Badano wpływ dymu tytoniowego, aniliny, relanium oraz diety niskobiałkowej na zdolność rozrodczą i potomstwo szczurów. Badania wykonano na sześciu miotach białych szczurów. W pierwszym etapie badano efekty działania dymu tytoniowego, którego stężenie kontrolowano drogą pomiaru tlenku węgla w powietrzu komory doświadczalnej.
The effect of tobacco smoke and low-protein diet on rat reproducibility and offspring was investigated in six litters of white rats. Smoke concentration was controlled by determining the air carbon oxide content which aberaged 1000 mg/m3. During the experiments blood carboxyhemoglobin level was measured; by the end of exposure, each generation displayed closely similar concentrations amounting to 23.9-26.6%. There were no diet-related differences in blood carboxyhemoglobin concentrations.

The induction of acidic proteins in seven potato cultivars (Drop, Atol, Sokół, Tarpan, Jagna, Irga, Darga) was analysed. It has been found that induction of the acidic pathogenesis-related (PR) proteins in potato leaves infected with a necrotic strain of potato virus Y (PVY-N Wi) is specific to each cultivar.

Four-month-old female Wistar rats were exposed for 20 days to tobacco smoke obtained from non-filter cigarettes. During the exposure, concentration of tobacco smoke was monitored indirectly by measuring the CO level (1500 mg/m3 air). The efficacy of exposure was assessed by measuring urine nicotine and cotinine levels. Cigarette smoke did not change total cytochrome P450 and b5 protein levels in any of the organs studied, and most of these organs did not show any changes in the activity of reductases associated with these cytochromes. Following exposure to tobacco smoke, fetal rat liver expressed CYP2B1/2 protein; in newborns (day 1) both liver and lung showed CYP2B1/2 protein expression and very low pentoxyresorufin O-dealkylase activity. Western blot analysis of adult liver, lung, heart, but not of brain microsomes, showed that tobacco smoke induced CYP2B1/2 in both nonpregnant and pregnant rats, though its expression was lower in the livers and hearts of pregnant females. In the rat and human placenta, neither rat CYP2B1/2 nor human CYP2B6 showed basal or tobacco smoke-induced expression at the protein level. This study shows clearly that the expression of CYP2B1/2, which metabolizes nicotine and some drugs and activates carcinogens, is controlled in rats by age-, pregnancy-, and tissue-specific regulatory mechanisms.

Inoculation of tobacco cv. Xanthi nc or bean plants with the mixtures of benzothiadiazole (Bion) and tobacco mosaic tobamovirus (TMV) or alfalfa mosaic virus (AIMV), respectively did not show any inhibition of the number and size of the local lesions. Protective treatment of plants with Bion caused a significant decrease in disease incidence. In the case of tobacco cv. Xanthi nc and TMV or bean plants and AIMV that protective effect increased day by day and 6-7 days after treatment the production of local lesions was inhibited almost completely. Bean plants treated with Bion demonstrated resistance ranging between 60-90% also in nontreated parts. Bean and tomato plants pretreated with 0.01% Bion were effectively (in 60-70%) protected against systemic infection by tomato black ring nepovirus (TBRV).
Badano możliwość zastosowania benzothiadiazolu (Bion) do ograniczania infekcji wirusowych przenoszonych na drodze mechanicznej. Bion dodany do inokulum przed inokulacją nie inhibitował ani liczby, ani średnicy plam nekrotycznych powodowanych przez wirus mozaiki tytoniu (TMV) i wirus mozaiki lucerny (AIMV) odpowiednio na tytoniu odmiany Xanthi nc oraz fasoli. Potwierdza to fakt, że Bion bezpośrednio nie oddziałuje na patogeny, w tym na wirusy. Prewencyjne zastosowanie Bionu powodowało znaczne ograniczenie infekcji wirusowych zarówno lokalnych jak i systemicznych. W przypadku tytoniu odmiany Xanthi nc i fasoli odporność indukowana przez Bion, przejawiająca się inhibicją tworzenia plam przez wirus, narastała z dnia na dzień, aby po 6-7 dniach osiągnąć maksymalny poziom. Efekt ten utrzymywał się przez dłuższy czas ( co najmniej kilkanaście dni). Bion indukuje odporność, nie tylko w traktowanych częściach rośliny, ale także poza nimi, systemicznie, która na fasoli wyrażała się 60-90% redukcją liczby plam nekrotycznych. Około 60% roślin fasoli i 70% pomidora traktowanych Bionem było wolnych od systemicznej infekcji wirusem czarnej pierścieniowej plamistości pomidora (TBRV). Powyższe wyniki świadczą o tym, że odporność wzbudzona przez Bion w roślinie jest skierowana także przeciwko wirusom. Dla praktycznego zastosowania bardzo istotne jest określenie w jakim stopniu aktywność Bionu przenosi się na infekcje roślin wywołane przez wirusy, które są przenoszone przez wektory, głównie mszyce będące podstawowym sposobem rozprzestrzeniania się wirusów w naturze.

