Chitosan is widely used to prepare films, hydro-gels, cryogels, sponges, fibers and other various biomaterials used in the tissue engineering field. It is one of the best processable polysaccharides used in biomedicine. However, its stability is generally lower as compared with others, due to its pH sensitivity and hydrophilic character. Using chitosan in combination with agarose may not only improve chemical and mechanical properties of the resultant material (by the formation of a biocomposite), but also lead to the formation of a gel imitating physical attributes of the extracellular matrix. Moreover, the combination of these two polysaccharides has a promising ability to improve the stability of chitosan and to increase fibroblasts’ affinity to agarose. Characteristic advan-tageous features of these natural polymers raise a wide interest in tissue engineering. The aim of this study was to develop and optimize a new method to produce a highly biocompatible foam-like chitosan/agarose wound dressing for skin healing applications. The production process optimization helped to obtain the absorbent foam-like biomaterial which is non-toxic to skin fibroblasts and does not conduce their adhesion. Employing sodium bicarbonate as the main agent in the foaming reaction not only led to obtaining the foam-like structure but also neutralized the acidic pH, making the material non-toxic and non-irritating to the skin. In conclusion, the new foam-like biomaterial has great potential for biomedical applications as the wound dressing accelerating the healing process of the damaged tissues.

An organic solvent and surfactant stable α-amylase was obtained from soybean seeds. The direct and indirect effect of various organic solvents (non-polar, polar protic, and polar aprotic) and surfactants on the activity and stability of free enzyme was determined. The enzyme showed a very high catalytic efficiency and stabilization against most of the organic solvents and surfactants tested, except for few. Those organic solvents and surfactants (like chloroform, dimethyl formamide, n-butanol, and Tween 20), which caused an inhibition in enzyme activity, were used to study their effects on immobilized enzyme. The inhibitory effect was found to be decreased in immobilized enzyme as compared to free enzyme indicating that immobilization imparted stability to the enzyme. Moreover, the possibility of reuse of the enzyme in the presence of the organic solvents and surfactants was increased upon immobilization. The stability of soybean α-amylase towards organic solvents and surfactants shows that it is a potential candidate for use in organic-solvent biocatalysis as well as in detergent industries.

Herein, an effort was made to investigate the antibacterial potential of agarose nanoparticles (ANPs) and poly(quaternary ammonium) modifi ed ANPs (mANPs) against Escherichia coli (gram-negative bacterium) and Staphylococcus aureus (gram positive bacterium) in liquid systems as well as on agar plates. ANPs were synthesized by nanoprecipitation technology and characterized by XRD, TEM, TGA, DTA and DLS. The particle size estimated was 30 nm while atomic force microscopy was used to observe the interaction of ligand on ANPs. Antimicrobial characterization was monitored by colony forming units (CFU) as a function of ANPs concentration on agar plates. It was observed that ANPs showed 15 x 109/ml CFU after 24 hours of incubation at 20 mM ANPs concentration while the modified ANPs exhibited 21 x 109/ml CFU under similar incubation conditions. Moreover, zone of inhibition (ZOI) was 2.9 and 3.8 cm, respectively for E. coli by ANPs at 0.2 and 0.4 mM, respectively while it was 3.2 and 3.8 cm respectively by modified ANPs under similar conditions. Similarly, ZOI for S. aureus by ANPs at 0.2 and 0.4 mM was observed at 3.1 and 4.0 cm, respectively, while these values were 3.5 and 4.1 cm, respectively for modified ANPs under similar incubation conditions.

Opis:
Chitosan is one of the most promising polymers for biomedical applications due to its unique properties, such as its biocompatibility, low toxicity, biodegradation, and the presence of reactive amino and hydroxyl groups. Analysis of physicochemical properties of chitosan and its derivatives is a time-consuming process and requires expensive equipment and large amounts of the sample. This paper proposes a method for express analysis of the molecular weight (MW), the degree of deacetylation (DD), the substitution degree (SD), and the charge of chitosan and its derivatives using agarose gel electrophoresis under acidic and neutral conditions with Coomassie staining. Positively charged chitosan samples required acidic Tris Acetate-EDTA (TAE) buffer to move in the gel. The electrophoretic mobility of chitosan depended on MW, DD, SD, and the chitosan charge. Based on the dependences obtained by the proposed method, the MW and DD of commercial chitosan samples were determined. Express analysis of chitosan and its derivatives in agarose gel can be used to monitor the reactions of chitosan modification and to analyse samples with unknown characteristics.

This paper presents a methodology for the mechanical characterization of agarose millimetric spheres using resonant principles. Detection of the modes of vibration was conducted using a low-cost experimental setup based on an electret microphone adapted with a thin latex elastic membrane for the sensing stage and a piezoelectric actuator driven by a conventional transformer for the excitation stage. The identification of vibration modes is supported through an ANSYS Finite Element model of the experimental setup. Experimental and numerical results demonstrate that two modes of vibration, known as Quadrupole and Octupole, appear in the amplitude spectrum and can be used to obtain stiffness values for the samples. Following this approach, Young’s modulus of 209 ± 19.80, 338 ± 35.30 and 646 ± 109 kPa for 2%, 3% and 4% agarose millimetric spheres were calculated.

There is a growing trend in the engineering of biomaterials, focusing on surface modifications of biomaterials to improve their mechanical strength, corrosion resistance, and biological properties. Cold plasma treatment may improve biological properties of biomaterials for biomedical applications by enhancing their integration with host tissue. This study investigated the influence of different cold plasma treatments on the surface properties of a polysaccharides- -based biomaterial to improve cell adhesion to its surface. The samples were subjected to plasma treatment using three different reactors operating at atmospheric pressure: gliding arc discharge (GAD) reactor, dielectric barrier discharge (DBD) plasma jet, and DBD surface reactor. Next, surface chemistry of the biomaterial after plasma treatment was determined by ATR-FTIR analysis. Furthermore, a cell adhesion assay on the samples was carried out using normal human skin fibroblasts (BJ cell line). The attenuated total reflection Fourier transform infrared analysis (ATR- -FTIR) showed that new potential functional groups could be formed on the material surface after plasma treatment. However, plasma treatment of the samples did not enhance cell adhesion to the surface of the polysaccharides-based biomaterial. Thus, the obtained results indicate that plasma treatment using GAD reactor, DBD plasma jet, and DBD surface reactor was not effective for surface modification and cell responses.
