The observation of a group of 168 children treated for toxocariasis shows that the most frequent symptoms at the time of the physical examination were abdominal pains - 41 %; headache was observed in 12% cases. Eosinophilia was found in 9.2% cases. The final diagnosis was based on the results of serological investigation. The level of IgG antibodies was established by the ELISA method using the excretory - secretory antigen of Toxocara canis larvae. A few years of studies lead to the observation that after treatment the return of the level of antibodies to normal is a very slow process - positive results are found in 57% of patients even after 4 to 7 years and the decrease of the mean titre of antibodics does not exceed 45%. The observations show that examination of the level of antibodies in the way hitherto applied does not provide a satisfactory indication of the positive effect of treatment. Therefore, it is necessary to develop a system of additional laboratory tests that will altow a faster evaluation of the effects of treatment of this disease.

An antibody (Ab), also known as immunoglobulin (Ig), is a large Y-shape protein consisting of two heavy chains and two light chains (Figure 1) which form a functionally bivalent monomer that is produced by B cells receptor used by the immune system to identify and neutralize foreign objects such as bacteria and viruses. Current widely used rapid detection systems exploit antibodies for recognition, identification and quantification of target analytes [68]. Abs played an important role in the advancement of diagnostic assays, making these indispensable in diagnostic tests that are commonly used routinely in clinics in classical immunological methods such as ELISA, dot blot immunobinding assays, electro-chemiluminescence, flow cytometry and several microscopic techniques like fluorescence, as well as in the construction of numerous immunosensors. Immunoglobulin M (IgM) usually exists as a pentamer in mammals that predominates in primary immune responses to most antigens, and is the most efficient complement fixing immunoglobulin. It comprises approximately 10% of all normal human serum Ig content. IgM is mainly produced by the immune system for protection against numerous viral, bacterial, fungal and parasitic infections.

Imaging and quantification of MAbs distribution by MRI after administration in vivo can improve patient outcomes, although it is not routinely used in the clinic hence the motivation for development of new MAbs imaging methodologies. Improvements in imaging and quantification of antibody distributions can be made by conjugating MAbs to “hotspot” atoms such as 19F as 19F Currently, research investigating the incorporation of 19F to antibody delivery systems has been limited to trastuzumab perfluorocarbon emulsions and this method has not been extended to other clinically approved antibodies. The methodologies of 19F MRI can improve current limitations of antibody immunotherapy such as quantitative visualization of targeted surface antigens expressed on tumor cells.

Opis:
Goose astrovirus (GoAstV) is a novel avastrovirus that typically causes gosling gout and results in 2 to 20% mortality. GoAstV capsid protein is the sole structural protein, which is responsible for viral attachment, assembly, maturation as well as eliciting host antibodies. However, the epitopes within capsid protein have not been well studied. In this study, a monoclonal antibody, named 1D7, was generated against GoAstV capsid protein by hybridoma technology. Western blot results showed that this MAb could react with recombinant capsid protein expressed in E. coli. Also, it recognized the precursor of capsid protein, VP90 and VP70, in GoAstV-infected cells. Besides, excellent specificity of MAb 1D7 was further demonstrated in indirect immunofluorescence assay and immunohistochemical analysis. Epitope mapping results revealed that MAb 1D7 recognized the epitope 33QKVY 36 within Cap protein. Sequence alignment indicated that 33QKVY 36 is a conserved epitope among the isolates of goose astrovirus type 2 (GoAstV-2), suggesting the potential for its use in GoAstV-2 specific diagnostic assay. These findings may provide some insight into a function of the GoAstV capsid protein and further contribute to the development of diagnostic methods for GoAstV infection.

