Members of Enterobacteriaceae are known to produce extended-spectrum beta-lactamases (ESBL) which hydrolyze the beta-lactam group of antibiotics. The existence of ESBL-producing Salmonella enterica (S. enterica) and Escherichia coli (E. coli) harbored by urban avifauna was investigated in this study. Dropping samples (n= 180) were collected from six different bird species in the district Jhang, Punjab province, Pakistan. Isolation and identification of ESBL isolates were made by using cefotaxime- (4 mg/L) supplemented MacConkey agar and double disc synergy test (DDST). Polymerase chain reaction (PCR) was performed for the detection of four different ESBL genes including bla CTX-M, bla TEM, bla SHV and bla OXA. A total of 42.69% isolates were confirmed as ESBL via DDST including 30.64% S. enterica and 49.54% E. coli. The incidence of ESBL S. enterica and ESBL E. coli was found highest in egret (Ardea alba) and pigeon (Columba livia) as 64.28% and 78.95%, respectively. The bla CTX-M gene was detected in 57.89% and 64.81% of isolates of S. enterica and E. coli, respectively. Among other genes in S. enterica and E. coli, bla TEM (21.05%, 20.4%); bla SHV (15.78%, 9.26%), and bla OXA (5.26%, 5.56%) were detected, respectively. All of the tested isolates were found resistant to at least one of the thirteen antimicrobial agents except meropenem. To the best of our knowledge, this is the first study reporting the incidence and genetic diversity of ESBL bacteria associated with urban avifauna in Pakistan. The urban avifauna can serve as a potential subject of bio-surveillance to monitor the emergence of antimicrobial-resistant bacteria.

Salmonella contamination in poultry feed is one of the main issues in poultry industry and public health. The aim of the present study was molecular detection and typing of Salmonella serotypes isolated from poultry feeds. Moreover, we determined the antibiotic resistance pattern and the ability of biofilm formation in the serotypes. To this end, eighty feed samples were collected from aviculture depots. Salmonella serotypes were identified by culture and PCR methods. For serological identification, a slide agglutination test was used. BOXAIR and rep-PCR methods were applied to evaluate the diversity of serotypes. The disc diffusion method was performed to evaluate the antibiotic susceptibility of serotypes to sixteen antibiotics. Biofilm formation was also assessed by the microtiter-plate test. From a total of 80 feed samples, 30 samples were contaminated with Salmonella spp., which were divided into 5 different serotypes belonging to B, C, and D serogroups. BOXAIR-PCR (D value [DI] 0.985) and rep-PCR (DI 0.991) fingerprinting of isolates revealed 23 and 19 reproducible fingerprint patterns, respectively. A higher antibiotic resistance was observed to ampicillin and doxycycline (100% each), followed by chloramphenicol (83.33%) and tetracycline (73.33%). Multidrug resistance (MDR) was detected in all Salmonella serotypes. Half of the serotypes possessed the ability of biofilm formation with varied adhesion strengths. These results revealed the high and unexpected prevalence of Salmonella serotypes in poultry feed with MDR and biofilm formation ability. BOXAIR and rep-PCR revealed a high diversity of Salmonella serotypes in feeds and subsequently indicated variation in the source of Salmonella spp. The unknown sources harboring high diversity of Salmonella serotypes indicated poor control, which could cause problems for feed manufacturing.

Antimicrobial resistance in Salmonella has been associated with the presence of integrons and many other resistance mechanisms contributing to the spread of antimicrobial-resistant genes within and between livestock and human populations. In this study, the presence of Salmonella serovars from broiler and cattle samples and their antimicrobial resistance, integrons, tet resistance, ESBL and resistance genes carriage were investigated. Total of 209 litter (broiler farms) and fecal samples (cattle farms) were examined by bacteriological procedures, susceptibilities against 18 antimicrobials and genes carriages were detected by singleplex and multiplex PCR. A total of 46/209 (22 %) Salmonella strains were isolated. Six different Salmonella serotypes from 46 Salmonella isolates were identified and the most common serotype was S. Infantis 38 (82.6%) from broiler litter; followed by S. Kitenge 3 (6.5 %) from fecal sample. The highest occurrence of resistance observed for penicilline (46/46, %100), lincomycin (43/46, 93.5%) and 42 isolates (43/46, 93.5%) exhibited MDR. The overall occurrence of class 1, 2 and 3 integrons carrying Salmonella in tested samples were 63.04% (29/46), 43.5% (20/46) and 84.8% (39/46) respectively. Out of the 27 isolates produced an ESBL, mostly CTX and TEM. On 46 Salmonella isolates, in 16 (34.8%) Tcr’ genes were determined. Genotypic and phenotipic detection of ESBL genes found within integrons from Salmonella isolates from different sources (broiler and cattle) can provide powerful information about health and economic risk associated with transferable multidrug resistance.

