Opis:
Multidrug resistance-associated protein (MRP1) is a transmembrane pump protein responsible for the efflux of chemotherapeutic drugs, an important cause of anticancer treatment failure. Trying to circumvent MRP-mediated resistance we designed and synthesized hairpin loops forming antisense oligodeoxyribonucleotides (ODNs), both phosphodiesters (PO-ODNs) and their phosphorothioate analogues (PS-ODNs), to reduce the protein expression by targeting its mRNA in a sequence specific manner. Melting temperature measurements as well as polyacrylamide gel electrophoresis supported the preferential formation of a secondary structure, which was expected to protect ODNs against 3'-exonuclease degradation. ODNs and PS-ODNs designed in this work were successfully tested as antisense inhibitors of the expression of MRP1 in the leukaemia HL60/ADR cell line. Foreseeing the necessity to perform clinical studies with such ODNs we investigated their stability against the 3'-exonuclease activity of fetal calf serum and human plasma. Under the conditions, corresponding to physiological ones, we observed high stability of hairpin loop forming ODNs, especially those containing longer (e.g. 7 base pair) stems. Comparative studies on the stability of chemically unmodified hairpin loop forming ODNs and their PS-counterparts indicated that endonuclease activity did not play any important role in the process of their nucleolytic degradation. Our studies provide strong evidence for high stability of chemically unmodified hairpin loop ODNs, making them an attractive alternative to phosphorothioate analogues commonly used in antisense strategy.

Opis:
Bovine neonatal pancytopenia (BNP), a newly emerged syndrome of discussed etiology in calves, has been diagnosed since 2006. Here we describe first cases of BNP in Poland. Between September 2008 and April 2011, 62 cases of BNP were diagnosed in dairy calves. Bleeding skin lesions were mostly pronounced in summer and early autumn. Severe thrombocytopenia was observed in all sick animals. All calves came from herds vaccinated against BVDV infection with PregSure BVD vaccine (Pfizer). Substitution of colostrum from dams of BNP positive calves with colostrum from dams from herds free of BNP was the only effective measure to avoid new cases in affected herds.

Comparison of calving course of Charolaise purebreds and their crossbreeds with Polish Holstein-Frisian cows. Calving course of cows of Charolaise breed covered by bulls of the same breed as well as cows of Polish Holstein Friesian breed inseminated with Charolaise bull’s semen was investigated. In purebred population the material was taken from the beef cattle recording scheme provided by the Polish Association of Beef Cattle Breeders and Producers. In case of commercial crossing the material was taken from the questionnaires collected by the technicians representing Mazovian Centre of Animal Breeding and Reproduction in Łowicz. The data base covered calving course degree and calf body weight at birth degree. It was proved, that the significantly higher ratio of deliveries when human help was needed was stated in the purebred Charolaise population (24.5%), in comparison to crossbreeding where the above mentioned ratio was only 12.2%. The most difficult calvings were observed for the calves of the highest birth weight. The high ratio of assisted calvings in commercial crossing as well as in purebred population of Charolaises suggest that all of the deliveries should be monitored by the breeders. It should be also stated that the ratio of difficult calvings in the commercial crossing is significantly lower than that observed in purebred PHF population. Because of so low ratio of assisted calvings in commercial crossing with Charolaise bulls it should be recommended to use the semen of bulls which pedigree show high body weight at birth and muscularity.
Porównanie przebiegu porodu u krów rasy charolaise i jej mieszańców z bydłem rasy polskiej holsztyńsko-fryzyjskiej. W pracy oceniono przebieg porodów 98 krów rasy charolaise krytych buhajami tej samej rasy oraz 123 krów rasy polskiej holsztyńsko-fryzyjskiej odmiany czarno-białej (phf) krytych w krzyżowaniu towarowym z buhajami mięsnej rasy charolaise. W hodowli czystorasowej materiałem do badań były wyniki oceny użytkowości bydła mięsnego prowadzonej przez Polski Związek Hodowców i Producentów Bydła Mięsnego. W przypadku krzyżowania towarowego materiałem do badań były dane zawarte w „Kartach przebiegu ocielenia krowy” prowadzonych przez specjalistów Mazowieckiego Centrum Hodowli i Rozrodu Zwierząt Sp. z o.o. w Łowiczu. Dane dotyczące 98 porodów krów rasy charolaise inseminowanych nasieniem buhajów tej samej rasy oraz 123 porodów krów phf inseminowanych nasieniem buhajów mięsnej rasy charolaise obejmowały ocenę przebiegu porodu oraz masę cielęcia przy urodzeniu. Uzyskane wyniki jednoznacznie pokazują, że znacznie wyższy i statystycznie istotny udział porodów wymagających pomocy człowieka stwierdzono w populacji czystorasowej bydła charolaise (24,5%), w porównaniu z krzyżowaniem towarowym krów mlecznych z buhajami tej rasy, gdzie odsetek trudnych ocieleń wyniósł zaledwie 12,2%. Analiza statystyczna wykazała istotny wpływ masy cielęcia na rozkład ocen przebiegu porodu w obu badanych populacjach. Najwięcej trudnych porodów zanotowano w grupie cieląt o najwyższej masie przy urodzeniu. Stosunkowo wysoki odsetek porodów wymagających udziału człowieka zarówno w krzyżowaniu towarowym, jak i w populacji czystorasowej charolaise, sugeruje potrzebę monitorowania przez hodowcę wszystkich porodów. Należy podkreślić, że odsetek przypadków trudnych ocieleń w krzyżowaniu towarowym z rasą charolaise jest znacznie mniejszy z ich częstością, jaką notuje się u bydła phf utrzymywanego w czystości rasy.

