Opis:
Cancer is a widespread and lethal disease. It is considered as the first leading cause of deaths in economically developed countries. Nonetheless, the search for an effective treatment of cancer is still a major challenge. In this review, we analyzed the core structure of compounds and their impact on antitumor activity. Diversity of heterocyclic ring in the innovatory anticancer substances showed the potential in the design and development of new anticancer drugs.

Opis:
Chronic exposure to environmental ultraviolet radiation (UVR) plays a key role both in photocarcinogenesis and induction of accelerated skin ageing. Sailors commonly experience a significant number of cutaneous problems, related to the exposure to environmental factors due to their working conditions. Among these factors, sun exposure is able to determine both acute and chronic skin damage, mostly linked to the effects of the ultraviolet (UV) radiation on epidermal and dermal structures. We report a case of sailor with cutis rhomboidalis nuchae, solar elastosis, actinic keratosis (AK), numerous squamous cell carcinoma (SCC) and basal cell carcinoma (BCC) of the head and trunk.

Squamous cell carcinoma of the pinna occurs primarily in older males who are exposed to sunlight for a prolonged period. Most squamous cell carcinomas of the pinna are well differentiated and present as a painless mass or a sessile nodule. We describe an interesting case of an adult woman with a non-healing wound located on the right pinna which was treated conservatively, and was later diagnosed as squamous cell carcinoma.

Cabozantinib, the latest available in Poland medication for the treatment of renal cell carcinoma, registered in this indication by the European Medicines Agency (EMA) in September 2016, has been available in several cancer centers in Poland since November 2016 as part of the expanded access program. Primary hypothyroidism is a common complication during thyrosine kinase inhibitors (TKI) treatment, although there are few reports of its occurrence during treatment with cabozantinib, which belongs to this medication group. We present a case of rapid development of clinically apparent hypothyroidism after cabozantinib treatment and report data on this complication in the group of our patients.

Objective: Results of second-line everolimus treatment in clear cell renal carcinoma. Material and methods: Clinical data of 32 patients of the Clinic of Systemic and Metastatic Malignancies, Centre of Oncology, Maria Sklodowska-Curie Memorial Institute (Krakow Branch) treated with second-line everolimus between 2011 and 2016 were analysed. Results: Median progression-free survival was 7 months. Stable disease was observed in 24 cases. Adverse events occurred in 59% of all patients, and the majority were mild and moderate (G1 and G2) according to CTCAE. The most frequent adverse events included anaemia, stomatitis, rash and fatigue. Conclusion: Molecularly targeted drugs offer the only effective and available therapy in Poland, which can be administered in patients with advanced renal cell carcinoma. Second-line everolimus is a valuable therapeutic option in the above mentioned group (following progression on first-line TKI therapy), especially when it is necessary to avoid the accumulation of similar adverse events after TKI treatment (different toxicity profiles).

Opis:
Merkel cell carcinoma (MCC) is a rare cutaneous malignancy, with aggressive behaviour. The discovery of MCC polyomavirus (MCPyV) provided a major insight into pathogenesis of MCC, as MCPyV is present in most MCC tumours. The primary treatment of locoregional MCC is surgery, and sentinel lymph node biopsy should be performed in all cases at clinical stage I-II. Adjuvant radiotherapy decreases disease recurrences in stage I-III of the disease. Chemotherapy is primarily used for palliation and provides overall response rates of approximately 70%; however, the disease often recurs within a few months. All cases of MCC should be managed in an experienced multidisciplinary setting.

Opis:
Skin cancers are one of the most common cancers in the Caucasian population. A constantly increasing number of nonmelanoma skin cancers and malignant melanomas is observed. The incidence of skin cancers is associated mainly with exposure to sunlight. Therefore, agricultural workers who work in open spaces are a particularly vulnerable group. Currently, studies on the pathogenesis of skin cancer focus on the molecular basis associated with ultraviolet radiation. This study is an attempt to summarize the current state of knowledge on this issue. There have been demonstrated mutations in different classes of genes associated with carcinogenesis, including protooncogenes, tumour suppressor genes, genes that control apoptosis, genes encoding transcription factors and DNA repair genes in patients with skin cancers. Mutations in the latter result in reducing the effectiveness of DNA repair and fixation of mutations. All changes at the gene level lead to structural changes, quantitative and dysfunction of proteins encoded by these genes. All these factors contribute to the process of carcinogenesis. Due to increasing number of skin cancers, it seems important to increase knowledge of the molecular basis of skin cancers. This knowledge could be crucial for predicting the course of the disease, and for the development of new therapeutic strategies.

