The aim of this paper is to assess the functioning of spatial planning in Poland in the context of public management objectives. Drawing on the concepts of New Public Management, we use the category of stakeholders and public value generated for stakeholders. Two groups of stakeholders in spatial planning are distinguished, namely public (national and local government) entities, and private entities (including individuals and businesses). We describe the mechanisms of stakeholders’ influence on spatial planning and the effects of this influence on the development of space. We found that the actual spatial planning mechanisms in Poland are aberrational from the perspective of public management. The ubiquity of these aberrations (in diverse municipalities) allows us to rule out the most obvious types of causes, particularly the incompetence or corruption of local authorities or administrative apparatus. Therefore, it is postulated that the causes of the aberrations are structural and inherent to the legal system that impacts spatial planning both directly and indirectly. We propose solutions to improve the situation through the appropriate management of relations with stakeholders in processes implemented by a public entity. The article presents a model of such management. A change in the legal context – above all an amendment of the law regulating spatial planning procedures – provides an opportunity to balance the impact of various stakeholders on spatial planning arrangements.

Objectives: The aim of this study was to investigate the genotoxicity of waterpipe smoking in the lymphocytes of waterpipe smokers using chromosomal aberrations (CAs) assay. Materials and Methods: Fifty waterpipe smokers and 18 healthy nonsmokers volunteered to participate in the study. Additionally, 18 heavy cigarette smokers were recruited for comparison. Chromosomal aberrations (CAs) assay was used to evaluate DNA damage in the lymphocytes. Results: The results showed that similarly to cigarette smoking, waterpipe smoking significantly increased the frequencies of CAs (p < 0.01). In addition, the frequencies of CAs increased with more waterpipe use. Conclusions: Waterpipe smoking causes DNA damage to lymphocytes and the damage increases with more waterpipe use.

Exposure to environmental pollutants often leads to an upsurge in the production of reactive oxygen species (ROS). ROS oxidize cellular fatty acids to produce lipid peroxyl radicals, subsequently transformed into lipid peroxides, which decrease membrane fluidity and increase the activity of various enzymes implicated in degenerative diseases and cancer formation. Edible plants that contain exogenous compounds like curcumeroid, β-carotene, turmeric, and so on, protect the aerobic cells from oxidation of free radicals. This study thus evaluates antioxidant and antimutagenic activities of ethyl acetate, aqueous and methanolic fractions of Holarrhena floribunda leaves. Inhibitory activities of the ethyl acetate fraction on Fe2+-induced lipid peroxidation in hen egg yolk; rat liver and brain tissues were also evaluated. The Allium cepa root assay was used to evaluate antimutagenic activity. Results showed that the ethyl acetate scavenged DPPH, OH•, and •O2- much stronger than other fractions, as evidenced by its lowest respective IC50 values. All the fractions displayed antimutagenic activities against cyclophosphamide-induced chromosomal aberrations. Likewise, all the fractions induced a reduction in mitotic index, a hallmark of cytotoxicity in the root meristem of Allium cepa. The decrease in mitotic index was most profound for the ethyl acetate fraction, which also demonstrated a significant lipid peroxidation inhibitory activity in the liver and brain homogenates, but not in egg yolk, compared with the ascorbic acid standard. In general, the results suggest that the ethyl acetate fraction might contain beneficial phytochemicals that should be explored as novel candidates for preclinical drug development.

Cytogenetics is complementary to genetic and molecular analysis of genome structure and function. From the beginning it has been mainly used for identification of chromosomes and karyotype construction. Most significant for the progress in cytogenetics was development of chromosome banding techniques and in situ hybridization, especially fluorescent in situ hybridization (FISH) and its different modifications which have become the most important techniques in molecular cytogenetics. FISH allows physical gene mapping and localization of different non-coding DNA sequences on chromosomes and interphase nuclei. Repetitive DNA sequences can generate unique FISH-signal patterns on individual chromosomes valuable for karyotyping and phylogenetic analysis. These studies have important implications for basic research and practical applications. The understanding of the structure, function, organization and evolution of genomes enabled many new cytogenetic applications to both medicine and agriculture, particularly in diagnosis and plant breeding.

