Recently, several attempts have been undertaken to correlate in vitro calibration curves with observed frequencies of chromosomal aberrations and micronuclei in lymphocytes of patients undergoing radiotherapy. The aim of such correlations is the search for a biological method to reconstruct the dose received during radiotherapy. While the in vitro dose-response curves are linear-quadratic, the observed in vivo relationship is usually linear and there is some controversy as to the nature of the observed linearity. We have, therefore, constructed a model to calculate the frequencies and distributions of chromosomal aberrations in lymphocytes of patients undergoing conventional radiotherapy. The model assumes that each fraction of radiation induces a certain number of Poisson-distributed aberrations in the irradiated blood volume. In addition, a simplified assumption is made that lymphocytes flow freely inside the body of the patient and no elimination of cells occurs. The model yields linear dose response curves. The steepness of the curves increases with increasing size of irradiated block of tissue (referred to as irradiated volume) and increasing dose per fraction. The distributions of aberrations become increasingly overdispersed with increasing dose per fraction but are independent of the number of radiation fractions. The modelled dose-response curves agree well with the majority of published experimental results. Given the simple assumptions made, this indicates that cell elimination, which occurs during radiotherapy does not bias the results obtained experimentally. The linearity of the dose response-curve results from the fractionated irradiation. Hence, great care should be applied when attempting to use standard, linear-quadratic calibration curves to estimate the doses received by patients during radiotherapy.

Opis:
The aim of the present paper was to find out by in vitro chromosomal aberration test using human lymphocytes whether cysteine has anticlastogenic properties towards a well-known mutagen - mechlorethamine. The lymphocytes tested were obtained from three healthy donors. Two doses of cysteine (1.0 and 2.0 μg/ml) and three doses of mechlorethamine (0.1,0.2 and 0.3 μg m⁻¹) were tested. It was found that cysteine had anticlastogenic properties and that it reduced the number of metaphases with chromosomal aberrations induced by mechlorethamine.

Opis:
The optical trapping forces of tightly-focused radially polarized circular partially coherent beams on Rayleigh particles are theoretically investigated. Numerical calculations are performed to study the optical trapping forces on Rayleigh particles for different initial coherent length of the incident circular partially coherent beams. The results show that the magnitude of the gradient force decreases with the reduction of the initial coherent length of the focused radially polarized circular partially coherent beams, while the balanced position (i.e., the position where the optical trapping forces becomes zero) stays constant. Moreover, the focused spot gradually elongates along the optical axis with the reduction of the initial coherent length, and the axial gradient force on Rayleigh particles also decreases gradually with the reduction of the intensity gradient in axial direction. As there exists an spherical aberrant in the focusing optical system, the focal spot in the direction of the optical axis becomes trumpet-shaped, and the optical trapping forces on Rayleigh particles change as well.

The present study was undertaken to contribute to the characterization of the degree of variability in baseline damage in white blood cells from control population, and to investigate how this variability is associated with external and internal factors. Altogether 170 healthy volunteers, randomly selected from the general population of the Republic of Croatia, participated in the study. Two sensitive tests: the alkaline comet assay and the chromosome aberration test were applied to study the background levels of DNA damage in their white blood cells. The results point to inter-individual differences, indicating different genome sensitivity. As revealed by both assays, the background levels of DNA damage were mostly influenced by smoking habit as well as medical exposure (especially to diagnostic X-rays). Sex and age of subjects did not significantly influence the values of DNA damage recorded in the white blood cells. Although higher levels of DNA damage were recorded in blood samples collected during winter and autumn, they were mostly influenced by medicinal exposure and smoking habit. Statistical evaluation of the data confirmed that a positive correlation exists between DNA migration and the number of long-tailed nuclei found with the comet assay and the total number of chromosome aberrations. The data obtained can serve as control values in forthcoming biomonitoring studies.

