The aim of the study was to isolate and identify the extent of Salmonella and Shigella induced diarrhoea in children under five and to determine their antibacterial susceptibility patterns. The method of investigation was a cross-sectional study. Samples were collected from children less than five years old afflicted with diarrhoea coming for treatment at Thika Level Five Hospital, Kiambu County. A total of 80 stool samples were collected. These were specifically examined for Shigella and Salmonella. In doing so, the samples were cultured in MacConkey and Xylose lactose deoxycholate (XLD) agar. The plates were subsequently incubated aerobically at 37 °C overnight. After incubation, suspected Salmonella and Shigella colonies were identified morphologically, marked and the colonies inoculated to biochemical tests for species identification as described in the Medical Laboratory Manual. Following this, colonies derived from purity plating through biochemical testing were sub-cultured onto nutrient agar to obtain pure colonies. The obtained pure colonies were then used to perform susceptibility tests to commonly prescribed antibacterial drugs, including Gentamicin, Ciproflaxicin and Erythromycin. Results showed 10 (12.5%) confirmed positive isolates, where 6 (60%) were Salmonella and 4 (40%) Shigella. With respect to age, both isolates were found to be concentrated more at ages of 1-4 years. Herein, Salmonella – 1-2 years (33.33%), - 2-3years (50%); Shigella – 1-2 years (50%), - 2-3 years (25%). From the study, sex distribution (male or female) of the host isolates was equal (50-50%). Moreover, Ciprofloxacin and Gentamycin were the most potent antibiotics, whereas Ampicillin, Erythromycin, Cotrimoxazole and Sulfamethoxazole were highly resisted. According to study results, Quinolones and Aminogylcosides are the antibiotics of choice for severe diarrhoea illnesses caused by Salmonella and Shigella.

Shigella species are intracytosolic Gram-negative invasive enteropathogenic bacteria, causing the rupture, invasion and inflammatory destruction of the human colonic epithelium. They utilize the host cytoskeletal components to form propulsive actin tails. The so-called invasive phenotype of Shigella is linked to expression of a type III secretory system (TTSS) injecting effector proteins into the epithelial cell membrane and cytoplasm, thereby inducing local but massive changes in the cell cytoskeleton that lead to bacterial internalization into non-phagocytic intestinal epithelial cells. The molecular and cellular bases of this invasive phenotype essentially encompass crossing of the epithelial lining, apoptotic killing of macrophages, entry into epithelial cells, and escape into the cytoplasm, followed by cell-to-cell spread. Intracellular colonization is likely to protect the micro-organisms from killing by humoral and cellular effectors of the innate immune response. Concurrently, the capacity of Shigella to reprogram invaded epithelial cells to produce proinflammatory mediators plays a major role in the strong inflammatory profile of the disease. This profile is likely to impact on the nature and quality of the adaptive response, which is dominated by humoral protection at the mucosal level. In recent years, a large amount of information has been generated regarding the host, pathogen and environmental factors that impact the pathogenesis of shigellosis at the cellular and molecular level. This review summarizes what is currently known about Shigella, detailing those factors that contribute to pathogenesis and examining the current progress in the development of a vaccine.

Shiga toxins belonging to a family of structurally and functionally related protein toxins serve as the key virulence factors for pathogenecity of the virulent enteric bacterial strains namely Shigella dysenteriae serotype 1 including Shiga toxin-producing Escherichia coli (STEC). S. dysenteriae type 1 isolates remain major public health concerns due to their widespread outbreaks and the severity of extra-intestinal diseases, including acute renal failure and also affects the central nervous system. Despite practicing improved hygienic conditions and regulating food and drinking water safety, the enteric pathogens are imposing a major threat in the well being of the human society. Shiga toxin on entry into host cells’ endoplasmic reticulum (ER) activates the stress response in ER and inhibits protein synthesis by catalytic inactivation of eukaryotic ribosomes. In many cell types shiga toxins trigger apoptosis. Recent studies have shown that shiga toxins induce autophagy which activates different signaling pathways in toxin-sensitive and toxin-resistant human cells. In this review the molecular basis of Shiga toxins’ effect on host cell leading to manifestation of the infection in affected individuals are discussed with an emphasis on recent findings.

SSOR, a clinical isolate of Shigella sonnei which exhibits a Salicin-negative phenotype, is unable to mutate to give rise to Sal^+ derivatives although a homolog of the Escherichia coli bgl operon is retained by the strain. This was correlated to the presence of an endogenous plasmid in the strain. A plasmid-cured derivative, AK711, could give rise to Sal^+ mutants in two steps. Introduction of the plasmid DNA, extracted from SSOR, into various strains of E.coli and S. sonnei, resulted in ampicillin resistant transformants. Interestingly, the presence of the plasmid suppressed the mutational activation of the bgl operon in the transformants. This was further substantiated by the observation that, transformants that have lost the plasmid regained the ability for mutational activation of the bgl operon. Preliminary characterisation of the plasmid indicated a size of 3.8 kb with an origin of replication resembling that of ColE1 replicons and the bla gene homolog of Tn3. Observations of the mutation frequency at the srl and lac loci in the presence of the plasmid indicate that there is a reduction in the mutation frequency, suggesting an antimutator activity associated with the plasmid.

