    We present the results of optimization of fixation and staining of the Chinese hamster lung fibroblast V79 cell line. The experiment included four methods of fixation and three methods of staining and enabled the selection of the best visualization technique for V79 cell lines culture. The Schiff method of colouring with Light Green contrasted well the cell nucleus and the cytoplasm, but the staining was light-sensitive and transient. The Giemsa staining made cell preparations suitable for microscopic evaluation, although the difference between the nucleus and the cytoplasm was not always easily recognisable.

    Opis:
    Environmental pollution causes a variety of health problems, including cancer. Many known pollutants have carcinogenic properties and polycyclic aromatic hydrocarbons (PAH) belong to this group. In this study, an in vitro culture of V79 cells of the Chinese hamster was subjected to three tested PAHs: 5-amino-2,3-dihydro-1H-cyclopenta-[c]phenanthrene (ACP[c]Ph), 5-amino-9-methoxy-2,3-dihydro-1H-cyclopenta[c]phenanthrene (AMCP[c]Ph) and cyclopenta[c]phenanthrene (CP[c]Ph). The in vitro micronucleus (MN) assay was applied in order to evaluate the genotoxic properties of the studied compounds. The highest genotoxic effect was observed for AMCP[c]Ph in a concentration of 0.02μg·ml-1. The genotoxic effect of the other two compounds was slightly lower.

    Kelvin probe microscopy is an experimental technique designed to investigate fluctuations of surface potential (work function per electron) related to distribution of electric charge or variations in composition. The paper describes principle and precision of measurements. The results obtained for group-III nitrides semiconductor heterostructures grown on c-plane sapphire by metal-organic vapour phase epitaxy are presented. The observations concerns defects: inversion domains for Ga- and N-polar layers, threading dislocations and effects of spontaneous polarization leading to 2D carrier gas. To achieve insight in the evolution of defects, bevelled and cross-sectioned samples were investigated along with surface of "as grown" layers. Applicability of standard Kelvin probe microscopy method was also extended by investigating dependence of the surface potential on variable wavelength illumination, offering opportunity for spectroscopy of individual defects.

    In this article we present the method of determining the tip-sample distance using advanced data processing of scanning field's optical picture. This feature can reduce both: the approach process time and the risk of damaging the tip or the sample. Experimental results will be also presented.

    We report recent advances in material characterization on the nanometer scale using scanning microwave microscopy. This combines atomic force microscopy and a vector network analyzer using microwave tip sample interaction to characterize dielectric and electronic material properties on the nanometer scale. We present the methods for calibration as well as applications. Scanning microwave microscopy features calibrated measurements of: (1) capacitance with attofarad sensitivity. For calibration a well characterized array of capacitors (0.1 fF to 10 fF) is used. The method is applied to determine the dielectric properties of thin organic films, (2) Semiconductor dopant density. Calibration is performed by imaging the cross-section of a standard sample with differently doped layers (dopant stair case) from $10^{16}$ atoms/$cm^3$ to $10^{20}$ atoms/$cm^3$.

    Biological applications of ion beams have recently become a new important research field using single ion hit facilities to study individual living cells and their response to the hit of a counted number of ions. One motivation is the search for a better understanding of the fundamental processes taking place in cells and organs as a result of irradiation. Another comes from the increasing interest in using high energy protons and heavy ions as a modality for radiotherapy of deep seated tumours. In the view of treatment efficiency, study of cell culture behaviour under controlled radiation experiments, and in different chemical environments at single ion hit facilities, is a first step towards a better understanding of the processes. Tomographic techniques are applicable to situations where you need information of the inside of an object but do not want to section it into thin slices or cannot do it. Using focused MeV ion beams for tomography restricts the sample size to the order of 10-100 μm, depending of the initial energy. On the other hand, the ability to focus at a sub-micrometer level makes ion beams well suited for analyses of small sized objects as cells, spores, etc. The scanning transmission ion microscopy mode of tomography gives the mass density and corresponding morphological structure of holes and pores. It can then be used to correct the results from the other mode, particle induced X-ray emission tomography. Here is discussed a porosity analysis of bentonite clay that is planned to form an important buffer zone around canisters filled with spent nuclear reactor fuel waste deposited 500 m underground in Sweden.

    A volumetric method was applied to study an adsorption coefficient of hydrogen molecules in a gas phase on super activated carbon surface. The investigations were focused on getting the best possible materials for the energy storage. Several treatments on raw samples were used to improve adsorption properties. The biggest capacities were obtained after high temperature treatment at reduced atmosphere. The adsorption coefficient at 77 K and 2 MPa amounts to 3.158 wt%. The charge transfer between lithium and carbon surface groups via the doping reaction enhanced the energy of adsorption. It was also found that there is a gradual decrease in the adsorbed amount of H$\text{}_{2}$ molecules due to occupation active sites by lithium ions.

    In the paper the electric properties and electronic structure of the intermetallic $Tb_{x}Gd_{1-x}Ni_3$ compounds are presented. The partial replacement of Gd by Tb atoms causes the decrease of the Curie temperature $(T_{C})$ and the increase of the residual resistivity. According to the Matthiessen rule the scattering mechanisms in ρ(T) have been analyzed. Moreover, the reduced form of the electrical resistivity $ρ_{Z} (T - T_{0})$ indicates a deviation from the linearity for x > 0.2. This kind of behaviour can be attributed to density of d states near by the Fermi level $(E_{F})$ which are dominated by Ni 3d states. The valence band spectra as well as the core level lines have been analyzed as the influence of Tb/Gd substitution on the electronic structure.
