22 representative antibiotics, including 8 quinolones (QNs), 9 sulfonamides (SAs), and 5 macrolides (MCs) were selected to investigate their occurrence and removal efficiencies in a Wastewater Treatment Plant (WWTP) and their distribution in the receiving water of the Chaobai River in Beijing, China. Water quality monitoring was performed in an integrated way at different selected points in the WWTP to explore the potential mechanism of antibiotics removal during wastewater treatment. Water quality of the Chaobai River wa s also analyzed to examine environmental distribution in a river ecosystem. The results showed that within all the 22 compounds examined, 10 antibiotics were quantifi ed in wastewater influent, 10 in effluent, and 7 in river. Sulfadiazine (SDZ, 396 ng/L) and Sulfamethazine (SMZ, 382 ng/L) were the dominating antibiotics in the influent. Both the conventional treatment and advanced Biological Aerated Filter (BAF) system was important for the removal of antibiotics from the wastewater. And the concentrations of selected antibiotics were ranged from 0–41.8 ng/L in the effluent-receiving river. Despite the fact that the concentrations were reduced more than 50% compared to effluent concentrations, WWTP discharge was still regarded as a dominant point-source input of antibiotics into the Chaobai River.

This review article describes the results of a 15-year cooperation between the Department of RNA Biochemistry at the Institute of Bioorganic Chemistry, Polish Academy of Sciences in Poznań and the Medical Chemistry Team of the Faculty of Chemistry at the University of Wrocław, headed by Professor Małgorzata Jeżowska- -Bojczuk. A wide spectrum of antibiotics and other low molecular compounds and their complexes with Cu2+ ions have been tested as potential inhibitors of the HDV ribozyme catalytic reaction. Unexpectedly, it has been found that a number of compounds, depending on the conditions, exhibit inhibitory or stimulatory properties, i.e. they act as modulators of the RNA catalysis process. It was found that the effect of stimulation / inhibition of the catalytic activity of the HDV ribozyme is closely related to the degree of protonation of the antibiotics under study in given conditions. Their ability to inhibit catalysis also increases after binding the Cu2+ cation. In an environment with a higher pH, antibiotics usually stimulate the cleavage reaction, as at least some of their nitrogen centers are allowed to participate in the catalysis reaction, as proton acceptors / donors or a catalytic metal ion coordination site. During the study of one of the antibiotics, bacitracin, it was also observed that it exhibits nucleolytic properties with RNA and DNA molecules. The discovery of the hydrolytic properties of bacitracin extended the potential use of this antibiotic in antiviral therapy with the aim to destroy undesired nucleic acids in the cell. To search for DNAzymes catalyzing RNA hydrolysis, the in vitro selection method was used. In the selection experiment aimed at obtaining DNAzymes active in the presence of Cd2+ ions, variants belonging to the family of DNAzymes 8–17 previously described in the literature were obtained. Analysis of their properties showed that not only Cd2+ but also Zn2+ and Mn2+ ions support catalysis, therefore the site of catalytic metal ion coordination is not highly specific. The DNAzymes obtained in the second selection experiment showed an optimum of catalytic activity in the pH range of 4.0–4.5 and were inactive at a pH higher than 5.0. Interestingly, they do not require the presence of any divalent metal ions as cofactors in the catalysis reaction. The obtained results broaden the repertoire of DNAzymes which operate under non-physiological conditions and bring new information on the possible mechanisms of reactions catalyzed by nucleic acids.

