In the yeast Saccharomyces cerevisiae the RER2 and SRT1 genes encode Rer2 and Srt1 proteins with cis-prenyltransferase (cis-PT-ase) activity. Both cis-PT-ases utilize farnesyl diphosphate (FPP) as a starter for polyprenyl diphosphate (dolichol backbone) formation. The products of the Rer2 and Srt1 proteins consist of 14-17 and 18-23 isoprene units, respectively. In this work we demonstrate that deletion or overexpression of SRT1 up-regulates the activity of Rer2p and dolichol content. However, upon overexpression of SRT1, preferential synthesis of longer-chain dolichols and a decrease in the amount of the shorter species are observed. Furthermore, overexpression of the ERG20 gene (encoding farnesyl diphosphate synthase, Erg20p) induces transcription of SRT1 mRNA and increases the levels of mRNA for RER2 and DPM1 (dolichyl phosphate mannose synthase, Dpm1p). Subsequently the enzymatic activity of Rer2p and dolichol content are also increased. However, the amount of Dpm1p or its enzymatic activity remain unchanged.

Opis:
Autocrine growth factors produced by epithelial cells mediate the development and proliferation of neoplastic human prostate tissue. Various approaches have been used to down-regulate neoplastic growth of prostate cancer using natural flavonoids, soluble receptors, pseudo-ligands, monoclonal antibodies and tyrosine kinase inhibitors (tyrphostins). Selected growth factor/growth factor receptor loops (mainly TGFα/EGFR and IGFs/IGFIR) have been proposed as regulators of prostate cancer cell growth. We have previously determined that blockade of IGFIR or VEGF2R signaling pathways by tyrphostin AG1024 and SU1498 inhibits autocrine growth and viability of DU145 cells in vitro. Recently, we compared the activity of AG1024 and SU1498 with the inhibiting effect of tyrphostin A23 (a selective inhibitor of EGFR). The results described in this paper confirm that DU145 cells do not produce IGFI or EGF. In contrast, DU145 cells produce a great amount of VEGF, much more than TGFα (about 60-fold), and VEGF may be the real autocrine growth factor of the investigated cells. The results indicate that the growth of DU145 may be regulated by at least three autocrine loops: TGFα/EGFR, IGFII/IGFIR and VEGF/VEGFR2. Neither AG1024 nor SU1498 affected the production of TGFα substantially, which excludes the possibility that IGFRs or VEGFR2 inhibitors arrest the growth of these cells by inhibition of synthesis and/or secretion of TGFα. The obtained data indicate that all tree investigated tyrphostins (AG1024, SU1498 and A23) inhibit signal transmission by Akt (PKB), ERK(1/2), Src and STAT in a similar manner. A comparison of the effects of the investigated tyrphostins indicates that TGFα, IGFII and VEGF stimulate cell growth by affecting the same signaling pathway. The hypothesis was confirmed by the effect of the investigated tyrphostins on activation of EGFR. All these inhibitors decreased phosphorylation of EGFR to the same extent, and after the same time of incubation with cell culture. These results strongly suggest that stimulation of EGFR kinase is the main step in the initiation of mitogen signaling in DU145 cells, regardless of the type of ligand (TGFα, IGFs or VEGF) and their specific receptors.

Calcium ions as second messengers play an essential role in many important cellular processes. In plants, transient changes in calcium content in the cytosol (calcium signatures) have been observed during growth, development and under stress conditions. Such diverse functions require many different calcium sensors. One of the largest and most differentiated group of calcium sensors are protein kinases, among them calcium-dependent protein kinases (CDPKs) which were identified only in plants and protists. CDPKs have a regulatory domain which is able to bind calcium ions. For regulation of CDPKs activities not only calcium ions but also specific phospholipids and autophosphorylation are responsible. CDPKs have many different substrates, which reflects the diversity of their functions. Potential protein substrates of CDPK are involved in carbon and nitrogen metabolism, phospholipid synthesis, defense responses, ion and water transport, cytoskeleton organization, transcription and hormone responses. Presently, participation of CDPKs in stress signal transduction pathways (e.g., cold, drought, high salinity, wounding) is intensively studied in many laboratories. An intriguing, but still not fully clarified problem is the cross-talk via CDPKs among different signaling pathways that enables signal integration at different levels and ensure appropriate downstream responses.

