mild (30%) and moderate (23%) severity. They were observed more often in people over 50 years old. Depression symptoms were significantly more common in the group of overweight and obese people (p=0.022, r(X,Y)=0.2490). Conclusions: The problem of depression increases with disease duration. People diagnosed with type 2 diabetes have lower levels of disease acceptance. Disease acceptance is a factor that eliminates the occurrence of depressive disorders of varying severity.

Opis:
Introduction: Postural instability in patients with Parkinson’s disease (PD) is one of the major factors contributing to an increasing number of falls and fall related complications. Diagnosis of balance disorder is based on assessment of postural control by the biomechanical assessment of the musculoskeletal system, coordinating speed and precision of movements. The condition for the performance of precise and smooth movement is coordination of motor activity and sensory signals fl owing to the motor system. Study aim: The aim of the study is to assess the impact of non-motor symptoms (depression and cognitive impairment) and the impact of the severity of motor symptoms on the process of course of movement planning and control in Parkinson’s disease. Material and methods: The study involved 40 patients with idiopathic Parkinson’s disease, diagnosed according to the criteria adopted by the United Kingdom Parkinson’s Disease Society Brain Bank (UKPDSBB), with stage II or III severity of symptoms of the disease according to the Hoehn-Yahr sclae. All the patients underwent physical examination with an evaluation using the UPDRS (Unifi ed Parkinson’s Disease Rating Scale) and screening clinical trials were performed, evaluating cognitive functions (Montreal Cognitive Assessment, MoCA) and depressive disorders (Beck Depression Inventory, BDI). Evaluation of mobility, stability and motor control was done using the Berg Balance Scale (BBS), the Tinetti test, the Up and Go test (TUG) and the Dynamic Gait Index (DGI). Results: There was a signifi cant correlation between the level of severity of movement disorders (according to UPRDS ON part III) and the parameters defi ning motor skills in the DGI (R= -0.49, p<0.001), the TUG test evaluating mobility (R=0.4, p<0.01), and static and dynamic balance disorder using BBS (R=-0.44, p<0.004). The level of cognitive functioning had signifi cant impact on the mobility and stability of patients, and the occurrence of depressive disorder did not affect the deterioration of basic postural reactions in the group of patients with PD. Materiał i metody: Do badania zakwalifikowano 40 chorych z idiopatyczną chorobą Parkinsona, rozpoznaną zgodnie z kryteriami United Kingdom Parkinson’s Disease Society Brain Bank (UKPDSBB), w stadium II lub III zaawansowania objawów choroby wg Hoehn-Yahra. U wszystkich chorych przeprowadzono badanie fizykalne wraz z oceną w skali UPDRS (Unified Parkinson’s Disease Rating Scale) oraz wykonano przesiewowe testy kliniczne, oceniające funkcje poznawcze (Montreal Cognitive Assessment, MoCA) i zaburzenia depresyjne (Beck Depression Inventory, BDI). Oceny mobilności, stabilności i kontroli ruchu dokonano za pomocą testów: Berg Balance Scale (BBS), testu Tinetti, testu Up and Go (TUG), Dynamic Gait Index (DGI). Wyniki: Wykazano istotną zależność pomiędzy poziomem nasilenia zaburzeń ruchowych (wg UPRDS ON cz. III) a parametrami definiującymi zdolności motoryczne w DGI (R= -0,49, p<0,001), testem oceny mobilności TUG (R=0,4, p<0,01) oraz zaburzeniami równowagi statycznej i dynamicznej w BBS (R=-0, 44, p<0,004).

Redukcja dawek w sadach półkarłowych i karłowych przynosi korzyści ekonomiczne.
Spray volumes applied in spatial crops, like apple orchards, should be adjusted to the size of trees. One of the method of spray volume adjustment to orchards of different types is tree row volume concept (TRV). The objective of the experiment was to verify biologically the TRV concept. The TRV based spray volumes proved to be efficient in apple scab (Venturia inaequalis) and powdery mildew (Podosphaera leucotricha) control. This was due to maintenance of sufficient spray deposit in the trees the spray volumes were adjusted to. The reduction of spray volumes in semi-dwarf and dwarf orchards is economically advantageous.
