Curcumin, known for thousands of years as an Ayurvedic medicine, and popular as a spice in Asian cuisine, has undergone in recent times remarkable transformation into a drug candidate with prospective multipotent therapeutic applications. Characterized by high chemical reactivity, resulting from an extended conjugated double bond system prone to nucleophilic attack, curcumin has been shown to interact with a plethora of molecular targets, in numerous experimental observations based on spectral, physicochemical or biological principles. The collected preclinical pharmacological data support traditional claims concerning the medicinal potential of curcumin and its congeners but at the same time point to their suboptimal properties in the ADME (absorption, distribution, metabolism and excretion) area.

Numerous adverse effects limit the applicability of mitoxantrone for the treatment of drug-resistant tumors, including carcinosarcoma. Here, we estimated the additive effects of mitoxantrone and curcumin, a plant-derived biomolecule isolated from Curcuma longa, on the neoplastic and invasive potential of carcinosarcoma cells in vitro. Curcumin augmented the cytostatic, cytotoxic and anti-invasive effects of mitoxantrone on the Walker-256 cells. It also strengthened the inhibitory effects of mitoxantrone on the motility of drug-resistant Walker-256 cells that had retained viability after a long-term mitoxantrone/curcumin treatment. Thus, curcumin reduces the effective doses of mitoxantrone and augments its interference with the invasive potential of drug-resistant carcinosarcoma cells.

Cytotoxic CD8+ cells play an important role in determining host response to tumor, thus chemotherapy is potentially dangerous as it may lead to T cells depletion. The purpose of this study was to elucidate the propensity of quiescent and proliferating human CD8+ cells to undergo cell death upon treatment with curcumin, a natural dye in Phase I of clinical trials as a prospective chemopreventive agent. Methods: We treated human quiescent or proliferating CD8+ cells with 50 µM curcumin or irradiated them with UVC. Cell death symptoms such as decreased cell viability, chromatin condensation, activation of caspase-3 and specific DFF40/CAD endonuclease and oligonucleosomal DNA fragmentation were analyzed using MTT test, microscopic observation, Western blotting and flow cytometry. Results: Curcumin decreased cell viability, activated caspase-3 and decreased the level of DFF45/ICAD, the inhibitor of the DFF40/CAD endonuclease. However, this did not lead to oligonucleosomal DNA degradation. In contrast, UVC-irradiated proliferating, but not quiescent CD8+ cells revealed molecular and morphological changes characteristic for apoptosis, including oligonucleosomal DNA fragmentation. Curcumin can induce cell death in normal human lymphocytes both quiescent and proliferating, without oligonucleosomal DNA degradation which is considered as a main hallmark of apoptotic cell death. Taking into account the role of CD8+ cells in tumor response, their depletion during chemotherapy could be particularly undesirable.

Quercetin and curcumin are plant polyphenolic antioxidants with proven pharmacological efficacy. Their use is, however, limited due to low bioavailability and oral administration route. The encapsulation of lipophilic compounds in liposomes enables to increase their bioavailability and to create an injectable form. The present study aimed to comparatively investigate the antioxidant activity of a complex liposomal preparation containing two lipophilic antioxidants (quercetin and curcumin) and their monopreparations in liposomal forms. This study was conducted on Wistar line rats with experimental model of ischemic heart disease. Oxidative stress markers such as total antioxidant activity, malondialdehyde, and peroxidized proteins were analyzed in blood serum and cardiac tissue. Ischemic heart disease is accompanied with lipid peroxidation and changes in the activity of the antioxidant system. The obtained results demonstrated the antioxidant activity of monopreparations of curcumin and quercetin and their complex in liposomal forms. Quercetin and curcumin showed different antioxidant activities in terms of oxidative stress markers. The complex of the two antioxidants showed the synergistic effect of their lipophilic compounds in liposomal forms, which led to the normalization of test parameters according to the level of the control sample.

