In this study, Aspergillus oryzae Β galactosidase was immobilized on concanavalin A layered calcium alginate-cellulose beads as a bioaffi nity support. Immobilized enzyme showed a remarkable broadening in temperature-activity profi les as compared to the native enzyme and exhibited 65% activity in the presence of 5% galactose. Michaelis constant (Km) was 2.57 mM and 5.38 mM for the free and the immobilized Β galactosidase, respectively. Crosslinked Β galactosidase showed greater catalytic activity in the presence of Mg2+ and was more stable during storage at 4°C for 6 weeks. Immobilized enzyme hydrolyzed 67% lactose in milk in 8 h and 85% lactose in whey in 9 h in the stirred batch process at 50oC. The continuous hydrolysis of lactose by crosslinked Β galactosidase in spiral bed reactor exhibited 93% and 88% hydrolysis of lactose at flow rate of 20 ml/h and 30 ml/h, after 1 month operation, respectively.

Mining effluents contain cobalt ions that can damage humans and flora. However, this meta also has high commercial value when recovered. The objective of this research work was to recover cobalt (Co2+) from diluted solutions using a biosorbent, specifically protonated dry alginate beads (PDAB). Experimental work was carried out in batch from an initial concentration of 22×10-6 kg dm-3 Co2+ and 80 mg alginate. Variables such as agitation, pH solution, experimental time, isotherm values, and temperature were analyzed. Maximum cobalt recoveries were obtained at pH values above 5.0, reaching 60.6×10-3 kg kg-1 of PDAB. Cobalt recovery occurred with ion exchange mechanisms from alginate carboxyl group proton release. Experimental data had excellent fit with both the Lagergren kinetic model (pseudo-first order) and the Langmuir isotherm model. As temperature increased, cobalt recovery increased. The calculated activation energy was 12.8 kJ mol-1. Compositional measurements obtained by scanning electron microscope and energy-dispersive X-ray spectroscopy for alginate crosssections showed uniform distributions of cobalt concentrations throughout the spherical alginate structure, independent of solution pH, contact time, or temperature. Furthermore, elution gave significant cobalt re-extraction (98.2%) and demonstrated PDAB reusability.

This research aimed to identify the copper ion removal mechanism when using protonated dry alginate beads. This mechanism was explained through ion exchange between Cu ions and the protons from the functional groups of the alginate beads. Copper removal increased with stirring velocity, reaching values of 97.5 mg g-1 (97.5×10-3 kg/kg of PDAB) of dry alginate at 200 rev min-1, at a solution pH of 6.0 and a run time of 360 min. For the lowest level of copper concentrations, at 10 mg dm-3 (10×10-6 kg dm-3), full removal was attained. The removal kinetics was represented by a pseudo- first order model. A value of 0.0131 min-1 was found for the velocity constant. Under equilibrium conditions, the experiment data was fit to the Langmuir adsorption model, and the highest removal values were 270.3, 222.2 (222.2×10-3 kg/kg of PDAB) and 49 mg g-1 (49×10-3 kg/kg of PDAB) for pH values of 5.0, 3.5 and 2.5, respectively. These are higher than most sorbents used in the literature for copper removal. Increased temperature leads to higher Cu removal. The activation energy was calculated at 9.3 kJ mol-1 for the temperature range of 283 to 343K. Observations using SEM and composition measurements of the alginate cross-section taken by EDS showed a uniform distribution of the copper concentration through the structure of the alginate beads, independent of the solution pH, contact time and temperature.

This study evaluates the feasibility of using tamarind (Tamarindus indica) seed powder for de-fluoridation of fluoride contaminated water. Batch study confirmed that tamarind seeds in dry powder form could remove 87% of fluoride from water. This bio-sorbent can be used effectively in areas where fluoride concentrations are above the permissible limits of 1.5 mg·l-1 as per WHO Standard, 1984. Tamarind seed powder was incorporated in a matrix of sodium alginate and made into gel-beads. The beads were tested for de-fluoridation efficiency by conducting column studies. The effect of various factors, such as flow rate, retention time, and the number of runs on the efficacy of fluoride removal was also studied. The results revealed that flow rate did not seem to have much effect on the percentage fluoride removal but the fluoride concentration decreased drastically upon greater retention time and multiple runs.

