Aegle marmelos leaves contained π-sitosterol, aegelin, lupeol, rutin, marmesinine, flavone, glycoside, Oisopentenyl halfordiol, marmeline, and phenylethyl cinnamides. The additional growth promoters show its effect on tissue culture of plant.

The supply of seed yams for intensive yam production is hindered by many constraints, including diseases and pest infestations as well as the unavailability of quality planting material. The combination of tissue culture and aeroponics system is perceived to be the way towards clean and adequate supply of seed yam for enhanced yam production. However, the system is considered as expensive for any individual to implement. In order to encourage private sector to participate in this venture, the economic analysis determining the feasibility and viability of using aeroponics in seed yam production was performed. Using data from established tissue culture and aeroponics system in Ghana, the fixed cost and variable cost parameters as well as production costs were obtained. Results revealed that total cost of building aeroponics structure amounted to GH₵ 94,178.00 (USD 17,938.70). Annual cost of aeroponics structure was GH₵ 9,417.82 (USD 1,793.87). Annual total cost of production totalled GH₵ 204,391.75 (USD 38,931.61). Annual net revenue was GH₵ 75,888.00 (USD 14,454.86). Payback period was 15 months and benefit cost ratio was 1.4. Aeroponics system for seed yam production is therefore profitable since short period would be needed to recoup investment. For food security and creation of workplaces, government could partner with the private sector in the establishment of aeroponics systems to increase yam production and export.

In vitro tissue culture technique, especially micropropagation, is attracting increasing attention in the production of ornamental plants. This technique will probably dominate the horticultural market in the near future. Light is one of the several factors affecting the success of in vitro plant tissue cultures. It directly affects the widely understood morphogenetic response of the explant, i.e., the ability of the explant to grow or regenerate, produce roots, etc. Lighting conditions provided during the in vitro stage may also greatly affect the plant vigor after transferring to nonsterile conditions. Moreover, the necessity of providing artificial light significantly contributes to the total cost of maintaining tissue cultures (related to energy consumption and the need to cool down the heat generated by lamps). Light quantity (intensity) and quality (spectral composition) are the two main parameters that determine its influence on in vitro cultures. This impact depends on the species and other accompanying factors. The aim of this mini-review is to summarize information on the influence of light on the morphogenetic and biochemical response of explants of some selected ornamental plant species grown under in vitro conditions.

Opis:
Microspheres (MS) made of biostable polymer, namely polystyrene, have been used as substrates for cell culture enabling rapid cell expansion in dynamic conditions. However, due to non-resorbability, polystyrene (PS) MS when repopulated with cells cannot be directly used in tissue engineering. Our concept was to produce MS from resorbable polymer – poly(L-lactide- -co-glycolide) (PLGA) as a support for adherent cells, e.g. osteoblasts. We hypothesize that such MS can be applied to the injured site to act as cell carriers or as modules for modular tissue engineering (MTE). In this article, we present the results of optimizing the PLGA MS manufacturing conditions via oil-in-water emulsification. Due to such a technique, MS with the required size, size distribution and properties suitable for cell culturing can be obtained. Three parameters of the oil-in-water emulsification were examined: the stirring speed of a water phase during MS manufacturing, the surfactant concentration, i.e. poly(vinyl alcohol) (PVA) in a water phase and concentration of PLGA in dichloromethane (DCM) as an oil phase. The results proved that the 7.5% PLGA concentration in DCM solution as an oil phase, the 0.5-2% concentration of PVA solution as a water phase and the stirring speed of water phase of 1000 rpm provided MS with the 160 μm mean diameter, which is suitable for cell culture. Moreover, the developed sieving and cleaning procedures were efficient to collect MS with the mean diameter of 280 μm, the more coherent size distribution and the ability to sink in the cell culture medium. The presence on the bottom of cell culture wells is crucial for MTE.

Opis:
The paper constitutes a brief and subjective review of polymeric materials in the contemporary health service. The range of applications of polymeric materials is discussed, special att ention being paid to such materials for the development of carriers of pharmaceutically active species, stents and vascular prostheses, amongst them to the application of „smart” materials for these purposes, layers and scaff olds for the growth of organs and tissues, antifouling layers. The authors try to turn the att ention of the reader to the research and intellectual eff orts necessary for the development of polymeric materials for the medicine, and conclude about the growing importance of such studies.
Artykuł stanowi skrótowy, subiektywny przegląd materiałów polimerowych wykorzystywanych we współczesnej ochronie zdrowia. W pracy skupiono się na zastosowaniach materiałów polimerowych do konstrukcji nośników leków, stentów i protez naczyń, w tym także na użyciu polimerowych materiałów „inteligentnych”, implantów ortopedycznych oraz podłoży i rusztowań do hodowli komórek lub tkanek, a także warstw zapobiegających porastaniu wszczepionych konstruktów. Autorzy zwracają uwagę na znaczny wysiłek badawczy i intelektualny, niezbędny w procesie opracowania materiałów polimerowych dla medycyny, i na stale rosnące znaczenie takich badań.

