The aim of the study was to determine if azoxystrobin and metconazole used for the control of wheat FHB at half, full, and quarter more the recommended dose rate may affect in differentiated way on the occurrence of Fusarium spp. and their ability to mycotoxin production in harvested grain, in wheat ears artificially inoculated with two DON-producing isolates of F. culmorum. Macroscopic evaluation showed high incidence of fusariosis. Plant health in the plots where the heads were artificially inoculated and fungicide was not applied was similar to the protected ones. Only increasing the dose metconazole resulted in a stronger reduction of fusariosis. The advantageous effect of azoxystrobin was not observed. Mycological analysis of harvested grain showed the presence of a number of F. culmorum, but from samples sprayed with metconazole it was isolated in smaller quantities. Also F. avenaceum, F. graminearum, F. poae and F. tricinctum were isolated. Molecular analysis showed the presence of F. culmorum in all samples of harvested grain. Also genes from Tri cluster were identified, involved in the synthesis of type-A and type-B trichothecenes - especially DON and 3Ac-DON. Chromatography revealed the presence of small quantities of mycotoxins. In all samples DON and 3Ac-DON were predominant. In general, F. culmorum isolate, which caused weaker symptoms of FHB and was less numerously isolated from grain that the other one, produced smaller amounts of mycotoxins. Samples protected with azoxystrobin contain the largest quantities of DON. Effect of different doses of fungicides on the number of mycotoxins was not clearly established...

The aim of the research was the laboratory evaluation of the fungistatic effect of the Zielony Busz (ZB) organic-mineral fertiliser on the linear growth of polyphagic phytopathogenic fungi Alternaria alternata, Colletotrichum coccodes, Fusarium avenaceum, Fusarium equiseti and Fusarium oxysporum. The preparation was applied to the medium (PDA potato-glucose agar) in concentrations of 2% and 5%, compared to 0.05% azox- ystrobin (relative control) and pure PDA medium (absolute con- trol). The strongest antifungal effect of the Zielony Busz fertilizer was found at the 5% concentration in reference to A. alternata (inhibition of colony growth at the level of 50–52% compared to the absolute control; 20–41.7% compared to azoxystrobin) and C. coccodes (18.2–53.3% growth inhibition compared to abso- lute control). Weaker inhibitory effect of 5% concentration of the fertilizer was noted against F. avenaceum (10.5% inhibition of colony growth compared to absolute control) and F. oxysporum (20% inhibition compared to absolute control), but only at the beginning of the experiment. The Zielony Busz fertilizer at a con- centration of 2% caused the surface growth of all tested fungi, especially F. equiseti. The best effects of the fungistatic effect of the fertilizer were obtained in the first days of the experiment (4th day) with a 5% concentration of the product in the substrate. The objective of these studies was to identify differences and similarities within the progeny of Rhizoctonia solani AG5, which arose from basidiospores produced by the maternal strain ID23. The following characteristics were analyzed: appearance of the mycelium (color, structure, zonation, and presence of sclerotia), growth rate (at 10°C, 20°C, and 30°C), fungicide sensitivity, and hyphal structure. The mycelial color of R. solani AG5 ranged from white/cream to light and dark brown. The structure of the mycelium may be compacted or flattened with visible zoning or fluffy with dark brown sclerotia on the colony surface. Homokaryons and heterokaryons derived from homokaryons were analyzed by constructing a phylogenetic tree using morphological data. Single basidiospore-grown isolates formed a separate subclade, the majority of which were grouped with a maternal isolate; however, heterokaryons derived from them created a separate subclade. In addition, isolates grown in basidiospores germinated at low temperatures created their own group, but with some exceptions. This shows a divergence in the morphological parameters of the subsequent generations and within generations. The optimal temperature for growth was found to be between 20°C and 30°C. The exceptions were strains obtained from basidiospores that germinated at refrigerated temperatures. For these samples, 10°C was found to be the optimal growth temperature. The hyphae of homokaryons were characterized by the presence of branching at an almost right angle and the presence of a septum at the site of constriction of the branch itself. The mean diameter of hyphae ranged 2.93–15.60 μm, depending on the age of hyphae. The fungicidal compounds at a concentration of 10 ppm had no significant effect on the activity of the tested strains, whereas a tenfold increase in the dose reduced the growth ability of the tested isolates. The activity of fungicides containing azoxystrobin, thiuram, or thiophanate-methyl on R. solani resulted in a reduction in the mycelial growth rate only in the case of azoxystrobin and thiuram, and in some cases, it was completely inhibited (thiophanate-methyl).