Opis:
Thin and lightweight achromatic focusing elements with F-number close to 1 are desirable in many practical applications. We present the idea to use diffractive structures designed to work for the substantially increased THz frequency range. The paper analyses mono- and multi-focal lenses forming point-like foci as well as axicon and light sword optical elements focusing THz radiation into line segments located along the optical axis. We consider diffractive elements in a form of the first and the second order kinoforms having various thicknesses. Designed and fabricated elements were numerically and experimentally examined to verify their achromatic functioning. We present point spread functions (XY scans) and 2D energy maps (XZ scans) for different THz frequencies. Moreover, a diagram of chromatic aberration is created by registering energy distribution along the optical axis for different frequencies. The distance corresponding to the highest energy is chosen for each frequency. Therefore, we can compare broadband working of designed structures. The spherical lens coded as kinoform of the second order provides the best broadband functioning, however it is two times thicker than structures providing extended depth of focus (light sword and axicon) working with slightly smaller efficiency but being much thinner.

Opis:
Ionizing radiation passing through living matter in the form of ion tracks causes DNA double strand breaks, which lead to chromosomal aberrations. On one hand, the number of aberrations is determined by the absorbed dose of radiation and on the other hand results from the efficiency of the repair mechanisms. These mechanisms are especially important for low doses, the study of which is particularly time-consuming.Here, a new method to estimate the effectiveness of repair mechanisms is presented. The method is based on the reduction of variance of observed chromosomal aberrations in relation to the values expected for the Poisson statistics. This effect is known in other fields of physics as the Fano factor and can be applied to various tissues and for both low and high LET (Linear Energy Transfer) radiation’s types.By exploiting the new method, distributions of chromosomal aberrations in peripheral blood lymphocytes observed for gamma and 150 MeV proton radiation, have been compared. Experiments using proton beams were carried out in the Joint Institute for Nuclear Research in Dubna, Russia.
Promieniowanie jonizujące, tworząc w materii ożywionej ślady jonowe i łamiąc podwójną helisę DNA, prowadzi do powstania aberracji chromosomowych. Ich liczba z jednej strony określona jest pochłoniętą dawką rodzajem promieniowania, a z drugiej strony wynika z wydajności mechanizmów reperacji komórkowej. Mechanizmy te są szczególnie ważne w przypadku niskich dawek, dla których badania są szczególnie pracochłonne.W niniejszej pracy przedstawiona zostanie metoda oszacowania efektywności mechanizmów reparacyjnych w oparciu o obserwowaną redukcję wariancji liczby otrzymanych aberracji chromosomowych w stosunku do zakładanej wartości według statystyki Poissona. Efekt ten znany jest w innych dziedzinach fizyki jako czynnik Fano i może być zastosowany do różnych tkanek oraz promieniowania jonizującego o niskim bądź wysokim LET (ang. LinearEnergy Transfer).W oparciu o nową metodę statystycznego oszacowania efektywności mechanizmów reperacji komórkowej porównane zostaną rozkłady aberracji chromosomów limfocytów krwi obwodowej obserwowane dla naświetlań zarówno kwantami gamma, jak i protonami o energii 150 MeV. Eksperymenty z użyciem wiązki protonowej przeprowadzone zostały w Zjednoczonym Instytucie Badań Jądrowych w Dubnej w Rosji.

Opis:
Acetylsalicylic acid (ASA) and α-2-pyrazylidene-α-cyano N-butyl acetamide (PD-101), a new antiaggregatory pyrazine derivative were tested for their genotoxicity in human lymphocytes in vitro using the sister chromatid exchange (SCE) technique. Both compounds were found to be inactive in inducing SCE in concentration from 1 µM up to 1000 µM. The agents displayed inhibitory effect on cell kinetics.