Curcumin is reported as an anti proliferative and chemopreventive compound. However, it shows poor water solubility, low bioavailability, and rapid metabolism.. To address these problems, curcumin derivatives substituted with polyether chain were synthesised to improve the hydrophilicity of curcumin and thus its bioavailability. The biological activity of modified curcumin molecules were studied with human normal B-lymphocytes GM 14667 and human leukemia cells HL60 and covered: the denotation of IC50 values for each new compound with use of MTT test and the studies on apoptosis induction observed as morphological changes and the expression of apoptotic proteins, BAX, BCL-2 and survivin with use of Western Blot analysis. All effects were referenced to native curcumin. The derivatives with modified polyether chains number and the position of methoxy- groups were selected for further studies. As was shown, the solubility of the compound does not directly correlate with its cytotoxicity. The maintenance of the methoxy group in the meta position as well as the introduction of hydrophilic polyether chains but not the number of polyether chains may affect the cytotoxic activity of curcumin derivatives against cancer cells. Modifications of the curcumin molecule structure can determine its impact on the profile of pro or anti apoptotic proteins.

BackgroundThe authors’ aim was to study the dynamics of oxidative stress in experimental exposure to silica dust, to evaluate the histopathological findings in the phase preceding the formation of fibrous/fibrohyaline pulmonary nodules, and to assess the effects of curcumin administration.Material and MethodsThe research was performed on 48 male Wistar rats with an average weight of 320 g. Overall, 38 rats were instilled with a single dose of 0.3 ml suspension containing 30 mg of a SiO2/ml saline solution, and were sacrificed 30, 90 and 120 days after instillation; 14 of those sacrificed on days 90 and 120 also received curcumin. The control group included 10 animals which were instilled with a saline solution. Malondialdehyde (MDA), carbonyl proteins (CPs), total thiolic proteins (TPs) and reduced glutathione (GSH) were determined in blood and the lung tissue. The standard technique for pulmonary toxicology developed by Porter was applied to semi-quantitatively assess the histopathological findings.ResultsIt was found that MDA had increased significantly early on in both biological environments and remained elevated, and adding curcumin proved beneficial, while CPs only increased moderately in the lung tissue without a curcumin impact. Moreover, TPs dropped abruptly, significantly and persistently in the lung tissue and blood, and were not influenced by curcumin. Finally, GSH decreased significantly and intensely in the lung tissue and blood, with curcumin lowering the levels towards those found within the control group. The histopathological examination identified nodules of a cellular type, without any fibrosis, but with spots of associated lipoproteinosis. The early lesions in the airways and vessels were suggestive of a remodeling process. Curcumin diminished the occurrence of alveolitis but not the remodeling process.ConclusionsThe study confirms the early onset of oxidative stress in experimental silicosis. It also simultaneously and dynamically researches markers of oxidative stress in blood and the lung tissue. Curcumin proved beneficial on oxidative stress and lesions in the alveolar epithelia, but ineffective in preventing vascular and airway remodeling. Med Pr. 2021;72(3):239–47

Natural polymers, like chitosan, collagen, and alginate, offer promising solutions for wound healing. Derived from natural sources, they exhibit biocompatibility and bioactivity, promoting tissue regeneration. These polymers can form scaffolds or dressings that accelerate wound closure while reducing infection risks. Their inherent properties make them promising options in the quest for effective wound care materials. In this work, composites based on polyvinyl alcohol (PVA), chitosan (Chi), and curcumin (Cur) were prepared. PVA, a synthetic water-soluble polymer, finds extensive use in biomedical and wound-healing applications. It is approved by the U.S. FDA for cosmetic, medical, and wound healing products. Chi, a polysaccharide, is widely used in biomedicine and possesses antibacterial properties. Both PVA and chitosan are biocompatible and exhibit good filming characteristics. Curcumin (Cur) with antibacterial and antioxidant properties is being explored for regenerative medicine. PVA, chitosan, and curcumin were blended. The structure was studied by FTIR, microscopic observations were done with optical and scanning electron microscopes, and the mechanical properties were assessed. FTIR revealed component interactions, while microscopy showed a flat film surface. The polymeric blend (PVA/Chi/Cur) had a Young’s modulus of 1.49 GPa, tensile strength of 47.69 MPa, stress value of 8.39 N, and 35.34% elongation at break. These properties make the blend suitable for consideration in wound healing applications.

