Opis:
Currently, globalization, urbanization and industrialization have led to several environmental issues. In many industries, particularly in textile industries, the extensive use of synthetic dyes has increased. Dye is an integral element used to impart color to textile materials. Wastes generated during the processing and treatment of the dye contain inorganic and organic compounds that are hazardous, thereby posing a serious threat to the ecosystem. It is therefore important to implement cost-efficient and successful measures against these emissions in order to preserve habitats and natural resources. In this context, biodegradation by fungi or mycoremediation of dyes using potential fungi is a fairly inexpensive and environmental friendly method for decomposing or mineralizing barely or less decaying dye compounds. Fungi play a crucial role in degrading and decolorizing organic dyes by enzymes and processes such as absorption, adsorption and aggregation of effluent colorants. The factors affecting the decolorization and biodegradation of dye compounds through fungal bioremediation, such as pH, temperature, dye concentration, agitation, effects of carbon and nitrogen sources, dye structure, enzymes, electron donor and redox mediators are discussed in this review. The review also includes a summary on the mechanism and kinetics of dye degradation as well as recent advances and future perspectives in mycoremediation of dyes.

Opis:
The current investigation is the first report of utilization of Aspergillus brasiliensis for the decolorization of melanoidin in distillery effluent. The effluent generated from alcohol distilleries is one of the most complex wastewater with a high biological oxygen demand (BOD) and other organic, inorganic, and toxic constituents. The effluent contains melanoidin, a dark brown compound, which is difficult to remediate by using conventional technologies. The disposal of spent wash in the natural environment is hazardous and can deteriorate land and water resources. The decolorization of spent wash through physical and chemical methods remains unsuitable, and the only alternative to decolorize spent wash is biological treatment. In the current study, three fungal strains were isolated from the distillery waste and screened for their ability to decolorize melanoidin. The isolate RS2 exhibited maximum decolorization of 83% and was identified as Aspergillus brasiliensis. Its optimum growth temperature was 37EC, and the maximum efficiency was recorded after 120 h of incubation. Nutritional sources were investigated for the fungi showing the maximum decolorization of melanoidin, and starch and peptone were found to be the best carbon and nitrogen sources, respectively. At 1.5% starch concentration and 1.5% peptone concentration, the decolorization level attained was 87.45% and 88.74%, respectively. A. brasiliensis exhibited a high potential to decolorize melanoidin. The decolorization percentage was high, which makes this fungus a potential candidate for use at the industrial scale for the bioremediation of spent wash.

Opis:
Autochthonous fungi are potential successful agents in the process of bioremediation through their efficient adaptation to pollutant toxicity and competition with other microorganisms that are present in the leachate treatment plant. The leachate from municipal waste landfills was an important source of fungi. Even though some of these fungi have the potential to be human pathogens, such strains wh en used in biolo gical treatment approaches may serve as a possible tool for integrating the leachate bioremediation process because of the extracellular enzymes produced by fungal strains. In this study, the leachate sample was tested with regard to four parameters, including total dissolved solids (TDS), electrical conductivity (EC), pH, and the decolorization of the leachate sample by a number of indigenous species of fungi, which were observed by use of the culture techniques. The scanning electron microscope coupled with energy dispersive x-ray analysis, UV-spectrum, and reflected and transmitted polarizing light microscope were used in the research project to o bserve and assess the obtained data from the experimental work. The results indicated the various levels of efficiency of the isolated fungal strains in eliminating color, lowering TDS, EC, and pH through their geoactivities potential in metal biosorption and immobilization by biomineralization of new minerals in the growth environment and eventually reducing the metal bioavailability and toxicity.

The knowledge about biological activities of constituents from medicinal mushrooms belonging to the genus Tricholoma is limited. A 59-kDa laccase has now been purified from fresh fruiting bodies of the mushroom Tricholoma matsutake. The purification protocol entailed ion exchange chromatography on DEAE-cellulose, affinity chromatography on Affi-gel blue gel, ion exchange chromatography on CM-cellulose, affinity chromatography on ConA-Sepharose, and gel filtration by fast protein liquid chromatography on Superdex 75. Of the various affinity and ion exchange chromatographic media employed, the laccase bound only on Con A-Sepharose. The activity of the laccase did not undergo major changes over the temperature range 20-80°C. However, all activity vanished following exposure to 100°C for 10 minutes. The enzyme activity varied only slightly over the pH range 3-5, with the optimal pH of 5, but exhibited a precipitous decline when the pH was increased to 6, and was undetectable at pH 8 and 9. The laccase showed activity in the decolorization of azo dyes without a mediator. Its N-terminal sequence demonstrated only slight resemblance to those of other mushroom laccases. The newly described laccase is distinctive from the previously isolated Tricholoma mushroom laccases in a number of aspects.