Lipoxygenase was localized in the primary leaves of Phaseolus coccineus (L.), seedlings treated with 25 µM Cd and in control plants using the immunogold method. The enzyme was localized mainly in the peripheral parts of protoplast of control plant cells. It was found in the cell wall, along the ER elements, at plastid lamellae and inside the mitochondria. In Cd-treated seedlings the elements of parenchyma cells showed an atypical inner structure. The immunolabelling of LOX was less intensive in comparison with control. The enzyme was found in the cytoplasm, at the cell wall area, vacuoles and in the plastid stroma as single gold particles. LOX activity optima were determined at pH 7.0 and 8.0 for both linoleic and linolenic acid used as substrates. After 2 days of seedlings exposure to Cd the activity of LOX decreased at pH 7.0 and 8.0 when linoleic acid was used as substrate, and strongly declined at pH 7.0 after 4 days of the metal treatment. When linolenic acid was the substrate LOX activity slightly increased after 2 days of the plants exposure to Cd, but after 4 days it rapidly decreased at pH 7.0. The changes in LOX activity are discussed.

Studies on serum interleukin (IL)-31 levels in dogs with atopic dermatitis (AD) and their correlation with disease severity are limited. To the author’s knowledge, there are no studies that measured serum IL-31 in dogs treated with lokivetmab injections, a selective inhibitor of this key cytokine in pruritus. The aim of the study was to evaluate serum IL-31 levels in dogs treated with lokivetmab and correlate it with the severity of canine atopic dermatitis using the pruritus visual analog scale (pVAS) and canine atopic dermatitis extent and severity index (CADESI-04). Ten client-owned dogs diagnosed with AD received two injections of lokivetmab four weeks apart. Disease severity was assessed using the pVAS and CADESI-04 scores before and after both injections. In addition, canine serum IL-31 levels were measured at the same moments. Serum IL-31 was detected in all dogs in the study. There was a significant reduction in pVAS scores and serum IL-31 after administrations. However, there was no difference in CADESI-04 scores, and there was no significant correlation between CADESI-04 scores and serum IL-31 in dogs diagnosed with AD. Nonetheless, a significant positive correlation was observed between the pVAS scores and serum IL-31 levels with lokivetmab therapy, which reinforces the role of IL-31 in the pathogenesis of pruritus in dogs with AD. The data presented here provide further evidence that IL-31 is directly involved in pruritus pathogenesis in dogs with AD. In addition, blocking IL-31 has a significant antipruritic effect, but has no influence on skin lesion severity and extension.

Opis:
Passive immunity is defined as a particular antigen resistance provided by external antibodies. It can be either naturally or artificially acquired. Natural passive immunization occurs during pregnancy and breast-feeding in mammals and during hatching in birds. Maternal antibodies are passed through the placenta and milk in mammals and through the egg yolk in birds. Artificial passive immunity is acquired by injection of either serum from immunized (or infected) individuals or antibody preparations. Many independent research groups worked on selection, verification and detailed characterization of polyclonal and monoclonal antibodies against the influenza virus. Numerous antibody preparations were tested in a variety of in vitro and in vivo experiments for their efficacy to neutralize the virus. Here, we describe types of antibodies tested in such experiments and their viral targets, review approaches resulting in identification of broadly neutralizing antibodies and discuss methods used to demonstrate their protective effects. Finally, we shortly discuss the phenomenon of maternal antibody transfer as a way of effective care for young individuals and as an interfering factor in early vaccination.

Following genetic immunization of laying ducks with a plasmid expressing Helicobacter pylori UreB (large subunit of urease), IgY against UreB were obtained from egg yolks. These polyclonal and monospecific IgY antibodies are of higher-titer and specifically recognize recombinant H. pylori urease purified from Escherichia coli. To our knowledge this is the first report describing generation of IgY antibodies directed against antigens of H. pylori by DNA-based immunization.

