Opis:
Plasmogenes are largely located in mitochondria or plastids and they can infl uence the inheritance of many plant characteristics. This phenomenon is called cytoplasmic inheritance and can be detected on the basis of the expression of a trait in progeny F1 obtained from single and reciprocal crosses. The aim of this study was to examine the cytoplasmic inheritance of in vitro productivity of morphogenesis in three genotypes of Fragaria x ananassa Duch., i.e. the cultivars ‘Dukat’, ‘Teresa’ and the breeding clone no. 590. Single and reciprocal crosses were done according to Griffi ng’s method 3. The value of general combining ability (GCA) indicated cv. ‘Teresa’ as the best maternal component for crossing and ‘Dukat’ as the worst. The negative reciprocal cross effects (rij) revealed the cytoplasmic inheritance for cv. ‘Dukat’ as maternal form and positive rij for the breeding clone no. 590 indicated the nuclear inheritance of morphogenetic ability. Cv. ‘Teresa’, as maternal component, showed nuclear inheritance of that trait in crossing with cv. ‘Dukat’ and with 590 cytoplasmic inheritance. The productivity of morphogenesis in strawberry depended on the parental combination and the direction of crossing.
Plazmogeny zlokalizowane są głównie w mitochondriach oraz plastydach i mogą wpływać na dziedziczenie wielu cech u roślin. Zjawisko to określa się mianem dziedziczenia cytoplazmatycznego, wykrywanego na podstawie ekspresji danej cechy w pokoleniu F1, uzyskanym z krzyżowań prostych i odwrotnych. Celem badań było określenie wpływu dziedziczenia cytoplazmatycznego na produktywność morfogenezy in vitro trzech genotypów Fragaria x ananasa Duch. tj. odmiany ‘Dukat’, ‘Teresa’ i klonu hodowlanego nr 590. Proste i odwrotne krzyżowania wykonano według trzeciej metody Griffi ng’a. Najlepszym komponentem matecznym określonym na podstawie ogólnej zdolności kombinacyjnej (GCA) była odmiana ‘Teresa’, natomiast najsłabszym odmiana ‘Dukat’. Negatywny efekt krzyżowań odwrotnych (rij) wskazał na cytoplazmatyczne dziedziczenie zdolności morfogenetycznych u odmiany ‘Dukat’ jako formy matecznej, natomiast pozytywny rij na dziedziczenie jądrowe u klonu hodowlanego nr 590. U odmiany ‘Teresa’ jako komponentu matecznego, w krzyżowaniu z odmianą ‘Dukat’, stwierdzono dziedziczenie jądrowe danej cechy a cytoplazmatyczne w krzyżowaniu z klonem hodowlanym nr 590. Produktywność morfogenezy badanych odmian i klonu truskawki zależała od kombinacji rodzicielskiej oraz kierunku krzyżowania.

Opis:
The biogenesis of plant mitochondria and plastids is a multistep process that depends on the expression of both, organellar and nuclear genes. A growing body of evidence suggests that the indispensable coordination of different steps in this process may be gained by participation of the non-coding RNAs. A plethora of non-coding RNAs of diverse length, both intraorganellar ones, as well as encoded by the nuclear genome (including microRNAs and short interfering RNAs), were also suggested to play a role in the stress response by regulating the expression levels of targeted genes important for organelle biogenesis. Selected points of current interest regarding the regulation of plant mitochondrial and plastid gene expression by diverse non-coding RNAs, also discussed in the aspect of abiotic stress conditions, are highlighted here.