The review was conducted from studies/reports of the performance of Holstein Friesian routinely kept by Heifer Project International (HPI) Farmer groups on imported Holstein Friesian herds on zero grazing in the western highland region of Cameroon. The review provides insight to the reports of the studied area, its climatic conditions, management of the animals and the methodology adopted by the researchers. Their findings and recommendations were also highlighted in order to inform policy makers and agencies with the responsibility to design programmes for rural development and youth empowment in tropical countries, the potentials of this technology to alleviate poverty, hunger, rural-urban migration and unemployment. The mean age at first calving was higher compared to other reports. The additive genetic variance of the sire that sired the dam of the heifer, the breeding techniques of maintaining a central bull for several farmers and other management techniques employed elongated age at first calving. Calving interval and calf birth weight were appreciable, though longer and lower respectively compared to existing reports. They were greatly affected by the management techniques employed by the farmers. The mean gestation length was low despite the poor management indicating that, this breed with improved management can be integrated into the tropical conditions. The percentage calf mortalities were very high. It varied across herd, indicating that there was no standard routine management practice adopted at the herds locations. The herd with better management recorded lower calf mortality. The effect of calf mortality rate on calf crop and selection for genetic improvement were worse and also varied across herds. In some herds, all the heifers produced must be retained to maintain herd size. In other herds the cow must calve many times before it is able to produce a heifer, that would replace it and lactate in the herd. The higher the number of calving that produced a lactating heifer, the longer the generation interval, and the lower the genetic progress achieved through selection when generation intervals are elongated. The genotypic diversity created through independent assortment, segregation and recombination of genes as random effects through gametogenesis and fertilization, respectively were lost to calf mortality and never presented to the environmental challenges of the tropics for selection and adaptation. Superior genotypes that could have tolerated the tropical condition were never exposed to build physiological mechanism that would help them cope with tropical stress. The few genotypes that survived, were introduced into the stressful conditions of the tropics often too early and sudden with no time to allow them develop and build their survival techniques to enhance their adaptation in the tropics. There were high incidences of disease prevalence which also varied across herds, locations, type of bedding materials used and floor type in the housing. There was no housing design directed towards herd health as participating farmers erect structures they can afford. This was why calf mortality rate were very high. The calves are being introduced into an already diseased hazard environment without adequate time to develop their own defence mechanisms. The survival of some calves, reproductive performance of the herd and their lactation thus indicate that, this breed can be integrated into the tropical conditions. There is however, need for interested countries and agencies to design a low cost housing type and insist that, their potential beneficiaries must provide this housing and adequate feed from the zero grazing plots first before they acquire their animals. Mud floor must be replaced with laterite floor for easy drainage of urine and water. This will improve herd health, introduce calves into the challenging tropical stress gradually and allow them enough time to build their body defence against prevalent diseases. There is also the need to reduce the number of farmers attached to a central bull, to ensure that all heifers attached to a bull have successful services.