The aim of this study was to overproduce, identify and apply novel laccase-like multicopper oxidases (LMCOs) from Myrothecium roridum in a dye removal process. LMCOs' production was enhanced by modifying the medium and adding copper ions. After purification, two proteins, LMCO1 and LMCO2, with molecular masses of 46.7 and 66.3 kDa were discovered. Peptide analysis by mass spectrometry revealed that they belong to the cupredoxin superfamily. Characteristic peptide sequences were obtained for MCOs and bilirubin oxidases. Crude enzymes were applied in a dye decolorization process. Supplementation with 1 mM of vanillin allowed an almost complete elimination of the Indigo carmine within 3 hours. The dye was removed from a solution containing metals, surfactants and organic solvents. The in-gel assessment of the activity and decolorization ability of MCOs, followed by protein extraction and SDS-PAGE, confirmed that only LMCO2 was responsible for the dye removal. MCOs produced by Myrothecium sp. have been poorly studied before. The obtained results broaden knowledge on this subject and may contribute to the development of an eco-friendly method of dye elimination.

Opis:
The role of fungi in the treatment of wastewater has been extensively researched. Many genera of fungi have been employed for the dye decolourization either in living or dead form. In this study, the removal of an acidic dye, Indigo Carmine (IC), from an aqueous solution by biosorption on dead fungus, Pleurotus ostreatus, was investigated. The effects of contact time, initial dye concentration, amount of dead biomass, agitation rate and initial pH on dye removal have been determined. Experimental results show that an increase in the amount of dead biomass positively affected the dye removal. The highest removal was obtained at 150–200 rpm. Slightly lower removing activities were found at lower agitation rates. The dye adsorption effi ciency was not affected by pH except minor variation in the pH of 2–8. Color removal was observed to occur rapidly within 60 minutes. The removal of dye by dead biomass of P. ostreatus was clearly dependent on the initial dye concentration of the solution. Dye removal was reduced from 93% to 64% as concentration was increased from 50 to 500 mg/L Indigo Carmine. This study showed that it was possible to remove textile dyes by dead biomass of P. ostreatus.

Synthetic dyes are a serious threat if they get into the environment, even in small quantities. Therefore, studies to improve the treatment of waste water containing dyes is still a challenge. One of the most frequently used groups of dyes in the dyeing are those belonging to the anthraquinone class. The aim of the study was to determine the possibility of using fungi in decolorization of dye belonging to this group as is Remazol Brilliant Blue R. The tests were carried out on solid media of different composition. Among the 9 strains tested, the highest activity was recorded for species: Pleurotus ostreatus (strain K4), Xerocomus badius (strain K45) and Hypholoma fasciculare (strain K47). They were effectively removing dye added to the medium rich in organic compounds, but reduction of nutrient concentrations in medium stimulated discoloration of the dye.

Starch-iodine assay method for the determination of α-amylase activity is also used in screening extracts for α-amylase inhibitors. However, there are indications that this method may not be appropriate for screening some classes of compounds or plant extracts. The present study investigated the limitation(s) of this method in screening plant extracts/compounds for α-amylase inhibition. A crude methanol extract (CR) of Dacryodes edulis, its solvent fractions (ethyl acetate (EA), aqueous methanol (AM), and hexane (HX)), quercetin (QC), and benzoic acid (BA) were used for this study. The phytochemical content and antioxidant activity were screened spectrophotometrically. α-Amylase inhibition (expressed in percentage and as IC50) was determined by starch-iodine method approach I and II (ST-ID I and ST-ID II, respectively) and dinitrosalicylic acid (DNSA) as the control method. The results showed that the extracts/compounds (AM, EA, and QC) with significantly high polyphenolic content, antioxidant activity, and starch-iodine complex decolorization effect yielded contrary results of α-amylase inhibition when the results of ST-ID I and II methods were compared to that of the DNSA method. The other test samples (CR, HX, and BA) yielded similar results for all the three methods. The result also showed the decolorization (%) of starch-iodine complex by the test samples correlated significantly (r = 0.877, P < 0.05) with DPPH reduction (%). In conclusion, the present study showed that the starch-iodine method is not appropriate for screening antioxidative extracts/compounds for α-amylase inhibitors – they decolorize the assay reagent in a manner similar to DPPH reduction and hence confound the result.