Conditions of achieving the maximal accumulation of sulfhydryl metabolites in the leaves of tobacco were explored. Simultaneous production of bacterial O-acetylserine (thiol)-lyase and serine acetyltransferase resulted in the increased thiols contents as compared to single transformants and controls. However, leaf discs feeding experiments differently affected thiols concentration in different plant groups and suggested that the most promising strategy to obtain plants with a high level of non-protein thiol-containing compounds might be sulfate feeding to plants overproducing serine acetyltransferase.Formation of cysteine from sulfide and O-acetyl-L-serine (OAS) is catalyzed by O-acetylserine (thiol)-lyase (OAS-TL) (EC 4.2.99.8) while OAS is synthesized by serine acetyltransferase (SAT) from acetyl-coenzyme A and serine. Molecular interactions between SAT and OAS-TL are involved in the regulation of the enzymatic activities of these proteins in bacteria (Mino et al., 2000) and in plants (Bogdanova & Hell, 1997; Droux et al., 1998). Experiments with Escherichia coli enzymes clearly indicated that OAS-TL activity was reduced to 30% in a complex (Mino et al., 2000). Similarly, a very dramatic decrease of the catalytic activity of OAS-TL bound in a complex has been reported for the plant enzymes (Droux et al., 1998). On the other hand, the bienzyme complex formation stabilizes bacterial SAT (Mino et al., 2000) and increases the apparent affinity for the substrates of plant SAT (Droux et al., 1998). The stability of the complex is negatively affected by OAS and positively by sulfide (Droux et al., 1998). Bacterial SAT is feedback regulated by cysteine, however, no relationship seems to exist between the complex formation and SAT sensitivity to this inhibition (Mino et al., 2000).Both enzymes, SAT and OAS-TL, are located in three compartments of the plant cell: the cytosol, chloroplasts and mitochondria. Different isoforms of these enzymes are in a different way regulated by sulfur nutrition (Nakamura et al., 1999; Takahashi et al., 1997; Warrilow & Hawkesford, 1998). The feedback regulation by L-cysteine of various isoforms of plant SAT has recently been studied (Inoue et al., 1999; Noji et al., 1998). According to the model proposed, the only role of the chloroplastic and mitochondrial isoforms (that are insensitive to the feedback inhibition) would be the production of OAS for cysteine biosynthesis. The cysteine-sensitive cystosolic isoform of SAT would, according to this model, have two roles: (i) OAS production for cysteine biosynthesis in the cystosol and (ii) control of OAS pool for regulatory purposes. The second postulated function is tightly connected with the fact that OAS acts as a positive regulator of genes whose expression is affected by sulfur status (Saito, 2000).Glutathione, the main low-molecular-mass thiol-containing compound in the plant cell, has multiple functions, including involvement in responses to various environmental stresses and maintenance of the redox homeostasis (Foyer & Rennenberg, 2000). Under non-stressing conditions the majority of glutathione is maintained in the reduced form (GSH) and its concentration is determined mainly by the rate of biosynthesis. GSH is produced from cysteine, glutamate and glycine in two steps catalyzed by γ-glutamylcysteinyl synthetase (γ-ECS) and glutathione synthetase (GS), respectively (Noctor et al., 1998). The biosynthesis and accumulation of GSH has been shown to depend on (i) the activity of γ-ECS, (ii) the availability of cysteine, and (iii) the light-dependent formation of glycine through the photorespiratory pathway (Foyer & Rennenberg, 2000).The main aim of this study was to identify the optimal conditions for the maximal accumulation of non-protein sulfhydryl metabolites in plant leaves. The transgenic tobacco plants with cytosolic production of bacterial enzymes of the cysteine biosynthesis pathway, SAT and OAS-TL, were obtained and analyzed for the transgenes expression. Additionally, leaf discs of either single or double transformants, as well as of control plants, were assayed for the thiol contents upon incubation in solutions of various compounds expected to have an influence on sulfur metabolism.