Opis:
Natural plant extracts offer a promising hope in the prevention/treatment of cancer arising from genetic mutations. This study evaluated in vitro and in vivo mutagenic and antimutagenic effects of aqueous fraction of Myristica fragrans (AFMF) leaves on TA100 strain of Salmonella typhimurium and Mus musculus (Male Swiss albino mice), respectively. The antioxidant activity of AFMF against 2,2-diphenyl-1-picrylhydrazyl (DPPH), total phenolic and flavonoid contents were determined, followed by its phytochemical elucidation using the Ultra Performance Liquid Chromatography technique (UPLC). The mutagenicity of AFMF at 4, 20, 50, 100, 200, 500, and 1000 µg/well was <2.0 in S. typhimurium and the induced micronucleated polychromatic and normochromatic erythrocytes at 500, 1000, 2000, and 4000 mg/kg were not significantly different from the negative control (p≥0.05). The mutagenic activity of benzo[a]pyrene and cyclophosphamide was significantly suppressed above 50.0% throughout the tested concentrations. Fifty percent of the free radicals from DPPH were scavenged by AFMF at 0.11 mg/ml. Total phenolic and flavonoid contents of AFMF were 51.0 mg GAE/g and 27 mg QE/g, respectively. Rutin was elucidated by the UPLC technique, and thereby suspected to be the phytochemical responsible for the observed antimutagenic activity. Thus far, AFMF seems to contain a promising chemotherapeutic agent for the prevention of genetic damage that is crucial for cancer development.

Opis:
The function of acetylcholinesterase (AChE) is the rapid hydrolysis of the neurotransmitter acetylcholine (ACh), which is involved in the numerous cholinergic pathways in both the central and the peripheral nervous system. Therefore, AChE measurement is of high value for therapy management, especially during the course of intoxication with different chemicals or drugs that inhibit the enzyme. Pyridinium or bispyridinium aldoximes (oximes) are able to recover the activity of the inhibited enzyme. Since their adverse effects are not well elucidated, in this study the efficiency of HI-6 oxime in protection and/or reactivation of human erythrocyte AChE inhibited by the antineoplastic drug irinotecan as well as its cyto/genotoxicity in vitro were investigated. HI-6 was effective in protection of AChE and increased its activity up to 30%; the residual activity after irinotecan inhibition was 7%. Also, it reactivated the enzyme previously inhibited by 50% irinotecan (4.6 µg/ml) applied at ¼ of the IC50 value. The tested concentrations of HI-6 exhibited acceptable genotoxicity towards white blood cells, as estimated by the alkaline comet assay, DNA diffusion assay and cytogenetic endpoints (structural chromosome aberrations and cytokinesis-block micronucleus assay). The results obtained warrant the further investigation of HI-6 in vivo, as well as its development for possible application in chemotherapy.

1) Background: the modeling, characterization, transformation and propagation of high-power CW laser beams in optical (including fiberoptic) trains and in the atmosphere have become hot topics in laser science and engineering in the past few years. Single-mode output is mandatory for high-power CW laser applications in the military field. Moreover, an unstationary, dynamic operation regime is typical. Recognized devices and procedures for laser-beam diagnostics could not be directly applied because of dynamic behavior and untypical non-Gaussian profiles. 2) Methods: the Wigner transform approach was proposed to characterize dynamically variable high-power CW laser beams with significant deterministic aberrations. Wavefront-sensing measurements by means of the Shack-Hartmann method and decomposition into an orthogonal Zernike basis were applied. 3) Results: deterministic aberration as a result of unstationary thermal-optic effects depending on the averaged power of the laser output was found. Beam quality determined via the Wigner approach was changed in the same way as the measurements of the beam diameter in the far field. 4) Conclusions: such an aberration component seems to be the main factor causing degradation in beam quality and in brightness of high-power CW laser beams.

To investigate the effectiveness of a rotating biological contactor (RBC) and a two-stage membrane bioreactor (MBR) for the treatment of coke wastewater, samples were collected three times (Batches I, II, III) from the “Jadwiga” coke plant in Zabrze, Poland at two-week intervals. The wastewater was then diluted with tap water (1:3 ratio, wastewater : tap water) and then treated at retention times of 4.1 days (RBC) and 7 days (MBR). For phytotoxicity and genotoxicity tests, the wastewater was sampled from various points in the treatment systems and further diluted to produce a range of concentrations. In the phytotoxicity tests (growth inhibition), Lemna minor and Vicia faba were used. A low concentration of wastewater (6.25%) often stimulated growth. Higher concentrations, however, inhibited L. minor growth completely. These tests indicated that the MBR generally reduced growth inhibition more effectively than the RBC. In the genotoxicity tests (chromosome aberrations and micronuclei formation) root meristem cells of V. faba were examined. The genotoxicity of the different batches varied, and neither system was particularly effective for reducing genotoxicity. The results of this study indicate that, because its composition is so variable, coke wastewater should be constantly monitored. Also, because of its potentially high genotoxicity, the ecotoxicological characteristics of coke wastewater should be monitored in addition to basic indicators of wastewater quality, such as COD, BOD, and content of nitrogen compounds.