Background. The aim of the study was to determine the hygienisation efficiency of the municipal waste composting process based on the inactivation rate of the bacteria Salmonella Senftenberg W775 H2S- and Ascaris suum eggs. Material and methods. The research was conducted in a composting plant located in northern Poland. The processed material was organic waste (kitchen waste) consisting of food waste (plant origin – approx. 50%, animal origin – approx. 20%) and grass and leaves (approx. 10%). Temperature generated in the compost heap was measured using a sensor in its upper part and a mobile probe. Results. The fastest elimination of Salmonella bacilli occurred after 18.5 hours in the upper part of the compost heap. In the middle part of the pile bacteria were not isolated after 24 hours, whereas at the bottom – after 48 hours. In the lateral part of the compost heap Salmonella were not isolated after 65 hours of composting process. Total inactivation of Ascaris suum eggs occurred after 18.5 hours in the middle part of the heap and 89 hours in the lateral part. Conclusion. Obtained results confirm the higienisation potential of the process of municipal waste composting in terms of inactivating pathogens. A correct course of the process guarantees microbiological safety of the final product use for agricultural purposes.
Celem badań była ocena efektywności higienizacyjnej procesu kompostowania odpadów komunalnych w oparciu o tempo inaktywacji pałeczek Salmonella Senftenberg W775 H2S- oraz jaj Ascaris suum. Uzyskane wyniki wskazują, że bakterie z rodzaju Salmonella ulegały najszybszej eliminacji w górnej warstwie pryzmy, gdzie nie wykrywano ich obecności już po 18,5 godz. Z części środkowej pryzmy nie izolowano ich po 24 godzinach, a w dolnej po 48 godzinach kompostowania. Najdłużej bakterie wskaźnikowe przeżywały w bocznej części pryzmy (do 65 godzin procesu kompostowania). Całkowita inaktywacja inwazyjnych jaj Ascaris suum następowała w czasie od 18,5 do 89 godz. kompostowania. Badania potwierdzają potencjał higienizacyjny procesu kompostowania odpadów komunalnych w zakresie inaktywacji drobnoustrojów patogennych. Prawidłowy przebieg procesu gwarantuje bezpieczeństwo mikrobiologiczne stosowania produktu finalnego do celów rolniczych.

Zoonotic serotypes of Salmonella enterica subsp. enterica are an important biological factor that poses a threat to public health, and broilers are often asymptomatic carriers of these bacteria. The aim of this study was to analyze the prevalence of Salmonella spp. and Salmonella serotypes in broiler chicken flocks in northern Poland in 2014–2016. The study was conducted on commercial flocks monitored by the Institutes of Veterinary Hygiene in Gdańsk and Olsztyn in 2014–2016. A total of 4,331 samples were investigated. Identification of Salmonella spp. was performed by a culture-dependent method followed by biochemical and serological confirmation tests, in accordance with Polish Standards. The total percentage of infected flocks in northern Poland reached 1.57%. The prevalence of Salmonella spp. in broiler chickens decreased from 2.19% in 2014 to 1.22% in 2016. The percentage of flocks infected with S. Enteritidis and S. Typhimurium was 1.18% and 0.12%, respectively (S. Enteritidis and S. Typhimurium were detected in 1.29% of flocks). The most common serotypes that had not been covered by the control programme were S. Mbandaka (0.14%), S. Infantis (0.07%) and, sporadically, S. Kentucky and S. Anatum. The percentage of infected flocks was higher (by 0.26%) in the Pomeranian Region than in the Region of Warmia and Mazury, and it increased over the analyzed period – the highest number of positive samples were found in 2016 (2.04%). In the Region of Warmia and Mazury, the percentage of infected flocks was highest in 2014 (3.08%); in 2014–2016, the number of positive samples decreased steadily to reach 0.41% in 2016. In the analyzed regions, the incidence of human salmonellosis was correlated with the percentage of infected broiler flocks.

