The aim of this paper it to review various scale approaches to the blood flow modelling. Blood motion may be described by three types of mathematical models according to the observed scales or resolutions, namely microscopic, mesoscopic and macroscopic descriptions. The above approaches are discussed together with their advantages and disadvantages. Several results of mesoscopic simulations are presented with particular attention paid to mesoscale semi-continuum models suitable for real-time blood flow visualisation.

This research was conducted from October 2018 until April 2019 in the Faculty of Fisheries and Marine Sciences, Universitas Padjadjaran. This study aims to determine the optimum potassium diformate dosage which was added to feed to increase the immune status of Pangasianodon hypophthalmus (Sauvage, 1878). The research method used is the experimental Complete Random Design (CRD) with 4 treatments and 3 replications. The treatments are A (control), B (0.3% KDF), C (0.5% KDF), and D (0.7% KDF). Observed parameters are survival rate, gross clinical signs, intestine pH levels, total leukocyte count (white blood cells), total erythrocyte count (red blood cells), and water quality. The results showed that the 0.5% KDF addition in feed is the most effective to improve immune status. This treatment also is the most effective to improve Pangasius immune status after being challenged with A. hydrophila with survival rate of 93.33%, leukocyte count 2.98×106 cells/mm3 and erythrocyte count 1.02×106 cells/mm3. Increased leukocyte count was 65.26% and erythrocyte count - 58.23%.

Purpose. The aim of the study was to assess the in vitro potency of pentoxifylline (PTX) and one of its most active metabolites lisofylline (LSF) to improve rheological properties of red blood cells (RBC) from healthy individuals and patients with chronic venous disease (CVD). Additionally, the study aimed to compare the effects of PTX and LSF on RBC deformability and aggregation. Methods. Blood samples were collected from healthy volunteers (antecubital vein) and from CVD patients (varicose and antecubital vein). Deformability and aggregation of RBC were assessed using Laser-assisted Optical Rotational Cell Analyser (LORCA). Results. PTX and LSF increased RBC elongation significantly. Additionally, RBC incubation with PTX resulted in a marked decrease in RBC aggregation. PTX reduced the tendency towards the formation of RBC aggregates and of their stability. The beneficial effect of PTX on RBC aggregation was most apparent for those cells whose aggregation tendency and aggregate stability was the greatest. Conclusions. In vitro addition of PTX or LSF effectively increased deformability of RBC from healthy donors and patients with CVD. Thus, LSF may contribute to the in vivo hemorheological effects of pentoxifylline. On the other hand, there was no significant effect of LSF on aggregation of RBC in vitro. Hence, LSF has no contribution to this particular effect of PTX. Additionally, the present study demonstrated the use of RBC with impaired deformability and aggregation for the evaluation of in vitro rheological activity of xenobiotics.

In recent years it has been discovered that long non-coding RNAs are important regulators in many biological processes. In this review, we summarize the role of lncRNA in erythropoiesis. lncRNA are crucial for regulation of gene expression during both, proliferation and differentiation stages of red blood cell development. Many are regulated by erythroid-specific transcription factors and some are expressed in a developmental stage-specific manner. The majority of individually studied lncRNAs are involved in regulating the terminal maturation stages of red cell differentiation. Their regulatory function is accomplished by various mechanisms, including direct regulation in cis or trans by the lncRNA product or by the cis-localized presence of the lncRNA transcription itself. These add additional levels of regulation of gene expression during erythropoiesis.

Fractal dimension of three dimensional red blood cell aggregates were determined by measurement of their size and sedimentation velocity. The sedimentation of the aggregates was investigated with red blood cells suspended in dextran 70 solutions at concentrations from 2 to 5 g/dL, at hematocrit 5% and 10%. The aggregate velocity and size were measured using an image analysis technique. The velocity vs. radius dependence of the aggregates exhibited a scaling behavior. This behavior showed the fractal structure of the aggregates. It is shown that the fractal dimension of the three dimensional red blood cell aggregates depends on the dextran concentration in the suspension. This parameter exhibited a minimum at dextran concentration between 3 and 4 g/dL. Thus the fractal dimensions increased as the aggregation extent decreased. The obtained results show that the sedimentation experiment together with image analysis is a promising technique to determine the fractal dimension of the three dimensional red blood cell aggregates.