The following article reviews the issue of antibiotic resistance of microorganisms to meropenem in intensive care units in Ukraine. An increase in meropenem inefficiency against microorganisms in intensive care units has been observed in the last years. The data analysis suggests a significant predominance of gram-negative flora: A. baumannii, Р. aeruginosa, Antimicrobial resistance happens when microorganisms change when they are exposed to antibiotics. Then, treatment becomes ineffective and infections persist in the body, increasing the risk of spreading to other persons. The new resistance mechanisms that are emerging and spreading globally cause that the so far applied methods of treatment do not work, threatening the human ability to resist common infectious diseases, which in turn results in prolonged infections or even death. Antimicrobial resistance occurs naturally over time, usually through genetic changes. However, the misuse and overuse of antimicrobials are accelerating this process. It has become common to overuse and misuse antibiotics both in people and animals, which are often prescribed without professional oversight. Antimicrobial resistance is a complex problem that affects all of society and is driven by many interconnected factors. Single, isolated interventions have limited impact. Coordinated action is required to minimise the emergence and spread of antimicrobial resistance.
Ze zjawiskiem antybiotykooporności mamy do czynienia w przypadku gdy bakteria nabierze oporności na dany antybiotyk. Stosowane wówczas leczenie staje się nieskuteczne a obecność samego drobnoustroju w ciele chorego może przyczynić się do jego rozpowszechnienia na inne osoby. Nowe mechanizmy nabywania oporności na antybiotyki sprawiają, że dotychczasowe sposoby leczenia wielu chorób zakaźnych przestają być skuteczne, czego następstwem są trwające znacznie dłużej zachorowania a nawet śmierć. Zjawisko nabywania oporności nie jest czymś nowym, gdyż wynika z naturalnych właściwości przystosowawczych bakterii jednakże niewłaściwe lub nadużywanie antybiotyków zarówno u ludzi jak i zwierząt znacznie przyspiesza ten proces. Problem antybiotykooporności stał się problemem ogólnoświatowym, dotyczącym wszystkich grup społecznych. Ze względu na złożoność problemu potrzebne są skoordynowane działania gdyż tylko takie mogą przyczynić się do zminimalizowania rozprzestrzeniania tego niekorzystnego zjawiska.

The „golden age” of antibiotics seems to be coming to an end. The effect of uncontrolled distribution of chemotherapeutic agents in animal production is the commonly growing phenomenon of dramatic drug resistance in bacterial populations. At the end of the 20th century, the repeated failures to effectively control of bacterial infections have initiated the introduction of legislative procedures to limit the use of antibiotics, especially as feed additives in animals. The next step was the developing of alternative for antibiotics to control bacteria, with particular emphasis on natural substances as extracts and oils obtained from plants, which have been used against Gram – positive and Gram – negative bacteria, as well as Candida fungi. These are cationic peptides stabilized by cysteine (defensins and cathelicidins) – identified both in prokaryotes and eukaryotes. Other examples are the nanoparticles, especially silver, gold or platinum. The use of bacteriophages as natural anti-bacterial agents, which bactericidal effect has been demonstrated in numerous experimental therapies in humans and animals, also deserves special emphasis. In this article scientific approaches to novel antibacterial therapies were presented and briefly discussed. As a consequence of the threat of widespread and global growth of multi-resistant bacteria there is a strong need to develop of alternative for antibiotics in elimination of multidrug resistant pathogens.

Antiseptics are a group of substances widely used in ophthalmology for many years. They are well established, especially in the prevention of perioperative infections. For decontamination in ophthalmology, a 5% povidone-iodine solution is used as a standard for the conjunctival sac. It is considered the most effective preoperative antisepsis. In patients with an allergy to iodine, a solution of 0.05% chlorhexidine is recommended. There are also attempts to use other antiseptics and antibiotics for perioperative prophylaxis. Recently, there has also been interest in potential use antiseptics for the treatment of conjunctivitis.

Metal ions are essential for numerous antibiotics. They play a crucial role in the mechanism of action and may be involved in specific interactions with cell membrane or target molecules, such as: proteins and nucleic acids. Due to the fact that complexes usually poses a higher positive charge than free ligands, they might interact more tightly with DNA and RNA molecules. However, complexes may also form during antimicrobial agents application, because a lot of them possess functional groups which can bind metal ions present in physiological fluids. Many recent studies support a hypothesis that drugs may alter the serum metal ions concentration. Moreover, it has been shown that numerous complexes with antibiotics can cause DNA degradation, e.g. bleomycin which form stable complexes with redox metal ions and split the nucleic acids chain via the free radicals mechanism. Therefore, it is widely used in cancer therapy.