Diarrhoea in developing countries is caused by an increasingly long list of bacterial, viral, and parasitic pathogens with rotavirus, Enterotoxigenic Escherichia coli, Campylobacter, Shigella, and Salmonella heading the list. Using methods to detect most of the known enteropathogens, one or more enteropathogen(s) is isolated in two-thirds of diarrhoeal illnesses in the developing world. Deoxyribonucleic acid probes have proved very useful in detecting pathogens such as enterotoxigenic (ETEC), enteroinvasive (EIEC), enteropathogenic E. coli (EPEC), and Shigella but have not yet proved to be particularly rapid or less expensive. Molecular biology has proved useful in epidemiological studies as a means of strain identification and detection of genome diversity. Since the introduction of ribonucleic acid gene restriction patterns as taxonomic tools in 1986, ribotyping has become an established method for systematics, epidemiological, ecological population and genome diversity studies of microorganisms including Shigella. The technological development culminated in the automation of ribotyping which allowed for high-throughput applications. PCR ribotyping has proved being a highly discriminatory, flexible, robust and cost-efficient routine technique which makes inter-laboratory comparison and build of ribotype databases possible, too. The aim of the present review is to determine the present status of ribotyping technique in detecting the diversity in Shigella isolates.

In Sri Lanka, among 2588 Salmonella positive cases, the highest incidences were recorded from Jaffna peninsula during 2005 to 2013. Therefore, the present study aimed to identify the microbiological and chemical contamination status of groundwater (40 well water) sources in Jaffna during November 2016. The total coliform, E. coli, Salmonella spp. and Shigella spp. along with some physico-chemical parameters of groundwater were studied. The results revealed that entire peninsula was contaminated with total coliform and E. coli bacteria and the parameters recorded were not within the WHO and SLS (Sri Lanka Standards) drinking water quality standards. 38% of sampling locations were positive for Salmonella spp. and among them six sampling locations were being used for drinking purposes. The results of the study correlates with the statistics of typhoid cases recorded in Jaffna. Results of the study also revealed that around 80% of wells were not within the values specified in guidelines of the SLS for drinking water quality on electrical conductivity. Further, 15% of wells recorded greater than 10 mg∙dm–3 nitrate, which is still below the SLS drinking water standards (45 mg∙dm–3). According to the water quality data, PCA analysis showed that Jaffna town, Nallur, Tellippalai and Kopay DS divisions has similar characteristics for water quality.
Spośród 2588 przypadków zatrucia salmonellą zanotowanych na Sri Lance największą liczbę zachorowań stwierdzono na półwyspie Jaffna w latach 2005–2013. Dlatego prezentowane badania miały na celu identyfikację chemicznego i mikrobiologicznego zanieczyszczenia wód gruntowych z 40 studni w listopadzie 2016 r. Analizowano całkowitą liczbę bakterii z grupy coli, Escherichia coli, Salmonella spp., Shigella spp. oraz wybrane parametry fizyczne i chemiczne wód gruntowych. Wyniki ujawniły, że cały półwysep jest skażony bakteriami z grupy coli i bakterią Escherichia coli, a analizowane parametry nie spełniają standardów WHO i standardów Sri Lanki co do jakości wody. W 38% badanych stanowisk stwierdzono obecność Salmonella spp., przy czym sześć z nich wykorzystywano jako źródła wody pitnej. Wyniki badań korelowały z rejestrowanymi przypadkami duru brzusznego na półwyspie. Stwierdzono także, że 80% studni nie spełniało wymogów stawianych wodzie pitnej przez normy Sri Lanki w odniesieniu do przewodnictwa elektrolitycznego. Ponadto stężenie azotanów w 15% studni było większe niż 10 mg∙dm–3, ale nie przekraczało granicznej wartości 45 mg∙dm–3 wyznaczonej przez normy dla wody pitnej. Analiza PCA wykazała, że miasto Jaffna oraz gminy Nallur, Tellippalai i Kopay mają wody o podobnych właściwościach.

