    Opis:
    The main goal in gene therapy and biomedical research is an efficient transcription factors (TFs) delivery system. SNAIL, a zinc finger transcription factor, is strongly involved in tumor, what makes its signaling pathways an interesting research subject. The necessity of tracking activation of intracellular pathways has prompted fluorescent proteins usage as localization markers. Advanced molecular cloning techniques allow to generate fusion proteins from fluorescent markers and transcription factors. Depending on fusion strategy, the protein expression levels and nuclear transport ability are significantly different. The P2A self-cleavage motif through its cleavage ability allows two single proteins to be simultaneously expressed. The aim of this study was to compare two strategies for introducing a pair of genes using expression vector system. We have examined GFP and SNAI1 gene fusions by comprising common nucleotide polylinker (multiple cloning site) or P2A motif in between them, resulting in one fusion or two independent protein expressions respectively. In each case transgene expression levels and translation efficiency as well as nuclear localization of expressed protein have been analyzed. Our data showed that usage of P2A motif provides more effective nuclear transport of SNAIL transcription factor than conventional genes linker. At the same time the fluorescent marker spreads evenly in subcellular space.

    When we investigated the genotoxicity of β-carotene cleavage products (CPs) in primary rat hepatocytes stimulated to proliferate, we observed dose-dependent increases of chromosomal aberrations, sister chromatid exchanges and micronuclei. In contrast to other genotoxic substances, however, this increased genotoxicity was not accompanied by increased cytotoxicity. As a consequence we observed metaphases showing massive chromosomal damage, indicating inhibition of apoptosis by CPs enabling these cells to proceed in the cell cycle. Since proliferative stimulation by growth factors may support this effect, the in vitro toxicological effects of CPs were studied on proliferatively quiescent primary rat hepatocytes. A significant increase of both apoptosis and necrosis was found. Supplementation with antioxidants did not significantly lower the level of apoptosis, while the level of necrosis was significantly reduced by Trolox and N-acetylcysteine at all concentrations tested as well as ascorbic acid (50 µM) and a combination of Trolox (50 µM) and ascorbic acid (50 µM). These observations indicate that a) the cytotoxic potential in combination with the genotoxic potential of CPs may promote the initiation of cells due to compensatory cell division in exposed tissues and may aggravate inflammatory processes under chronic exposure, and b) the applied antioxidants may protect from cytotoxicity primarily via the detoxification of aldehydic β-carotene cleavage products.

    Seeing sociology visually adds a sense of realness to the viewer compared to only reading sociological texts. In this paper, I aim to provide an example of how a single scene from a feature film can be utilized as a practical and meaningful means to analyze a social situation and to help students of sociology to grasp key features of Goffman’s theory of interaction order. More precisely, the main aims of the paper are 1) to illustrate Goffman’s theory of the interaction order by identifying acts of disruption and alignment in interaction through a film clip; and 2) to attempt to analyze, in a Goffmanian sense, what is really going on in the situational interaction. The scene is from the 2013 American movie August: Osage County and follows a dinner of immediate family in the wake of the funeral of the hostess’s late husband. The normative and civilized interaction of the meal is, however, jeopardized by the hostile and provocative mood of the hostess, as she repeatedly disrupts the interaction order with attempts to mock and/or uncover the hidden and vulnerable truths of the immediate members of her family, exemplifying her power status in the particular situation. The dinner guests, however, try to overlook and resist the provocation of the hostess and stick to their predetermined roles to save and sustain their idealized selves (their faces) and the interaction order (the faces of others), In doing so they, on the one hand, discard the uncomfortable truths acclaimed by the hostess and, on the other, explain the hostess’s provocative actions in terms of their claim that she is unwell and in need of medical attention. Thus, the attacked dinner guests in the scene align more alliance to the interaction order than to truth itself.

    In this paper, the cellular automata model is applied to analyse cleavage and ductile fracture in front of a crack in three-point-bend specimens made of Hardox-400 steel. The research programme was composed of experiments followed by fractographic and numerical analyses. On the basis of microscopic observations, the sizes of cells used in the automata were determined. The algorithm enabled mapping of the two-dimensional crack surface as well as a simulation of temperature-dependent failure mechanisms by defining transition rules based on the modified Ritchie-Knott-Rice cleavage fracture criterion. The critical stress values were estimated and verified by the cellular automata model.

The cleavage takes 4 days, being terminated on the 5th day of embryogenesis, when the cells start to invaginate and differentiate. The karyomeres play a role in the initial mitotic divisions. Cellularization occurs on the 2nd and 3rd day of embryogenesis. The blastoderm cells maintain their potential for division, and as from the 5th day cellular differentiation starts.

    Artykuł porusza kwestię teoretycznie ważną, ale empirycznie zaniedbaną, a mianowicie czy dostrzegamy strukturyzacje i konsolidacje polskiego systemu partyjnego. Po ćwierćwieczu demokratycznej praktyki oczekiwanie takie jest zasadne. Odpowiedzi na powyższe pytanie poszukujemy wykorzystując klasyków – teorię rozłamów socjopolitycznych Seymoura Lipseta i Steina Rokkana oraz dorobek ich następców. Wykorzystujemy do tego gromadzone przez ostatnie dwie dekady dane Polskiego Generalnego Studium Wyborczego. Nasze analizy wskazują, że dopiero od roku 2005 możemy mówić o symptomach owej konsolidacji i pojawianiu się objawów polityki rozłamów socjopolitycznych. Konkluzja jest jednak ostrożna – dostrzegamy właśnie symptomy takiej strukturyzacji, natomiast nie twierdzimy, że mamy do czynienia z rozłamami pełnymi, z jakimi mieliśmy do czynienia w Europie Zachodniej w połowie, i nieco później, XX wieku.
    This article addresses the important yet neglected issue of whether the Polish party system has become more structured and consolidated. After twenty-five years of democracy, it would be reasonable to expect such an outcome. To answer the question, we based our analysis on the classic socio-political-cleavage theory of Lipset and Rokkan and the work of those who were influenced by them. Our analysis, which makes use of data collected over the last two decades by the Polish National Election Study, shows that it has been possible to discern symptoms of consolidation and the emergence of socio-political cleavages only since 2005. Our conclusions are therefore tentative – we can observe a degree of structuring, but we cannot claim that we have found full cleavages akin to those observed in Western Europe in the second half of the twentieth century.