The effect of alginate treatments with or without salicylic and oxalic acid as post-harvest coating in extending the postharvest life of plums (Prunus salicina L. cv. ‘Black amber’) and maintaining their quality were investigated. Plums were treated with 2% alginate coating with or without salicylic (1.0 mM) and oxalic acid (1.0 mM), and then stored at 0−1°C and 90 ±5% relative humidity for 40 days. The quality of plums was assessed at 10-day intervals by evaluating the following quality parameters: weight loss, soluble solids content, titratable acidity, firmness, respiration rate, ascorbic acid content, total anthocyanin content, total phenolic content and antioxidant activity. The respiration rate, weight loss and changes in quality parameters were much lower in coated plums as compared with the control. Alginate coating resulted in a significant reduction in weight loss of fruits. Alginate treatments with or without salicylic and oxalic acid were effective on delaying the evolution of parameters related to postharvest ripening, such as soluble solids content, softening and reducing respiration rate. At the end of the storage period, the edible coatings showed a positive effect on maintaining higher concentration of total phenolics, total anthocyanin content and antioxidant activity, which decreased in control plums as a result of over-ripening and senescence processes. The results suggested that the use of alginate enriched with salicylic acid could maintain considerably higher quality of fruits and level of bioactive compounds than other coating treatments during 40 days of storage at 0−1°C.

The study examines the physical and chemical description of magnesium alginate obtained from the brown alga Cystoseira barbata, existing in the coastal area of the Black Sea. We have separated a magnesium alginate by a specific method, which involves the precipitation of the alginate with an alcohol solution of magnesium chloride. The study is focused on three directions: determining the concentration of certain metals, confirming the structure of the alginate type through IR spectral analysis and the examination of the ratio of fixation of magnesium on the alginate. Results confirmed the alginate type structure as well as the presence of certain metals, among which Fe, Cd, Mn, which are to be found in the alga. The presence of Mg is to be noticed, which was fixed on the alginate during the process of obtaining the alginates. The magnesium was fixed in the ratio approximately 80% from the added magnesium quantity in the phase of precipitation of the magnesium alginate.

Alginate encapsulation is a viable alternative for the preservation of probiotics along the gastric route or within a food product during its shelf life. Furthermore, co-encapsulation with a vegetal material could act as a prebiotic and enhance the viability of the encapsulated probiotic. The rheological properties of dressing-type foods could be altered by adding an ingredient that would affect the quality of the final product. In this investigation, alginate beads loaded with Lactiplantibacillus plantarum and beetroot extract were obtained by two methods (emulsification and extrusion). They were characterized by size and morphology, encapsulation efficiency, and bacteria viability under simulated gastrointestinal conditions. Finally, they were added in an oil-in-water emulsion model food for which rheological properties and probiotic survival were monitored. The encapsulation efficiency ranged from 85.26 to 88%. Morphology and size of capsules varied depending on the method of encapsulation applied. No significant changes were evidenced in the rheological properties of the model food; the viscosity, the particle size (d3,2), and the coalescence rate remained stable after the addition of the capsules. Survival of L. plantarum was significantly higher in the capsules with beetroot extract. These results suggest a prebiotic effect conferred by the beetroot extract when co-encapsulated. It is worth mentioning that the incorporation of capsules with beetroot extract does not cause any destabilization of the model food.