Shigella is one of the most common bacterial pathogens that are isolated from patients with diarrhea. Various attempts are being made worldwide with encouraging observations; still the emergence of multidrug-resistant Shigella strains and a continuous high disease incidence imply that shigellosis is an unsolved global health problem which can probably be solved only by developing a proper vaccine and a vaccine regime for the disease. The need of the hour is to foster the development of an effective vaccine which should not only serve to improve hygiene but also should be able to curb infections by the pathogen. This goal can only be achieved by gaining proper detailed knowledge underlying Shigella pathogenesis. The analyses of the Shigella invasion proteins which have been long been targeted to be potential candidate vaccines remains an open ended problem and forms the core of this present computational study which identifies the fact that long regions in the structure of the proteins are disordered having no distinct structural conformation; multiple alignments however, did not show any conserved stretches in the disordered regions. The results probably explain the ability of these proteins to interact with multiple cellular proteins and perform a diverse array of functions leading to successful pathogenesis.

The prevalence of antimicrobial resistance among food pathogens has increased during recent decades. In this work, pathogenic bacteria such as Staphylococcus aureus, Salmonella spp., Shigella spp., and E. coli were isolated following standard methods. The bacterial isolates were then tested for their sensitivity to common antibiotics using the disc diffusion method on Mueller-Hinton Agar. All of the pathogenic bacteria were found to be resistant to erythromycin and almost all were sensitive to penicillin.

Improperly prepared fresh fruit and vegetable juices are recognized as an emerging cause of food borne illnesses. Therefore, this study was aimed at evaluating the microbiological safety of fresh fruit juices marketed in Debre Markos town and their hygienic conditions of preparations. Thirty-six fruit juices samples were collected from 6 cafés and restaurants of Debre Markos town and analyzed for total aerobic viable bacterial count (TAVBC), total staphylococcal count (TSC), aerobic spore forming bacterial count (ASFBC), total coliform count (TCC), fecal coliform count (FCC), yeast and mould count (YMC). The spread plate method was used for the isolation of microorganisms on appropriate selective media. All isolates were characterized following standard methods. Bacterial and fungal species were isolated following standard methods, while questionnaires were distributed to 30 juice makers to obtain preliminary information on their hygienic and safety practices. Results show that the mean TAVBC, ASFBC, TSC, yeast and mould, TCC and FCC of mango were 2.2±0.48×106, 0.13±0.04×105, 0.004×105, 1.1±0.2×106, 0.15±0.05×105, 5.7±3.73×104 and 0.06±0.04×104, cfu/ml, respectively. Moreover, the mean of TAVBC, ASFBC, TSC, YMC, TCC, and FCC of avocado juice were 3.6±0.6×106, 0.08±0.02×105, 0.27±0.07×105, 1.2±0.4×106, 0.02±0.01×105, 6.46±3.7×104, and 0.2±0.1×104 cfu/ml, respectively. The bacterial isolates were identified as S. aureus, E. coli, Klebsiella spp. B. cereus, Enterobacter spp., Enterococcous spp., Streptococcus spp., and Serratia spp., while the identities of the fungal isolates were Fusarium spp., Mucor spp. and S. cerevisiae. The results also showed that the microbial loads of most of the fruit juices were higher than the specifications set for fruit juices sold in the Gulf region and other parts of the world. Most venders obtained fruit from the open market and all juice makers lacked special training in food hygiene and safety. Therefore, regular training and health education on food hygiene and safety is recommended for juice handlers to improve the quality of fresh fruit juices in the study area.

Various types of biological weapons have been known and practiced throughout history, including the use of biological agents such as microbes and plants, as well as biotoxins and the venoms that can be derived from them. In ancient civilisations, the attempt was to infect and kill enemies by throwing cadavers into water wells. Emperor Barbarossa during the battle of the Italian town, Tortona, in 1155, did the same. In modern times, America and the Soviet Union also undertook biological warfare and anti-biological warfare protection activities. This even intensified after WWII. When the Soviet forces captured and interrogated some Japanese scientists in 1945, they utilized the obtained information in their own biowarfare program and their research accelerated in 1946. Following this, a series of new biowarfare study centres and production facilities was constructed in the 1950s. The Soviet biowarfare program included tularemia, anthrax, brucellosis, plague, glanders, marburg virus, smallpox virus, and VEE virus. During the time of the Korean War, it was believed that biowarfare agents were used by America against Soviet Union. The Americans had began their own program in Fort Detrick (former Camp Detrick) in 1943 and a new production facility at Pine Bluff Arsenal in Arkansas was constructed. The United States of America started producing tons of Brucella suis in 1954. In the peak year of their program, they involved about 3,400 people and a number of agents: Bacillus anthracis, Francisella tularensis, Brucella suis, Coxiella burnetti, Venezuelan equine encephalitis virus, yellow fever, botulin, Staphylococcal enterotoxin, and the anti-crop agents Pyricularia oryzae and Puccinia graminis. Due to public pressure, President Nixon declared a unilateral halt in 1969 to biological weapon projects. The only permitted research was defensive, such as diagnostic, vaccines, and chemotherapies tests – as evidenced in the UK where the base in Porton Down was converted into a defence institution.