The natural wound healing process consists of four basic phases: homeostasis, inflammation, proliferation, and remodelling. Macrophages play an important role in the body’s response to biomaterials, as they are modulators of the wound healing process and can polarize into different phenotypes capable of inducing both deleterious and beneficial effects on tissue repair. Curcumin (CU) is known for its anti-inflammatory properties and has the potential to treat diabetic foot ulcers, but it should be delivered to wounds in a controlled manner. In this study, the encapsulation of curcumin in polymeric microparticles based on poly(sebacic anhydride) (PSA) was developed using an emulsification method. PSA-based microparticles containing different concentrations of CU were obtained: 0% weight (wt). CU (unloaded microparticles), 5, 10, and 20 wt% CU. CU encapsulation efficiency and loading were determined using a fluorescence-based calibration curve method and semi-quantitative Fourier-transform infrared spectroscopy (FTIR) analysis. The potential cytotoxicity of the obtained biomaterials in contact with primary human macrophages and their susceptibility to polarization from the M1 (pro-inflammatory) phenotype to the M2 (antiinflammatory) phenotype were evaluated. The morphology of cells cultured in contact with polymeric microparticles was evaluated using phalloidin red and 4′,6-diamidino2-phenylindole (DAPI) staining. Macrophage phenotype was assessed using flow cytometry. The obtained biomaterials showed no cytotoxic effect on primary human macrophages. Flow cytometry studies showed enhanced polarization of macrophages into anti-inflammatory M2 phenotype when exposed to microparticles loaded with CU and CU powder as compared to unloaded microparticles

Nanotechnology holds significance in all fields of research, and the formation and surface alterations of nanomaterials are particularly important in this discipline. Nanoformulations synthesized with bioactive plant components play a crucial role in the improvement of several therapeutics and diagnostics. In the present study, we reported the synthesis of a curcumin nanoformulation (CN) by using curcumin and D-α-tocopheryl polyethylene glycol 1000 succinate (TPGS). The synthesized CN was characterized using dynamic light scattering, UV-Visible spectrophotometry, Fourier-transform infrared spectroscopy, field-emission scanning electron microscopy, and X-ray diffraction. Furthermore, it was evaluated for solubility, drug loading, encapsulation efficiency, stability, in vitro release, and anticancer potentials. The role of TPGS in the synthesis of CN was validated. The synthesized CN exhibited a size of 6.2 ± 1.9 nm, needle-shaped morphology, a polydispersity index of 0.164, and zeta potential of -10.1 ± 3.21 mV, as determined by characterization techniques. Its water solubility was 2.5 × 104 times higher than that of pure curcumin. The encapsulation efficiency and curcumin loading efficiency of the synthesized CN were found to be 80 and 10%, respectively, with storage stability exceeding 30 days. Moreover, the synthesized CN demonstrated significant in vitro anticancer activity against the colorectal cancer cell line HCT-116, with an IC50 value of 12.74 ± 0.54 μM at 24 h.