We report an integrated micro-actuator for independent control of the displacement in three orthogonal directions. Electrostatic comb drives are employed for controlling the displace- ment in the x and y directions while a parallel plate-type actuation is used for driving the displacement in the z direction. The three actuators are mechanically coupled, but are elec- trically isolated. The calculation models are established for investigating the operation char- acteristics of the micro-actuator. The calculated results are in good agreement with those obtained by the finite element method in Comsol Multiphysics 4.3. The results of modal analysis show that the displacement in the three orthogonal directions can be independently controlled with low mode cross-talk.

Cross-talk between two signal transduction pathways leads to a negative regulation of androgen-induced ornithine decarboxylase (ODC) gene expression in the mouse kidney. One pathway is triggered by testosterone via the intracellular androgen receptor, AR, and the other is induced by antifolate CB 3717 or folate via hepatocyte growth factor and its cell membrane receptor c-Met. Here we report the studies of the expression of AR and c-Myc transcription factors involved in ODC transactivation. Administration of CB 3717 or folate decreased the expression of AR. In contrast, testosterone did not modify AR mRNA content but augmented the AR protein. Furthermore, we demonstrate that administration of folate, but not testosterone, increases c-Myc transcript and protein level. We also document that activation of both examined pathways does not decrease the testosterone-induced AR protein level, but markedly increases c-Myc protein which is nearly 2-fold up-regulated compared to its level evoked solely by testosterone. We suspect that this pronounced increase of c-Myc protein might have functional consequences mirrored by down-regulated expression of AR target genes, among them ODC.

In this review we summarize the present status of our knowledge on the enzymes involved in the extracellular metabolism of nucleotides and the receptors involved in nucleotide signalling. We focus on the mechanism of the ATP and ADP signalling pathways in glioma C6, representative of the type of nonexcitable cells. In these cells, ATP acts on the P2Y2 receptor coupled to phospholipase C, whereas ADP on two distinct P2Y receptors: P2Y1 and P2Y12. The former is linked to phospholipase C and the latter is negatively coupled to adenylyl cyclase. The possible cross-talk between the ATP-, ADP- and adenosine-induced pathways, leading to simultaneous regulation of inositol 1,4,5-trisphosphate and cAMP mediated signalling, is discussed.

The article concerns the author’s concept of a specific kind of social research. “Networking research” rely on connecting field work research with the intention of making social change. The aim of the networking research is “networking”,ie. the strengthening of interpersonal relations, inter-organizational and cross-sector relations in the local community. This article presents “exploratory walk” (or “walk & talk”) as an example of application the concept of networking research. The article indicates the cognitive and practical benefits of using this approach, with particular emphasis on the revitalization process. The article presents the theoretical foundations of the networking research concept. It indicates a source of inspiration and basic assumptions. It justifies choosing the exloratory walk technique as a way of learning and simultaneously changing social reality. It presents some rules of conducting the “walk & talk” technique in the context of diagnosing a local community. It also presents how to use the method in during a process of preparing of revitalization programs. It also explains, presenting selected examples, what is the “networking” nature of the research.
Artykuł dotyczy autorskiej koncepcji specyficznej odmiany badań społecznych. Badania sieciujące polegają na łączeniu poznawania rzeczywistości w terenie z intencją dokonywania zmian społecznych. Celem badań siecujących i połączonych z nimi interwencji jest „sieciowanie”, czyli wzmacnianie relacji interpersonalnych, międzyorganizacyjnych i międzysektorowych w społeczności lokalnej. Celem artykułu jest przedstawienie spacerów badawczych jako przykładu zastosowania koncepcji badań sieciujących. W artykule wskazano korzyści poznawcze i praktyczne wynikające ze stosowania tego podejścia, ze szczególnym uwzględnieniem procesu rewitalizacji. W artykule przedstawiono teoretyczne podstawy koncepcji badań sieciujących. Wskazano źródła inspiracji i podstawowe założenia. Uzasadniono także wybór techniki spacerów badawczych jako sposobu poznawania i jednoczesnego zmieniania rzeczywistości społecznej. Przedstawiono kilka zasad dotyczących sposobu prowadzenia badań techniką „walk & talk” w kontekście diagnozowania społeczności lokalnej, w związku z opracowaniem programu rewitalizacji. Pokazano też, na wybranych przykładach, na czym ma polegać „sieciujący” charakter owych badań.