Opis:
Human peripheral blood cells stimulated by phytohemagglutinin (which serve as a model of cancerous cells) and resting cells were incubated in dimethyl sulfoxide solutions of various phthalocyanines. In order to diminish the influence of atmospheric oxygen the cells were embedded in a polymer (polyvinyl alcohol) film. Fluorescence spectra of the samples were measured over two regions of excitation wavelengths: at 405 nm (predominant absorption of the cell material) and in the regions of strong absorption of phthalocyanines (at about 605 nm and 337 nm). The intrinsic emission of cell material became changed as a result both of cells' stimulation and of incubation of cells in dye solution. In most cases the stimulated cells when stained by dye exhibited higher long wavelength fluorescence intensity than resting cells. This suggests higher efficiency of dye incorporation into cancerous cells than into healthy cells. The absorption spectra of samples were also measured. The spectra of various phthalocyanines in incubation solvent, in polymer and in the cells embedded in polymer, were compared. The comparison of properties of the cells stimulated for different time periods enabled to establish the conditions of stimulation creating a population of cells incorporating a large number of sensitizing molecules.

In the paper we present some results concerning application of simple shape parameters that are successfully used to distinguish between normal and pathological blood cells. All descriptors are based on shape area and its perimeter. We use five parameters that are automatically calculated for objects pointed out in the analyzed image with the help of our software. The experiments performed on the same set of tested images as in [3] let us draw the same conclusions as those reported in [3] where fractal analysis to shape has been used.

In the paper we present some results concerning the usage of Fourier analysis which we can successfully apply to distinguish between normal and pathological blood cells. We use special area function and different number of Fourier descriptors that are automatically calculated for all objects pointed out in an image. Calculations are performed with the help of the software made by the first author. Experiments performed on the same set of tested images as in [3] and [5] draw as to the same conclusions as there in [3] where the fractal analysis to shape has been used and in [5] where simple geometrical shape parameters have been applied. This paper presents an analysis of use of ultrasonic standing wave in cell separation from bodily fluids based on the example of erythrocyte separation from plasma. It describes movement of red blood cells in plasma under the influence of the acoustic field (whose forces result from interaction of red blood cells with plasma as the vibrating medium) and under the influence of resistance forces in Stokes’ and Oseen’s approximation. The general properties of solutions of the motion equation are given. The solutions for the parameters of the ultrasonic wave and blood cells which are interesting in terms of practical applications in medical diagnostics are discussed. Time constants of the cell transportation to the regions of stable equilibrium in the field of ultrasonic standing wave are estimated. The formulas which determine the time needed to obtain the assumed concentration increase in plasma in nodes and/or anti-nodes of the standing wave are derived.

INTRODUCTION: Acute coronary syndromes (ACS) and sudden death cause most ischemic heart disease (IHD)- related deaths, which represent 1.8 million deaths per year, with similar numbers of men and women dying from coronary artery disease (CAD). It’s known that inflammation plays crucial role in atherosclerotic plaque formation and its destabilization. The purpose of this study is to evaluate of white blood cells count in its subpopulation in patients with ACS and modifiable cardiovascular risk factors – arterial hypertension and 2 type Diabetes Mellitus (DM). MATERIAL AND METHODS: In this observational cohort trial we observed of 184 patients with ACS. All patients were randomized into four groups: 1st group - 42 patients with ACS without arterial hypertension (AH) or DM; 2nd group – 56 patients with ACS and previous AH; 3rd group – 42 patients with ACS and 2 type DM; and 4th group – 44 patients with ACS and AH and DM. We studied of leukocytes count and their subpopulations in blood. RESULTS: The mean white blood cells count was significant higher in patients with ASC, compared with control group: 8.23 [6.50; 9.40] vs 5.49 [5.20; 5.70] (p<0.001). Similarly, ACS caused increase of leukocytes subpopulation count in blood. The significant higher count of white blood cells was observed in patients with ACS and co-morbidities: 2 type DM and its association with AH. In patients with ACS and previous AH we observed significant lower neutrophils count (p<0.05), but increased quantity of lymphocytes, compared with patients with ACS without co-morbidities (p<0.001). DM and its association with AH was characterized of neutrophils, lymphocytes and monocytes counts growth. CONCLUSIONS:ACS is characterized of raised white blood cells count and its population, especially in cases of association with 2 type Diabetes Mellitus.