Analysis of a fusion between rat glioma cells and biomimetic liposomes with encapsulated diamond nanoparticles or curcumin. Liposomes are used as carriers for different bioactive agents, both hydrophilic, which are encapsulated in water core of the liposome, and hydrophobic, which are entrapped within liposome walls. The walls are built from phospholipid bilayer, therefore their structure resembles cell membrane. It was hypothesized that if the wall is made of set of lipids typical for a cell, the liposome will be eagerly consumed by the cell. We performed the experiments using C6 rat glioma cells as an example, since central nervous system cells are extremely rich in lipids, including the unique ones. Since all cancer cells have high proliferation potential, they need to absorb precursors to build cell membrane around new cells, therefore such biomimetic liposomes may be one of the most effective way to deliver anticancer agents into the cell. Analysis of physicochemical properties of obtained liposomes, as well as in vitro tests, showed that obtaining such liposomes is possible and that the liposomes are biocompatible, stable carrier both for hydrophilic and hydrophobic agents. Encapsulation of diamond nanoparticles did not affect the liposomes, whereas entrapping of curcumin, which is a spice known in traditional Asian medicine for its anticancer properties, significantly increased its activity. Obtained results showed that biomimetic liposomes can be effective, individually-tailored carriers for bioactive agents.
Analiza fuzji pomiędzy komórkami glejaka szczurzego i liposomami biomimicznymi z enkapsulowanymi nanocząstkami diamentu lub kurkuminą.Liposomy są wykorzystywane jako nośniki dla biologicznie aktywnych związków, zarówno hydrofilowych, które są enkapsulowane w wodnym rdzeniu liposomy, jak również hydrofobowych, które mogą być zamykane w jego ścianie. Ściany liposomów zbudowane są z dwuwarstwy fosfolipidowej, przez co strukturalnie przypominają błonę komórkową. Założono, że jeżeli taka ściana zostanie stworzona z zestawu lipidów charakterystycznych dla danej komórki, liposomy będą chętnie pobierane przez taką komórkę. Wykonano doświadczenie, w którym jako przykład użyto komórek glejaka szczurzego linii C6, ponieważ wiadomo, że komórki centralnego systemu nerwowego są wyjątkowo bogate w lipidy, włączając w to także te nietypowe. Ze względu na to, że wszystkie komórki nowotworowe charakteryzuje wysoki potencjał proliferacyjny, muszą one pobierać prekursory niezbędne do budowania błony komórkowej wokół nowopowstających komórek. Dlatego też liposomy biomimiczne mogą być jednym z najbardziej skutecznych sposobów na dostarczenie związków przeciwnowotworowych do takich komórek. Analiza właściwości fizykochemicznych, jak również testy in vitro, pokazały, że otrzymanie takich liposomów jest możliwe i że liposomy te są biozgodnym, stabilnym nośnikiem zarówno dla związków hydrofilowych, jak i hydrofobowych. Enkapsulacja nanocząstek diamentu nie wpłynęła na wytworzone liposomy, podczas gdy zamknięcie w ścianie kurkuminy, która jest znana w tradycyjnej medycynie azjatyckiej ze względu na woje właściwości przeciwnowotworowe, istotnie podniosło jej aktywność. Otrzymane wyniki pokazały, że liposomy biomimiczne mogą być efektywnym, indywidualnie dopasowanymi nośnikami dla związków biologicznie czynnych.

Żywność zawiera potencjalnie mutagenne substancje, takie jak PhIP, heterocykliczna amina aromatyczna powstająca podczas obróbki cieplnej mięsa, jak również substancje mogące działać w sposób ochronny poprzez m.in. modulowanie odpowiedzi antyoksydacyjnej i przeciwzapalnej, jak np. kurkumina, która jest obecna w kurkumie i mieszance curry. Materiał i metody: W przedstawionych badaniach analizowano rolę PhIP i kurkuminy na uszkodzenia DNA w limfocytach izolowanych z krwi pacjentów z rakiem jelita grubego. Limfocyty osób z rakiem jelita grubego (10 pacjentów) oraz zdrowych (kontrola, 6 osób) traktowano PhIP i/lub kurkuminą, następnie poziom uszkodzeń DNA porównano metodą kometową. Wyniki: Wstępne wyniki sugerują, że limfocyty chorych na nowotwór jelita grubego mają większe podstawowe uszkodzenia DNA niż te od osób zdrowych. Uszkodzenia wywoływane PhIP są liniowo zależne od zastosowanej dawki i mogą być zredukowane w obecności kurkuminy (zwłaszcza w stężeniu do 5 μM). Wnioski: Dane wskazują, że kurkumina może wywierać działanie ochronne przed uszkodzeniami DNA indukowanymi mutagenem obecnym w żywności.
Food contains substances that are potentially mutagens such as PhIP, heterocyclic aromatic amine, produced during heat treatment of meat, as well as substances that may act in protective manner, inter alia by modulating the antioxidant and anti-infl ammatory response, such as curcumin, which is present in turmeric and curry. Material and methods: This study investigated the effect of PhIP and curcumin on DNA damage in lymphocytes isolated from the blood of colorectal cancer patients. Lymphocytes from individuals diagnosed with colorectal cancer (N=10) and healthy individuals (control, N=6) were treated with PhIP or/and curcumin, followed by analysis of DNA damage using single cell gel electrophoresis (comet assay). Results: Preliminary results suggest that lymphocytes of patients with colorectal cancer have a greater baseline DNA damage than those from healthy individuals. PhIP-induced DNA damage is dose-dependent and can be reduced in the presence of curcumin, especially at low concentration (up to 5 μM). Conclusions: The data show that curcumin may exert a protective effect against DNA damage induced by mutagens present in food.

