Ceratopteris thalictroides is a water fern very sensitive to both dehydration and low temperature. This study focuses on the cryopreservation of this species by encapsulation-dehydration technique, in particular on the effects of pre-culture step, alginate bead size and the physical conditions of culture on the cryopreservation efficiency. Encapsulated and non-pre-cultured gametophytes did not survive cooling with liquid nitrogen. When cryopreservation was preceded by a 2-week period of pre-culture, regrowth reached 42.1%. Reduction in the size of the alginate bead, and culture in total darkness resulted in improved gametophyte regrowth capacity (75.5% or 81.7%, respectively). The best results (91.3%) were obtained when all factors tested occurred simultaneously. The gametophytes recovered very quickly and sporophytes were formed within 4 weeks after rewarming. These simple improvements can be used, not only for the cryopreservation of gametophytes in cryptogams but also for some recalcitrant species of seed plants.

Artykuł jest poświęcony problematyce kriokonserwacji ludzkich embrionów. Stosowanie tej techniki pozostaje w ścisłym związku z prokreacją wspomaganą. Stwarzane celem zwiększenia skuteczności procedury in vitro nadliczbowe embriony zostają zamrożone, aby następnie można było je wykorzystać w kolejnych próbach transferu. Autor przedstawia techniczny wymiar zamrażania i ryzyko z nim związane. W ostatniej części Autor analizuje wymiar etyczny tych technik. Podkreśla, że chociaż sama prokreacja wspomagana jest niemoralna, to jednak moralnie słuszne jest dążenie do naprawienia tego zła przez matkę, która jest gotowa przyjąć do swojego łona stopniowo rozmrażane embriony. Dobrym rozwiązaniem wydaje się być adopcja porzuconych embrionów. Takiego rozwiązania nie da się jednak usprawiedliwić z punktu widzenia etycznego. Jeżeli rozmrożenie zostaje wymuszone przez prawo lub wymówienie umowy przez klinikę przechowywującą te embriony, możliwa jest zgoda na ich obumarcie. Z obowiązku transferu zwalnia również stan zdrowotny embrionów uniemożliwiający ich rozwój. Ponadto niemoralna jest selekcja eugeniczna embrionów oraz transfer wyłącznie tych embrionów, które mają pożądane cechy fenotypowe czy płeć.
The article discusses the issues of cryopreservation of human embryos. The use of this technique is related to assisted procreation. The supernumerary embryos created in order to increase the effectiveness of the in vitro procedure are frozen so that they can be used in subsequent transfer attempts. The author presents the technical dimension of freezing and the risks associated with it. In the last part, the author analyzes the ethical dimension of these techniques. He emphasize that although assisted procreation is immoral itself, it is morally right to strive to correct this evil by the mother, who is ready to accept into her womb the thawed embryos. The adoption of abandoned embryos seems to be a good solution. However, this solution cannot be justified from the ethical point of view. If it is forced by law to defrost or terminate the contract by the clinic that stores the embryos, it is possible to consent to their death. The health condition of the embryos, which makes their development impossible, releases from the obligation to transfer. Moreover, it is immoral to select embryos by eugenics and transfer only those embryos that have the desired phenotypic or sex characteristics.

Opis:
The purpose of the research is to prepare a mathematical and numerical model for the phenomenon of heat transfer during cryopreservation. In the paper, two popular methods, slow freezing and vitrification, are compared. Furthermore, the basic model of thermal processes is supplemented by the phenomenon of phase transitions. To determine the temperature distribution during cryopreservation processes, one uses the heat transfer equation proposed by Pennes. An integral part of the energy equation is the substitute thermal capacity (STC) performed according to the concept named one domain method (fixed domain method), The numerical model is developed using the finite difference method (FDM) connected with directed interval arithmetic. The final part of the article contains the results of numerical simulations.

Opis:
The discovery of conifer somatic embryogenesis (SE) and the subsequent development of SE protocols for a range of genera and species have opened new research opportunities to forest biotechnologists and a means towards mass clonal propagation for the forest industry. This paper provides a general description of the conifer SE process, followed by a review of protocols developed specifically for several conifer species in the Pinaceae family for which production of somatic trees has been demonstrated. Additionally, future research needs, including approaches for developing markers to optimize SE process and the production of high quality embryos, are discussed.

Cryopreservation (storage in liquid nitrogen, –196°C) is a technique that ensures safe, long−term conservation of plant species with recalcitrant seeds, vegetatively propagated species and biotechnology products such as somatic embryos, cell lines and genetically transformed material. The aim of cryostorage is to keep plant tissue in a safe way, which does not cause changes during storage at neither phenotypic, cytological and histological, biochemical, genetic nor epigenetic level. Therefore, before cryostorage will be applied for long−term preservation it should be thoroughly assess if it does not cause any injuries in plant material.

