Opis:
A cysteine-specific methanethiosulfonate spin label was introduced into yeast iso-1-cytochrome c at three different positions. The modified forms of cytochrome c included: the wild-type protein labeled at naturally occurring C102, and two mutated proteins, S47C and L85C, labeled at positions 47 and 85, respectively (both S47C and L85C derived from the protein in which C102 had been replaced by threonine). All three spin-labeled protein derivatives were characterized using electron paramagnetic resonance (EPR) techniques. The continuous wave (CW) EPR spectrum of spin label attached to L85C differed from those recorded for spin label attached to C102 or S47C, indicating that spin label at position 85 was more immobilized and exhibited more complex tumbling than spin label at two other positions. The temperature dependence of the CW EPR spectra and CW EPR power saturation revealed further differences of spin-labeled L85C. The results were discussed in terms of application of the site-directed spin labeling technique in probing the local dynamic structure of iso-1-cytochrome c.

The effects of ozone at different concentrations (10, 30, 45 g/m3) on fluidity and thermotropic properties of erythrocyte membranes were investigated by EPR using two spin probes: 5-doxylstearic acid (5-DSA) and 16-doxylstearic acid (16-DSA). The effect of ozone on the erythrocyte membrane fluidity was a dose-dependent process. The ozone at concentration of 10 g/m3 caused rigidization of the membrane while at concentration of 45 g/m3 increased fluidity both on the surface and in the deeper hydrocarbon region of the membrane. Temperature transitions close to the polar heads region (monitored by 5-DSA) were not sensitive to an increase in ozone concentration. In the case of 16-DSA, low temperature thermotropic transition (around 20°C) gradually decreased with the increase of ozone concentration. High temperature transition (around 40°C) significantly differed at the ozone concentration of 10 g/m3 and 45 g/m3, being higher and lower, respectively, as compared to untreated cells. For the ozone concentration of 45 g/m3 the disappearance of the low temperature break and the appearance of two breaks at 37°C and 16°C were observed.

Opis:
Unsaturated lipids are rapidly oxidized to toxic products such as lipid hydroperoxides, especially when transition metals such as iron or copper are present. In a Fenton-type reaction Fe2+ converts lipid hydroperoxides to the very short-lived lipid alkoxyl radicals. The reaction was started upon the addition of Fe2+ to an aqueous linoleic acid hydroperoxide (LOOH) emulsion and the spin trap in the absence of oxygen. Even when high concentrations of spin traps were added to the incubation mixture, only secondary radical adducts were detected, probably due to the rapid rearrangement of the primary alkoxyl radicals. With the commercially available nitroso spin trap MNP we observed a slightly immobilized ESR spectrum with only one hydrogen splitting, indicating the trapping of a methinyl fragment of a lipid radical. With DMPO or 5-diethoxyphosphoryl-5-methyl-1-pyrroline N-oxide (DEPMPO) adducts were detected with carbon-centered lipid radical, with acyl radical, and with the hydroxyl radical. We also synthesized lipophilic derivatives of the spin trap DEPMPO in order to detect lipid radical species generated in the lipid phase. With all spin traps studied a lipid-derived carbon-centered radical was obtained in the anaerobic incubation system Fe2+/LOOH indicating the trapping of a lipid radical, possibly generated as a secondary reaction product of the primary lipid alkoxyl radical formed. Under aerobic conditions an SOD-insensitive oxygen-centered radical adduct was formed with DEPMPO and its lipophilic derivatives. The observed ESR parameters were similar to those of alkoxyl radical adducts, which were independently synthesized in model experiments using Fe3+-catalyzed nucleophilic addition of methanol or t-butanol to the respective spin trap.

Alloxan can generate diabetes in experimental animals and its action can be associated with the production of free radicals. It is therefore important to check how different substances often referred to as free radical scavengers may interact with alloxan, especially that some of these substance may show both pro- and antioxidant activities. Using the alkaline comet assay we showed that alloxan at concentrations 0.01-50 μM induced DNA damage in normal human lymphocytes in a dose-dependent manner. Treated cells were able to recover within a 120-min incubation. Vitamins C and E at 10 and 50 μM diminished the extent of DNA damage induced by 50 μM alloxan. Pre-treatment of the lymphocytes with a nitrone spin trap, α-(4-pyridil-1-oxide)- N-t-butylnitrone (POBN) or ebselen (2-phenyl-1,2-benzisoselenazol-3(2H)-one), which mimics glutathione peroxides, reduced the alloxan-evoked DNA damage. The cells exposed to alloxan and treated with formamidopyrimidine-DNA glycosylase (Fpg) and 3-methyladenine-DNA glycosylase II (AlkA), enzymes recognizing oxidized and alkylated bases, respectively, displayed greater extent of DNA damage than those not treated with these enzymes. The results confirmed that free radicals are involved in the formation of DNA lesions induced by alloxan. The results also suggest that alloxan can generate oxidized DNA bases with a preference for purines and contribute to their alkylation.