The effects of long-term administration of a standardised extract of Curcuma longa on the explorative activity in aged 24-month-old male Wistar rats were estimated in a hole-board test. The animals received the extract orally for two months in rodent chow at doses of 10 and 50 mg/kg/day. The correlations between concentration of some neurotransmitters and amino acids in selected brain regions, as well as the level of corticosterone in plasma (described previously), and the parameters of exploration were calculated. The antioxidant processes (GSH, GST, CAT and MDA levels) in heart and skeletal muscles were also investigated. The results suggested that the explorative activity of plant extract-treated rats was enhanced; however, no correlations between brain neurotransmitter concentration or plasma corticosterone level and the parameters of explorative activity were observed. In biochemical investigation, chronic C. longa extract administration influenced antioxidant processes (CAT, GST and MDA levels) in skeletal muscles of aged rats, but not in the heart muscle.

Microparticles (MPs) made of fast biodegrading biomaterials, loaded with drugs, are considered a superior treatment method for pulmonary infections. One of the promising biomaterials for obtaining such a drug delivery system (DDS) is poly(sebacic anhydride) (PSA) due to its favourable degradation kinetics and mechanism. In this paper, we present a study of manufacturing MPs from PSA loaded with curcumin (CU) for pulmonary purposes. MPs were manufactured by oil-in-water emulsification; their morphology and size distribution were evaluated using optical microscopy, while the encapsulation efficiency and drug loading were obtained by the fluorometric assay. The cytotoxicity of the MPs, both the empty ones and loaded with CU, was analysed by in vitro tests with BEAS-2B human lung epithelial cells. To this end, metabolic activity by AlamarBlue assay and fluorescent staining (DAPI/ eosin) of the cells were performed. The MPs produced were round, regular in shape with diameters in the range of 1-5 µm and of yellow colour originating from CU. The CU encapsulation efficiency ranged from 42% to 55% and decreased with a higher CU ratio. The drug loading ranged from 4% to 11% and increased at a higher CU ratio. Both empty and CU-loaded MPs did not show a cytotoxic effect at concentrations up to 10 µg/ml.

Proces zapalny jest podstawowym zjawiskiem w przebiegu infekcji i korzystnym, gdy trwa krótko. Przewlekłe zapalenie jest z kolei stanem patologicznym, które leży u podłoża licznych chorób, w tym coraz częstszych chorób autoimmunizacyjnych. Obecnie wiadomo, że sposób odżywiania oddziałuje na mikrobiotę jelitową. Ta natomiast, jako kluczowy element układu immunologicznego, jest ściśle powiązana z funkcjami obronnymi organizmu. Odporność i sprawność zwalczania infekcji mogą być skutecznie wspierane poprzez odpowiednie składniki pożywienia. Ponadto naturalne produkty immunostymulujące wykazują duży potencjał w prewencji oraz łagodzeniu przewlekłego stanu zapalnego. Celem pracy jest przybliżenie zjawiska procesu zapalnego i zakresu możliwości modulowania go poprzez odżywianie, zarówno w stanach ostrych, jak i przewlekłych.
The inflammation process is essential in the course of developing infection and beneficial when it lasts for a short period. Conversely, chronic inflammation represents a pathological condition that lies at the bottom of numerous disorders, including the increasingly common autoimmune diseases. Nowadays it is commonly known that our eating habits affect the gut microbiota. Being the key element of the immune system, it is thus highly relevant to the organism’s defensive functions. Immunity and the ability to fight infections can be efficiently enhanced by nutrients. Furthermore, natural immunostymulants show a high potential of prevention and alleviation of chronic inflammation. The aim of this paper is to present the phenomenon of inflammation and the range of possibilities to modulate it through nutrition, both in acute and chronic conditions.