Opis:
Hemagglutinin (HA), as a major surface antigen of influenza virus, is widely used as a target for production of neutralizing antibodies. Monoclonal antibody, mAb6-9-1, directed against HA of highly pathogenic avian influenza virus A/swan/Poland/305-135V08/2006(H5N1) was purified from mouse hybridoma cells culture and characterized. The antigenic specificity of mAb6-9-1 was verified by testing its cross-reactivity with several variants of HA. The mimotopes recognized by mAb6-9-1 were selected from two types of phage display peptide libraries. The comparative structural model of the HA variant used for antibody generation was developed to further facilitate epitope mapping. Based on the sequences of the affinity- selected polypeptides and the structural model of HA the epitope was located to the region near the receptor binding site (RBS). Such localization of the epitope recognized by mAb6-9-1 is in concordance with its moderate hemagglutination inhibiting activity and its antigenic specificity. Additionally, total RNA isolated from the hybridoma cell line secreting mAb6-9-1 was used for obtaining two variants of cDNA encoding recombinant single-chain variable fragment (scFv) antibody. To ensure high production level and solubility in bacterial expression system, the scFv fragments were produced as chimeric proteins in fusion with thioredoxin or displayed on a phage surface after cloning into the phagemid vector. Specificity and affinity of the recombinant soluble and phage-bound scFv were assayed by suitable variants of ELISA test. The observed differences in specificity were discussed.

Opis:
Chlamydophila abortus is one of the most important causative agents of enzootic abortion and other chlamydial infections of sheep and goats. The presence of specifi c serum antibodies to Chlamydophila abortus was studied in sheep and goats breeding in the Slovak Republic by the complement fi xation test. 22,040 sheep and goats were examined during 5 years. Specifi c anti-Ch. abortus antibodies were found in 2,360 out of 20,878 sheep sera examined (11.7%), and in 85 out of 1,162 examined goats (7.7%). The occurrence of antichlamydial antibodies indicates the importance of performing screening examinations in commercial breeding with the aim of reducing the spread of this disease between animals, and also interrupting the spread and transmission from animals to human.

Introduction. Bordetella pertussis is a Gram-negative rod which attacks the respiratory system and causes whooping cough disease. Despite the high percentage of childhood vaccination amongst the population an increasing number of pertussis cases have been observed over time, especially among adolescents and young adults. Objective. The aim of the study was to analyze the concentration of IgG anti-PT Bordetella antibodies in the serum of young adults in the aspect of maintaining immunity. Materials and method. The study group consisted of 30 patients, medical students aged 21–26 years. They were interviewed with a questionnaire, which asked about their general health, medications taken, basic information about whooping cough, or vaccination status against B. pertussis. The anti-PT Bordetella levels were measured in the collected serum by a commercial ELISA kit (enzyme-linked immunosorbent assay). Results. All patients had previously received a mandatory vaccination for pertussis as a child, and 36.6%had received a booster dose after the age of 18. The total average antibody level was 13.977 IU/mL (range: <5.00 – 50.74) and 40% of samples had a very low concentration (<5 IU/mL). Conclusions. Current findings suggest that post-vaccine immunity decreases over time, while infection susceptibility among younger age groups who have not yet been fully immunized has increased. Methods that could prevent and enable risk stratification of whooping cough include the screening test for anti-PT Bordetella antibody, the improvement of diagnostics, and an additional booster dose.

Opis:
Hypericum perforatum is well known for its antidepressant and anti-inflammatory activities, for which hypericin and its derivatives are indicated to be the most active compounds. Hypericin synthase (Hyp-1) is the only protein proven to catalyze the synthesis of hypericin. In this study, the full-length cDNA of Hyp-1 was chemically synthesized according to the Hyp-1 sequence in GenBank (accession no. AY148090) and then cloned into the plasmid pET22b. Hyp-1 was expressed in Escherichia coli BL21 (DE3) and purified with a Ni-NTA column. The purified protein was used to immunize New Zealand white rabbits, from which an antiserum was purified by protein G affinity chromatography. The polyclonal antibody against Hyp-1 provides a valuable tool for the study of hypericin biosynthesis in H. perforatum. Expression of Hyp-1 and the cellular distribution of hypericin were analyzed in different organs of red-pigmented H. perforatum plantlets. The black glands were not the only site of hypericin accumulation and the results indicated that hypericin might be synthesized in mesophyll cells or in tissues of the root and/or stem and then transported to the glands. This work provides a foundation for further investigation of the regulatory mechanism of hypericin synthesis during the development of H. perforatum.