Two isostructural series of polycrystalline compounds: Er2-xYxFe14B and Er2-xCexFe14B have been studied by 57Fe Mössbauer spectroscopy in the temperature range 80-370 K. The spin reorientation phenomenon (a transition from basal plane to axial easy magnetisation direction) has been studied extensively by a narrow step temperature scanning in the vicinity of the transition. Using the procedure of subtracting the Mössbauer spectra taken for the same compound at different temperatures, it was possible to follow the influence of transition on the shape of spectra. From this procedure it was concluded that in the region of transition each subspectrum splits into two Zeeman sextets, which are characterised by different hyperfine magnetic fields and quadrupole splittings. The consistent way of describing the Mössbauer spectra was proposed. The spin reorientation temperatures have been established for all compositions and compared with the values obtained from theoretical calculations of spin orientation angle based on phenomenological model. The spin arrangement diagrams have been constructed.

Samples of natural alfa-Fe doped randomly with Os impurities up to about 9 at.% have been investigated by means of the 14.41 keV Mössbauer absorption spectroscopy of 57Fe vs. Os concentration and at room temperature. It has been found that the nearest neighbor single Os impurity reduces hyperfine magnetic field on the iron nucleus by 3.97 T, while the second neighbor single Os impurity decreases this field by 1.11 T. More distant impurities slightly increase iron hyperfine field. There are no discernible effects of the Os-Os interaction on the magnetic field seen by iron. On the other hand, the electron charge density on the iron nucleus is increased by the nearest neighbor Os atom as seen via changes in the isomer shift, i.e. via changes in the spectral shift. The second neighbor impurity has the opposite effect on the charge density. More distant impurities tend to decrease the charge density as well. The effect of the first and second neighbor impurities decreases with increasing overall Os concentration indicating some Os-Os interaction influencing a charge density of the conduction electrons seen on the iron nucleus.

Opis:
Electron configuration changes of Fe(III) ions in porphyrin complexes with chloride axial ligands caused by the successive nitrogen substitution of CH methine bridges at meso positions of the porphyrin ring is discussed on the basis of Mössbauer spectroscopy results. It was shown that increase of a number of nitrogen atoms at the meso positions changes the character of quantum-mechanically mixed spin state of Fe(III) ions (S = 5/2 + 3/2) by the increase of the intermediate-spin (S = 3/2) contribution. This feature is reflected in Mössbauer spectra by an increase of quadrupole splitting values and the decrease of the asymmetry of quadrupole doublets, when the number of nitrogen atoms at the meso positions increases. Isomer shifts remain practically unchanged. These peculiarities are discussed in the light of spin relaxation mechanisms and the occupancy of d orbitals in Fe(III) ions coordinated to the porphyrin ring and chloride ligand. It has been noticed that Mössbauer parameters correlate qualitatively with EPR data.

The Er2 xGdxFe14B (x = 0.5, 1.0, 1.5) polycrystalline compounds have been investigated with 57Fe Mössbauer spectroscopy in the 80 330 K temperature range. The spin reorientation phenomena (changes from planar to axial spin arrangements) have been studied extensively by a narrow step temperature scanning in the neighbourhood of the spin reorientation temperature. From the analysis of the spectra it was deduced that in the region of transition each subspectrum was split into two Zeeman sextets, which were characterized by different hyperfine magnetic fields and quadrupole splittings. A consistent way of describing the Mössbauer spectra in the wide range of temperatures was proposed. The composition and temperature dependencies of hyperfine interaction parameters and subspectra contributions were derived from experimental spectra. The transition temperatures were determined for all the compounds studied.

The presence of melanin in spleens of black C57BL/6 mice has been known for long. Although its origin and biological functions are still obscure, the relation of splenic melanin to the hair follicle and skin pigmentation was suggested. Here, we demonstrated using for the first time electron paramagnetic resonance spectroscopy that black-spotted C57BL/6 spleens contain eumelanin. Its presence here is a "yes or no" phenomenon, as even in the groups which revealed the highest percentage of spots single organs completely devoid of the pigment were found. Percentage of the spotted spleens decreased, however, with the progress of telogen after spontaneously-induced hair growth. The paramagnetic properties of the spleen eumelanin differed from the hair shaft or anagen VI skin melanin. The splenic melanin revealed narrower signal, and its microwave power saturability betrayed more heterogenous population of paramagnetic centres than in the skin or hair shaft pigment. Interestingly, the pigment of dry hair shafts and of the wet tissue of depilated anagen VI skin revealed almost identical properties. The properties of splenic melanin better resembled the synthetic dopa melanin (water suspension, and to a lesser degree - powder sample) than the skin/hair melanin. All these findings may indicate a limited degradation of splenic melanin as compared to the skin/hair pigment. The splenic eumelanin may at least in part originate from the skin melanin phagocyted in catagen by the Langerhans cells or macrophages and transported to the organ.