Opis:
Models are proposed to describe the heart’s action potential. A system of stochastic differential equations is used to recreate pathological behaviour in the heart such as atrioventricular nodal reentrant tachycardia (AVNRT) and also AVNRT with conduction aberration. Part of the population has abnormal accessory pathways: fast and slow. An additional pathway is not always induced, since the deterministic model is not proper due to a stochasticity in this process. Introduction of a stochastic term allows modelling a pre-excitation perturbation (such as unexpected excitation by premature contractions in atrium (PAC)) which triggers the mechanism of AVNRT. Also, a system of AVNRT with additional conduction aberration, which is a rare type of arrhythmia, is considered. The aim of this work is to propose a mathematical model superior to the deterministic one that recreates this disease better and allows understanding its mechanism and physical dependencies, which may help to propose a new therapy of AVNRT. Results are illustrated with numerical solutions.

Opis:
Five newly synthesised monophosphates of two pyrimidine acyclonucleoside series, namely 1-N-[(2’-hydroxy)ethoxymethyl] and l-N-[(l’,3’-dihydroxy)- 2’-propoxymethyl] derivatives of 5- and 5,6-alkylated uracils were tested in vitro for chromosome aberrations and sister chromatid exchanges (SCE). Metaphase plates were obtained via microculture of human lymphocytes from heparinized peripheral blood. The compounds were tested in doses: 10, 20, 40, 80 and 150 µg per mL of culture. The tested compounds induced mainly chromatid gaps, less frequently chromosome gaps. A low number of mitoses with chromatid and chromosome breaks, acentric fragments, dicentric chromosomes and exchange figures were also observed. The tested compounds in doses: 40, 80 and 150 µg per mL, doubled or tripled the percentage of cells with chromatid gaps and chromosome gaps as compared to the control. The percentage o cells with aberrations (excluding gaps) induced by the tested compounds in all doses did not exceed 2%. The tested compounds induced a higher number of SCE per cell but less than double frequency as compared to the control. SCE frequencies and replication index (RI) values varied depending on the examined compounds. For the highest dose of the tested compounds (150 µg per mL) a significant decrease in RI values was observed for l-N-[(2’-hydroxy)ethoxymethyl]-5,6-tetramethyleneuracil monophosphate and for l-N-[(2’-hydroxy)ethoxymethyl]-5,6-dimethyluracil monophosphate. So far, the results have indicated potential clastogenicity of all the tested compounds except acycloguanosine monophosphate.

The mitotic effect of physical mutagen gamma rays was observed in the root tip cells of Ashwagandha. The Chromosome analysis has been showed as an important tool for establish variability of the plant seed by use of physical mutagen gamma rays. The gamma rays have of low wavelength and high penetrable power. The plant has tremendous medicinal values and it is known from ancient times. The dry and well matured seeds of ashwagandha were irradiated with different doses of gamma rays viz., 5, 10, 15, 20, 25, 30, 35, 40, 45 and 50 KR respectively. The chromosome number of control plant is 2n = 48. The gamma rays affect the normal cytological behavior of ashwagandha species. The chromosomal aberrations increase with increase in the doses of gamma rays to optimum level of 30KR, because it causes changes in the chromosome structure, cellular structure and metabolism of plants. The chromosome aberration like, Sticky metaphase, Precocious moment chromosome, Fragments, Anaphasic bridge, Anaphasic laggard, Telophasic laggard. The present investigation was carried out to study the cytogenetic analysis of the species Withania somnifera. The chromosomal aberration increases with increase in the doses to optimum level (50 KR) of physical mutagen gamma rays.

Opis:
Based upon the example of geometrical optics and  geometrical optics of electrons, as well as the theory of the electric field and the theory of the flow of the perfect liquid in two dimensions, the analogy of the laws of physics have been discussed. In the awareness of physicists, the analogy is a syntactical identity, of the physically interpreted mathematical formulas, describing the laws of physics. In the paper, it has been presented some relevant laws of the two pairs of the theories being compared. For the three cases, it has been provided the appropriate photographs presenting the results of the empirical tests of some of the relevant laws.
