Opis:
The content of amino acids, except tryptophan, was determined in fresh, sterilized and frozen broad beans. The cultivars investigated were Windsor Bialy, Comprimo RS, Green Hangdown and Threefold-white at the maturity stage corresponding to 30% dry matter content in seeds. At all stages of the investigation the limiting amino acids were methionine and cystine. An average CS index of these amino acids was 32 for fresh seeds, 33 and 28 for the whole seeds and the cotyledons, respectively, in canned sterilized products, 32 and 30 for the whole seeds and the cotyledons, respectively, in frozen-and-cooked seeds. The respective EAA indices were 69, 60, 61, 62 and 64. The cotyledons were poorer than the whole seeds in phenylalanine, threonine, cystine, aspartic acid and serine, and also in methionine in the frozen seeds.

From the previous studies it is known that faba bean albumins occur as a nucleo-glyco-metalloprotein complex. The purpose of this work was to determine the rate constant for the first stage of glycation of faba bean albumins, i.e. the formation of aldimine. The variation of glycation rate constant of faba bean albumin complex was stated. Most probably it resulted from the participation of nucleic acid amino groups which were not considered in the model of glycation kinetics assumed. The results obtained show that the glycation of faba bean albumin complex is the reaction of fractional order.

This work presents electrophoretic seed globulin data obtained for 173 accessions representing nine Vicia species of the section Faba, which were already investigated for electrophoretic seed albumin patterns (PRZYBYLSKA, ZIMNIAK-PRZYBYLSKA, in press). Electrophoretic analysis of seed globulins was performed using sodium dodecyl sulphate - polyacrylamide gel electrophoresis (SDS-PAGE). In the examined material totally 38 well-defined polypeptide bands, in the relative molecular mass range of 19-61 kDa, were distinguished. The presence/absence of particular bands was recorded for each analysed individual. The electrophoretic data were used for hierarchical grouping of the examined taxa, based on calculated Bhattacharyya distances. V. bithynica proved to be the most distinctive member of Vicia section Faba. In turn, V. faba was clearly different from species of the V. narbonensis complex. Taxa of this complex formed two clusters: one consisting of V. narbonensis varieties and another - of the remaining species. In the latter group, V. serratifolia appeared to be rather distantly related to V. eristalioides, V. galilaea, V. hyaeniscyamus, V. Johannis and V. kalakhensis. The obtained results are compared with the corresponding seed albumin data and discussed with reference to taxonomie relationships in Vicia section Faba.

Opis:
The effects of fusarial toxins: DAS, T-2, DON, 3 Ac-DON, MON and ZEA, on actively dividing root tip cells of rye, wheat and field bean were investigated. Three concentrations: 1, 5 and 10 µg mL⁻¹ were applied for 24 hours. Nuclei and chromosomes were stained using the Feulgens method. It has been found that trichothecene mycotoxins (DAS, T-2, DON, 3 Ac-DON) had a profound effect on mitosis, as they decreased the mitotic index, produced excessive condensation of pro- and metaphase chromosomes, C-metaphascs and C-anaphases, and caused an accumulation of metaphases. The main effect of trichothecene mycotoxins, probably caused via their influence on protein synthesis, was abnormal functioning of the mitotic spindle.

W 1993 roku z Pracowni Zasobów Genowych IW w Skierniewicach uzyskano nasiona populacji bobu nr 179510. Otrzymana populacja odznaczała się dużym zróżnicowaniem nasion pod względem wielkości i barwy. Nasiona posegregowano według barwy okrywy nasiennej: o barwie ciemnofioletowej, różowo-fioletowej, jasnozielonej, beżowej i ciemno-beżowej. W latach 1994-1997 zastosowano hodowlę wsobną w pokoleniach o nasionach ciemno-fioletowych i różowo-fioletowych. W wyniku zastosowanej hodowli wsobnej uzyskano 7 linii hodowlanych, w tym 5 linii wyrównanych o nasionach granatowo-czarnych, które zostaną wykorzystane do wyhodowania nowej odmiany bobu. Otrzymane linie bobu charakteryzują się szerokim, grubym strąkiem, skośnie ustawionym na roślinie. Średnia liczba strąków na roślinie wynosi od 19 do 27 szt. Natomiast średnia masa 1000 suchych nasion waha się od 1500-1800 g a średnia liczba wykształconych nasion w strąku jest od 1,4 do 1,7 w zależności od linii.
The population no. 179510 of faba bean seeds has been ibtained in 1993 by Plant Genetic Resources Laboratory of RIVC, Skierniewice during an expedition in the south part of Poland. The original seed sample was the mixture of large seeds, very variable in colour. The seeds were separated into groups according to seed colour: dark-violet, rose-violet, light-green, beige, dark-beige. During 1994-1997 the inbred was applied in the generations of dark-violet and rose-violet seeds. As a result of inbred 7 breeding lines were obtained, among them 5 lines with uniform dark blue-black seed colour. Derived lines were the maternal materials for breeding new cultivar of faba bean. Mentioned lines of faba bean are characterized by large and thick pods. Average number of pods per 1 plant ranged from 19 to 27. Average weight of 1000 dry seeds ranged from 1500 g to 1800 g and the numbers of good quality seeds per 1 pod were from 1.4 to 1.7, depending on the line.