Every year there has been a growing increase in infections caused by strains of bacteria resistant to multiple drugs. This prompts scientists to search for new antibiotics that would be able to fight these infections. New therapeutics used in medicine, which offer greater hopes are nucleoside antibiotics. They represent a large family of natural compounds exhibiting a variety of biological functions [1]. These include antifungal, antiviral, antibacterial, insecticides, immunosuppressants or anticancer properties. These broad-spectrum antibiotics can be divided into three main groups: • antibacterial nucleoside antibiotics, responsible for the inhibition of bacterial translocation of phospho-N-acetylpentapeptides, involved in the biosynthesis of peptidoglycan cell wall of bacteria; • antifungal nucleoside antibiotics, which role is to inhibit chitin synthase, or stopping construction of the cell wall of fungi; • antiviral antibiotics nucleoside, their mechanism of action is mainly based on blocking the biosynthesis of proteins by peptide inhibition transferase. In recent years much attention has been focused on the construction, mechanism of action and biosynthesis of antibiotics [1–3]. The development of genetic engineering has opened the way for combinatorial biosynthesis and obtaining new or hybrid compounds. In this work we would like to discuss some of bioactive naturally occurring nucleoside antibiotics, such as tunicamycin (Fig. 6) [19–22], mureidomycin (Fig. 8) [31–34], muramycin (Fig. 9) [36] or capuramycin (Fig. 10) [38].

Due to constantly increasing antibiotic resistance of pathogens and participation of the biofilms they make in various types of infections, a development of alternative therapeutic strategies becomes an urgent need. Taking advantage of the biological activity of plant-derived compounds can solve this problem. In this study antimicrobial, including those synergistic with classic antibiotics, and cytotoxic properties of newly-obtained extracts from Vaccinium myrtillus leaves (VLE) and Frangula alnus bark (FBE) were evaluated. Both tested extracts exhibited relevant antistaphylococcal activity (MIC range 0.75-1.5 mg/mL) accompanied by a relativly low cytotoxic effect on mammalian cells (BI > 1). Phytochemical analysis of the extracts tested showed a high total content of phenolic compounds with the predominance of hydroxycinnamic acids in VLE and hydroxybenzoic acids and flavanols in FBE. Widely described in the literature antimicrobial properties of phenolics were probably connected with the biological activity of the extracts tested. We also report that the presence of VLE or FBE at sub-MIC concentrations enhances biocidal potential of vancomycin and linezolid. Therefore, we are considering a possibility of an alternative therapy for local infections caused by S. aureus by combining classic antibiotics with plant-derived extracts.

Mastitis is an increasing problem in dairy farms and in the dairy industry. The paper presents changes in susceptibility of strains of bacteria isolated from quarter milk cows from mastitis in 2011, 2012, 2013, 2014, 2015 in the Factory Laboratory of Dairy Cooperative in south-eastern Poland. In total, 3409 strains of staphylococci, 2774 strains of streptococci and 423 strains of E. coli were isolated in these years. Susceptibility to antibiotics was examined by the disc diffusion method on Mueller-Hinton base. On the basis of the research it can be concluded that the highest sensitivity of staphylococci in all study periods is found for cefguin (from 91.2% to 96.1%) and cefapirin (92.2% to 96.2%), and the smallest for tetracycline (18 %), streptomycin (11.9%) and amoxicillin (9.3%). The highest sensitivity of bacteria from the Streptococcus family are cefapirin (97%), cefquinome (96.1%), amoxycycline (95.6%) and cefalexin (89.9%) on average in all years of research. The highest resistance was found for neomycin and streptomycin. They show that during the period under study over 90% of Escherichia coli were sensitive to streptomycin. Sensitivity at the level of 70%-80% was noted for amoxicillin, neomycin and cefapirin. The highest resistance of E. coli bacilli evolved for cefallaxin and tetracycline. Accordingly, early diagnosis of a pathogenic pathogen, and then selecting the appropriate form of treatment, gives you the opportunity to minimize the loss of mastitis in dairy cows.