There are three main approaches in cancer treatment: surgery, chemotherapy, and radiotherapy. Recently, the use of bioengineered bacteria as therapeutic agents has been shown to have some valuable properties in the treatment of cancer, which do not exist in conventional approaches. Bacteria in particular can target tumors, and they can preferentially proliferate and accumulate within tumors and inhibit the growth of cancer cells by inducing cytotoxicity. Thus, bacteria can be easily detected in tumor sites. Moreover, bacteria-derived factors exert an immunostimulatory effect. Over the past decade, Salmonella, Clostridium, and other bacterial genera have been shown to inhibit tumor growth and promote the survival rate in animal models. Clinical trials for cancer treatment with bacteria have shown improved results by combination with other therapeutic methods such as chemotherapy or radioactive agents. This review is an effort to introduce the use of healthy bacteria in tumor therapy. We specifically focus on Salmonella, which has been extensively used in tumor therapy. Therefore, in this review study, we discuss the merits, mechanisms, and attenuated strains of a combination therapy compared to other therapeutic approaches in Salmonella-mediated cancer therapy.

Nano-biotechnology represents the intersection of nanotechnology and biotechnology which is an emerging field in creation, productivity and utility of nanoscale structures for advanced biotechnology. Plant and plant extract are considered as green and effective paths in the synthesis of gold and silver nanoparticles. The aim of the present study is to analyze the interaction of silver nanoparticles synthesized from herbal source (Cinnamon zeylanicum) with pathogenic bacteria Salmonella typhi (S. typhi) in order to know the microbe pathogenicity. The micro-nano interface disturbed the growth of microbes. Under the influence of different concentration of nanoparticles (50 µL to 1,000 µL), the microbes exhibited slow growth as compared to control. The viable cell count also decreased as the nanoparticles concentration increased. This shows major difference in the bacterial culture of with and without nano particle. Under normal culture, without nanoparticles intervention, the microbes show lag phase at 0-4 h, the log phase begins and its exponential growth occurs between 4-8 h and after 12 h, stationary phase was attained. There was a slow growth observed when the culture was exposed to silver nanoparticles. This clearly shows the antimicrobial effect of nanoparticles. In this study we used green synthesized nanoparticles, which have shown a good antibacterial efficacy against the pathogenic microorganism Salmonella typhi.

The prevalence of antimicrobial resistance among food pathogens has increased during recent decades. In this work, pathogenic bacteria such as Staphylococcus aureus, Salmonella spp., Shigella spp., and E. coli were isolated following standard methods. The bacterial isolates were then tested for their sensitivity to common antibiotics using the disc diffusion method on Mueller-Hinton Agar. All of the pathogenic bacteria were found to be resistant to erythromycin and almost all were sensitive to penicillin.

The survival of potentially infectious enteropathogens in digestate from two-phase mesophilic anaerobic digestion (Two-phase MAD) technique raises concern on handling and application of the resulting digestate as far as hunan health and environment safety is concerned. In this research, temperature validation and hygienization potential of Two-phase MAD operated as a completely stirred-tank reactor with semi-continuous feeding was investigated. High enteropathogens survival up to 6.4 log E. coli and 6.1 log Salmonella Senftenberg was observed in digestate resulted from anaerobic digestion of potato peels artificially spiked with 8.7 log Salmonella Senftenberg and 8.8log E. coli simulating extra high pathogens contamination in biowaste used as feedstock. Even though an average decrease in pH by 2.3 units from the original value of 6.5 observed in hydrolysis reactor created unfavorable environment on the survival of both E. coli and Salmonella Senftenberg causing a 3.6 log and 3.8 log reduction, yet the alkaline condition in the methanation reactor pH 7.7 boosted the surviving rate by 1.3 log in E. coli and 1.2 log in Salmonella Senftenberg confirming the inefficiency of Two-phase MAD set-up at providing a completely hygienized digestate. This renders the biosafety of the resulting final digestate questionable and limit the possibility of farmapplication for nutrients recycling due to the involving environmental threat and health risks in case the digestate is to be used. Temperature-time combination at 65 °C for 30min was confirmed sufficient at completely elimination of E. coli and effecting >1 log cycle die-off equivalent to 90% die-off in Salmonella Senftenberg with z-Value of 11 ºC validating the temperature-time regime of 70 ºC for 60min proposed in European Commissions Regulation (EC) No. 208/2006.

