Opis:
We investigated the effect of daunorubicin on glutathione content and activity of GSH-related enzymes in cultured normal and diabetic human fibroblasts. Cells were incubated with 4 μM daunorubicin (DNR) for 2 h followed by culture in drug-free medium for up to 72 h. Treatment of diabetic cells with the drug caused a time-dependent depletion of intracellular GSH and a decrease of the GSH to total glutathione ratio. GSH depletion was accompanied by apoptotic changes in morphology of the nucleus. Analysis of GSH-related enzymes showed a significant increase of the activities of Se-dependent and Se-independent peroxidases and glutathione S-transferase. In contrast, glutathione reductase activity was reduced by 50%. Significant differences between normal and diabetic cells exposed to DNR were observed in the level of GST and Se-dependent glutathione peroxidase activities. These findings indicated that daunorubicin efficiently affects the GSH antioxidant defense system both in normal and diabetic fibroblasts leading to disturbances in glutathione content as well as in the activity of GSH-related enzymes.

Opis:
Some sterically hindered N-substituted derivatives of daunorubicin are known to be poor substrates for NADH dehydrogenase, NADPH cytochrome P450 reductase and xanthine oxidase. In consequence, poor oxygen radical generation by these compounds is observed. In this study we examined a new family of sugar-N-substituted derivatives of daunorubicin bearing a bulky substituent introduced on the nitrogen atom through the amidine spacer. These compounds were found to be very active in radical formation catalyzed by all three studied enzymes. Thus, the introduction of a heterocyclic ring, even if it is bulky but flexible, on the nitrogen atom of daunosamine moiety through the one-atom spacer (amidine group), does not induce the steric hindrance effect on the interaction of daunorubicin derivatives with these flavoprotein enzymes.

Opis:
The aim of this study was to compare the physico-chemical and biological properties of polymeric nanoparticles obtained from poly(DL-lactide-co-glycolide) (PLGA) with different ratios of monomers loaded with daunorubicin (DNR). Methods: DNR-loaded nanoparticles (NPs) were prepared with use of modified simultaneous double-emulsion solvent evaporation/diffusion technique. NPs were characterized using dynamic light scattering, atomic force microscopy, transmission electron microscopy, scanning electron microscopy, and differential scanning calorimetry and Fourier transform infrared spectroscopy. Results: NPs with DNR were differing in size and zeta potential, depending on the type of polymer. The data obtained show that total content of DNR correlates with the values of the binding constant of DNR with polymers. The release of DNR from NPs proceeds predominantly for polymers with lower binding constants. The in vitro study of NPs on the MCF-7 cells showed similar activity of particles and substances while for the anthracycline-resistant MCF-7Adr cells the cytotoxicity of the nanoparticles was 3 to 7 times higher depending on the type of copolymer. Conclusions: PLGA DNR-loaded nanoparticles can be used to overcome multidrug resistance (MDR) as well as for reducing the frequency of DNR reception due to the prolonged effect, which allows maintaining the concentration of the drug at the required level. The usefulness of binding constant calculations for obtaining nanoparticles with the maximum drug loading was proven. The rate of drug administration and the frequency of administration can be calculated based on the DNR release profiles and release parameters that depend on polymer type.
