Opis:: Immunosenescence is viewed as a remodeling process with the exhaustion of naïve T cells and filling up of the immunological space with memory cells. In this study some phenotypic changes of CD8+ human cells during in vivo ageing were compared with those observed in long term cultures of lymphocytes derived from cord blood or from peripheral blood from donors of different age. Both in vivo and in vitro a significant decrease of the fraction of CD8+CD28+ cells was observed. Comparing the proportions of other T cell subpopulations (the CD4/CD8+ ratio, CD56, CD57, CD27) made it possible to conclude that replicative senescence in vitro partially reflects in vivo ageing.

Opis:: An interesting field of research in tumor immunology are T-lymphocytes and NK-cells in the area directly adjacent to tumor, i.e. tumor-infiltrating lymphocytes (TIL). An important indicator of immune control of cancer cells is the correlation between intensity of TIL infiltrate and clinical course of the disease. The aim of this paper is a review of particular types of immunocompetent cell infiltrating the tumor and a review of English literature as to data concerning prognostic significance of TIL subpopulations in ovarian, endometrial, cervical and vulvar cancer. Material and method: the PubMed database was searched using key words: CD8+, CD4+, FOXP3+, CD57+, CD56+, NK, combined with phrases: cervical cancer, vulvar cancer, ovarian cancer, endometrial cancer and prognosis. Results: Most important TILs include cytotoxic T-lymphocytes, mostly represented by the CD8+ population, helper T-lymphocytes, designed as CD4+ cells, regulatory lymphocytes Treg, where the most specific maker is transcription regulator forkhead box P3(FOXP3), and NK/NKT cells (naturalkiller/natural killer T-lymphocytes), identified by CD56 and CD57. A close correlation was found between patients’ survival and intensity of infiltrate by CD8+ cells and indicators CD8+/FOXP3+, CD8+/CD4+ and ovarian cancer, endometrial cancer and cervical cancer. An exception to this is vulvar cancer, where CD8+, CD4+ and FOXP3+ have no prognostic significance. Conclusions: Severity of local cytolysis of tumors and inhibition of immunosuppressive activity of regulator cells may be used in future therapeutic strategies in patients with ovarian, endometrial and cervical cancer. In vulvar cancer we should rather focus on non-specific immune response for tumor control.
W immunologii nowotworów szczególną uwagę zwraca się na limfocyty T i komórki NK z bezpośredniego otoczenia guza, tzw. limfocyty naciekające guz, czyli komórki TILs (tumor-infiltrating lymphocytes). Ważnym dowodem na nadzór układu odpornościowego nad komórkami raka jest korelacja pomiędzy intensywnością nacieków komórek TILs a przebiegiem choroby. Celem pracy było scharakteryzowanie poszczególnych typów komórek immunokompetentnych, które mogą uczestniczyć w naciekaniu guzów nowotworowych, oraz przeanalizowanie publikacji anglojęzycznych pod kątem danych dotyczących znaczenia prognostycznego subpopulacji TILs w raku jajnika, trzonu macicy, szyjki oraz sromu. Materiał i metoda: Baza PubMed została przeszukana przy użyciu słów kluczowych: CD8+, CD4+, FOXP3+, CD57+, CD56+, NK w połączeniu z frazami: cervical cancer, vulvar cancer, ovarian cancer, endometrial cancer oraz ze słowem prognosis. Wyniki: Do najważniejszych TILs należą cytotoksyczne limfocyty T, w większości reprezentowane w badaniach przez populację komórek CD8+, pomocnicze limfocyty T, oznaczane jako komórki CD4+, limfocyty regulatorowe Tregs, dla których najbardziej specyficznym markerem jest regulator transkrypcji forkhead box P3 (FOXP3), oraz komórki NK/NKT (natural killer/natural killer T lymphocytes), identyfikowane za pomocą CD56 i CD57. W raku jajnika, endometrium oraz szyjki macicy wykazano silny związek pomiędzy czasem przeżycia pacjentów a intensywnością nacieków z komórek CD8+ oraz wskaźnikami CD8+/FOXP3+, CD8+/CD4+. Wyjątek stanowi rak sromu, w którym komórki CD8+, CD4+, FOXP3+ nie posiadały znaczenia prognostycznego. Wnioski: Nasilanie lokalnej cytolizy guzów, a także hamowanie immunosupresyjnego działania komórek regulatorowych może być w przyszłości wykorzystane w strategiach terapeutycznych w raku jajnika, endometrium oraz szyjki macicy. W raku sromu należy ocenić udział komórek nieswoistej odpowiedzi immunologicznej w zwalczaniu guzów nowotworowych.

