One of the main scopes of modern cell engineering is development of cellular models that can replace animals in drug screening and toxicological tests, so called alternative methods. Construction of the alternative model is a very challenging task due to a richness of factors creating the in vivo environment. The monolayer cell culture — cultivation of adhesive cells on artificial surfaces such as glass or polymer — lack most of the in vivo-like interactions, but still is the only tool for the majority of applications. One of the most prospective approaches on mimicking in vivo environment is “Lab-on-a-chip” technology. Microfluidic devices offer lots of advantages over traditional in vitro culture, e.g. much higher cell volume-to-extracellular fluid volume ratio or possibility of regulation of hydrodynamic stress. This presentation aims to introduce latest advances of our team in microfluidic cell culture devices. Our novel approach is to cultivate three dimensional multicellular aggregates (spheroids) in microenvironments arranged in a microfluidic system. The geometry and materials of the system allow for cultivation, observation and analysis of multicellular spheroids. The results presented concern multicellular tumor spheroids (MCTS) rising from human cancer cells, which are considered to represent most of the conditionings of cancer tumor in vivo. The fully developed MCTS microdevice will be a reliable tool for anticancer drug screening, as the results most likely will be in a close accordance with the results obtained in vivo.

As an important component of blood cells, the red blood cell plays a vital role in many diseases such as malaria and so on. Although quantitative phase imaging techniques can be used for homogeneous cellular thickness distribution to obtain ideal results, they cannot achieve 3D morphological distribution. In this paper, a new method is presented to get a 3D morphology image of red blood cell. With this method, only two cellular quantitative phase images obtained from two orthogonal directions are needed as original information. By using the grid method, the sample is divided into many small phase cubes, and then we take a layer’s cubes into calculation so that the 3D problem could be transformed into a 2D problem to elaborate. Then it can be applied to the tomographic imaging combined with the maximum entropy method according to the two orthogonal phase images. This method has been proved by a simulation of red blood cell. The results show that cellular morphological distribution can be achieved in detail very well just based on only two orthogonal phase images.

Lung cancer is one of the most lethal types of cancer due to a lack of proper treatment. The rare presence of molecular therapy targets forces the use of platinum-based drugs. Cisplatin, approved by the USA as an anticancer therapy in the 1970s, is still one of the most prominent therapies against lung cancer. Unfortunately, the biggest limitation of cisplatin-based therapy is the development of cisplatin resistance. Cancer cells overcome the vast DNA damage caused by the drug in a variety of ways such as detoxication and extracellular transport of the drug, enhanced repair mechanisms, omitting apoptosis and epigenetic alterations. Chemotherapy resistance is an issue that so far cannot be dealt with. Nevertheless, better understanding of the molecular pathways behind cisplatin resistance brings hope for better therapy outcomes in lung cancer patients.

Introduction: A variety of benign and malignant tumours involving the nasal cavity and paranasal sinuses (PNS) are commonly encountered in clinical practice. The presenting features and symptomatology of all sinonasal masses are similar. It is impossible to determine clinically what pathology lies underneath. So a detailed history, clinical examination, proper imaging, and thorough histopathologic evaluation are essential to reach a diagnosis. Objectives The purpose of this study was to classify various types of Benign and malignant lesions presenting as nasal and paranasal mass and characterize their clinico-pathological profile in a tertiary care center. Materials and Methods: This was a prospective study where 120 cases of nasal and paranasal masses were included over a period of 24 months (sep 2013-sep 2015) Clinico-pathological study was carried out in these cases. A provisional diagnosis was made after clinical assessment and radiologic investigations, but final diagnosis was made after histopathologic examination. Results: The number of benign lesions were more than the malignant lesion, 66.66% versus 33.33% respectively. All age groups were involved and the mean age of presentation was benign tumours 29.5 years and malignant tumours 50.25 years. Male to Female ratio was 3:1 for benign tumours and 1.7:1 for malignant tumours. In our study, among benign lesions the occurrence of Angiofibroma was highest seen in 37.5% cases. In malignant lesions, Squamous cell carcinoma was most common seen in 67.5% cases. Carcinoma nasal cavity was the commonest malignant lesion seen in 70% cases. Nasal obstruction was the most common (91.6%) presenting complaint followed by intermittent epistaxis (69.16%) and nasal discharge (58.3%). Conclusion: We concluded that for proper evaluation of a sinonasal mass, clinical, radiologic, and histopathologic evaluation should be carried out conjointly in all the cases. Histopathology always gives a confirmatory diagnosis but in few cases immuno-histocytochemistry becomes the ultimate diagnostic technique for correct and timely intervention.

