Opis:
The non-enzymatic reaction between reducing sugars and proteins, known as glycation, has received increased attention from nutritional and medical research. In addition, there is a large interest in obtaining glycoconjugates of pure well-characterized oligosaccharides for biological research. In this study, glycation of bovine serum albumin (BSA) by d-glucose, d-galactose and d-lactose under dry-heat at 60°C for 30, 60, 120, 180 or 240 min was assessed and the glycated products studied in order to establish their biological recognition by lectins. BSA glycation was monitored using gel electrophoresis, determination of available amino groups and lectin binding assays. The BSA molecular mass increase and glycation sites were investigated by mass spectrometry and through digestion with trypsin and chymotrypsin. Depending on time and type of sugar, differences in BSA conjugation were achieved. Modified BSA revealed reduction of amino groups' availability and slower migration through SDS/PAGE. d-galactose was more reactive than d-glucose or d-lactose, leading to the coupling of 10, 3 and 1 sugar residues, respectively, after 120 minutes of reaction. BSA lysines (K) were the preferred modified amino acids; both K256 and K420 appeared the most available for conjugation. Only BSA-lactose showed biological recognition by specific lectins.

Baltic blue mussels Mytilus trossulus were collected from the Gulf of Gdańsk (southern Baltic Sea) in order to isolate ferritin from its soft tissues, as well as to purify and characterise this protein. Proteins were isolated from the inner organs of M. trossulus (hepatopancreas, gills and soft tissue residue) by thermal denaturation (70◦C) and acidification (pH 4.5) of the homogenates, followed by ammonium sulphate ((NH4)2SO4) fractionation. The ferritin was then separated by ultracentrifugation (100 000×g, 120 min.). The protein content in the purified homogenates was determined by the Lowry method using bovine serum albumin (BSA) and horse spleen ferritin (HSF) as standards. PAGE-SDS and Western blotting analysis permitted identification of ferritin in the purified preparations. Additionally, the purified homogenates and mussel soft tissue were analysed for their heavy metal contents (especially cadmium and iron) in a Video 11 E atomic absorption spectrophotometer, following wet digestion of the samples (HNO3/HClO4). The electrophoregrams showed that the inner organs of M. trossulus contained ferritin, which, like plant ferritin, is characterised by the presence of subunits in the electrophoregram in the 26.6–28.0 kDa range. The highest ferritin content was recorded in the hepatopancreas, followed by the gills and the soft tissue residue. With regard to the sampling stations, the highest content of ferritin was noted in the animals sampled off Sopot (station D3), and in those collected by a diver off Jastarnia (W1) and Gdynia (W4). Ferritin isolated from the inner organs of mussels collected from these stations also contained the largest quantities of heavy metals (Cd and Fe). Ferritin isolated from the inner organs of mussels collected by a diver from wrecks – sites where the concentrations of iron and other trace metals in the sea water are high – contained higher quantities of heavy metals (Cd and Fe) than the ferritin isolated from the inner organs of mussels collected with the drag. This confirms that ferritin is a protein able to store and transport not only iron, but also, though to a lesser extent, some other heavy metals, including cadmium.

Sialic acids are important constituents of animal tissue glycoconjugates and are also present in the antigens of some bacterial strains. Capsular polysaccharides with sialic acid (NeuAc) have been extensively studied with regard to sensitivity to the bactericidal action of serum, whereas little is known in this regard about lipopolysaccharides (LPS) which contain NeuAc. Strains of Salmonella 048, able to infect animals and containing the same structures of LPS with NeuAc, were examined for their susceptibility to the bactericidal action of normal bovine serum (NBS). The strains showed varied sensitivity to the bactericidal action of NBS, which indicates that the expression of LPS containing NeuAc residues is not critical for the strains' resistance to the serum's activity. In this study the mechanisms of complement activation responsible for killing serum-sensitive Salmonella 048 rods by NBS were also established. Three such mechanisms were distinguished: activation of the classi- cal/lectin pathways, important (decisive) in the bactericidal mechanism of complement activation, parallel activation of the classical/lectin and alternative pathways, and independent activation of the classical and lectin or the alternative pathway.

Candida albicans is a major human fungal pathogen especially as an etiologic agent of opportunistic oral and genital infections. Moreover, C. albicans can be involved in the deep infections and recent evidence suggests that the majority of diseases produced by this pathogen are associated with biofilm growth. The aims of this study were to evaluate biofilm production ability of C. albicans strains isolated from different sources, and to evaluate the effect of serum for enhancement the growth of biofilm. The strains used in this study were obtained from three sources; 12 from feces of patients with gastrointestinal disturbances, 13 from the oral cavity of patients with oral candidiasis, and 16 from the vagina of patients with Candida vulvovaginitis (CVV). Polystyrene 96-well plates were used to grow biofilms and crystal violet (CV) staining method was used to evaluate the growth. There were no differences in biofilm growth expressed as CV absorbance between C. albicans strains from different origins neither in Yeast Nitrogen Base broth (YNB) or in bovine serum (BS) (ANOVA, P=0.1648, P=0.5106, respectively). In the BS, the biofilm production was greater than in YNB medium for all samples (ANOVA, P=0.0003).