Opis:: Normal human T lymphocytes growing in culture undergo replicative senescence. Previously, we have shown that in our conditions polyclonal T cells cease proliferation after about three weeks (Radziszewska et al., 1999, Cell Biol. Int. 23, 97-103). Now we present results of a more detailed analysis of in vitro growth as well as phenotypic changes of T cells. Cell cycle analysis showed that about 20% of cells were in the S phase untill the 17th day of culture (young cells). The highest number of mitotic cells (phase G2/M; 10%) was observed during the first week of culture. All not dividing senescent cells were stopped in the G1 phase (after the 30th day of culture). The sub-G1 fraction which represents apoptotic cells did not exceed 8% during the whole period until the 30th day of culture. During in vitro T-cell growth, a rather rapid selection to CD3+CD8+ cells occurs. In the presenescent (between the 17th and 30th day) and senescent populations the majority of cells (above 90%) were CD8 positive. We also have checked the expression of α-chain interleukin-2 (IL-2) receptor (CD25). In young and presenescent cells about one third of cells was CD25 positive, but only 15% in the pool of senescent cells. Immunoblotting analysis of p16 protein recognized previously as a marker of senescent T cells, showed its highest and transient expression in presenescent cells. A critical review of the polyclonal T cell replicative senescence model is presented.

Opis:: α1,6-fucosyltransferase (FUT8) attaches fucose residues via an α1,6 linkage to the innermost N-acetylglucosamine residue of N-linked glycans. Glycans with this type of structure are present in GpIIb/GpIIIa complex (CD41a) which is present on megakaryocytes (Mks) and platelets. CD41a is the earliest marker of megakaryocytopoiesis. The aim of this study was to analyse the morphology, phenotype, ploidy level and activity of FUT8 during induced differentiation/maturation of Mk progenitor cells in ex vivo culture. We used SU6656, a selective inhibitor of Src tyrosine kinases, as differentiation-inducing agent for Mks. The addition of SU6656 to the culture system of megakaryocytic progenitors from cord blood CD34+ cells and Meg-01 cell line induced their maturation towards later stages of Mk differentiation with increased activity of FUT8. We suggest FUT8 as a candidate for an early marker of differentiation and possibly of the ploidy level of Mks. We confirm a special status of FUT8 in megakaryocytopoiesis.

Worm expulsion in intestine was faster than in the control (opposite results in experiment II). The composition of cells infiltration in the muscle was in both experiments similar to the control, however, fewer Cn4⁺ lymphocytes were observed in larva capsule and there were fewer muscle larvae. Therefore CD8⁺ cells are believed to take part in restricting only the muscle stage of trichinellosis.

Opis:: INTRODUCE The aim of the study was to determinate the effect of low X-ray doses on the percentage and number of peripheral blood lymphocytes T (CD3+), T helper (CD4+), T cytotoxic (CD8+), lymphocytes B (CD19+), activated T lymphocytes (CD3+CD25+) and activated T helper lymphocytes (CD4+CD25+) as well as serum immunoglobulins (IgM, IgG and IgA) concentration in workers of an interventional cardiology ward. MATERIAL AND METHODS The study was performed among a group of 40 workers (28 women and 12 men) and a control group of 35 persons (25 women and 10 men). Flow cytometry was used for the analysis of lymphocytes and the turbidimetric method for assessing serum immunoglobulin concentrations. RESULTS The study showed a decreased percentage and number of CD3+CD25+ and CD4+CD25+ cells as well as IgG concentration in interventional cardiology ward staff in comparison to the control group. A negative correlation between the length of employment and percentage of CD3+CD25+ was found. CONCLUSION The study indicates suppressive impact low doses of ionizing radiation on activated T cells, activated T helper cells as well as IgG production that is adverse in relation to the existing risk of infections caused by different microorganisms.
WSTĘP Celem pracy była ocena oddziaływania niskich dawek promieniowania jonizującego na odsetek i liczbę limfocytów T (CD3+), limfocytów T pomocniczych (CD4+), limfocytów T cytotoksyczych (CD8+), limfocytów B (CD19+), aktywowanych limfocytów T (CD3+CD25+) i aktywowanych limfocytów T pomocniczych (CD4+CD25+) oraz stężeń immunoglobulin IgM, IgG i IgA u pracow-ników oddziału kardiologii interwencyjnej. MATERIAŁ I METODY Badanie przeprowadzono w grupie 40 zatrudnionych (28 kobiet i 12 mężczyzn) oraz w grupie kontrolnej 35 osób (25 kobiet i 10 mężczyzn). Badania limfocytów krwi obwodowej przeprowadzono z wykorzystaniem cytometrii przepływowej, a stężenia immunoglobulin w surowicy oznaczono metodą turbidymetryczną. WYNIKI W badanej grupie pracowników – w porównaniu z grupą kontrolną – wykazano znamienne obniżenie liczby i odsetka limfocytów CD3+CD25+ i CD4+CD25+ oraz stężeń immunoglobuliny IgG. Wykazano także istnienie negatywnej korelacji między okresem zatrudnienia badanych pracowników a odsetkiem limfocytów CD3+CD25+. WNIOSEK Badanie wskazuje na supresyjne oddziaływanie niskich dawek promieniowania jonizującego na aktywowane limfocyty T, aktywowane limfocyty T pomocnicze i produkcję IgG, co nie jest korzystne w odniesieniu do istniejącego ryzyka zakażeń drobnoustrojami.