Mandevilla sanderi is an important commercial ornamental pot plant. Traditional vegetative propagation is limited due to the low rate, therefore there is a need to develop an alternative, more efficient method. There is an interest in development of micropropagation technology for the species, as it allows to obtain a lot of offsprings in a relatively short time. The aim of the present work was to estimate an influence of explants type and position on regeneration of Mandevilla sanderi in tissue culture. Four different types of explants (leafy shoot tips, decapitated leafy shoot tips, defoliated shoot tips, decapitated and defoliated shoot tips) were used in the experiment, which were placed on the media vertically, while defoliated shoot tips were placed horizontally or vertically upside down. The explants were cultivated on a Murashige and Skoog (MS) medium supplemented with 1 mg·dm–3 benzyladenine (BA) and 0.5 mg·dm–3 indole-3-butyric acid (IBA). It was noted that both explants orientation and positioning, influenced the multiplication rate. Defoliated shoot tips placed horizontally were characterized by higher multiplication rate (6.8) in comparison to upside down vertical positioning (3.2). It was also observed that removal of shoot apex improved axillary branching, while defoliation of shoots placed in a normal position reduced multiplication rate.

The aim of the work was to evaluate the influence of cytokinins on in vitro propagated dahlia and their consequent effect on acclimatization. Plant material consisted of shoot tips and nodes. Among the three cytokinins, benzyladenine, kinetin and 2-isopentenyl-adenine, only BA effectively stimulated the shoot multiplication from axiliary buds. The highest multiplication rate was obtained from nodes in the presence of 0.25– 0.5 mg·dm–3 BA. Higher concentrations shortened the internodes and decreased the leaf blades and growth of callus. 1 mg·dm–3 of KIN and 2iP positively influenced the shoot growth and size of leaves. Gibberellic acid (GA3) used with BA increased the number of auxillary shoots. The best quality shoots and the highest multiplication rate were obtained when 2 mg·dm–3 BA was used with 5 mg·dm–3 GA3. Cytokinins affected the rooting and acclimatization ex vitro. Dahlia shoots multiplicated in the presence of 1 mg·dm–3 KIN or 2iP rooted faster in the soil and 100% survived in field, while those from 1 mg·dm–3 BA media rooted slowly, had shorter shoots and only 60% of them survived. Plants bloomed after 11–12 weeks in the field. Dahlia plants that had been multiplicated in the presence of KIN had larger diameter and fresh weight in the field. BA and 2iP positively influenced the flower diameter, length of flower stalk and a number of the first-order shoots.

Opis:
Initiation of tissue culture of many plant species is a very difficult stage due to appearance of many contaminations. The other problem might be a choice of media for regeneration. Initiation of grass species tissue cultures are thought to be very difficult. Therefore, a research was undertaken to evaluate the use of nano-silver particles for plant material disinfection and to estimate a medium Pennisetum alopecuroides. The plant material were buds and nodal explants that were disinfected in 2% NaOCl for 30 min or 0.1% HgCl2 for 1 min. Half of the explants disinfected with NaOCl were soaked in 50, 100 or 250 mg·dm–3 Ag NPs for 1 hour. Explants not soaked in nano-silver were placed on media with Ag NPs at concentrations of 4, 8 or 16 mg·dm–3. An influence of growth regulators on Pennisetum alopecuroides was evaluated in vitro. Regenerated shoots were placed on MS media with: 3 mg·dm–3 BA + 0.3 mg·dm–3 IBA, 3 mg·dm–3 KIN + + 0.3 mg·dm–3 IAA, 1 mg·dm–3 BA + 0.1 mg·dm–3 IBA. It was observed that the use of nano-silver particles lowered the level of contamination. The best results were obtained when Ag NPs was used at concentration of 100–250 mg·dm–3 alone or as a supplementation of the media, at concentration of 4 mg·dm–3 for nodes and 16 mg·dm–3 for adventitious buds. The use of nodal explants allowed to obtain less contamination. Regeneration depended on a media content. The most regenerated shoots were obtained on the MS media supplemented with 1 mg·dm–3 BA and 0.1 mg·dm–3 IBA.

Sucrose phosphate synthase (SPS) is a key enzyme catalyzing sucrose metabolism in plants. In this study, we isolated the SPS cDNA from Saccharum spontaneum and designated as SsSPS (GenBank accession no. MF398541). The full-length of SsSPS cDNA was 4153-bp with an opening reading frame (ORF) of 3132 nucleotides, which encoded a 1043-amino acid protein. The nucleotide sequences alignment showed that it had 98%, 97% and 87% homology with S. officinarum, Setaria italica and Lolium perenne, respectively. Moreover, the SsSPS was detected to express in leaf and stem tissues of S. spontaneum and exhibited a predominant expression in the stem tissue. However, there was no significant difference in the expression level of SsSPS between young leaves and mature ones. Additionally, we generated transgenic S. spontaneum using Agrobacterium-mediated transformation. Our data will provide a valuable foundation for further study of the potential role of SPS in plants.

Opis:
A mathematical model of a hybrid culture system supported with a stationary layer of liquid perfluorochemical (PFC) as a source of O2 for cells which grow in the aqueous phase of culture medium has been developed and discussed. The two-substrate Monod kinetics without inhibition effects, i.e. the Tsao-Hanson equation, has been assumed to characterise the biomass growth. The Damköhler number which relates the growth rate to the mass transfer effects has been used to appraise the regime (i.e. diffusion-limited or kinetics) of the whole process. The proposed model predicted accurately previously published data on the submerged batch cultures of Nicotiana tabacum BY-2 heterotrophic cells performed in a culture system supported with a stationary layer of hydrophobic perfluorodecalin as a liquid O2 carrier. Estimated values of the parameters of the model showed that the process proceeded in the kinetics regime and the growth kinetics, not the effects of the mass transfer between aqueous phase and liquid PFC, had essential influence on the growth of biomass.
