Great economic losses to the dairy industry are associated with bovine mastitis, which results in poor milk quality and high treatment costs. Anti-inflammatory proteins play an important role in the suppression of the immune response against invading pathogenic microorganisms and are therefore being studied for possible use in the early diagnosis of mastitis. In our study, we used milk samples from 15 cows of Holstein Friesian breed with different health status (5 healthy, 5 subclinical, and 5 clinical animals), and tested them using immunohistochemical (IHC) analysis to evaluate the presence of IL-2, IL-10, TGF-β1, βDEF-2, DEF-3, and Cathelicidin LL37 proteins. The calculation of positively and negatively stained cells for each biomarker was performed using the semiquantitative counting method. We found the presence of all factors with the exception of Cathelicidin LL37, which was almost absent in milk samples of all animal groups. The significant decrease of IL-10, β-def2, and β-def3 expression levels within the 3 days of sampling, found in the milk of animals with sub- and clinical mastitis, indicates the loss of antiinflammatory protection of the affected cow’s udder. In contrast, the stable increase of IL-2 and TGF-β1 positive cells observed in the milk of mastitis-affected cows, and the similar expression of these factors in the milk of healthy animals, indicate the possible lack of involvement of these cytokines at an early stage of udder inflammation.

Introduction. Bovine tuberculosis is a chronic contagious disease caused by Mycobacterium bovis or Mycobacterium caprae. Before widespread action conducted in Poland between 1959–1975 to combat bovine tuberculosis (BTB), about 40% of all tuberculosis cases in pigs was caused by the bovine bacillus. At the present time, correctly carried out, long-term control of cattle has resulted in cases of bovine tuberculosis in pigs and humans being extremely rare and sporadic. In pigs, tuberculosis is most often caught in a slaughterhouse during slaughter. Materials and method. Samples came from pigs kept on the farm. Traditional bacteriological methods on solid media (Stonebrink, LJ with pyruvate) supported by the semi-automatic, liquid indicative culture method (MGIT) and PCR test were applied in targeted studies. The GenoType Mycobacterium MTBC and CM tests (Hain Lifescience, Germany) were used to additionally confirm that isolated strains classification was used. Results. Strains of mycobacteria were isolated from all examined pigs. Mycobacterium bovis was determined by real time PCR and Hain Genotype methods. Conclusions. In order to effectively fight against BTB, all animals on farms should be tested, regardless of species, while the milk of suspected cows should be utilized without being used for feed. It is important to adapt the current legal regulations to the current epidemiological situation.

BACKGROUND: The aim of the present paper was investigation of the temperature dependence of the activation energy of viscous fl ow for some proteins in aqueous solutions. MATERIAL AND METHODS: The viscosity of hen egg-white lysozyme, bovine -lactoglobulin, and human, bovine and porcine IgG immunoglobulin aqueous solutions was measured at temperatures ranging from 5oC to 55oC and in a wide range of concentrations. The measurements were performed with an Ubbelohdetype capillary microviscometer. RESULT S : The average value of the activation energy of viscous flow ΔE can be experimentally obtained from the slope of the line that represents the dependence of the liquid viscosity η (in logarithmic scale) versus a reciprocal of the absolute temperature (T-1). The functional dependence of ΔE on temperature can be obtained from strict defi nition ΔE = R[dlnη/d(T-1)], where R is the gas constant and from a three parameters modifi ed Arrhenius formula which gives an analytical function describing the viscosity-temperature dependence for proteins solutions in a wide range of temperatures. As appears, ΔE obtained in such a way decreases with increasing temperature according to a square function. The parameters of this function have been obtained for all studied proteins. CONCLUSIONS: The obtained results show that square function describes the temperature dependence of ΔE both for water, solutions and proteins themselves. One of the main factor which infl uence the activation energy is a molecular mass of protein. However, the results obtained for the studied immunoglobulins IgG show that this factor is not the only one.
WSTĘP: Celem niniejszej pracy było zbadanie temperaturowej zależności energii aktywacji przepływu lepkiego dla kilku białek w roztworach wodnych. MATERIAŁ I METODY: Lepkość wodnych roztworów lizozymu jaja kurzego, β-laktoglobuliny bydlęcej oraz ludzkiej, bydlęcej i świńskiej immunoglobuliny IgG zmierzono w zakresie temperatur od 5oC do 55oC i w szerokim zakresie stężeń. Pomiary wykonano przy pomocy kapilarnego mikrowiskozymetru typu Ubbelohde’a. WYNIKI: Średnią wartość energii aktywacji przepływu lepkiego ΔE można doświadczalnie otrzymać z nachylenia prostej przedstawiającej zależność lepkości cieczy η (w skali logarytmicznej) od odwrotności temperatury bezwzględnej (T-1). Funkcyjną zależność ΔE od temperatury można otrzymać z dokładnej definicji ΔE = R[dlnη/d(T-1)], gdzie R jest stałą gazową oraz z trójparametrycznego zmodyfi kowanego równania Arrheniusa, które daje analityczną funkcję opisującą temperaturową zależność lepkości dla roztworów białek w szerokim zakresie temperatur. Jak się okazuje, ΔE otrzymana w ten sposób maleje ze wzrostem temperatury zgodnie z funkcją kwadratową. Parametry tej funkcji zostały wyznaczone dla wszystkich badanych białek. WNIOSKI: Otrzymane wyniki pokazują, że funkcja kwadratowa opisuje temperaturową zależność ΔE zarówno dla wody, roztworów jak i samych białek. Jednym z głównych czynników wpływających na energię aktywacji jest masa molowa białka. Jednakże, wyniki otrzymane dla badanych immunoglobulin IgG pokazują, że nie jest to czynnik jedyny.