The present study was undertaken to contribute to the characterization of the degree of variability in baseline damage in white blood cells from control population, and to investigate how this variability is associated with external and internal factors. Altogether 170 healthy volunteers, randomly selected from the general population of the Republic of Croatia, participated in the study. Two sensitive tests: the alkaline comet assay and the chromosome aberration test were applied to study the background levels of DNA damage in their white blood cells. The results point to inter-individual differences, indicating different genome sensitivity. As revealed by both assays, the background levels of DNA damage were mostly influenced by smoking habit as well as medical exposure (especially to diagnostic X-rays). Sex and age of subjects did not significantly influence the values of DNA damage recorded in the white blood cells. Although higher levels of DNA damage were recorded in blood samples collected during winter and autumn, they were mostly influenced by medicinal exposure and smoking habit. Statistical evaluation of the data confirmed that a positive correlation exists between DNA migration and the number of long-tailed nuclei found with the comet assay and the total number of chromosome aberrations. The data obtained can serve as control values in forthcoming biomonitoring studies.

This studyis a contribution to research on the biomedical and clinical applications of ultrasound. Our research concerns the procedure for the separation of human blood fractions - erythrocytes. Ultrasonic waves can be used for the separation of cells in human blood. From a physical point of view, the human blood is a suspension of liquids and solids (cell elements), and behaves like a non-Newtonian fluid. Our work is devoted to the problem of the motion of red cells in human blood under the influence of ultrasonic wave. It defines a range of the applicability of approximation consisting in neglecting the nonlinear term in the friction force. It also analyzes the general properties of the equation of motion of the cell in the case of large attenuation constants, corresponding to the values of the drift forces for the cells with radii of a few μm. Finally, it defines the applicability criterion of the so-called King-St Clair approximation consisting in the assumption of equilibrium between the drift and the Stokes viscosity forces, neglecting the term representing inertia. This approximation permits analytical estimation of the time constants for the cell transport to points of stable equilibrium in an ultrasonic standing wave field.

The interactions of two metal-free phthalocyanines [(H2Pc) and Solar Pc (with four peripherical groups: SO2N(CH2CH2OH)2)] and of one metal substituted dye (CoPc) with resting and stimulated human peripheral blood mononuclear cells (PBMC) were compared. The absorption, fluorescence, photoacoustic and EPR spectra of both resting cells and cells stimulated by phytohaemagglutinin, incubated in dimethyl sulfoxide (DMSO) with very low or 95% water content and with or without dye addition, were measured. The fate of the light absorbed by the samples was investigated. It is known that singlet oxygen production is crucial for photodynamic action of dyes. Thermal deactivation and luminescence emission compete with this process, so investigation of these alternative paths of sensitizer deactivation provides information about photodynamic action. The incorporation of the investigated dyes into cells and the perturbation of the cell structure caused by the dyes, the incubation solvent and the activator were investigated by comparing the spectral properties of PBMC before and after stimulation and incubation. Incubation of the cells for 1 h in a solution of Solar Pc in 99.5% aqueous DMSO, resulted in an efficient dye incorporation which was highly selective. Solar Pc being introduced much more efficiently into stimulated cells than into resting cells.

Peppers (Capsicum spp.) are a rich source of different bioactive compounds with potential health-promoting properties. Pepper fruits have numerous uses in culinary preparations that make this one of the most important vegetables and spices. This study investigated the evaluate the antioxidant activity of green chili pepper extracts on human red blood cells by comparison of different extraction method and treatments. Two solvents (water and methanol) was used to extract from green pepper compounds (fresh and after boiling process). The cytotoxic effect of sodium benzoate and fresh and boiled green chili pepper on human red blood cells was investigated. The obtained results show the protective effect of the extract from hot chili peppers on the negative effect of sodium benzoate. This effect was significantly dependent (p<0.05) on the type of source (fresh or boiled), not on the concentration of extract. Used type of solvent had statistical significant matter in C3 concentration of fresh vegetable. The highest protection gave fresh chili pepper extract.