Wprowadzenie. Zespół pierścieniowego chromosomu 21 należy do grupy chorób zależnych przede wszystkim od konstytucji genetycznej. Cel. Celem tej pracy jest usystematyzowanie i synteza wiadomości dotyczących różnorodności genotypowej i fenotypowej występującej w tym zespole. Materiał i metoda. Dokonano systematycznego przeglądu najnowszego piśmiennictwa odnoszącego się do etiopatogenezy oraz obrazu klinicznego zespołu pierścieniowego chromosomu 21. Wniosek. Zespół pierścieniowego chromosomu 21 charakteryzuje się zarówno różnorodnością genotypową, jak i fenotypową. U podłoża zespołu leży złożony i przypadkowy proces rearanżacji genomu. Posiada on równocześnie cechy zespołu malformacyjnego i dysplastycznego.
Introduction. The ring chromosome 21 belongs to a group of diseases dependent on the genetic constitution of an individual. Aim. The aim of this paper is systematization and synthesis of the knowledge related to genotypic and phenotypic variability occurring in this syndrome. Material and method. The author has carried out a systematic review of current literature related to etiopathogenesis and clinical picture of the ring chromosome 21 syndrome. Conclusion. The ring chromosome 21 syndrome is characterized by genotypic and phenotypic variability. It possesses traits of both the malformed and dysplastic syndrome.

Polycyclic aromatic hydrocarbons (PAHs) result from the incomplete combustion of natural or synthetic organic materials. The working environment at a coke plant can negatively affect the employed workers who were exposed to coke oven emissions containing PAHs, which formed and released into the environment by the process of pyrolysis of coke. This study aims to analyze the relationship between the exposure of PAHs and the risk of genetic damages such as chromosomal alteration (CA), micronucleus (MN), and DNA damage (PCR-RFLP) in peripheral blood lymphocytes of 27 coke oven workers and equal number of control subjects. The exposed subjects and controls were divided into two groups based on their age (group I<35 years and group II ≥35 years). The exposed subjects were further classified into two groups based on the exposure period (<12 years and ≥12 years). The frequencies of CA and MN in exposed subjects are relatively high with respect to controls. The XRCC1 399 Arg/gln polymorphism showed a substantial smaller difference in allele frequencies between exposed and control subjects. Based on present data, it was concluded that coke oven workers under risk should be monitored for adverse effects of the any long-term exposure.

We report methodology of three-dimensional laser beam shaping. The analytical and numerical analysis is presented. A strategy for designing optical system involves the controlled application of aberrations which is realized by specific aspheric lens shaping. The goal is to obtain the desired optical power density distribution in space, which is optimal for selected application. The proposed method is discussed with regard to the design of optical transmitter for laser shooting simulator, however the developed methodology can be used for a number of other light shaping applications.

The Computer Aided designing Tools for Electromagnetic Lenses (CADTEL) is a new software as application package of programs concept to aid both users and professionals in symmetric and asymmetric electromagnetic lenses with single, double, and multipole piece in electron microscope of electron optics field. The CADTEL software has been designed to run on several computer platforms and includes two types of design procedure. The first one named, analysis procedure (which is based on trial and error) where consist of three sorts called; H1 programs for magnetic scalar potential by solving Laplace's equation [1, 2], H2 and H3 programs for magnetic vector potential by solving Poisson's equation [3, 4] in linear and non linear media respectively. While the second part, named H4 programs for the synthesis procedure (inverse design). The previous types differ by the obtaining the magnetic flux density, and poles shape, while it is analog by compute and plot the lens properties which are operating at zero, low, high, and infinite magnification conditions (operation modes). CADTEL software consist of computational and plot steps of magnetic field, equipotential surfaces, flux lines, objective and projector properties, and poles shape for proposed lens design which are appear in auto visual interfaces, that are coded in visual basic language [5].