Electron paramagnetic resonance (EPR) spin-labeling methods were used to study the effects of carotenoids on the physical properties of saturated phosphatidylcholine (PC) membranes to evaluate the contribution of the terminal hydroxyl groups of xanthophyll molecules to the carotenoid-membrane interaction. Effects of the dipolar, terminally dihydroxylated carotenoid lutein on membrane phase transition, fluidity, order, and polarity were compared with those of monopolar (β-cryptoxanthin) and nonpolar (β-carotene) carotenoids. These effects were monitored at the membrane center as a function of the amount of the carotenoid added to the sample and as a function of temperature for fluid-phase membranes. PC membranes with different thickness (from 12 to 22 carbons in alkyl chains) were used. Carotenoids shifted to lower temperatures and broadened the main phase transition of PC membranes. They decreased the membrane fluidity and increased the order of alkyl chains. Carotenoids also increased the hydrophobicity of the membrane interior. These effects were the strongest for lutein, significantly weaker for β-cryptoxanthin, and negligible for β-carotene. They decreased with the increase of the membrane thickness. Presented results suggest that anchoring of carotenoid molecules at the opposite membrane surfaces by polar hydroxyl groups is significant in enhancing their effects on membrane properties. This manuscript also shows the ability of EPR spin-labeling methods to monitor different membrane properties that can be applied in biotechnological studies with the use of liposomes.

Temperature dependence of asymmetry as well as isomer shift and quadrupole splitting values of Mössbauer spectra of trivalent iron complexes coordinated to porphyrin ligands such as octaethylporphyrin, monoazaethioporphyrin, diazaoctaethylporphyrin and phthalacyanine is discussed in this paper. All the above mentioned complexes have the chloride axial ligand at the fifth coordination site. Different temperature changes of asymmetry and other parameters of Mössbauer spectra of these complexes can be correlated with a number of methine bridges CH substituted by nitrogen atoms in the porphyrin macrocycle. The form of this asymmetry is such that the higher-energy component of the doublet is more broadened than the lower-energy one, while the areas (intensities) under each of two components are the same. The most distinct asymmetry is observed at room temperature. When temperature is decreasing, the spectrum becomes successively more symmetrical and at liquid helium temperature linewidths of both components of the quadrupole splitting are very close one another. The Mössbauer spectrum of the FeCl-monoazaethioporphyrin complex exhibits reversed asymmetry at liquid helium temperature in comparison with the spectra of the remaining complexes i.e. the lower-energy component of the doublet is slightly more broadened than the higher-energy one. The spin-spin and spinlattice relaxation mechanisms in the context of quantum mechanically mixed spin states S = 5/2 + 3/2 of FeIII ions are considered as a source of the observed asymmetry.

Site-directed spin labeling and electron paramagnetic resonance spectroscopy have recently experienced an outburst of multiple applications in protein science. Numerous interesting strategies have been introduced for determining the structure of proteins and its conformational changes at the level of the backbone fold. Moreover, considerable technical development in the field makes the technique an attractive approach for the study of structure and dynamics of membrane proteins and large biological complexes at physiological conditions. This review focuses on a brief description of site-directed spin labeling-derived techniques in the context of their recent applications.

Effective technology transfer can be a source of sustainable competitive advantage and thus is key for economic growth. The aim of the guide is to shed light on the area with big potential and still few success stories in Poland – bringing scientific discoveries to the market at large profit. In most cases of new discoveries patenting is the best way of protecting intellectual property (IP); however there are exceptions from this rule. A new patented technology can be commercialized in three ways: a) selling/assigning ownership to the third party; b) licensing and c) via a start-up company owning the technology (spin-off). First option is the fastest, but usually gives the least reward. Breakthrough technologies can be a foundation of great companies, the shares of which can get very valuable quickly. How much of the company’s shares belong to the inventor depends on technology’s value (measured by ability to generate cash) and scale of investments needed to build the company. There is capital available for spin-off with viable technologies, both from public and private sources. Choosing the right one depends on the phase of company development and its needs. Early stage enterprises should seek partners among seed funds that have specific know-how of building new companies and can properly complement the competence of the inventor. Introduction: Spinal injury with neurological deficit usually results in a neurogenic bladder disorder. The problem may range from total lack of micturition during the spinal shock phase to various forms of detrusor-sphincter dyssynergia. Typical consequences include increased intra-vesical pressure, inability to effectively empty the bladder, increased risk of vesico-ureteral reflux, infection, bladder or kidney stones, neoplastic complications within the urinary tract and renal failure. Proper diagnostics of urinary tract function and morphology enables choosing adequate bladder emptying strategy, early diagnosis and effective treatment of urinary complications in patients after spinal injury with neurological deficit.Study purpose: Presentation of contemporary methods of functional, imaging and endoscopic diagnostic tests of the lower urinary tract in patients with spinal injury with neurological deficit. We discuss clinical value and accuracy of particular diagnostic methods in prevention and monitoring of the therapy of urinary complications.Study form: Literature review.Conclusions: Although urodynamic studies serve as the principal tool among the modern methods of the lower urinary tract function assessment, simple functional tests such as measurement of residual urine volume measurement or cystometrogram may be helpful in clinical practice, particularly during the early post-traumatic phase. Ultrasonographic examination is the first choice imaging study. Clinical validity of screening cystoscopy in spinal injury with neurological deficit patients remains doubtful, but this procedure is of unquestionable value in cases with haematuria.