The results of ultrafiltration tests carried out in a cross-flow mode with BSA model solutions using a laboratory scale setup equipped with fiat ceramic membranes are presented. The influence of operating parameters such as TMP, NaCl concentration and pH on membrane permeability was investigated. A decline in permeate flux with the increase of TMP and NaCl concentration and the decrease of pH was observed during tests. Membrane permeability for pure water was found in order to determine the membrane resistance RM for different TMP values. A short analysis of fouling phenomenon was performed based on the calculated flux decline and fouling resistance.
Przedstawiono wyniki badań ultrafiltracji w układzie krzyżowym (crossflow) modelowych roztworów BSA przy użyciu instalacji laboratoryjnej z płaskimi membranami ceramicznymi. Badano wpływ parametrów operacyjnych takich jak TMP, stężenie NaCl i pH na przepuszczalność membrany. Dla wyższych wartości TMP oraz stężenia NaCl oraz niższych wartości pH obserwowano spadek strumienia objętościowego permeatu. W celu ustalenia wielkości oporu membrany RM dla różnych wartości TMP wyznaczono przepuszczalność membrany dla wody. Przeprowadzano krótką analizę zjawiska foulingu na podstawie obliczonego spadku strumienia objętościowego permeatu oraz oporu foulingu.

The results of ultrafiltration tests carried out with model BSA and myoglobin solutions using ceramic 50 and 150 kDa membranes have been presented. Membrane permeability and selectivity were investigated in function of pH, transmembrane pressure (TMP) as well as molecular modelling data for proteins such as size, geometrical parameters and pH of minimal free energy of folding. The study has shown that the permeate flux J(v) depends on TMP, whereas the protein rejection is mainly influenced by pH. The results demonstrated that molecular modelling data are not sufficient to explain the membrane behaviour and the membrane–protein charge interactions and fouling effects must be also considered to explain the rejection mechanisms.

The binary and ternary complex formations of poly(methyl vinyl ether-co-maleic anhydride) (PMVEMA) with copper ions and with bovine serum albumin (BSA) in the presence of copper ions in phosphate buffer solution at pH = 7 were examined by the techniques of UV-visible, fluorescence, dynamic light scattering, atomic force microscopy measurements. In the formation of binary complexes of PMVEMA-Cu(II), the addition of copper ions to the solution of PMVEMA in phosphate buffer solution at pH = 7 forms homogeneous solutions when the molar ratio of Cu(II)/MVEMA is 0.5. Then the formations of ternary complexes of PMVEMA-Cu(II)-BSA were examined. Study analysis revealed that the toxicities of polymer-metal and polymer-metal-protein mixture solutions depend on the nature and ratio of components in mixtures.

Chemiczna modyfikacja struktury barwników skwaryliowych daje nowe układy chromoforowe o szerokim zakresie właściwości optycznych, takich jak absorpcja światła i emisja fluorescencji, z zakresu światła od widzialnego do bliskiej podczerwieni. Otrzymano nowe funkcjonalne barwniki, w których pod wpływem bodźca zewnętrznego lub oddziaływań chemicznych, zachodzą zmiany w rozmieszczeniu ładunku elektronowego w chromoforze skwaryliowym. Z uwagi na wyraźne zmiany ich właściwości spektroskopowych pod wpływem połączenia z substancją analizowaną, mogą zaleźć praktyczne zastosowanie, jako markery fluorescencyjne do oznaczania biocząsteczek.
Chemical modification of the structure of squarylium dyes gives a new chromophoric systems that have a wide range of optical properties, such as the light absorption and fluorescence emission in the range from the visible light to the near infrared one. The new functional dyes in which the changes of the electron charge distribution in chromophore under the influence of an external stimulus or chemical undergo have been obtained. The interaction between dye and analyte leads to the significant changes of their spectroscopic properties. It can give the possibility of practical application of these new dyes as the fluorescent markers for biomolecules labeling.

The targeted drug delivery has been studied as one of the main methods in medicine to ensure successful treatments of diseases. Pharmaceutical sciences are using micro or nano carriers to obtain a controlled delivery of drugs, able to selectively interact with pathogens, cells or tissues. In this work, we modified bovine serum albumin (BSA) with lactose, obtaining a neoglycan (BSA-Lac). Subsequently, we synthesized glyconanoparticles (NPBSA-Lac) with the premise that it would be recognized by microbial galactose specific lectins. NPBSA-Lac were tested for bio-recognition with adhesins of E. coli K88 and Ricinus communis agglutinin I (RCA). Glycation of BSA with lactose was analyzed by electrophoresis, infrared spectroscopy and fluorescence. Approximately 41 lactoses per BSA molecule were estimated. Nanoparticles were obtained using water in oil emulsion method and spheroid morphology with a range size of 300-500 nm was observed. Specific recognition of NPBSA-Lac by RCA and E. coli K88 was displayed by aggregation of nanoparticles analyzed by dynamic light scattering and atomic force microscopy. The results indicate that the lactosylated nanovectors could be targeted at the E. coli K88 adhesin and potentially could be used as a transporter for an antibacterial drug.