Opis:: Cytotoxic CD8+ cells play an important role in determining host response to tumor, thus chemotherapy is potentially dangerous as it may lead to T cells depletion. The purpose of this study was to elucidate the propensity of quiescent and proliferating human CD8+ cells to undergo cell death upon treatment with curcumin, a natural dye in Phase I of clinical trials as a prospective chemopreventive agent. Methods: We treated human quiescent or proliferating CD8+ cells with 50 µM curcumin or irradiated them with UVC. Cell death symptoms such as decreased cell viability, chromatin condensation, activation of caspase-3 and specific DFF40/CAD endonuclease and oligonucleosomal DNA fragmentation were analyzed using MTT test, microscopic observation, Western blotting and flow cytometry. Results: Curcumin decreased cell viability, activated caspase-3 and decreased the level of DFF45/ICAD, the inhibitor of the DFF40/CAD endonuclease. However, this did not lead to oligonucleosomal DNA degradation. In contrast, UVC-irradiated proliferating, but not quiescent CD8+ cells revealed molecular and morphological changes characteristic for apoptosis, including oligonucleosomal DNA fragmentation. Curcumin can induce cell death in normal human lymphocytes both quiescent and proliferating, without oligonucleosomal DNA degradation which is considered as a main hallmark of apoptotic cell death. Taking into account the role of CD8+ cells in tumor response, their depletion during chemotherapy could be particularly undesirable.

Opis:: Cyclodextrin (CD) is a cyclic compound with a spatial structure in the shape of a toroid. It is characterized by specific properties. The outer portion of the structure has hydrophilic properties, while the cavity of the toroid is hydrophobic. This enables cyclodextrin to form inclusion complexes with the enclosure of lipophilic molecules in the interior. This paper presents the results of attempts to form a complex of γ-CD/CL-20. This study determined unambiguously that a γ-CD with CL-20 complex in a mole ratio 1:1 is formed as a result of mixing of solutions of γ-CD and CL-20. This conclusion was corroborated by FTIR, 1H NMR, UV-Vis spectroscopic techniques and by measurement of the density of the complex obtained. The resulting complex of γ-CD/CL-20 is characterized by much lower sensitivity to friction and impact than CL-20 itself.

Opis:: The aim of the study was to determine whether treatment with recommended doses of meloxicam or flunixin had an effect on the apoptosis of peripheral blood T lymphocytes in calves. The study was carried out on 4-5 months old calves (n = 24, 8 per group). Experimental animals were injected subcutaneously with a single dose of 0.5 mg . kg-1 of meloxicam or intravenously with 3 doses of 2.2 mg . kg-1 day-1 of flunixin. The non-treatment animals served as control. Blood samples were taken at day 0 and at days 1, 2, 3, 5, 7 and 14 after the first NSAIDs injection. Apoptosis was determined by flow cytometry using Annexin V-PE/7-AAD staining. The kinetic analysis of apoptosis in the total lymphocyte population, as well as in the CD4+ and CD8+ subsets did not reveal significant differences in the frequency of early apoptotic cells between control and experimental groups throughout the period studied. Although, 24 h after administration of the first dose of NSAIDs, late-stage apoptosis/necrosis was significantly increased in the total lymphocyte population (the meloxicam group), as well as in the CD4+ (the meloxicam group and the flunixin group) and CD8+ (the flunixin group) subsets of T cells. However, this disturbance was transient, relatively poorly expressed and, thus, unlikely to be of clinical significance. Our results indicate that the use of meloxicam or flunixin in accordance with the recommended dosage regimen in cattle do not have a clinically significant influence on apoptosis of peripheral blood T cells.

Opis:: Introduction. Results of studies analyzing the role of immunocompetent cells in tumor environment and whole peripheral blood indicate their responsibility for aggressiveness of neoplasm, prognosis and therapeutic effect. Atcivation of lymhocytes T is connected with expression the markers (antigens) on their surface. The aim of this study was the analysis of activation antigens expression on lymphocytes T in patients with laryngeal carcinoma and the connection with clinicomorphological features. Material and methods. Analysis of activation antigens expression CD69, CD71 and CD25, CD26, HLA/DR on lymphocytes T CD4+ i CD8+ in 33 patients with squamous cell carcinoma of the larynx was performed. Flow cytometry-based analysis of activation antigens in T cell cultures with and without PHA stimulation was used. The connection of these molecules and clinicomorphological features was examined (pT, pN, G, Anneroth, Batsakis and Lunas’ classifi cation). Results. The significant correlation between chosen markers of activation and tumor features were noted: pT with HLA/DR/CD4, CD69CD8, CD71CD8, pN with CD26CD8, G with CD25CD8, CD71CD8, ABL score with CD25CD4. Conclusion. Our data indicated the connection of immunocompetent cell activity and spread of neoplasm in patients with laryngeal carcinoma.