Opis:
Modification of commercial cellulose products commonly used in homes and hospitals was the main purpose of the research herein presented. Enhanced moisture sorption and antimicrobial properties were conferred upon regular commercial gauze. Nanoparticles of microcrystalline chitosan and complex chitosan/alginateNa/Ca were used in the modification. A method was elaborated for the preparation of polymeric materials with a particle size below 1 μm by means of an ultrasonic reactor (Hielscher UP 200S). Modified commercial dressing materials were obtained charecterised by a largely increased absorption capacity, thus easing the transportation of moisture to the outside of the dressing and providing an environment optimal for wound healing. Thanks to the internal surface developed and adequately selected composition, the modified cellulosic materials exhibit antibacterial and antifungal properties.
Głównym celem przeprowadzonych badań była modyfikacja komercyjnego materiału celulozowego w postaci gazy opatrunkowej, stosowanej w codziennym użytku jak i w szpitalach, celem nadania im zwiększonej zdolności sorpcji wilgoci jak i właściwości przeciwmikrobowych. Do modyfikacji surowca wykorzystano nano-cząsteczki mikrokrystalicznego chitozanu oraz kompleksu chitozan/alginanNa/Ca. Metodę otrzymywania polimerów o rozmiarach cząstek o wielkości poniżej 1 μm opracowano przy wykorzystaniu reaktora ultradźwiękowego. Otrzymano modyfikowane handlowe materiały opatrunkowe o kilkukrotnie wyższej zdolności sorpcyjnej, które ułatwiają odprowadzanie wilgoci na zewnątrz opatrunku i zapewniają optymalne środowisko gojenia się ran. Modyfikowane materiały celulozowe dzięki rozwiniętej powierzchni wewnętrznej i odpowiednio dobranym składzie charakteryzują się właściwościami przeciwbakteryjnymi oraz przeciwgrzybowymi.

The main objective of the work was to design and fabricate an injectable biomaterial with osteoconductive properties that can be used in dental applications in peri-implant therapy concerning guided bone regeneration. For that purpose, a self-setting biomaterial consisting of chitosan/tricalcium phosphate microparticles and sodium alginate was formulated. The obtained material was characterized regarding microsphere and formed agglomerates morphology and microstructure. Physical properties, relating to setting time and mechanical properties, were also investigated. Finally, in vivo response to implanted biomaterial was studied on a rat model and compared with commercially available alloplastic material. The results showed that designed injectable biomaterial fulfi lled main requirements for guided bone regeneration application.

Przeprowadzono studia porównawcze nad możliwością zastąpienia w badaniach adhezji bakterii mlekowych modelu kultury komórek nabłonka jelitowego innymi uproszczonymi modelami. Badano przyczepność bakterii probiotycznych Lactobacillus casei Shirota ATCC 39539, Lactobacillus acidophilus LC1 oraz Lactobacillus rhamnosus GG ATCC 53103 do membran wykonanych z alginianu, karagenu, żelatyny, kolagenu, szkła, polistyrenu i karboksymetylocelulozy. Zaobserwowano największy stopień przyczepności bakterii probiotycznych do membran wykonanych z karboksymetylocelulozy i kolagenu. W większości wariantów doświadczeń liczba przyczepionych komórek bakteryjnych wzrastała wraz ze wzrostem czasu ich kontaktu z powierzchnią stałą. Wykazano także różnice w przyczepności komórek do hydrożeli o różnych stężeniach. Na podstawie przeprowadzonych badań można stwierdzić, że stopień adhezji bakterii probiotycznych do różnych powierzchni organicznych i nieorganicznych jest cechą zaleną od szczepu mikroorganizmu. Żaden z prostych modeli adhezyjnych, zastosowanych w tej pracy, nie charakteryzował się adhezyjnością porównywalną z komórkami nabłonka jelitowego Caco-2 i HT-29, stosowanego obecnie jako główny model in vitro do badań adhezji bakterii probiotycznych.
Comparative studies were performed on the possibility of replacing intestinal epithelial cells by some other simplified models for the purpose of examining the adhesion of probiotic bacteria. The adhesion of Lactobacillus casei Shirota ATCC 39539, Lactobacillus acidophilus LC1, and Lactobacillus rhamnosus GG ATCC 53103 to surfaces of alginate, carrageenan, gelatin, collagen, glass, polystyrene, carboxymethylcellulose, and to intestinal epithelial cells (Caco-2 and HT-29 lines) were tested. Among the examined solid surfaces, the highest degree of adhesion was observed in the case of carboxymethylcellulose and collagen. In the majority of experimental variants, the number of bacterial cells adhering to surfaces increased parallel to the time of their contacting a solid surface. Additionally, there were stated differences in the cell adhesion to hydrogels showing varying concentration rates. On the basis of the experiments performed, it is possible to state that the adhesion degree of probiotic bacteria to various inorganic and organic surfaces is a parameter that depends on the strain of a microorganism. Among all the simplified models applied in the studies in question, no one was marked by an adhesion rate comparable to the adhesion of intestinal epithelial cells Caco-2 and HT-29, which are, nowadays, used as a major model in vitro while investigating the adhesion of probiotic bacteria.