Introduction: Studies based on the ISAAC questionnaire suggest a correlation between the use of antibiotics and the prevalence of asthma and allergy in children aged 6-7 years. The number of courses of antibiotic therapy is an important factor. Objectives: To investigate the relationship between the use of antibiotics during the fi rst years of life and the prevalence of allergy and asthma among children (aged 6-8 years) in the urban population of Poland. Materials and Methods: A survey-based study with a self-completed questionnaire. The respondents were parents of children aged 6-8 years living in Warszawa, Poland. 1461 completed questionnaires were collected. Results: Asthma was declared in 4.3% of the children. Wheezing and/or sibilant rhonchi within 12 months before the study was observed in 13.5% of the cases. Asthma medication was taken by 21.8% of the children. Allergic rhinitis was declared in 18.7% of the children. Problems with sneezing, rhinorrhea, and nasal congestion not associated with cold or fever were observed in 40.7% of the children. The a nalysis of the odds ratios between the use of antibiotics and the symptoms of allergic diseases revealed a clear correlation. The highest odds ratio was observed between the completion of over three courses of antibiotic therapy prior to the age of 12 months and the declaration of one of the following: asthma (OR = 5.59, 95% CI: 2.6-12.01), wheezing and/or sibilant rhonchi (OR = 4.68, 95% CI: 3.01-7.27) and taking medicines for breathlessness (OR = 5.12, 95% CI: 3.42-7.68). Conclusions: There is a direct relationship between antibiotic use in the fi rst 3 years of life and asthma and allergy symptoms in children aged 6-8 years old.

Nontuberculous mycobacteria (NTM) have recently emerged as important bacterial pathogens of both animals and humans. In this study, we aimed to evaluate the effect of a combination of ten antibiotics with an inhibitor of efflux pumps (EPI), i.e. berberine (BER), against 6 strains of NTM. Our results showed that the BER potentiated the anti-mycobacterial activities of the antibiotics. Overall, our findings show the importance of BER in increasing the efficacy of antibiotics in NTM.

The most common cause of ophthalmological consultations in an ophthalmological clinics are conjunctivitis, often caused by bacteria. The main form of its treatment is topical antibiotic therapy. The paper presents the current possibilities of treatment of conjunctivitis, with particular emphasis on the problem of development of the antibiotic resistance and the practical principles of antibiotic therapy. In the light of current knowledge, the best results in the treatment of bacterial conjunctivitis are achieved by the use of IV- and then III-generation of fluoroquinolones. These drugs have the best spectrum of action, and additionally, they have a very good penetration into the conjunctiva and are at the lowest risk of developing antibiotic resistance.
Najczęstszą przyczyną zgłaszania się pacjentów do poradni okulistycznej są zapalenia spojówek, których przyczyną często są bakterie. W takim przypadku podstawową formą leczenia jest miejscowa antybiotykoterapia. W pracy przedstawiono aktualne możliwości leczenia tych schorzeń, ze szczególnym uwzględnieniem problemu rozwoju antybiotykooporności. Szczególny nacisk został położony na praktyczne zasady antybiotykoterapii. W świetle aktualnej wiedzy najlepsze efekty w terapii bakteryjnego zapalenia spojówek daje zastosowanie fluorochinolonów IV, a następnie III generacji. Leki te mają najlepsze spektrum działania, a dodatkowo odznaczają się bardzo dobrą penetracją do spojówki i są obciążone najmniejszym ryzykiem rozwoju antybiotykooporności.

Urinary tract infections (UTI’s) are a bacterial infection that affects any part of the urinary tract. Urinary tract infection is one of the major diseases that affect people of all age groups and sexes and can be separated into asymptomatic and symptomatic. The normal urinary tract is sterile but gets infected with the normal flora and opportunistic pathogens. Urinary tract infections (UTIs) are a serious health problem affecting millions of people each year. This is the second most common type of infection in the body. In the present study 20 morphologically different bacterial isolates were recovered from 50 different urine samples from UTI’s patient. The prevalence of the isolates was varied with E. coli (40%), followed by Staphylococcus spp., Pseudomonas spp. and Klebsiella spp. each of (20%) respectively. All the isolates of different bacteria were tested for antibiotic sensitivity patterns against common antibiotics viz. Ampicillin, Tetracycline, Chloramphenicol, Vancomycin, Nitrofurontione and Ciprofloxacin with potencies of 30 μg each. The sensitive pattern of the isolates towards antibiotic Nitrofurontoine was (75%) followed by Tetracycline (55%) and Ciprofloxacin (45%). Intermediate activity was reported against Ampicillin (45%), Chloramphenicol and vancomycin (40%). Resistant pattern shown against vancomycin (60%), Chloramphenicol (45%), while both tetracycline and Ciprofloxacin shown (30%) against the UTI’s isolates. Therefore, drugs like Nitrofurontoine, Tetracycline and Ciprofloxacin are the most effective choices against the common UTI’s isolates.