Towards the end of June of 1997 a massive outbreak of blue mold caused by Peronospora tabacina on the experiment plots of the Institute of Soil Science and Plant Cultivation gave an opportunity to observe and record the response to that disease of 192 different N. tabacum genotypes held at the institute’s tobacco germplasm collection. Both the date of the onset of the first symptoms and the intensity of the symptoms varied. Symptoms were from very severe (e. g. Virgin Mutant A, Burley White) to severe, moderate, mild and none. Less than 10% of the accessions scored for blue mold were symptom-free. They belonged to the flue-cured type (15 out of 71) and dark air-cured (5 out of 49). All the remaining entries including orientals and semi-orientals, burleys and cigar tobaccos were affected to some extent. Except the very susceptible Virgin A Mutant systemic symptoms were rare probably because the fungus attacked relatively late in the season when the majority of accessions had already grown out of the most vulnerable stage and were at either budding or flowering.

As continuation of investigations in to the mechanism of the role of the H1 histone, which is a crucial protein component chromosomes of all eukaryotes, transgenic tobacco plants with different levels of the H1 histone variants were examined. Tobacco has six sequential variants of the H1 histone: two major ones (H1A and H1B), constituting ca. 90% of all H1, and four minor ones (H1C, H1D, H1E and H1F), occurring in very small quantities. The following groups of plants were examined: K - control group with a full set of histone variants; -AB -with the A and B variants removed; -ABCD - with the A, B, C and D variants removed; and -CD - with the C and D variants removed. The analysis of microsporogenesis in those plants, based on preparations squeezed in acetoorcein, revealed the asynchronous course of meiosis in -AB and -ABCD plants, occurrence of chromosomal aberration, and, consequently, the formation of sterile pollen grains (accordingly: 84,4% and 81,4%). In -CD plants, the percentage of aberration and sterile pollen grains was similar to the control material. Electron microscope observations of microsporogenesis showed ultrastructural changes. In -AB and -ABCD plants, a major portion of the pollen grains were degraded. The smallest number of degraded pollen grains, in comparison with the control, was found in the -CD group.

It was analytically examined how great amount of total iron can be absorbed during cigarette smoking. For this purpose, tobacco taken from cigarettes of selected brands (Marlboro, L&M, and Popular) was carefully analyzed before and after smoking. In addition, the cigarette filters were checked in terms of iron content. In all samples iron was determined by means of spectrophotometric method with sulfosalicylic acid. The method was realized with the aid of an original flow system allowing the analytical calibration to be performed with high reliability. As a result iron present in a sample in trace amount was able to be found with very good accuracy and with precision not exceeding 1.5% (in RSD). It was stated that typical amount of iron in a single cigarette is ranged between 0.3 and 0.4 mg. When smoking the risk should be taken into account that even 99% of this amount is transferred to human body without any chance to be limited by the cigarette filter.

We applied the yeast two-hybrid system for screening of a cDNA library of Nicotiana plumbaginifolia for clones encoding plant proteins interacting with two proteins of Escherichia coli: serine acetyltransferase (SAT, the product of cysE gene) and O-acetylserine (thiol) lyase A, also termed cysteine synthase (OASTL-A, the product of cysK gene). Two plant cDNA clones were identified when using the cysE gene as a bait. These clones encode a probable cytosolic isoform of OASTL and an organellar isoform of SAT, respectively, as indicated by evolutionary trees. The second clone, encoding SAT, was identified independently also as a "prey" when using cysK as a bait. Our results reveal the possibility of applying the two-hybrid system for cloning of plant cDNAs encoding enzymes of the cysteine synthase complex in the two-hybrid system. Additionally, using genome walking sequences located upstream of the sat1 cDNA were identified. Subsequently, in silico analyses were performed aiming towards identification of the potential signal peptide and possible location of the deduced mature protein encoded by sat1.

W pracy przedstawiono wyniki badań nad efektywnością zagospodarowania zbrykietowanego pyłu tytoniowego w procesie kompostowania z dojrzałym kompostem (z komunalnego osadu ściekowego i słomy pszennej) i świeżym osadem czynnym po stabilizacji tlenowej. Celem badań było określenie możliwości uzyskania stabilnego kompostu z przeznaczeniem na cele nieprzemysłowe oraz sprawdzenie tempa rozkładu zbrykietowanego pyłu tytoniowego charakteryzującego się bardzo dużą twardością. Doświadczenie obejmowało dwie pryzmy kompostowe i prowadzone było przez pięć miesięcy. Na podstawie przeprowadzonych badań stwierdzono, iż proces kompostowania odpadu tytoniowego z dojrzałym kompostem i osadem ściekowym w przyjętych proporcjach stosunku C:N stanowi korzystną alternatywę zagospodarowania tego odpadu w stosunku do dotychczas stosowanych metod.
This study undertook investigations on the elaboration and determination of the utilization efficiency of briquetted tobacco dust in the composting process using mature compost (with municipal sewage sludge and wheat straw) and fresh municipal aerobic sewage sludge. The objective of performed investigations was to ascertain possibilities of obtain of a stable compost intended for non-industrial purposes and to check up on rate of biodegradation of briquetted tobacco dust characterized by exceptional hardness. The experiment comprised two compost piles and was conducted for five months. The performed studies indicated, that composting of tobacco waste with mature compost and sewage sludge in estimated C:N ratio is advisable alternative of this waste management to till now using methods.