The aims of this study were 1) to determine the variability in the flowering phenology of Scots pine (Pinus sylvestris L.) clones in a seed orchard and 2) to compare the genetic structure and genetic markers (13 isozyme loci and 5 chloroplast and 3 nuclear DNA microsatellite loci) among groups of clones that are differentiated by flowering phenology. Using the timing of male inflorescence development, 57 plus trees represented by their clones in a seed orchard were classified into three phenological groups: early-, intermediate-, and late-flowering. The microsatellites showed no significant differences in the genetic structure of the analyzed phenological groups. However, the frequency of allele 2 at the shikimate dehydrogenase A locus (ShDH A 2) differed significantly between the groups of early- and late-flowering trees and between the groups of intermediate- and late-flowering trees. In addition, a significant difference in the frequencies of the genotype ShDH A 11 was observed between the intermediate- and late-flowering groups. Nei’s genetic distance indicated that the late-flowering group was the most genetically distant among the phenological groups. These results suggest that the ShDH A locus might be considered as isoenzymatic marker that differentiates these flowering groups of Scots pine clones. At several isozyme and DNA loci, the presence of private alleles in each group of pines was observed. However, these alleles cannot serve as markers of Scots pine flowering time because of their low frequencies.

Opis:
Beech decline phenomenon has been observed in Poland since the 1980s. The reasons are still unclear but the discovery of pathogens genus Phytophthora shed a new light on it. Stems as well as roots of beech trees were severely affected by P. citricola and P. cambivora. In order to find out the genetic background of infection, three loci of chloroplast microsatellite DNA were investigated in the genetic differentiation study of beeches growing in the Siewierz Forest District in Poland. Parameters of genetic diversity (h Nei) and differentiation (GST) in the chloroplast genome were estimated and compared between healthy and damaged trees. Healthy beeches were more heterozygous (h=0.243) than the damaged ones (h=0.113), and the distribution of cpDNA alleles was different between these two groups. Healthy trees were nearly 50% more genetically diversed than declining ones.

Four cpDNA regions were analyzed with the use of PCR-RFLP technique and nucleotide sequences of two mtDNA regions were characterized in order to find P. sylvestris and P. mugo species specific markers useful for studies of the species hybridization. The difference in the restriction fragment patterns of trnV-rbcL region after digestion with MvaI endonuclease was detected. The analyses of the species representatives from various geographic regions revealed that the observed polymorphism is species specific. No differences have been disclosed in the analyzed trnS-trnT, trnK1-trnK2, trnC-trnD cpDNA regions. The P. sylvestris and P.mugo mtDNA sequences of orf25 and coxI regions proved to be identical.

Scots pine (Pinus sylvestris L.) is one of the most widespread forest trees in the world, ranging from southern Mediterranean mountains to eastern Siberia. 30 trees of Scots pine from the Miłomłyn Forest District were analyzed in terms of 7 nuclear and 8 chloroplast microsatellite DNA loci as well as 7 morphological needle traits. Nuclear microsatellites (Simple Sequence Repeats) have proved to be useful in studying phylogeographic and gene flow patterns in conifers and are being used to infer the demographic history of tree species. A population’s genetic diversity, in fact, represents an important criterion which could be applied in planning future forest management and breeding. Additionally, many elements of Scots pine morphology, including needle traits, are subject to environmental modification. However, the adaptability of those traits remains, to some extent, under genetic control. The trees we investigated here are called Tabórz pine located in the northeast of Poland. This population is described as one of the best in Poland and even in Europe regarding bio mass production, ductility and wood quality. The aim of the study was to determine variation and genetic structure within this population. An analysis of phenotypic differentiation showed that the traits which differed the most between individuals were needle length, the number of serrations per 2 mm of needle length along the right edge and the number of stomatal rows on the convex side of the needles. In Tabórz pine, we detected 30 different genotypes with 87 alleles as well as 28 haplotypes with 30 alleles. We also revealed a slight excess of homozygotes, but the population is still in Hardy-Weinberg equilibrium. The Scots pine population from Tabórz furthermore showed a higher level of genetic diversity compared to stands from other Polish and European regions. This diversity may be the main factor impacting on the population’s wood quality and its breeding value.

RAPDs, ISJs, ISSRs, ITS and katGs were applied to determine genetic relationships between common Sphagnum species of the section Acutifolia. Twenty populations were genotyped using ten ISJ primers, 12 pairs of katG primers, 10 ISSR and 10 RAPD primers, and a restriction analysis of ITS1 and ITS2. ISSR and katG markers revealed the greatest number of species-specific bands. An analysis of ITS1 and ITS2 regions with restriction enzymes also proved to be a highly effective tool for species identification.
