Opis:
Obesity is one of the major causes of metabolic diseases such as diabetes and heart attack, and, hence, can lead to low quality of life. Elaborating adipocyte differentiation is very crucial for formulating the treatment and prevention of obesity. The objective of this study is to investigate the seeding cell number required to obtain optimum lipid accumulation during adipocyte differentiation using the 3T3-L1 cell line. Two sets of 5.48×104 (for Day 0 and Day 8 of differentiation), of 10.96×104 (for Day 8) and of 21.92×104 (for Day 8) of 3T3-L1 cells were seeded in each wells of a 12-well plate. Isobuthylmethylxanthine (IBMX), Dexamethasone, and Insulin-containing differentiation cocktails was added into the medium at Day 0 for 48 hours. The medium was changed every two days. Day 0 and Day 8 samples were then stained using Oil Red O and were examined under the microscope to observe the lipid droplets (red-coloured). The lipid droplets were quantified by measuring the absorbance at wavelength of 550 nm. In the study, seeding the number of 10.96×104 cells produced very significantly higher lipid accumulation, as compared with seeding the number of 5.48×104 cells. However, doubling the seeding number into 21.92×104 cells did not increase the lipid droplets significantly. This study found that the optimum seeding number to obtain the maximum lipid droplets during 3T3-L1 adipocyte differentiation was 10.96×104 cells.

Opis:
Chimeric antigen receptor T (CAR-T) cell therapy has been proven to be successful against different leukaemias and lymphomas. Its success has led, in recent years, to its use being tested for different solid tumours, including glioblastoma, a type of primary brain tumour, characterised by aggressiveness and recurrence. This paper presents an analytical study of a mathematical model describing the competition of CAR-T and glioblastoma tumour cells, taking into account their immunosuppressive capacity. The model is formulated in a general way, and its basic properties are investigated. However, most of the analysis considers the model with exponential tumour growth, assuming this growth type for simplicity. The existence and stability of steady states are studied, and the subsequent focus is on two different types of treatment: constant and periodic. Finally, protocols for CAR-T cell therapy of glioblastoma are numerically derived; these are aimed at preventing the tumour from reaching a critical size and at prolonging the patients’ survival time as much as possible. The analytical and numerical results provide theoretical support for the treatment of glioblastoma using CAR-T cells.

Opis:
Przeciwwirusowe terapie dla pacjentów z wirusem HIV są stale ulepszane. Podawanie kombinacji dwóch lub więcej leków powoduje spadek liczby wirusów poniżej poziomu detekcji u wielu pacjentów. W szczególności połączenie wysoce efektywnej terapii antyretrowirusowej (HAART) z aktywatorem immunologicznym (interleukiną 2, IL-2) wydaje się stanowić skuteczną metodę długookresowej kontroli replikacji wirusa  HIV in vivo.W naszym artykule zaproponowaliśmy matematyczny model odpowiedzi odpornościowej dla pacjentów z wirusem HIV przy zastosowaniu połączonej terapii  HAART i IL-2. Zbadaliśmy dynamikę modelu w poszukiwaniu warunków, przy których układ odpornościowy odnawia się dzięki takiej terapii. Z~analitycznego punltu widzenia oznacza to globalną stabilność stanu stacjonarnego odpowiadającego zdrowemu organizmowi. Analiza matematyczna została uzupełniona symulacjami komputerowymi.
Antiviral therapy for HIV-infected patients has greatly improved in recent years. Administration of drug combinations consisting of two or more different drugs can reduce and maintain virus load below detection level in many patients. Cyclic administration of the immune activator interleukin-2 (IL-2) in combination with highly active antiretroviral therapy (HAART) has been suggested as an effective strategy to realize long-term control of HIV replication in vivo. In this article, we formulate a mathematical model of immune response for HIV-infected individual in the presence of HAART and IL-2. We look  for the conditions under which the immune system recovers by applying IL-2 as an immune activator along with HAART. From analytical point of view this means global stability of the disease-free equilibrium. Comprehensive numerical simulations are presented to illustrate the analytical results.

The technology applied for the cloning of the open reading frame (ORF) I and II reflecting DNA fragments of the I-18 C gene of Chironomus tentans in regard to the preparation of the competent E. coli cells of the XL-1 strain needed for the transformation with the ligation reaction products, is presented. Also the transformation of these bacterial cells with the bluescript plasmid and its isolation for these cloning experiments, are described.
Opisano technologię, którą zastosowano do klonowania fragmentów DNA odzwierciedlających translacyjną otwartą ramę odczytu I i II genu I-18 C Chironomus tentans w zakresie: przygotowania kompetentnych komórek E. coli szczepu XL-1, poddawanych następnie transformacji produktami reakcji ligacji. Przedstawiono także zastosowaną technologię transformacji wymienionych komórek bakteryjnych przy użyciu plazmidu - blueskrypt oraz izolację tego plazmidu, w celu jego zastosowania w wymienionych eksperymentach klonowania.