Opis:
Several fusarial toxins (DAS, DON, FUS-X and MON) were assayed for their genotoxic activity with Vicia faba var. minor and Pisum sativum used as eukaryotic, whole-organism, test systems. Four concentrations: 1, 5, 10 and 20 µg mL⁻¹ were applied for 24 hours, and three fixing times: 24, 48 and 72 hours after the beginning of the treatment. Nuclei and chromosomes in mitotic cells were stained by the Feulgen method. Generally, fusarial toxins in plant cells produced three types of effects: prevented proliferating cells from entering mitosis, caused mitotic alterations and were cytotoxic. Mitotic alterations included C-mitoses, a higher incidence of metaphases/anaphases, excessive condensation of chromosomes, multipolar spindles, disturbed anatelophases and micronuclei. Both type and range of each effect were related to plant species, toxin concentration and duration of the recovery period. The results obtained from our plant bioassays were compared with those reported from other assay systems.

Opis:
The results of plant breeding trials with populations of fodder pea strains and broad bean hybrids were the basis of consideration on the interrelationship between some traits - the yield structure elements. Developed by Eaton, a relatively new method of yield component analysis called the two-dimensional partitioning method (TDP) was applied to analyse the data. The method, which combines multiple regression and ANOVA, allows for concise tabular presentation and simple interpretation of the distribution of traits in one direction and the sources of variance according to ANOVA model in the other direction. Additionally, the interpretation of the results was supported by such standard statistical techniques as ANOVA, simple and multiple regression and path analysis. The main components of pea yielding were plant height and the number of pods per plant. Among the analysed characters of broad bean the number of nodes with pods on the main stem, which turned out to be the determinant of broad bean yielding, might be strongly affected by environmental conditions. The number of nodes with pods might be considered a selecting character of high potential yielding of broad bean genotypes.

Opis:
In carbohydrate-starved root meristems of Vicia faba subsp. minor, the expression of two Principal Control Points located at the final stages of the G1 (PCP1) and G2 (PCP2) phases has been found to be correlated with a marked decrease of protein phosphorylation within cell nuclei, nucleoli and cytoplasm. Adopting the same experimental model in our present studies, monoclonal FITC conjugated antibodies that recognize phosphorylated form of threonine (αTPab-FITC) were used to obtain an insight about how the indole-3-acetic acid (IAA), benzyl-6-aminopurine (BAP), and the mixture of both phytohormones influence the time-course changes in an overall protein phosphorylation during sucrose-mediated PCP1→S and PCP2→M transitions. Unsuspectedly, neither IAA, BAP, nor the mixture of both phytohormones supplied in combination with sucrose did up-regulate protein phosphorylation. However using the block-and-release method, it was shown that root meristems of Vicia provided with sucrose alone indicated higher levels of αTPab-FITC. Contrarily, phytohormones supplied in combination with sucrose induced apparent decline in phosphorylation of cell proteins, which - when compared with the influence of sucrose alone - became increasingly evident in time. Thus, it seems probable, that a general decline in the amount of αTPab-FITC labeled epitopes may overlay specific phosphorylations and dephosphorylations governed by the main cell cycle kinases and phosphatases.

Opis:
Reaction of 25 conventional and 63 determinate forms of faba bean to artificial inoculation with A. fabae and B. fabae was evaluated over five years. The severity of infection of faba bean with A. fabae was significantly higher than with B. fabae. This was observed for the both morphological types. Most of the conventional forms were less infected than the determinate forms, but it was more distinct for the A. fabae infection. Variable resistance reaction of cultivars was observed depending on pathogen and growth habit. In all years, conventional as well as determinate cultivars differed significantly in resistance to A. fabae. Significant variability of resistance to B. fabae was found only in one year for the conventional types and in another year for the determinate types of faba bean. It was found that some determinate cultivars showed higher resistance to A. fabae or B. fabae than most of the conventional forms, and some conventional forms were in the group of the most susceptible cultivars.