The rough mutants of Gram-negative bacteria are widely used to induce protective antisera but the nature of the target epitope for such antibodies is not precisely defined. Endotoxin is one of several antigens present on the surface of bacterial cells, which are able to elicit specific antibodies. We studied the specificity of antibodies produced against a conjugate of E. coli J5 endotoxin core oligosaccharide with tetanus toxoid. The use of chemically defined antigen for immunisation excludes the possibility of production of antibodies against other cell surface antigens. A comparison of this monospecific anti-endotoxin serum with antiserum against E. coli J5 whole cells was performed in order to distinguish the role that endotoxin core oligosaccharide plays in the interaction with humoral host defences from that of other potentially important Gram-negative bacterial surface antigens. The reactivity of both sera with smooth and rough lipopolysaccharides was determined in ELISA, immunoblotting and by flow cytometry. Both antisera reacted with similar specificity with most lipopolysaccharides of identical or related core type. Less distinct reactions with endotoxins of the antibacterial serum in comparison with the anti-conjugate serum were found in all serological tests. LPS of E. coli O100 that showed the strongest reactions with both sera was used to stimulate IL-6, TNFα and nitric oxide production by the J-774A.1 cell line. Both sera were used to inhibit that stimulation and no inhibitory effects of the examined sera in comparison with non-immune serum were observed.

Human polyclonal antibody (HIgG) was successively labeled with 111In-indium chloride after residulation with freshly prepared cyclic DTPA-dianhydride. The best results of the conjugation were obtained by the addition of solid DTPA-dianhydride (0.1 0.3 mg) to 100 mi l of the HIgG solution (0.2 0.4 mg/ml) at pH = 6 in phosphate buffer media at 25°C with continuous stirring for 30 min. Radio-thin-layer chromatography showed an overall radiochemical yield of 96 99% at optimized conditions (specific activity = 300 500 MBq/mg, radiochemical purity >98%). The final isotonic 111In-DTPA-HIgG complex was checked by radio-TLC to ensure the formation of only one species followed by filtration through a 0.22 mi filter. Preliminary long-term in vivo studies in turpentine-oil induced inflammation in rat model was performed to determine late complex distribution of the radioimmunoconjugate. The target/skin and target/ blood ratios were 27 and 51 after 24 h, and 23 and 51 after 110 h, showing a high selectivity of the radiopharmaceutical for inflammatory lesions.

The practical inability to diagnose Trichinella spiralis antibodies in man before day 20 post infection (dpi) has stimulated interest in the development of immunodiagnostic test to detect circulating antigens. Our previous experience showed that soon after infection immune complexes as well as uncomplexed parasite antigens in sera of infected rats could be detected. To diagnose the presence of antigen in urine, double sandwich-capture ELISA was applied using a peroxidase-conjugated rabbit immunoglobulin to T. spiralis larval antigens. The plates were coated with metabolic (AES) or somatic (AS) larval antigens. Mice were infected with 500 T. spiralis larvae. The urine samples from experimentally infected mice taken from 1 to 41 dpi. and the urine samples from patients of the Clinical Hospital in Białystok taken from 3 to 120 dpi were examined. Before testing, the urine samples were heated for 6 min. at 100°C and centrifuged for 6 min. at 5000 g, supematants were used in ELISA. The presence of T. spiralis antigens in mice urine samples was detected between 6-26 days post infection (dpi) using double sandwich-capture ELISA. All samples taken later were negative as samples taken from uninfected mice. 3 from 9 human urine samples taken 3-10 dpi were positive, the remaining samples taken 3-10 and 10-30 dpi showed values near to "cut-off". In both mice and human urine samples the higher level of antigens was detected in ELISA when somatic larval antigen was used. The T. spiralis antigens were present in urine of infected men and mice in the first phase of infection.