Apoptosis, as the programmed cell death, plays a significant role in proper functioning of an organism, both in the postnatal period and during embryogenesis. Disturbances in this process can lead to the occurrence of several dysfunctions, e.g. cancer, stroke, Alzheimer’s disease and others. Apoptosis can be triggered by factors such as oxidative stress, free radicals, UV radiation and cytotoxic drugs, but also mycotoxins, e.g. aflatoxins, ochratoxin A and trichothecenes. These toxins are produced primarily by fungi of Aspergillus, Penicillium, Fusarium and Stachybotrys genera. The aim of the study was to investigate the effect of mycotoxins on the occurrence of apoptosis in the swine kidney (SK) epithelial cells. For this purpose, trichothecene T2 toxin was used at a concentration of 2.5 µM and 25 µM, aflatoxin B1 at a dose of 10 µM and 30 µM, and ochratoxin A concentrations of 50 µM and 80 µM. The results were assessed using flow cytometer Muse Cell Analyzer (Merck). Studies have shown high sensitivity of the cell line SK on mycotoxins. Apoptosis was caused by all kinds of toxins and depended on the dose of examined substance. T2 toxin at a concentration of 2.5 µM caused apoptosis in 6.9% of the cells, whereas at a concentration of 25 µM in 26.35% of the cells. Aflatoxin B1 used at concentrations of 10 µM and 30 µM caused apoptosis in 13.5% and 40.6% of the cells, respectively. The use of ochratoxin A in concentrations of 50 µM and 80 µM caused the occurrence of apoptosis respectively in 68.51% and 60.41% of the cells.

Psoriasis vulgaris is a common, worldwide autoimmune skin disorder characterized by T-cells mediated hyperproliferation of keratinocytes. The feature of T-cells arbitrated psoriatic lesions is the epidermal infiltration of oligoclonal CD8+ T-cells and also of CD4+ T-cells in the dermis. Psoriatic scratches are identified by red and enlarged lesions along with silver whitish scales. In this article, we propose a mathematical model for psoriasis, involving a set of differential equations, concerning T-cells, dendritic cells and epidermal keratinocytes. We introduce T-cell proliferation in the system, where T-cells are generated through expansion of accessible CD4+ T-cells from precursors. We are interested in observing how the cell biological system develops through T-cell proliferation in presence of control with respect to T-cells and keratinocytes. We study the model in both implicit and explicit ways and measure the effect of drug on the system through impulsive drug therapy.

One major reason for the lack of clinical success of Staphylococcus aureus vaccine candidates is the inability of the antigens to develop a CD4+ T cell-mediated immune response. Hence, it is important to identify CD4+ T cell antigens from S. aureus. CD4+ T cells are activated following the presentation of epitopes derived from exogenous proteins on HLA class II molecules. Fifty-nine secretory proteins of S. aureus were analyzed computationally for the presence of HLA class II binding peptides. Fifteen-mer peptides were generated, and their binding to 26 HLA class II alleles was predicted. The structural feasibility of the peptides binding to HLA-II was studied using molecular docking. Of the 16,724 peptides generated, 6991 (41.8%) were predicted to bind to any one of the alleles with an IC50 value below 50 nM. Comparative sequence analysis revealed that only 545 of the strong binding peptides are non-self in the human system. Approximately 50% of the binding peptides were monoallele-specific. Moreover, approximately 95% of the predicted strong binding non-self peptides interacted with the binding groove of at least one HLA class II molecule with a glide score better than !10 kcal/mol. On the basis of the analysis of the strength of binding, non-self presentation in the human host, propensity to bind to a higher number of alleles, and energetically favorable interactions with HLA molecules, a set of 11 CD4+ T cell epitopes that can be used as vaccine candidates was identified.

Introduction and objective. The study presents the problem of immune disturbances in pregnant women with vulvar carcinoma in situ (VIN3). Material and methods. NK cell and T reg activity in the study patient were analysed using flow cytometry. Results. Decreased NK cell activity and but increased T reg activity were observed after vulvectomy, with subsequent successful pregnancy outcome. Conclusions. Although vulvar cancer may influence immune cell activity, this issue merits further study.

Studies were undertaken to evaluate by parasitological techniques the course of intestinal and muscular invasions in mice infected with Trichinella spiralis and treated with TFX-Thymomodulin (TFX-Th). Studies were conducted on 120 mice of BALB/c and SWISS strains, infected each with a mean invasive dose of 300 T. spiralis larvae. TFX-Th (Thymoorgan GmbH Pharmazie Co. KG, Vienenburg, Germany) was administered subcutaneously at 30 mg/kg body weight. Depending upon duration of TFX-Th administration and stages of intestinal and muscular invasions, three experiments were distinguished in the study. The results indicated that TFX-Th promoted eradication of larvae from muscles, whether administered at the early of the late stage of T. spiralis invasion.