Improperly prepared fresh fruit and vegetable juices are recognized as an emerging cause of food borne illnesses. Therefore, this study was aimed at evaluating the microbiological safety of fresh fruit juices marketed in Debre Markos town and their hygienic conditions of preparations. Thirty-six fruit juices samples were collected from 6 cafés and restaurants of Debre Markos town and analyzed for total aerobic viable bacterial count (TAVBC), total staphylococcal count (TSC), aerobic spore forming bacterial count (ASFBC), total coliform count (TCC), fecal coliform count (FCC), yeast and mould count (YMC). The spread plate method was used for the isolation of microorganisms on appropriate selective media. All isolates were characterized following standard methods. Bacterial and fungal species were isolated following standard methods, while questionnaires were distributed to 30 juice makers to obtain preliminary information on their hygienic and safety practices. Results show that the mean TAVBC, ASFBC, TSC, yeast and mould, TCC and FCC of mango were 2.2±0.48×106, 0.13±0.04×105, 0.004×105, 1.1±0.2×106, 0.15±0.05×105, 5.7±3.73×104 and 0.06±0.04×104, cfu/ml, respectively. Moreover, the mean of TAVBC, ASFBC, TSC, YMC, TCC, and FCC of avocado juice were 3.6±0.6×106, 0.08±0.02×105, 0.27±0.07×105, 1.2±0.4×106, 0.02±0.01×105, 6.46±3.7×104, and 0.2±0.1×104 cfu/ml, respectively. The bacterial isolates were identified as S. aureus, E. coli, Klebsiella spp. B. cereus, Enterobacter spp., Enterococcous spp., Streptococcus spp., and Serratia spp., while the identities of the fungal isolates were Fusarium spp., Mucor spp. and S. cerevisiae. The results also showed that the microbial loads of most of the fruit juices were higher than the specifications set for fruit juices sold in the Gulf region and other parts of the world. Most venders obtained fruit from the open market and all juice makers lacked special training in food hygiene and safety. Therefore, regular training and health education on food hygiene and safety is recommended for juice handlers to improve the quality of fresh fruit juices in the study area.

Opis:
Celem pracy była statystyczna analiza związków pomiędzy źródłem izolacji 66 szczepów bakterii fermentacji mlekowej (LAB), ich przynależnością filogenetyczną do określonego rodzaju i gatunku a wykazywanymi właściwościami przeciwdrobnoustrojowymi w stosunku do bakterii Listeria monocyto- genes, Salmonella enteritidis i Escherichia coli. Analizę przeprowadzono na podstawie danych zebranych w trakcie badań w latach 2014 - 2019, prowadzonych w Zakładzie Higieny i Zarządzania Jakością Żywno- ści Katedry Technologii Gastronomicznej i Higieny Żywności Szkoły Głównej Gospodarstwa Wiejskiego w Warszawie. Zebrane dane obejmowały średnice stref zahamowania wzrostu drobnoustrojów wskaźni- kowych obserwowane w metodzie dyfuzyjnej studzienkowej, w której jako czynnik hamujący wykorzy- stywano pełne hodowle LAB (WBC), bezkomórkowy płyn pohodowlany (CFS) oraz zneutralizowany, traktowany katalazą, płyn pohodowlany (CFN). Spośród analizowanych szczepów LAB najwyższą ak- tywność przeciwdrobnoustrojową wykazywały szczepy gatunku Lactobacillus plantarum, a najniższą – Lb. fermentum. Szczepy wyizolowane z serów regionalnych (a szczególnie z oscypka) charakteryzowały się największym antagonizmem w stosunku do L. monocytogenes, zjawisko to występowało zarówno w WBC, CFS, jak i CFN. Wykazano, że badane szczepy LAB charakteryzują się zróżnicowanymi właści- wościami przeciwdrobnoustrojowymi, przy czym zróżnicowanie to jest zależne od takich czynników, jak źródło izolacji oraz przynależność filogenetyczna.