Human cystic echinococcosis (CE), caused by Echinococcus granulosus, is one of the most important and widespread parasitic zoonoses. As one of the problems that can be encountered after treating CE patients is the risk of postsurgical relapses or treatment failure, a long-term clinical and serological follow-up is required to evaluate the success and failure of therapy. Therefore, the aim of the present study was to identify the best diagnostic and prognostic ELISA markers in patients with CE. The cohort comprised 50 patients with symptomatic CE treated with antihelminthic drugs and surgery, who were followed up clinically and radiologically for a mean of 6 years (range 4–8 years). The results clearly indicate that the hydatid specific antibodies of IgE, IgG1 and IgG4 are the most important antibodies for the serological diagnosis of cystic echinococcosis during the active stage of the disease. None of the serum samples from healthy controls gave a non-specific reaction with IgE, IgG1 or IgG4, and a considerably reduced cross-reaction was observed with these antibodies. During post-operative follow-up, the IgM, IgE, IgG1, IgG2 and IgG4 antibody response provided the best correlate of disease activity. The detection of total IgG and IgG3 subclass antibody response for the assessment of post-treatment disease activity among CE patients was insensitive. All patients responded to treatment except 2 women (32 and 36 years old), in whom multiple cysts (12 and 7 cysts) were detected in the liver and lung two years after the first operation. Hence, it can be concluded that the CE-specific antibodies of IgE, IgG1 and IgG4 are the best immunological markers for diagnosis and prognosis of CE patients.

The human leukocyte antigen (HLA) system plays an important role in the acceptance of renal graft. Long and better graft survival has been reported in patients with HLA-identical siblings and a nonreactive cytotoxicity assay (CDC). New methods of HLA-typing and anti-HLA antibody detection techniques such as flow cytometry, solid-phase immunoassays, or antigen bead assays have further improved the outcomes of renal transplant recipients. In the present review, the explicit details of these methodologies are discussed in detail.

The aim of the study was to investigate the occurrence of serum antibodies to C. psittaci in workers at 8 stock farms located in a rural area of eastern Sicily. Serum samples from 188 workers and 160 controls were tested for immunoglobulin IgA, IgG and IgM direct against C. psittaci by microimmunofl uorescent test (MIF). Seropositive subjects were defi ned as those with IgG titres of 1:16-1:256, and seronegative subjects as those whose titre was <1:16. To rule out the presence of cross reactive antibodies, testing was also performed for C. trachomatis and C. pneumoniae. A total of 28 (14.9%) subjects studied met the criteria for seropositivity to C. psittaci. Two of the 188 (1.06%) had an IgG titre of 1:32, 16 (8.51%) an IgG titre of 1: 64, 6 (3.19%) an IgG titre of 1:128 and 4 subjects (2.12 %) demonstrated an antibody titre of 1:256. The high prevalence rate of C. psittaci antibodies among farmers suggests that this infection is spread in those subjects living in areas with an high percentage employed in rearing activities. The authors stress the importance of carrying out health surveillance in subjects working in close contact with animals receptive to infection, and confirm the need to adopt a serological test, such as MIF as a preventive measure for activities at risk.