The aim of the study was the analysis of the effect of calving season on the course of parturition and rearing results of Polish Holstein-Friesian calves of Black-and-White strain from birth to 3 months of age. Easy calvings (1;2) predominated on the farm. In spring, they accounted for 91.2% of all calvings occurring in this season. The respective values for summer, winter and autumn were 89.7%, 88.6% and 87.6%. The highest mean birth weight was characteristic of calves born in winter (40.8 kg and 44.2 kg for heifer calves and bull calves, respectively). The highest mean daily body weight gains in the whole rearing period were achieved by heifer calves born in winter (704 g) and bull calves born in spring (750 g). In the group of heifer calves and bull calves, statistically significant differences during the analysed rearing period were found (P≤0.01, P≤0.05). The highest percentage of ill calves was found in the autumn-winter season and the lowest one in spring and summer.
Celem pracy była analiza wpływu sezonu urodzenia na przebieg porodu i wyniki odchowu cieląt rasy polskiej holsztyńsko-fryzyjskiej odmiany czarno-białej od urodzenia do 3. miesiąca życia. W gospodarstwie przeważały porody łatwe (1;2).Wiosną stanowiły one 91,2% wszystkich porodów przypadających na ten sezon, latem 89,7%, zimą 88,6%, zaś jesienią 87,6%. Największą średnią masą ciała po urodzeniu charakteryzowały się cielęta z porodów zimowych (cieliczki 40,8 kg; buhajki 44,2 kg). Największe średnie przyrosty dobowe w całym okresie odchowu osiągnęły cieliczki urodzone zimą (704 g) i buhajki wiosną (750 g).W grupie cieliczek jak i buhajków stwierdzono statystycznie istotne różnice w analizowanym okresie odchowu (P≤0,01, P≤0,05). Największy procent cieląt chorych stwierdzono w okresie jesienno-zimowym, najmniejszy wiosną i latem.

Opis:
Twenty-two elements have been identified as essential to the growth and health of animals. They include 7 macroelements and 15 microelements, which play four key functions in the body: structural, physiological, catalytic and regulatory. Mineral deficiencies can result from low quality feed, impaired absorption or assimilation in the body or increased demand for minerals during intensive growth, pregnancy and lactation. Mineral-deficient feed and diets with an unbalanced mineral content impair the growth and development of young animals, decrease appetite, lower nutrient absorption, decrease immunity and increase susceptibility to contagious diseases. This paper discusses the consequences of low levels of macronutrients and micronutrients that are required for the optimal growth of calves, lambs and kids, including calcium, phosphorus, magnesium, selenium, cobalt, iron, zinc, copper, sodium, potassium and chloride.
Przyjmuje się, że 22 pierwiastki są niezbędne do prawidłowego funkcjonowania organizmów zwierzęcych. Należy do nich 7 makro- i 15 mikroelementów, które w organizmie pełnią 4 zasadnicze funkcje: strukturalną, fizjologiczną, katalityczną oraz regulacyjną. Przyczyną niedoboru pierwiastków mineralnych może być niewłaściwa jakość paszy, nieodpowiednia absorpcja lub asymilacja pierwiastków w organizmie lub zwiększone zapotrzebowanie zwierzęcia na składniki mineralne (okres wzrostu, ciąża, karmienie potomstwa). Niedobór pierwiastków mineralnych w paszy lub ich nieodpowiedni stosunek przyczynia się w głównej mierze do: zahamowania wzrostu i rozwoju młodych zwierząt, zmniejszenia apetytu oraz gorszego wykorzystania składników pokarmowych lub spadku odporności i większej zapadalności na choroby zakaźne. Celem artykułu jest omówienie skutków niedoboru najbardziej istotnych makro- i mikroelementów, takich jak: wapń, fosfor, magnez, selen, kobalt, żelazo, cynk, miedź, sód, potas oraz chlor, niezbędnych do prawidłowego rozwoju cieląt, jagniąt i koźląt.