Trzyletnie doświadczenie polowe (1996-1998) miało na celu określenie składu chemicznego liści tytoniu odmiany ‘Wiślica’ w warunkach zróżnicowanego nawożenia azotem. Na podstawie badań stwierdzono, że w warunkach niedoboru azotu liście tytoniu charakteryzują się niską zawartością N oraz wysokim poziomem K, Ca i suchej masy. Zastosowanie azotu w nawozach mineralnych, szczególnie w postaci saletry amonowej, powodowało wzrost zawartości N i stopniowe obniżanie poziomu pozostałych badanych składników. Skład chemiczny liści tytoniu ulegał zmianie w trakcie wegetacji roślin: zwiększała się zawartość suchej masy, malała koncentracja potasu, zaś stężenie azotu i wapnia miało charakter zmienny.
The aim of the three years lasting (1996-1998) field experiment was focused to define tobacco (cultivar ‘Wiślica’) leave chemical composition under different nitrogen fertilization conditions. The research proved that with nitrogen deficiency in soil, tobacco leaves were characterised by a low nitrogen content and high potassium, calcium and dry matter content. Nitrogen mineral fertilizers, particularly ammonium nitrate, caused an increase of nitrogen content and progressive decrease of other monitored components. Chemical composition of tobacco leaves changed during vegetation period: dry matter content incread, potassium content decreased and nitrogen and calcium concentration had variable characteristics.

The phenomenon of male sterility has often been observed in investigations on the role of histone H1 in regulation of morphogenetic and cytological processes in transgenic tobacco plants. These changes were accumulated by disturbances in flower development, consisting in lengthening of the pistil style in relation to stamen heads. This prevented pollination and production of seeds. As similar abnormalities occurred also in the present investigations (depending on combination, the sterility% was 84.4 to 19.9, at only 8.1 in the control), the main problem of our investigations was an attempt to explain their reasons. It is commonly known that one of the conditions for formation of fertile pollen is the properly functioning tapetum. Here, we carried out observations of ultrastructure of anther tapetum control cells in respect of abnormalities which occurred during microsporogenesis of transgenic plants with inactivated expression of two major (A, B) and two minor (C, D) histone H1 variants. The investigations were carried out on the following groups of plants: (1) control group with a full set of histone variants (K), (2) with inactivated A and B variants (-AB); (3) with inactivated A, B, C and D variants (-ABCD), (4) with inactivated C and D variants (-CD). It was found that tapetal development was normal in all the investigated groups of plants, and the sequence of changes was similar as in the control. However, certain ultrastructural differences appeared when tapetum functioned as secretory tissue, and in the degeneration phase. In tapetal cell cytoplasm, with participation of rER, lipid bodies were formed, which, having penetrated to the cell surface and to locules, took part in formation of pollen grain sporoderm. Both in the control and in the remaining combination, excluding -ABCD, these bodies looked similar: they were grey, homogenous and surrounded by black jagged deposits. In -ABCD plants, these bodies were more translucent, slightly rarefied, and not surrounded by the deposits. Moreover, in -CD plants, large lipid deposits were frequently observed between remainders of degraded tapetal cells. They did not occur in the control and the remaining combinations.

In contrast to mRNAs, ribosomal RNAs are generally not considered to be polyadenylated. Only a few recent reports describe non-abundant polyadenylated rRNA-related transcripts that have been detected and characterized in yeast and in human cells. Here we depict the phenomenon of 26S rRNA polyadenylation and degradation that was observed in shoots of Nicotiana tabaccum plants grown in the presence of cadmium. Fragments corresponding to 26S rRNA were identified using suppression subtractive hybridization during screening for genes induced in tobacco plants upon a three-week exposure to 15 µM cadmium chloride. Extracts prepared from the above-ground tissues of cadmium-treated tobacco plants were supposed to contain exclusively polyadenylated mRNAs. Surprisingly, numerous polyadenylated fragments matching parts of 26S rRNA were identified and their presence was confirmed by Northern blot and cDNA amplification techniques. To our knowledge this is the first report on rRNA polyadenylation in plants.