In the course of trichinellosis in mice the numerous cells inside larval capsules were observed. Beginning on 14 day post infection (dpi) they were seen in about 7% of infected muscles fibers but at 21 dpi the percentage of capsules with the cells amounted to 77. In the next stage of infection the number of capsules containing cells continued to increase and reach almost 100% at 60 dpi. The identification of the cells was carried out by the indirect immunoenzymatic method using anti-CD4⁺, anti-CD8⁺ and anti-macrophage (Mac-1 antigen) monoclonal antibodies. In course of observation the level of CD4⁺ cells decreased from 11.3 to 0.7, of CD8⁺ from 17.7 to 3.0 and of macrophages from 71.0 to 23.1 %. The most numerous CD4⁺ cells were seen at 14 dpi while the CD8⁺ and macrophages at 21 dpi. Beginning on 28 dpi negatively reacting cells inside larval capsules were also observed. The number of these cells increased from 23.8 to 73.2% at 60 dpi and their identification requires further investigations. The role and the function of all these cells are discussed.

Experimental evidence from the recent years indicates that low-level irradiations with X- or gamma rays may inhibit development of both primary and secondary tumours and stimulate the activity of natural anti-tumour immune mechanisms. Natural killer (NK) cells play an important role in anti-tumour defence of the host. In the present investigation cytotoxic activity, production of interferon-ă, and expression of the Fas ligand (FasL) were estimated in the NK splenocytes collected from BALB/c mice whose whole body was pre-exposed to irradiation with 0.1, 0.2, or 1.0 Gy X-rays. The results indicate that cytotoxic activity of the irradiated NK cells was significantly stimulated compared to that of the NK effectors obtained from the sham-exposed mice. This effect was totally abrogated by injection of the anti-asialo GM1 antibody. In addition, compared to the control mice, NK cells obtained from the irradiated animals exhibited reduced surface expression of FasL. Collectively, the obtained results suggest that the inhibitory effect of the low-level irradiations with X-rays on the development of pulmonary tumour nodules may be directly associated with stimulation by such exposures of anti-neoplastic functions mediated by NK cells.

Opis:
The aim of this paper is to study periodic solutions of Marchuk's model, i.e. the system of ordinary differential equations with time delay describing the immune reactions. The Hopf bifurcation theorem is used to show the existence of a periodic solution for some values of the delay. Periodic dynamics caused by periodic immune reactivity or periodic initial data functions are compared. Autocorrelation functions are used to check the periodicity or quasiperiodicity of behaviour.

Opis:
The laboratory diagnostics of T. gondii infections both in humans and animals relies mainly upon detection of specific antibodies. We studied the influence of the host genetic factors on the level and repertoire of antibodies produced in Toxoplasma infection employing as an experimental model two inbred mouse strains with different innate susceptibility to toxoplasmosis. Using polyvalent antigen preparations in ELISA and microagglutination we found no differences in the antibody levels in both strains. By comparison of the antibody profiles in immunoblot we determined mouse strain-specific and common toxoplasmosis markers.

Opis:
Sposób izolacji przeciwciał z surowicy w znacznym stopniu wpływa na ich jakość, a tym samym może wpływać na jakość zestawów diagnostycznych. Istnieje wiele sposobów wyodrębniania przeciwciał z surowicy (chromatografia jonowymienna na DEAE celulozie, chromatografia powinowactwa z białkiem A, itd.). Różnią się one pomiędzy sobą m.in. efektywnością, czasem wykonania oraz kosztem. Niniejsza praca przedstawia chromatografię tiofilną jako tańszą i wydajniejszą alternatywę dla innych metod uzyskiwania przeciwciał z surowicy króliczej.
A procedure used for purification of antibodies from serum has a significant effect upon their quality and therapy their suitability for diagnostic tests. The commonly used approaches include: (i) ion-exchange chromatography (both on anion and cation exchangers, depending on conditions applied) and (ii) affinity chromatography (on resins charged with the protein A or with the specific antigen). These methods differ in efficacy; amount of time needed and costs for antibodies purification. The study presented herein estimates the suitability of thiophilic chromatography as a cost and time-saving alternative approach for purification of antibodies from rabbit serum.

In the course of trichinellosis in mice the numerous cells inside larval capsules were observed. Beginning on 14 day post infection (dpi) they were seen in about 7% of infected muscles fibers but at 21 dpi the percentage of capsules with the cells amounted to 77.