The aim of this study was to investigate the effect of platelet activating factor (PAF) on the quality characteristics of cryopreserved canine spermatozoa. Cryopreserved semen of 5 mixed-breed dogs was treated with different concentrations of exogenous PAF (1 × 10⁻³M, 1 × 10⁻⁴M, 1 × 10⁻⁵M and 1 × 10-6M) and examined at different time intervals (0, 30, 60 and 120 min). Cryopreserved semen treated without PAF was used as the control. Sperm quality was evaluated for motility (computer-assisted semen analysis, CASA), mitochondrial function (JC-1/PI assay) and plasma membrane integrity (SYBR-14/PI assay and Hoechst 33258). Also, ATP content of spermatozoa was determined using a bioluminescence assay. Treatment of cryopreserved semen with 1 × 10⁻³ M PAF at 120 min of incubation resulted in significantly higher total sperm motility compared with the control. It was observed that PAF-improved total sperm motility was concurrent with enhanced sperm motility patterns after treatment of cryopreserved semen. Treatment of cryopreserved semen with PAF did not improve either sperm mitochondrial function or plasma membrane integrity, as monitored by different fluorescent membrane markers. Furthermore, ATP content of cryopreserved spermatozoa was significantly higher when PAF was used at a concentration of 1 × 10⁻³ M compared with the control and other PAF treatments, regardless of the incubation time. The findings of this study indicated that treatment with 1 × 10⁻³ M PAF at 120 min of incubation rendered better quality of cryopreserved canine semen, which was associated with improved sperm motility parameters and ATP content. It can be suggested that exogenous PAF addition is beneficial as a supplement for canine semen extender used for cryopreservation.

The aim of this study was to evaluate the influence of sucrose concentration in the preculture medium, as well as the duration of osmotic dehydration on the efficiency of chrysanthemum ‘Richmond’, ‘Lady Orange’ and ‘Lady Salmon’ cryopreservation by encapsulation-dehydration technique. For all cultivars tested, the best regrowth of cryopreserved shot tips was recorded with 0.25 M sucrose concentration and 10 µM ABA during a two-week preculture, followed by a 4- or 7-day osmotic dehydration. The survival rate ranged from 56.8–58.0% (the Lady group) to 63.6% (‘Richmond’). However, the ability to grow was smaller and reached 18.2–50.7%. It was found that higher sucrose concentration during the preculture slowed the growth of chrysanthemum shoot tips and led to an increased formation of multiple shoots (by activating axillary buds) or deformed adventitious shoots (incapable of further growth). The frequency of tissue hyperhydricity also increased, while the rhizogenesis efficiency decreased when higher sucrose concentration in the preculture medium was applied. The influence of osmotic dehydration duration on the explants morphogenetic response was cultivar-dependent.

Willows produce fast germinating and short-lived seeds, difficult to store in the long-term under controlled conditions. The aim of this study was to examine the feasibility of storage of three Salix spp. at controlled temperatures (3°, −10°, −196 °C). We also analyzed the effect of spermidine (Spd) as an antioxidant factor in desiccated seeds. Collected seeds were either desiccated or hydrated to obtain 10 levels of moisture content (between app. 4% and 2%) and subjected to storage at temperatures 3°, −10°, or −196 °C (liquid nitrogen; LN). After two months, seeds were germinated on the light at 20 °C. Seeds desiccated below a safe range of moisture content were further tested and germinated on filter paper with additions of 0.25 mM Spd solution. After 7 days seedlings were examined for hydrogen peroxide content (H2O2) and total antioxidant capacity (TAC). Fresh seeds of three Salix species: Persian willow (S. aegyptiaca L.), heartleaf willow (S. cordata Michx.) and crack willow (S. ×fragilis L.) were successfully stored at temperature −10° and −196 °C for two months. After cryopreservation seed of S. aegyptiaca, S. cordata, and S. ×fragilis germinated without viability loss in moisture content ranging from 4.4–15.9%, 6.4–18.5%, and 7.1–11.5% respectively. The addition of Spd during germination of desiccated seed did not affect germination capacity. However, seedlings of S. aegyptiaca had lower hydrogen peroxide content in comparison with control (germination on water). Seedlings of S. cordata showed an increase in hydrogen peroxide content in control after storing in LN. In seedlings of Crack willow Spd increased hydrogen peroxide content. Seeds of tested species differ in response to storage conditions. Salix seeds can be stored successfully for two months at −10° or −196 °C without losing viability in the safe range of moisture content. Storing at 3 °C can be used for storage in the narrower range of seeds’ moisture content, however, seedlings stored at this temperature produce a higher level of reactive oxygen species. Germinating seeds in Spd did not increase their germination, however in S. aegyptiaca and S. cordata decreased hydrogen peroxide content.

Shoot tips from in vitro plants of four rose species were cryopreserved by the droplet vitrification method. Optimized conditions involved exposure to loading solution for 20 min, then treatment with plant vitrification solution (PVS2) for 20 min (Rosa agrestis, R. canina and R. dumalis) or 30 min (R. rubiginosa) followed by freezing in liquid nitrogen. Survival rate ranged from 78.3 to 95.1%, depending on the species. Regrowth rate of shoot tips was 50.5% for R. agrestis, 63.2% for R. rubiginosa, 71.4% for R. dumalis and 78% for R. canina. The preculture of donor plants in a medium with 0.25 цЫ sucrose facilitated the isolation of shoot tips and increased regrowth rate after cryopreservation. Plant regeneration was carried out in Murashige and Skoog medium with 1 цЫ 6-benzylaminopurine, 1.5 цЫ gibberellic acid and 0.087 M sucrose. Plants regenerated from cryopreserved shoot tips did not display morphological alterations in comparison with non-cryopreserved shoot tip - derived plants.
