Aims: To evaluate the phytoconstituent and antitrypanosomal effects of aqueous extracts of Ginger and Garlic bulbs in mice experimentally infected with Trypanosoma brucei brucei. Design: A total of 30 adult male mice (weighing 25-40 g) were randomly grouped into six groups (I, II, III, IV, V, and VI) of 5 animals each. Five Groups (II-VI) were intraperitoneally injected with T. brucei brucei (5×105 cells/ml). Methods and Material: Aqueous extract of ginger and garlic bulbs were obtained using the procedure described by Wabo Pone and the extracts were subjected to phytochemical screening using the standard screening method of Silva. Also, each mice was inoculated with 0.1 ml of blood containing approximately 5×105 cells/ml as described by Herbert and Lumsden. Finally, the aqueous extracts were tested on the inoculated mice. Statistical analysis used: Data obtained were expressed as mean ± standard error of the mean (SEM), and subjected to one-way analysis of variance (ANOVA), SPSS 17.0 statistical software. p<0.05 was considered significant. Results: The aqueous extracts increased the survival time, packed cell volume, rectal temperature, and body weight (Ginger extract only) of mice infected with Trypanosoma brucei brucei. Phytochemical analysis revealed alkaloids, steroids, cardiac glycosides, phenol, and saponins in both Ginger and Garlic. Conclusions: Aqueous extracts of Ginger and Garlic bulbs do not have an anti-trypanosomal effect on Trypanosoma brucei brucei. Consequently, geographical location and time of collection of plants are factors that accept therapeutic on ginger and garlic on tryps and should be considered when testing the plants' efficacy on Trypanosoma brucei brucei.

Opis:
Selenoproteins have been described in all three domains of life and their function has been mainly associated with oxidative stress defense. Canonical elements required for selenoprotein production have been identified in members of the kinetoplastid group supporting the existence of a complete selenocysteine synthesis pathway in these organisms. Currently, nothing is known regarding the selenocysteine pathway in Trypanosoma evansi. In this study, we identified the expression of the elements selB, selC, selD, PSTK and selTRYP at the mRNA level in T. evansi. All translated proteins (selD, PSTK, selTRYP and selB) have the domains predicted and higher identity with Trypanosoma brucei. gambiense. The selenophosphate synthetase protein was localized in the cytoplasm. Our results support the existence of an active selenocysteine pathway in T. evansi.

Opis:
Trypanosoma evansi is a blood parasite principally transmitted by mechanical vectors (tabanids and stable flies) in large animals such as livestock. However, in other types of hosts, such as carnivores and rodents, oral transmission may be more important. In this experiment, attempts were made to infect rats and mice by the peroral route using infected blood, and ticks engorged on infected rats, in order to evaluate the potential role of ticks as passive vectors of trypanosomes. A strain of Trypanosoma evansi isolated from a cow in Thailand was grown in a rat and blood was collected at the peak of parasitaemia. In the first experiment, 5 rats and 5 mice were fed respectively with 1 ml and 0.5 ml of blood containing 107 Trypanosoma evansi/ml. In the second experiment, adult ticks belonging to the species Rhipicephalus sanguineus, which had fed on parasitaemic rats, were given as food to 3 healthy rats. For both experiments, the presence of parasites in the blood of the rats and mice was checked daily for 10 days, then every 2 days for the following 20 days. Within an average of 4.5 days post blood ingestion (from 4 to 5), 80% (CI95% 29–99) of the rats exhibited parasites by direct microscopic examination of the blood. Similarly, with an average of 4.7 days post ingestion (from 4 to 6), 60% (CI95% 15–95) of the mice exhibited blood parasites. After tick ingestion, no parasites were found in the blood of the rats fed with infected engorged ticks. Consequently, in this experiment, as in others, rats and mice appeared to be receptive by the oral route, but the possible role of ticks as a passive vector could not be demonstrated. Other models could be explored, involving the cattle tick (Rhipicephalus (Boophilus) microplus), to investigate the link from large to small animals.

20 individuals of Abramis brama from Gosławskie Lake and 10 individuals from Gopło Lake, central Poland, were investigated on the presence of trypanosomes. The infections of Trypanosoma abramidis LAVERAN and MESNIL, 1904 were detected in three breams from Gosławskie Lake and two in Gopło Lake. The question of the distinctivity of T. abramidis from T. carassii MITROPHANOW, 1883 found in other Cyprinidae in Poland is discussed.

The studies were carried out in Kosewo Górne in the Mazurian District (North-East region of Poland), in September 1995, September 1996 and between May 1997 and June 1998 each month. The animals were trapped in live traps, and after study they were marked and released. The infection of trypanosomes was detected using microhaematocrit centrifugation technique and in blood smears. The intensity of infection had the range from 50 to 150000 individuals in 1ml of blood. The maximal prevalence of infection was in August and September and there were 45% and 38% of infected voles respectively. The infection was detected also in May 1998, in other months the prevalence had low level. There were no individuals of bank vole infected in succecding months. The females of bank vole are more often infected with trypanosomes than males.