The non-enzymatic reaction between reducing sugars and proteins, known as glycation, has received increased attention from nutritional and medical research. In addition, there is a large interest in obtaining glycoconjugates of pure well-characterized oligosaccharides for biological research. In this study, glycation of bovine serum albumin (BSA) by d-glucose, d-galactose and d-lactose under dry-heat at 60°C for 30, 60, 120, 180 or 240 min was assessed and the glycated products studied in order to establish their biological recognition by lectins. BSA glycation was monitored using gel electrophoresis, determination of available amino groups and lectin binding assays. The BSA molecular mass increase and glycation sites were investigated by mass spectrometry and through digestion with trypsin and chymotrypsin. Depending on time and type of sugar, differences in BSA conjugation were achieved. Modified BSA revealed reduction of amino groups' availability and slower migration through SDS/PAGE. d-galactose was more reactive than d-glucose or d-lactose, leading to the coupling of 10, 3 and 1 sugar residues, respectively, after 120 minutes of reaction. BSA lysines (K) were the preferred modified amino acids; both K256 and K420 appeared the most available for conjugation. Only BSA-lactose showed biological recognition by specific lectins.

Reproductive failure in lactating dairy cows is a significant limitation to the dairy industry. Fertilization failure, Embryonic or fetal losses are major contributors to that limitation. The objective of this review was to determine cause of fertilization failure and early embryonic mortality in cattle. Fertilization failure and embryo mortality is a major cause of economic loss in dairy production systems. Direct effects of fertilization failure and embryo mortality are reflected in reduced conception rates with consequent effects on efficiency, production and profitability. Embryonic loss can be early or late .The major component of embryo loss occurs before day 16 following breeding with emerging evidence of greater losses before day 8 in high producing dairy cows. Late embryo loss causes serious economic losses because it is often recognized too late to rebreed females. Various factors can be a cause for fertilization failure and embryonic mortality. The major cause includes genetic, environmental, endocrine, infectious nutritional factors. More balanced breeding strategies with greater emphasis on fertility, feed intake and energy must be developed. There is a range of easily adoptable management factors that can either directly increase in embryo survival or ameliorate the consequences of low embryo survival rates. The correction of minor deficits in several areas can have a substantial overall effect on herd reproductive performance.

Early bovine embryos (two - to sixteen blastomeres) produced in vitro were cytogenetically analysed to determine the incidence of haploidy. Follicular oocytes were matured in vitro and inseminated with sperm prepared using the swim up method. After 2-3 days of culture, chromosome slides were prepared according to the air-drying technique and stained with Giemsa. Altogether 202 embryos produced metaphase spreads. Of these 4.5% were pure haploid embryos and 5.5% displayed a haploid cell line within mosaic embryos (n/2n and n/3n). The occurrence of haploid embryos observed in this study was compared to results of other studies and a possible origin of haploidy was discussed.

The strain of tuberculous mycobacteria called Mycobacterium caprae infects many wild and domestic animals; however, because of its zoonotic potential and possibility of transmission between animals and humans, it poses a serious threat to public health. Due to diagnostic limitations regarding identification of MTB strains available data regarding the incidence of M. caprae, human infection does not reflect the actual size of the problem. Despite the fact that the possible routes of tuberculosis transmission are known, the epidemiological map of this zoonosis remains underestimated. The progress in diagnostic techniques, application of advanced methods of mycobacterium genome differentiation and cooperation between scientists in the field of veterinary medicine and microbiology, have a profound meaning for understanding the phenomenon of bovine tuberculosis and its supervise its incidence. This is the first bacteriologically confirmed case of human infection of M. caprae in Poland.

Various methods of xenogeneic tissues stabilization have been proposed for the purpose of preparing many tissue-derived biomaterials. One of the most important treatments that may lead to obtaining the good-quality tissue biomaterials seems to be decellularization of such tissues. This process may contribute to the reduction of the most frequent failures resulting from the tissues stabilization. The aim of this work was to determine nanostructure of trypsin-treated bovine pericardium, using atomic force microscopy (AFM). The treatment of bovine pericardium with trypsin in EDTA solution resulted in non significant changes in tissue’s morphology. Demonstrated AFM studies of these tissues revealed no failures on the fibers’ surface in the nanoscale. Thus, our results confirm the expec-tation that decellularization may be considered as one of the most promising methods of the allogeneic and xenogeneic tissues stabilization.
