To determine the effect of coconut extract on callogenesis of Betula pendula, Roth stem barks were cultured in NT (Nagata and Takebe) basic culture media in two individual experiments: i) cultivation explant in different treatments of coconut extracts combined with 1 mg l-1 2, 4-D (2, 4-Dichlorophenoxyacetic acid) and ii) callogenesis in NT media containing 1.5 mg l-1 2,4-D and 0.5 mg l-1 BAP (6-Benzylaminopurine) and then cultivation under the first experiment treatments. The first experiment demonstrated that not all concentrations of coconut extracts lead to callus induction individually, but callus induction increased 84% in a culture containing 5% coconut extract plus 1 mg l-1 2, 4-D. Based on the results of the second experiment, this treatment also significantly increased the wet and dry weights of the produced calluses. The possibility of increasing the betulinic acid and betulin by ultrasound was also studied. Samples cultivated in the selected culture medium were exposed to ultrasound waves in two forms of 1) one exposure and 2) twice exposure (repetition with 24 hr interval) in steps of 20, 60, 100, and 160 sec, and one treatment as the control. Betulin and the betulinic acid amount were determined using HPLC (High-Performance Liquid Chromatography). The maximum betulinic acid and betulin were obtained in 100 sec in once exposure (2.3 mg g-1 DW) and 160 sec in the twice exposure (0.75 mg g-1 DW) to ultrasound. The results showed that betulinic acid content was more than betulin.

Betulin and its derivatives have been reported to affect several key genes of cell-cycle regulators. However, their specific targets haven’t yet been discovered. As an important cell cycle regulator, Cyclin-dependent kinase-2 (CDK2) has become a potential target for cancer therapy. Here we describe the design, synthesis and antitumor activities in vitro of eleven new betulin nitrates. The results revealed that compound (20) possesses potent antitumor activity against MCF-7 cell lines (IC50 < 10 μM). In order to investigate potential protein target, betulin nitrates were subjected to docking studies with CDK2. Compound (20) showed very good binding affinity for CDK2 via hydrogen bonding interactions. Thus, the CDK2 inhibitory potential could make compound (20) possible candidate as antitumor agent.

Opis:
White birch bark extract, used in traditional medicine, has many medical properties. Its main components are betulin and betulinic acid - lupane type pentacyclic triterpenes. The discovery of the anti-tumor and anti-viral properties of betulinic acid has become an impulse to modify the structures of these compounds in order to obtain better activity. Over twenty years of research, several derivatives have been selected that have significantly better anti-tumor or antiviral properties than their precursors. This paper presents all new derivatives of betulin and betulinic acid, which have been tested in phase I, II or III of clinical trials. Clinical experiments on animals with natural cancers have also been described. The results obtained by various research teams give hope for the potential use of these compounds in therapy. At the same time, the presented analysis shows that the implementation of the new drug on the market is a process that lasts 10-15 years, which, despite the involvement of huge financial and human resources, does not guarantee success.

Bacteria, viruses, and other pathogens in the surrounding environment are biological threat to human health and life. The development of various diseases in the world, as well as the pandemic caused by the rapid spread of the SARS-CoV-2 virus, have increased the demand for the use of upper respiratory protection devices. Out of concern for the natural environment, the aim of this work was to develop an innovative solution i.e. the FFP3 filtering membrane made of an organic compound. First, preliminary tests of the FFP2 mask were carried out to assess the chemical composition, morphological structure - fibers geometry, thickness, density, and arrangement. The FTIR analysis study was conducted to confirm that the main chemical in the mask was polypropylene (PP) and high-density polyethylene (HDPE). Optical, confocal microscopy and computer microtomography studies showed the fibers structure. They were densely arranged and their thickness was less than 1 den. The fiber structure of the FFP2 mask was also compared before and after immersing in betulin, an organic compound obtained from birch bark via the Soxhlet extraction. In addition, the assessment of microbiological activity was made on the reference strain Escherichia coli ATCC 25922, and the anti-inflammatory activity on normal human skin fibroblasts on polycarbonate with betulin. The studies showed that betulin supported the material antibacterial and anti-inflammatory properties.

