Rhodococcus equi
Opis:
Rhodococcus equi is an important etiologic agent of respiratory- and non-respiratory tract infections, diseases of animals and humans. Therapy includes the use of various group of chemotherapeutic agents, however resistance acquirement is quite common. To date there is no preferred treatment protocol for infections caused by isolates resistant to macrolides and rifampicin. The resistance acquirement is a result of many molecular mechanisms, some of which include alterations in the cell envelope composition and structure, activity of the efflux pumps, enzymatic destruction or inactivation of antibiotics, and changes in the target site. This paper contains an overview of antimicrobial susceptibility of R. equi, and explains the possible molecular mechanisms responsible for antimicrobial resistance in this particular microorganism.

A series of fourteen derivatives of adamantane was synthesised. The new compound 4-(adamant-1-ylmethoxycarbonyl)phthalanhydride obtained from 1-adamantane- methanol and trimellitic anhydride chloride appeared very useful for preparation of a number of N-substituted phthalimides. Antimicrobial activity of the newly obtained derivatives such as, for example, 4-(adamant-1-ylmethoxycarbonyl)-N-(5-carboxypentamethylene)phthalimide or 4-(adamant-1-ylmethoxycarbonyl)-N-(L-alanyl)phthalimide was tested against Staphylococcus aureus, Bacillus sp., Micrococcus flavus and Enterococcus faecium. The minimal inhibitory concentration (MIC) for these compounds against S. aureus were 0.022 and 0.05 μg/ml, respectively.

Antibiotic resistance has become a global public health concern in the last few years. Given the widespread rate of recurrence, increasing attention is being turned toward environmental pathways that potentially contribute to antibiotic resistance genes (ARGs) dissemination outside the clinical realm. In this study, a metagenome analysis of intestinal virus-like particle fraction (VLPs) from a wild coyote (Canis latrans) revealed for the first time, multiple ARGs, such as B-lactamases and multidrug efflux pumps. Description of ARGs presence in natural environments is critical to understand the emergence of resistant strains.

Opis:
The emergence of resistance in microorganisms on a global scale has made it necessary to search for new antimicrobial factors. Antimicrobial peptides (AMPs) seem to meet these expectations. AMPs are produced by bacteria, viruses, plants, and animals, and may be considered as a new class of drugs intended for the prophylaxis and treatment of both systemic and topical infections. The aim of this study is to review the results of studies on the use of peptides to combat infections in vivo. Antimicrobial peptides may be applied topically and systemically. Among the peptides used topically, a very important area for their application is ophthalmology. AMPs in ophthalmology may be used mainly for the protection of contact lenses from ocular pathogens. Many AMPs are in clinical trials for application in the therapy of local infections. There may be mentioned such preparations as: pexiganan (magainin analogue), MX-226 (based on indolicidin), NEUPREX (isolated from human BPI (bactericidal/permeability-increasing) protein), IB-367 (variant of porcine protegrin), P113 (based on histatin), daptomycin, polymyxins, as well as peptidomimetics. In the combat against systemic infections are used such peptides as: P113D (modified P113 peptide containing D-amino acids), colistin, peptoids, and peptides containing non-typical amino acids or non-peptide elements. AMPs are also used as antiprotozoal, antifungal, antitoxic and immunostimulatory agents. The limitations in the use of peptides in the treatment of infections, such as susceptibility to proteolysis, and resistance of microorganisms to the peptides, are also discussed. AMPs are a promising strategy in the fight against microbial infections.

Opis:
Nano-biotechnology represents the intersection of nanotechnology and biotechnology which is an emerging field in creation, productivity and utility of nanoscale structures for advanced biotechnology. Plant and plant extract are considered as green and effective paths in the synthesis of gold and silver nanoparticles. The aim of the present study is to analyze the interaction of silver nanoparticles synthesized from herbal source (Cinnamon zeylanicum) with pathogenic bacteria Salmonella typhi (S. typhi) in order to know the microbe pathogenicity. The micro-nano interface disturbed the growth of microbes. Under the influence of different concentration of nanoparticles (50 µL to 1,000 µL), the microbes exhibited slow growth as compared to control. The viable cell count also decreased as the nanoparticles concentration increased. This shows major difference in the bacterial culture of with and without nano particle. Under normal culture, without nanoparticles intervention, the microbes show lag phase at 0-4 h, the log phase begins and its exponential growth occurs between 4-8 h and after 12 h, stationary phase was attained. There was a slow growth observed when the culture was exposed to silver nanoparticles. This clearly shows the antimicrobial effect of nanoparticles. In this study we used green synthesized nanoparticles, which have shown a good antibacterial efficacy against the pathogenic microorganism Salmonella typhi.

Lactoferrin (LF) is an iron-binding glycoprotein from the transferrin family, which has been identified in most biological fluids as secretions from exocrine glands and the content of specific granules of neutrophils. It has been reported to have numerous functions. Due to antimicrobial and anti-inflammatory activity, the lactoferrin plays significant role in host defense against infection and extreme inflammation. Recent studies have also demonstrated that LF can protect against cancer in experimental animals and has anticarcinogenic activity in many human tumors. At the cellular level, LF modulates the proliferation, differentiation, maturation, activation, migration and function of immune cells. This review presents the multifunctional roles and specific beneficial properties of lactoferrin.

Opis:
This paper deals with the determination of the antimicrobial efficiency of plain plated weft knits antimicrobial treated with commercial silver chloride (AgCl (iSys Ag, Germany)) dispersed in a reactive organic-inorganic binder (iSys MTX, Germany). Antimicrobial treatment was performed by the exhaustion method. The morphology of untreated and antimicrobial treated knits was analysed using Scanning Electron Microscopy (SEM) to confirm the presence of silver particles on the fibre surface. The antimicrobial activity was estimated using the Agar diffusion Test (according to standard EN ISO 20645:2004 (qualitative method)) and antibacterial finishes in a Textile Materials Test (according to the AATCC 100–1999 standard (quantitative method)). Two strains of bacteria - E. coli and S. aureus were used in this research. The results obtained proved that the fibre type and raw composition have an important influence on the relationship between the fibre surface and antimicrobial effect of the material formation and optimal antimicrobial efficiency obtained. It was found that pure fibre cotton and man-made bamboo knits had the highest antimicrobial activity for both types of bacteria (E. coli and S. aureus). Meanwhile knits of 100% polyester fibre and blended knits containing polyester fibre had an insufficient effect on the bacterial growth. It was also established that antimicrobial treated knits with the same fibre composition and very similar total linear density of folded yarns but with a different number of folded yarns in the structure have different antimicrobial activity. This is because the higher number of folded yarns in the knitted loop has a larger surface area and, herewith, larger antimicrobial acting area.
W artykule oceniano właściwości antymikrobowe dzianin lewo-prawych platerowanych nasycanych komercyjnym chlorkiem srebra.

Azurocidin is an inactive serine protease homolog with primary sequence similarity to neutrophil elastase, cathepsin G, and proteinase 3. The aim of this study was to investigate possible consequences of differential glycosylation of azurocidin in regard to its secretion, protein stability as measured by susceptibility to proteolysis, and antibacterial activity. Site-directed mutagenesis was employed to generate mutant azurocidin variants lacking individual N-glycosylation sites. Our results show that N-linked glycans may play a role in proper azurocidin folding and subsequent secretion by insect cells. We also demonstrate that N-linked glycosylation contributes to azurocidin stability by protecting it from proteolysis. The lack of N-glycosylation at individual sites does not significantly influence the azurocidin antibacterial activity.