The aim of the study was to assess the toxicity of azoxystrobin, a fungicide belonging to the strobilurin class, for selected saprophytic fungi (Saccharomyces cerevisiae and Penicillium sp.) and for radish (Raphanus sativus L.) The parameters of fungi growth and the early development stages of radish were analysed. Based on the sensitivity of the organisms and their physiological processes to azoxystrobin, they can be arranged in the following order: growth of sod1 S. cerevisiae mutant, growth of wild-type S. cerevisiae, growth of Penicillium sp., respiration of germinating radish seeds, early seed germination, elongation of roots and seedlings, late seed germination. The mechanism of azoxystrobin toxicity seems to be associated with cellular antioxidant status.
Celem badań była ocena toksyczności azoksytrobiny, fungicydu należącego do klasy strobiluryn dla saprofitycznych grzybów (Saccharomyces cerevisiae i Penicillium sp.) oraz rzodkiewki (Raphanus sativus L.). Analizowano wzrost drożdży szczepu dzikiego i jego mutanta bezdysmutazowego sod1, grzyba strzępkowego Penicillium sp. oraz parametry biochemiczne i fizjologiczne kiełkujących nasion rzodkiewki i powstałych z nich siewek. Najbardziej wrażliwym na azoksytrobinę okazał się wzrost drożdży (mutanta sod1, następnie szczepu dzikiego wt) w następnej kolejności: wzrost grzybni Penicillium, oddychanie kiełkujących nasion rzodkiewki, proces kiełkowania oznaczany po 24 godzinach od wysiewu nasion, wydłużanie korzeni, wydłużanie siewek, proces kiełkowania określany po 96 godzinach. Mechanizm toksyczności azoksytrobiny wydaje się być powiązany z aktywnością komórkowego systemu antyoksydacynego.

Reduction of colony diameter using two DMI-fungicides and one strobilurine fungicide was evaluated in the Septoria tritici population. In an in vitro assay, 57 strains of S. tritici originating from different parts of the Czech Republic were grown on agar plates containing different concentrations of fungicides. The mean EC₅₀ values of strains were analysed for flusilazole (Capitain), metconazole (Caramba) and one strobilurine fungicide: azoxystrobin (Amistar). ED 50 values for metconazole were low. There were highly significant correlation in reaction of pathogen strains to metconazole and flusilazole. Some isolates showed significant higher level of resistance to azoxystrobin. The data will be used in a continuing survey of resistance development in the S. tritici population originating from the territory of the Czech Republic.
Na podstawie redukcji średnicy kolonii grzyba Septoria tritici oceniano odporność populacji grzyba na dwa fungicydy z grupy IBE i jeden fungicyd strobilurynowy. 57 szczepów (izolatów) Septoria tritici, zebranych z różnych rejonów Republiki Czech, testowano „in vitro” na szalkach Petriego z agarem z dodatkiem fungicydów w różnych koncentracjach. Dla substancji aktywnych: flusilazol (Capitain), metconazol (Caramba) i azoxystrobina (Amistar) wyznaczono średnią wartość ECW. Wartość EC₅₀ dla metconazolu była niska. Obserwowano bardzo wysoką korelację reakcji szczepów patogena na metconazol i flusilazol. Kilka z badanych izolatów wykazywało wyraźnie wyższą odporność na metconazol. Poszukiwania odporności na fungicydy w populacjach S. tritici na terenie Czech będą kontynuowane.