Introduction. Neoplastic diseases are a common cause of death, especially in developed countries. Various methods are used in cancer therapy including natural substances. Terpenes and terpenoids play a special role, including betulin and betulinic acid with the pentacyclic structure of lupane. They have a wide range of pharmacological activity, among which anti-cancer activity attracts the greatest attention. Abbreviated description of the state of knowledge. The review discusses the pharmacological effects of betulin and its derivatives on tumours, and focuses on the potential of its application in the treatment of melanoma. The review study also presents future prospects for the use of this natural substance. Betulin and its derivatives have the ability to influence the processes of apoptosis, angiogenesis and autophagy. Additionally, they have the ability to sensitize, increasing the sensitivity to commonly used methods of cancer treatment. To-date, numerous studies have been carried out on the synthesis of betulinic acid derivatives, which would show greater polarity and activity compared to the starting compound. Due to their greater bioavailability, the modified compounds showed a stronger anti-tumour effect. Conclusion. The results of the above-mentioned studies confirm the anti-cancer properties of betulin. Regarding melanoma, betulin shows significant therapeutic efficacy. This gives new perspectives for the treatment of the most dangerous skin cancer.

Postęp w rozwoju technik analitycznych umożliwia obecnie oznaczanie bardzo dużej liczby zanieczyszczeń organicznych występujących w środowisku naturalnym. Część tych zanieczyszczeń jest pochodzenia naturalnego. Aktualnie badania ukierunkowane są głównie na zanieczyszczenia specyficzne, których obecność jest wynikiem antropogenicznego oddziaływania na środowisko naturalne. W celu oceny występowania specyficznych zanieczyszczeń i mikrozanieczyszczeń w środowisku naturalnym, tj. wybranych kwasów tłuszczowych nienasyconych - oleinowego i linolowego oraz kwasu nasyconego - palmitynowego, a także steroli stigmasterolu i β-sitosterolu oraz betuliny, opracowano techniki ich wydzielania i ich chromatograficznej analizy. Analizę jakościowo-ilościową zarówno kwasów tłuszczowych, jak i steroli wykonywano przy użyciu chromatografu GC / ion trap / MS (model Saturn 2100 T firmy Varian) wyposażonego w kolumnę chromatograficzną VF-5ms (Varian). Do wydzielania związków zastosowano różne techniki ekstrakcji. W przedstawionych w pracy badaniach prowadzono monitoring występowania zanieczyszczeń w wodach powierzchniowych Górnego Śląska (Wójtowianka (Doa), Potok Rokitnicki, rzeka Bytomka, rzeka Kłodnica i Kanał Gliwicki) w okresie od stycznia do czerwca 2015 roku.
Nowadays, advances in the analytical techniques allow the determination of hundreds of organic pollutants present in the environment. Some of organic pollutants are of natural origin. Although the current study are mainly aimed at specific pollution, the presence of which is the result of anthropogenic impacts on the environment. In order to assess the appearance of specific pollutants and micropollutants in the environment ie the selected unsaturated fatty acids: oleic, linoleic, saturated acid: palmitic and sterols: stigmasterol, β-sitosterol and betulin, the techniques for their isolation and analysis have been developed. Quality and quantity analysis of fatty acids and sterols was carried out using GC/ion trap/MS chromatographic system (model Saturn 2100 T, Varian) equipped with a chromatography column VF-5ms (Varian). Various extraction techniques have been used for isolation of the pollutants. In this study the monitoring of level of contamination in surface water of Upper Silesia (Wójtowianka (Doa) Stream, Rokitnica Stream, Bytomka River, Klodnica River and Gliwice Channel) was conducted in the period from January to June in the 2015.