Introduction. Clear cell carcinoma, not otherwise specified/hyalinising clear cell carcinoma of the salivary gland (HCCC) is a malignancy that arises in minor salivary glands. It rarely leads to distant metastases or cancer-related death but has the potential for recurrence and focal metastases. Aim. A case is reported. Description of the case. A 72 years old female patient has reported to the Clinic of Otolaryngology with a tongue lesion. The patient had no history of malignancy. The lymph node has been surgically removed for further examination. Cords and nests of clear cells and cells with eosinophilic cytoplasm in a hyalinized stroma were identified within the lymph node. After the diagnosis the patient has been transferred to another Oncology Hospital for further treatment. Conclusion. The diagnosis of clear cell carcinoma may be challenging because many of it‘s features frequently overlap with other salivary gland lesions.

Opis:
Introduction. The most common malignancy of the biliary tract is gall bladder carcinoma and the main subtype according to the histological classification is Adenocarcinoma. Pure squamous cell carcinoma of the gall bladder is very rare entity accounting for only 1.1-3.7% of the gall bladder carcinomas. It is highly malignant with poor prognosis due to high proliferative rate and local invasiveness to the adjacent organs. The patients are usually diagnosed at an advanced stage with a bulky tumor owing to its aggressive behavior. Aim. In this paper, we describe a female patient with primary pure squamous cell carcinoma of the gall bladder. Description of the case. A 42-year old female patient presented with chief complaints of pain in abdomen associated with nausea and vomiting and gradually progressive jaundice since 02 months. Contrast Enhancing Computed Tomography (CECT) abdomen showed an enhancing mass lesion in gall bladder involving adjacent organs for which she underwent extended cholecystectomy with pancreaticoduodenectomy. Conclusion. Diagnosis as well as the management of this exceptionally rare type of tumour is undoubtedly challenging because of non-specific clinical as well as imaging findings. This case report is an attempt to add to the literary evidence for better pathological as well as clinical understanding of this rare and aggressive entity thereby providing additional material for the early diagnosis as well as the development of effective targeted therapies which will certainly help in increasing the lifespan of these patients.

Opis:
Background: Clear cell carcinoma in scars after cesarean section is extremely rare, with only 22 cases reported in the literature. Management of this condition needs to be further explored. Here, we report of a patient with clear cell carcinoma of the abdominal wall that developed 35 years after cesarean section. Case Report: The material of the study was a group of 61 patients divided into two groups. Group I – 35 deaf or with profound sensorineural hearing loss children (the pupils of the deaf and hard of hearing school), aged 5–17 years (average 9,2 years), 14 males, 21 females, II – control group comprised 26 normal hearing patients, aged 5–16 years (average 10,4 years), 14 males, 12 females (patients of Department of Pediatric Otolaryngology, Audiology and Phoniatrics, Medical University of Lodz). In both groups, exon 2 sequencing of GJB2 gene was performed. Results. A 58-year-old woman was admitted to our department due to abdominal pain and a progressively growing mass in the abdominal wall. Based on biopsy, a preliminary diagnosis of clear cell carcinoma was made. A wide surgical excision of the tumor with clear margins, hysterectomy with bilateral salpingo-oophorectomy, and abdominal wall reconstruction using synthetic mesh were performed. The patient was discharged in good condition after fifteen days of hospitalization. The patient remained recurrence-free 6 months after the treatment. Conclusions: T Lack of standardized management of rare malignant transformations hinders patient care. Due to a growing number of cesarean deliveries, we can expect clear cell carcinoma prevalence of the abdominal wall to increase. Therefore, patients and clinicians should attend to any pain, itching, or change in the size of abdominal wall scars.