Opis:
The article presents an analysis of the market potential in Poland for different applications of medical products made of alginate fibres. Emphasis is especially put on wound dressings; however, such issues as tissue engineering or alginate fibres for other specific medical use are also considered. A SWOT (Strenghts-Weaknesses-Opportunities-Threats) analysis of the Polish market for medical products made of alginate fibres, a comparison of prices of different wound dressings produced with the use of such fibres, and predictions for the future for such products in Poland are presented, as well as an analysis of the own research of the authors of the market potential of specialist alginate wound dressings conducted among Polish surgeons during the 65th Congress of the Polish Surgeons' Association.
Artykuł prezentuje analizę potencjału rynku w Polsce dla różnorodnych zastosowań produktów medycznych zrobionych z włókien alginianowych. Szczególny nacisk jest położony na opatrunki, jednak produkty związane z takimi zagadnieniami jak inżynieria tkankowa czy włókna alginianowe dla innych specjalistycznych zastosowań są również rozpatrywane. W artykule zaprezentowana jest analiza SWOT (Silne/Słabe strony, Możliwości/Zagrożenia - ang. Strengths-Weaknesses-Opportunities-Threats) polskiego rynku dla produktów zrobionych z włókien alginianowych, a także porównanie cen różnych opatrunków wyprodukowanych z takich włókien, pewne przewidywania na przyszłość dla takich produktów w Polsce oraz analiza badań własnych autorów potencjału rynku dla specjalistycznych opatrunków alginianowych.

Recently, the attention has been drawn to complex systems – biomicroconcretes composed of a bone cement matrix and resorbable granules or microspheres. This paper presents novel bone substitutes composed of α-tricalcium phosphate (α-TCP; cement matrix), calcium sulphate dihydrate granules (GCSD; aggregates in biomicroconcrete) and various polymers (chitosan, sodium alginate, methylcellulose) used for the improvement of material properties. The aim of this work was to study α-TCP-GCSD-polymer interactions and to compare the impact of organic additives on the physicochemical properties of biomicroconcretes. Methods: Scanning electron microscopy (SEM), mercury intrusion porosimetry (MIP), X-ray diffractometry (XRD) as well as universal testing machine (INSTRON), Gilmore apparatus and pH/ conduct-meter were used. Results: The chemical bonding between α-TCP matrix and CSD granules resulted in a compressive strength appropriate for low-load bearing applications (7–12 MPa) and clinically relevant setting times (8–33 min). Biomicroconcretes consisting of sodium alginate possessed the highest mechanical strength (12 ± 2 MPa). It has also been found that the dissolution-precipitation reactions of the α-TCP were retarded with the addition of chitosan and acetic acid. This effect was not observed in the case of methylcellulose and sodium alginate. Chemical stability and bioactivity of materials were demonstrated during in vitro studies in simulated body fluid. Conclusions: Materials containing calcium sulphate-based granules were surgically handy, possessed promising physicochemical properties and are supposed to ensure desired macroporosity as well as gradual resorption in vivo. It has been demonstrated that the presence of CSD granules and polymers influenced the physicochemical properties of composites.