Vacuoles play very important physiological roles in plant cells. Pea root nodules, which exhibit distinct zonation (meristematic zone and central tissue zones), may serve as a good experimental model for the investigations of vacuole development and its importance to cell and tissue functioning. Moreover, the nodule central tissue is composed of both infected and uninfected cells which play different physiological roles and differ in the level of vacuolation. Cytological observations revealed that central vacuoles of the infected cells of the effective nodules expand toward cell walls. Thus only thin layers of the cytoplasm separate each central vacuole from plasma membrane and cell wall. This finding is discussed from the viewpoint of improved exchange of solutes and water between the central vacuole and apoplast of the infected cell. Three-dimensional reconstruction of the vacuoles of infected cells within a fragment of effective nodule central tissue, showed their spatial arrangement. Possible advantages coming from the spatial arrangement of vacuoles within the central tissue are discussed. A comparative study of the central tissue (bacteroidal tissue) and meristem vacuolation of the effective and ineffective pea root nodules is also presented. Morphometric measurements revealed that the effective nodule central tissue was more vacuolated than the ineffective one. It was proved that maturation of the infected cells involves dynamic changes in their vacuolation. Having numerous fixing nitrogen bacteroids, the infected cells of effective central tissue were less vacuolated than uninfected cells. On the other hand, both infected and uninfected cells of the effective central tissue showed a much higher level of vacuolation in nitrogen-fixing zone than cells of the same type in ineffective tissue. These results indicate that vacuolation is an important factor in development and functioning of pea root nodule central tissue.

This paper studies the environmental impact of two different forms of solar power generation in Thailand – that of multicrystalline silicon solar cells, and that of thin film amorphous silicon solar cells. It takes as its study two of the largest solar cell power plants of their kind in Thailand; a multicrystalline silicon plant in the north (generating 90 MW) and a thin film amorphous silicon plant in the centre (generating 55 MW). The Life Cycle Assessment tool (LCA) was used to assess the environmental impact of each stage of the process, from the manufacture of the cells, through to their transportation, installation and eventual recycling. The functional unit of the study was the generation of 1 kWh of power transmitted and distributed by the Electricity Generating Authority of Thailand (EGAT) and Provincial Electricity Authority (PEA). The environmental impact results were calculated in terms of eco-points (Pt) per functional unit of 1 kWh. The characterised data for 1 kWh of solar power generation was then compared with data for 1 kWh of combined cycle and thermal power generation (both in Thailand), using the same set of characterisation factors. After analyzing the results, both forms of solar power energy generation were found to impact upon the studied categories of Human Health, Ecosystem Quality and Resource Depletion, whilst also highlighting the importance of the solar cell module recycling process in decreasing the overall environmental impact. When the two solar cell technologies were compared, the overall impact of the multicrystalline silicon solar cell was found to be higher than that of the thin film amorphous silicon solar cell. Furthermore, when assessing the overall impact against non-renewable power generating technologies such as combined cycle and thermal power generation, the thin film amorphous silicon solar cells were found to have the lowest environmental impact of all technologies studied.

In the recent times, lot of research work carried out in the field of fuel cells explicitly divulges that it has the potential to be an ultimate power source in upcoming years. The fuel cell has more storing capacity, which enables to use in heavy power applications. In these applications, power conditioning is more vital to regulate the output voltage. Hence, we need a dc-dc converter to provide a constant regulated output voltage for such high-power system. Currently, many new converters were designed and implemented as per the requirement. This paper has made comparative study on several topologies of the quadratic high gain dc-dc converter and the applications where these topologies can be used when the fuel cell is given as a source. Also, we have compared various parameters of all the converters considered and generated the results with steady-state and dynamic study. In this article, we briefed the types of analysis carried on the dc-dc converter to study its performance. Moreover, various application of fuel cell is presented and discussed. This paper will be a handbook to the researchers who start to work on high gain dc-dc converter topologies with quadratic boost converter as a base. This article will also guide the engineers to concentrate on the fuel cell components where it needs to be explored for optimizing its operation.