Opis:
Application of reactive dyes is very popular in textile industry as these dyestuffs are characterized by good fastness properties. Constapel et al estimated the production of this type dyes for over 140,000 Mg/year. The reactive dyes are mostly (50%) employed for coloration of cellulosic fibers, however they can be also applied on wool and nylon. Unfortunately, they possess a low degree of fixation (50÷90%), since the functional groups also bond to water, creating hydrolysis and the excess of dyes applied cause a colored pollution of aqueous environment. Moreover, dyeing process requires the use of: electrolytes in the form of aqueous solutions of NaCl or Na2SO4 in the concentration up to 100 g/dm3, alkaline environment (pH > 10) and textile auxiliary agents (among others detergents). Therefore, the wastewater generated during the reactive dyeing processes are characterized by high salinity, pH value and color, and due to low value of the ratio BOD5/COD are non-biodegradable. The successful methods of the textile wastewater treatment could be an advanced oxidation processes (AOPs), amongst which the Fenton reagent seems to be the most perspective as it is the cheapest and easy in use. Based on the newest literature survey it was found that many successful tests with Fenton reaction were performed mainly in decolorization. However, not enough attention was devoted to decolorization of real industrial wastewater containing dyes, detergents and salts NaCl, or Na2SO4. The experiments carried out in lab scale were focused on the impact of NaCl and textile auxiliary agent (liquid dispersing and sequestring agent) on an inhibition of decolorization process by Fenton reagent. The objects of the investigation were the synthetic mixtures simulating the composition of real textile wastewater as well as the real industrial wastewater generated in the reactive dyeing. The inhibition of the Fenton decolorization in the presence of NaCl and liquid dispersing and sequestering agent was demonstrated.
To sprawia, iż ścieki generowane w procesach barwienia reaktywnego charakteryzuje: wysokie zasolenie i zabarwienie, wysoka wartość pH, obecność SPC. Niewiele natomiast uwagi poświęcono odbarwianiu ścieków rzeczywistych czy symulowanych mieszanin zbliżonych składem do ścieków rzeczywistych, czyli zawierających barwniki oraz NaCl, Na2SO4, SPC, (Sun i in. 2009; Arslan-Alaton i in. 2008).

In this study, the effectiveness of using natural bio-coagulants and bio-flocculants to treat textile wastewater through the coagulation-flocculation method was examined. These bio-based agents have several advantages over chemical agents, including biodegradability, natural abundance, low toxicity, and low cost. A bio-coagulant (holm oak acorn (HOA)) and a bio-flocculant (cactus juice) were used to investigate the capacity for turbidity removal and decolorization of textile wastewater. The UV spectrophotometer was used to characterize the discharges before and after treatment, and the chemical oxygen demand (COD) and biological oxygen demand (BOD5) levels were calculated. Box-Behnken design (BBD) coupled with response surface methodology (RSM) were utilized to optimize the process and reduce turbidity and decolorization in textile wastewater. The obtained results show that under the optimal conditions (0.5 g•L-1 of HOA, 15 mL•L-1 of cactus juice, and a pH of 7), decolorization and turbidity removal were achieved at 69% and 90%, respectively. This study demonstrates the potential of using bio-coagulants and bio-flocculants in the treatment of textile wastewater.

Opis:
Sugar beet molasses vinasse is a high-strength distillery wastewater. It contains colored substances which significantly affect the degree of pollution and toxicity of vinasse. This study aimed to optimize the medium composition and the process condition of sugar beet molasses vinasse decolorization by Lactobacillus plantarum MiLAB393. The research was conducted in two stages: the shake-flask stage in the 250 cm3 Erlenmeyer flasks and the batch experiments in the 5 dm3 working volume stirred-tank bioreactor. During the study, the concentrations of glucose and yeast extract were optimized using experimental design of experiments (DOE). The influences of the initial value of pH and pH control, temperature, stirrer speed and glucose concentration on decolorization were tested. The highest color reduction of 24.1% was achieved for an experiment in which 24.93 g/dm3 of glucose was added to the medium and stirrer speed was 200 rpm. This efficiency of 30% v/v sugar beet molasses vinasse decolorization was obtained at non-controlled pH 6.0 and at 35.8°C. It was found that pH control determines vinasse decolorization. When the pH was controlled, decolorization did not exceed 9%. The glucose and yeast extract concentration and the stirrer speed have a great influence on the process. Changes in these parameters may increase biomass growth while decreasing the decolorization.