212 freshwater fishes, belonging to 7 species, were examined for the presence of trypanosomes. The fishes were coming from breeding ponds in Gołysz (Silesia). The blood smears were fixed with methyl alcohol and stained with Giemsa method. Trypanosoma carassii MITROPHANOW, 1883 was found in blood of roach Rutilus rutilus (L.) (in 4 out of 19 examined specimens) and tench Tinca tinca (L.) (in 2 examined fishes). In trypanosomes from blood of both species of host there was observed a considerable variety of forms and size of body. Morphological description is completed with morphometrical values in the Tab. 2. Our own morphological description and morphometrical values have been compared with those of other authors. The authors of this paper agree with the opinion of LOM and DYKOVA (1992) that Trypanosoma tincae LAVERAN et MESNIL, 1904 is the synonym of Trypanosoma carassii.

The Yucatan Peninsula of Mexico is endemic with Chagas disease. The main vector responsible for Trypanosoma cruzi transmission is Triatoma dimidiata which is abundant in domestic, peridomestic and sylvan cycles. The abundance of vectors favours T. cruzi transmission and is a high risk for developing chronic chagasic cardiomyopathy (CCC). In the past 10 years, little information was available on parasite seroprevalence and the prevalence of CCC in the Yucatan Peninsula. In the present work, we studied two Mayan communities with a high abundance of T. dimidiata and a random serial sample of 233 patients with an altered electrocardiogram or cardiac failure admitted to the Regional Hospital. A homemade enzyme-linked immunosorbent assay and indirect immunofluorescence standardized techniques were used to detect anti-T. cruzi IgG. In addition, Mayan volunteers were monitored by electrocardiography. In the Mayan communities, 4.8% (3/63) subjects were positive for T. cruzi antibodies none of them presented electrocardiographic alterations, however in seronegative subjects were detected right or left ventricle hypertrophy in 25% (16/63). A remarkable finding was that 90% of the Mayan population recognized the vector and 65% of them had experienced contact with triatomines bites. At the Regional Hospital 0.42% (1/233) were positive for T. cruzi antibodies showing compatible diagnosis with CCC; the most frequent pathology in this population was hypertension in 65% (151/233) and the less frequent was dilated myocardiopathy 6% (14/233). In conclusion, the prevalence of T. cruzi infection and CCC can be considered low in Yucatan, Mexico.

Opis:
The studies were carried out in Kosewo Górne in the Mazurian District (North-East region of Poland), in September 1995, September 1996 and between May 1997 and June 1998 each month. The animals were trapped in live traps, and after study they were marked and released. The infection of trypanosomes was detected using microhaematocrit centrifugation technique and in blood smears. The intensity of infection had the range from 50 to 150000 individuals in 1ml of blood. The maximal prevalence of infection was in August and September and there were 45% and 38% of infected voles respectively. The infection was detected also in May 1998, in other months the prevalence had low level. There were no individuals of bank vole infected in succecding months. The females of bank vole are more often infected with trypanosomes than males.

Opis:
Two from three brown rats (Rattus norvegicus) catched in Milanówek near Warsaw, and one catched in the centre of Warsaw were infected with Trypanosoma lewisi. The morphometric features of trypanosomes were in accordance with the features of T. lewisi gived by other authors. This is first note about infection of free living rats with T. lewisi in Poland.

Opis:
The application of biochemical and molecular techniques in parasitological studies has provided increasing evidences of genetic polymorphism among parasite populations. This review presents possible origins of genetic variation within populations of various protozoan species. Since the mode of reproduction has an important influence on genetic polymorphism within parasite populations these considerations refer mainly to some protozoan parasites which have various life cycles, e.g. Giardia, Trypanosoma, Cryptosporidium, Toxoplasma. Also other factors associated with parasites (such as: transmission and passage history in laboratory conditions; occurrence in different hosts or geographic regions; selective pressure of drugs; competitive interactions between populations) that affect parasite genetic diversity are discussed. However, the number of examined isolates of parasites and genetic markers, assortment of methods, probes, primers and reagents used is also of significance. The significance of genetic variability in parasite populations is still the subject of much interest and controversy. A simple interpretation of such variation is impossible because of the complexity of host-parasite interactions. The knowledge of parasite diversity at the nucleic acids level has continually increased, but a corect interpretation of this phenomenon requires at least the same knowledge of genetic variability in host populations. Nevertheless, genetic variability in protozoan parasites has many important implications, e.g. for taxonomy, epidemiology, control and evolution. Genetic differences within parasite populations might also be associated with phenotypic variability, e.g. virulence, antigenicity, infectivity, drug sensitivity, hostpreference etc.

The scope of this work is the presentation of new theories concerning the role of complement (C) in the resistance of high organism to the infection caused by protozoa. The mechanisms protecting of Entamoeba histolytica, Leishmania spp., Trypanosoma cruzi, Toxoplasma gondii and Naegleria fowleri from the action of C are also described.