Discrete cluster microforms, or simply clusters, in gravel streams result from organization of particles found in the surface layer of the gravel bed into disconnected patches. Clusters are the outcome of feedback interaction between flow, sediment and stream planform geometry. The complexity of this interaction results in several different cluster shapes, i.e. line, rhomboid and triangular. The objective of this research is to provide a quantitative characterization of cluster shape. To achieve this, we employed a novel method based on fractal theory and used for the shape description of clusters. Our novel method utilized the cell-counting method for the estimation of the areal fractal dimension, for two major datasets, namely fabricated clusters with well-defined shapes, and clusters developed in the laboratory. The principal finding of this research is that the proposed method successfully characterized cluster shape in quantitative terms. Specifically, it was shown that the new approach could identify clusters of different shapes 84% of the time, under different arrangements. This finding is of great importance for bed pattern recognition studies of stream reaches with superimposed roughness elements such as clusters. The findings of the current work could also assist numerical modellers in the development of more representative models of flows over roughness features such as clusters and in the interpretation of results from such models.

The completed SOFCs based Combined Heat and Power, CHP, system model was validated by data18 and numerical results obtained at steady-state mode showing its high-fidelity. A parametric study with respect to key operating parameters including changes in fuel utilization, lambda number and current density values was conducted. The global CHP system dynamic response, in term of the current/voltage delivered by two-stage SOFC stacks, under a fi xed fuel utilization, has been determined resulting in greater variations in the voltage of a single cell in the first stack in comparison to the corresponding values in the second stack.

This paper describes a miniaturized model of a small size solar-hydrogen power installation with a reserve electricity source in the form of a fuel cell. The cell was powered with hydrogen generated in the electrolyzer in time periods when the photovoltaic panel works in suitable insolation conditions. Hydrogen was stored in a special tank, containing hydride releasing hydrogen in convertible reactions. The tests shown were performed in the summer and in various weather conditions. Conclusions confirmed the usefulness of the model for the design of full sized installations dedicated to summer cottages and similar objects. Modifications of the model were proposed with the aim to obtain quantitative test results needed for designing more efficient installations.
W artykule przedstawiono miniaturowy model malej elektrowni stoneczno-wodorowej, w której zastosowano rezerwowe źródło zasilania prądem elektrycznym w postaci ogniwa paliwowego. Ogniwo zasilane jest wodorem wytwarzanym w elektrolizerze w okresach pracy ogniw fotowoltaicznych przy wystarczających warunkach nasłonecznienia. Wodór magazynowany jest w specjalnym zbiorniku zawierającym wodorki uwalniające wodór w procesie odwracalnym. Przedstawiono przebieg testów, którym poddano model w okresie letnim, przy zróżnicowanych warunkach pogodowych. We wnioskach potwierdzono przydatność modelu przy projektowaniu pełnowymiarowych instalacji dla obiektów letniskowych. Zaproponowano również jego modyfikacje w celu uzyskania ilościowych wyników testów, przydatnych przy projektowaniu bardziej wydajnych instalacji.

Solar energy is widely available in nature and electricity can be easily extracted using solar PV cells. A fuel cell being reliable and environment friendly becomes a good choice for the backup so as to compensate for continuously varying solar irradiation. This paper presents simple control schemes for power management of the DC microgrid consisting of PV modules and fuel cell as energy sources and a hydrogen electrolyzer system for storing the excess power generated. The supercapacitor bank is used as a short term energy storage device for providing the energy buffer whenever sudden fluctuations occur in the input power and the load demand. A new power control strategy is developed for a hydrogen storage system. The performance of the system is assessed with and without the supercapacitor bank and the results are compared. A comparative study of the voltage regulation of the microgrid is presented with the controller of the supercapacitor bank, realized using a traditional PI controller and an intelligent fuzzy logic controller.

Opis:
Research about exploitation the potential of waste and sludge increased drastically in the recent years. One of the most promising alternative methods of waste management is Microbial Fuel Cell (MFC), which generate clean bio-electricity using microorganisms. Organic compounds, sewage, municipal solid waste could be used as a source for microbial nutrition. The construction of MFC is one of the most important parameter in laboratory studies and during scale-up. The efficiency of MFC depends on many factors including type of membrane. To obtain optimization in terms of various operating conditions, a prototype of Microbial Fuel Cell with exchangeable membrane was projected and fabricated by additive manufacturing (AM) technology. This novel device allows to research effects of different types of separator membranes. Preliminary research showed possibility to produce 3D printed MFC systems.