The aim of the study was to determine whether treatment with recommended doses of meloxicam or flunixin had an effect on the apoptosis of peripheral blood T lymphocytes in calves. The study was carried out on 4-5 months old calves (n = 24, 8 per group). Experimental animals were injected subcutaneously with a single dose of 0.5 mg . kg-1 of meloxicam or intravenously with 3 doses of 2.2 mg . kg-1 day-1 of flunixin. The non-treatment animals served as control. Blood samples were taken at day 0 and at days 1, 2, 3, 5, 7 and 14 after the first NSAIDs injection. Apoptosis was determined by flow cytometry using Annexin V-PE/7-AAD staining. The kinetic analysis of apoptosis in the total lymphocyte population, as well as in the CD4+ and CD8+ subsets did not reveal significant differences in the frequency of early apoptotic cells between control and experimental groups throughout the period studied. Although, 24 h after administration of the first dose of NSAIDs, late-stage apoptosis/necrosis was significantly increased in the total lymphocyte population (the meloxicam group), as well as in the CD4+ (the meloxicam group and the flunixin group) and CD8+ (the flunixin group) subsets of T cells. However, this disturbance was transient, relatively poorly expressed and, thus, unlikely to be of clinical significance. Our results indicate that the use of meloxicam or flunixin in accordance with the recommended dosage regimen in cattle do not have a clinically significant influence on apoptosis of peripheral blood T cells.

Blastocystis spp. is a parasite that causes intestinal infection in humans and other animals. A few studies have been performed in Turkey on the distribution of Blastocystis in cattle. In this study, fecal samples were collected from 100 calves and subjected to analysis based on an SSU rRNA gene fragment. The overall prevalence of the disease was determined as 15% (15/100). This rate was 14.04% for females and 16.28% for males. In addition, three Blastocystis subtypes were identified: ST10, ST14, and novel subtypes ST25. To our knowledge, the ST25 subtype was reported with this study for the first time in Turkey. The nucleotide sequences (OM920832-OM920839) obtained in this study were deposited in GenBank. The results obtained will be useful for a better understanding of the epidemiology of Blastocystis spp., and its effects on public health.

Opis:
The aim of this study has been to determine the effect of a diet containing β-carotene and omega-3 fatty acids on the biochemical and nonspecific humoral immunity indicators and on the results of rearing calves to day 60 of life. The study was conducted in 2011-2012, on a herd of Polish Holstein-Friesian cows. 54 calves from the herd were divided into 3 groups, 18 animals in each: CTR, TRT1 and TRT2. During the colostral period lasting for 5 days after birth, calves were drinking the mother’s colostrum. From 6 to 60 days, calves were watered milk replacer formulation. The control group (CTR) consisted of calves after the colostral period given milk replacer formulation without any supplementation. In the two other groups, calves received milk replacers with the supplementation of b-carotene (TRT1 group) in the amount of 25 mg/calf/day or a suspension of liver omega-3 oil (TRT2 group) in the amount of 5g/calf/day. All calves received solid feed ad libitum. The research showed that the parameters of the metabolic profile of calves (ALT, ASP, CHOL, GLU, UREA, ALP, TG, CRE) were within the normal range, indicating good health of the calves. Moreover, the supplementation had a positive impact on the chosen immunological parameters, including a higher IgG concentration, especially at the end of the milk drinking period. The result of B-carotene in a diet was the highest daily gain in the period from 30 to 60 days of life among the analysed groups of calves. No such effect was achieved in the TRT2 group of calves, as their body weight was the lowest. However, noteworthy is the beneficial impact of beta-carotene and omega-3 oil on the non-specific humoral immunity parameters, which was manifested by fewer cases of clinical diarrhoea and upper respiratory tract diseases.
